  • The interaction between nalidixic acid and probenecid was studied pharmacokinetically in rabbits. The blood level and the area under the concentration curve(AUC) of nalidixic acid administered orally in dose of 100mg/kg was elevated by the coadministration of probenecid. Probenecid inhibited both the urinary excretion and the biliary excretion of nalidixic acid. Therefore, biological half-life of nalidixic acid was prolonged by the coadministrarion of probenecid. It was considered that the coadmini-stration of probenecid is more desirable than the single administration of nalidixic acid for the therapeutic effect.

  • Microencapsulation of nalidixic acid using Eudragit RL, a methacrylic acid copolymer was investigated. Microcapsules were prepared by dispersing the drug solution in liquid paraffing using aluminium tristearate as dispersing agent. The preparation of the microcapsules showed high reprodulibility in particle size, shape and the drug content. The dissolution rates of Nalidixic acid from the these microcapsules considerably decreased as compared with that from Nalidixic acid powder and Nalidixic acid-Eudragit RL solid dispersions. The release of Nalidixic acid increased with increasing percentage of aluminium tristearate added to the microcapsules.

  • The adsorption of nalidixic acid on human erythrocytes was found to expressed by Freundlich's isotherm. The amount of adsorption of nalidixic acid on erythrocytes increased with an increase of pH. The adsorption of nalidixic acid on human plasma was found to expressed at Scatchard's equation by the equilibrium dialysis method. An influence of pH on adsorption of nalidixic acid to human plasma proteins was studies at pH 4-10. It was found that the degree of adsorption increase with the increase of pH from 4-6, but descreased above pH 9.

  • Amides from amino acid ester and nalidixic acid were synthesized. The solubility characteristics and partition coefficient of the compounds were studied. The hydrolysis of the compounds was studied in the simulated gastric fluid and simulated intestinal fluid. Some compounds showed better antibacterial activity than nalidixic acid.

  • Binding of nalidixic acid which is used primarily in the treatment of urinary infection and probenecid which is used as a uricosuric agent to bovine serum albumin were studied using difference spectrophotomeric method. 2-(4'-Hydroxybenzeneazo) bcnzoic acid as a spectrophotometric probe was used for measuring the binding of nalidixic acid and probenecid to bovine serum albumin. The association constants of nalidixic acid and probenecid were $1.58{\times}10^{4}M^{-1}$ and $1.70{\times}10^{4}M^{-1}$, respectively.

  • In order to elucidate the effects of Panax Ginseng on the pharmacokinetic parameters of nalidixic acid in a patho-physiological changes, the kinetics of the disappearance of the drug from the blood, appearance in the bile and urinary excretion were studied in $CCl_4-toxicated$ rabbits. The pharmacokinetics of the drug nalidixic acid in rabbits were modeled by a two compartment. Total saponin, water extract from Panax Ginseng, significantly decreased biliary and urinary excretion of nalidixic acid.

  • For the purpose of investigating the effect of nalidixic acid on the nucleic acid synthesis in chloroplast isolated from Chlorella ellipsoidea, cells were cultured in the media treated with nalidixic acid(20ppm) for 5 days. Aliquots cells were taken out at the inoculation and at intervals during the culture and growth rate of Chlorella cells measured. After extraction of nucleic acids in chloroplast isolated from these cells, their contents were analyzed by the base composition and the effect of nalidixic acid on the nucleic acid synthesis interpreted to compare with those of the control. 1. It was showed that the inhibitory concentration affected by nalidixic acid on the growth of Chlorella cells were 20ppm. 2. Because nalidixic acid had depressed the DNA replication in isolated chloroplast as well as whole cell system, these contents were markedly decreased in comparison with those of the control. 3. In the isolated chloroplast as well as in the whole cell system, nalidixic acid was decreased contents of base in the RNA by preventing RNA transcription.

  • The interaction between probencid and nalidixic acid was studied pharmacokinetically in rabbits infused with or without acidic soiution (5% $NH_4Cl$). The results were as fellows. The blood level and the area under the blood concentration curve of nalidixic acid administered intravenously was elevated by coadministration of probenecid and more elevated in rabbits infused with acidic solution. Probenecid inhibited the urinary excretion of nalidixic acid in rabbits infused with adidic solution. Therefore, biolgcal half-life of nalidixic acid was prolonged by coadministration of probencid.

  • Nalidixic acid and six other drugs were studied for in vitro effectiveness against 200 strains of Escherichia coli isolated recently from healthy persons and bactericidal activity of ampicillin against one respective strain of Escherichia coli and Salmonella typhi isolated were also studied. The resutlts obtained by the plate dilution method showed the following percentage of resistance: kanamycin, 2.5%; streptomycin, 12.0%; ampicillin, 13.5%; tetracyclin, 15.5%; chloramphenicol, 17.5%; colistin sulfate, 19.5%. No strains were resistant to nalidixic acid, clearly indicating that nalidixic acid is the most effective drug tested. Ampicillin, measured by test-tube diltion method, was highly bactericidal against Salmonella typhi at the concentration of 2.5mcg/ml and against Escherichia coli at 5mcg/ml.

  • The base composition of DNA of Aspergillus phoenicis, Rhizopus acidus and Candida albicans treated with cycloheximide and nalidixic acid during the culture was analyzed to compare with the control. The contents of base in the DNA were inhibited by cycloheximide, 20.4% of adenine, 43.1% of thymine, 40.9% of cytosine, 35.3% of guanine, 32.2% of purine, and 42.7% of pyrimidine for A. phoenicis. In R. acidus, 34.2% of adenine, 42.1% of thymine, 38.0% of cytosine, 18.1% of guanine, 24.1% of purine and 40.0% of pyrimidine were depressed by cycloheximide. In the antibiotic treatment of C. albicans, 58.3% of adenine, 58.5% of thymine, 58.1% of cytosine, 42.4% of guanine, 46.8% of purine and 58.8% of pyrimidine were inhibited to compare with the control. The nalidixic acid treatments were showed that, in A. phoenicis 41.6% of adenine, 47.1% of thymine, 59.3% of cytosine, 46.3% of guanine, 45.6% of purine and 57.2% of pyrimidine were inhibited. When R. acidus was treated with nalidixic acid, 59.1% of adenine, 54.7% of thymine, 35.3% of cytosine, 37.4% of guanine, 45.9% of purine and 44.9% of pyrimidine decreased. In treatment of nalidixic acid, the content of DNA was depressed 60.1% of adenine, 68.6% of thymine, 60.7% of cytosine, 40.0% of guanine, 45.8% of purine and 63.5% of pyrimidine for C. albicans In the DNA synthesis of three fungal cells, cycloheximide and nalidixic acid treatments were analyzed obviously that the biosynthesis of pyrimidine was depressed than that of purine. Therefore, it was showed that the DNA contents in the various fungal cells were inhibited remarkably in nalidixic acid treatment than cycloheximide.
