Peptidylprolyl cis-trans isomerase (PPlase) catalyzes the cis-trans isomerization of prolyl peptide and facilitates the folding of cellular proteins and peptides. PPlase consists of two distinct immunophilins, each specifically binding to the immunosupressive drug cyclosporin A (CsA) or FK506, respectively. A PPlase was isolated and partially purified from porcine spleen. The molecular weight of porcine spleen PPlase was determined to be ~14,000 on the basis of SDS-PAGE. The purified enzyme was strongly inhibited by FK506, but not by CsA. The inhibition constant and the true concentration of enzyme preparations were determined by active site titration using the tight binding inhibitor FK506: $K_{i}=18.7$ nM and $E_{t}=172$ nM. The equilibrium ratio of conformer. [cis]/[trans], of prolyl peptide substrates (N-Suc-Ala-Xaa-Pro-Phe-p-NA) in anhydrous trifluoroethanol/LiCl solvent system varied from 0.24 to 0.85 depending on the nature of Xaa. Overall. in this solvent-salt system, the populations of the cis conformer of substrates in equilibrium are higher than in an aqueous solution so that the substantial error caused by high background absorption can be reduced. The reactivities of porcine spleen PPlase are shown to be highly sensitive to changes in the structure of substrates. Thus, $k_{cat}/K_m$ value for the most reactive substrate (Xaa Leu) is $4.007+10^{6}M^{1}s^{1}$ and, is 2,636 fold higher than that for the least reactive peptide substrate tested, Xaa=Glu.
Korean red ginseng (KRG) has been used worldwide as a traditional medicine for the treatment of various reproductive diseases. Gonadotropin releasing hormone (GnRH) neurons are the fundamental regulators of pulsatile release of gonadotropin required for fertility. In this study, an extract of KRG (KRGE) was applied to GnRH neurons to identify the receptors activated by KRGE. The brain slice patch clamp technique in whole cell and perforated patch was used to clarify the effect of KRGE on the membrane currents and membrane potentials of GnRH neurons. Application of KRGE (3 ${\mu}g$/${\mu}L$) under whole cell patch induced remarkable inward currents (56.17${\pm}$7.45 pA, n=25) and depolarization (12.91${\pm}$3.80 mV, n=4) in GnRH neurons under high $Cl^-$ pipette solution condition. These inward currents were not only reproducible, but also concentration dependent. In addition, inward currents and depolarization induced by KRGE persisted in the presence of the voltage gated $Na^+$ channel blocker tetrodotoxin (TTX), suggesting that the responses by KRGE were postsynaptic events. Application of KRGE under the gramicidin perforated patch induced depolarization in the presence of TTX suggesting its physiological significance on GnRH response. Further, the KRGE-induced inward currents were partially blocked by 6-cyano-7-nitroquinoxaline-2,3-dione (CNQX; non-NMDA glutamate receptor antagonist, 10 ${\mu}M$) or picrotoxin (PIC; $GABA_A$ receptor antagonist, 50 ${\mu}M$), and almost blocked by PIC and CNQX mixture. Taken together, these results suggest that KRGE contains ingredients with possible GABA and non-NMDA glutamate receptor mimetic activity, and may play an important role in the endocrine function of reproductive physiology, via activation of $GABA_A$ and non-NMDA glutamate receptors in GnRH neurons.
The corrosion behavior of Cr-N coated steels with different phases (${\alpha}-Cr$, CrN and $Cr_2N$) deposited by cathodic arc deposition on Hl3 steel was investigated in 3.5% NaCl solution at ambient temperature. Potentiodynamic polarization test, electrochemical impedance spectroscopy (EIS) and scanning electron microscopy (SEM) were the techniques applied to characterize the corrosion behavior. It was found that the CrN coating had a lower current density from potentiodynamic polarization test than others. The porosity, corresponding to the ratio of the polarization resistance of the uncoated and the coated substrate, was higher in the $Cr_2N$ coating than in the other Cr-N coated steels. EIS measurements showed, for the most of Cr-N coated steels, that the Bode plot presented two time constants. Also, the $Cr_2N$ coating represents the characteristic of Warburg behavior after 72hr of immersion. The coating morphologies were examined in planar view and cross-section by SEM analyses and the results were compared with those of the electrochemical measurement. The CrN coating had a dense, columnar grain-sized microstructure with minor intergranular porosity. From the above results, the CrN coating provided a better corrosion protection than the other Cr-N coated steels.
The mechanisms of the electrode reaction of chlorate ion were investigated, using Pt-electrode, by means of the technique of galvanostatic double pulses. This study was prepared to observe some enzyme activities in the liver of mice treated with extracts of Ginseng anticancer compound, separated from the petroleum ether extracts by silicic acid chromatography, has the cytotoxic activity against cancer cells. Swiss mice, 72 heads were used (or this experiment and they were divdied into control, test group I and test group If, that test group I was injected crude extract and test group II was injected anticancer compound, while the control group was injected 0.9% NaCl solution. The injections were carried out 1,2,4 and 8 times once a day for 1-8 day, respectively. The liver was removed carefully from the mice at 24 hours after drugs injected, and homogenized at 4$^{\circ}C$ for enzyme study. The activities of glutamic oxaloacetic transaminase (GOT) and glutamic pyruvic transaminase(GPT) were determined by Reitmen and Frankel method and lactic acid dehydrogenase activity was determined by Wroblewski methods in vitro. The results obtained are summarized as follows; 1. The GOT activity was increased 26%, crude extract and 16%, anticancer compound than those of control at 1st injected groups and decreased gradually according to increase of injection time, at 8th injected groups, the GOT activity was decresed by 16%, crude extract and 12%, anticancer compound. 2. The GPT activity was not changed significantly at 1st and 2nd injected groups, but, at 4th injected groups, the GPT activity was decreased 20%, crude extract and 14%, anticancer compound. While the GPT activity was recovered to normal value at 8th injected groups. 3. At 1st injected groups, the LDH activity was increased 17%, anticancer compound, while those of crude extract was shown normal value. At 2nd injected groups, the LDH activity increased 35yo:, crude extract while those of anticancer compound was showed normal value. And the LOH activity was recovered gradually at 4th and 8th injected groups.

Extent of reduction in protein digestibility by Godulbaegi(lxeris sonchifolia H.) kimchi powder appeared to be related to the kimchi weight-to-protein weight ratio, the kind of protein sources, part of plants and fementation period. In vitro digestibilities of protein were significantly(p<0.05) reduced as the freeze dried kimchi powder weight-to-protein ration increased from 0.5 : 1.0 to 1.0 : 1.0 for all Godulbaegi samples and protein sources. Overall digestibility of protein sources in the presence of kimchi powder, the reduction ranked in the following order : soybean>casein>beef>squid. Lower(p<0.005) reduction occurred for each protein source when raw plant was exchanged for kimchi products. Some greater reduction of digestibility was noted in young plants and leaf samples than ripe or root samples. Trypsin inhibitor, which expressed as soybena trypsin inhibitor, was inversely related(r=0.8437) to in vitro protein digestibility of casein in the presence of Godulbaegi kimchi powder. More than three times of total polyphenols contained in leaves than in roots. Young leaves had 30% more total polyphenols(37.64mg/g sample) than that in ripe ones. Soaking in 5% NaCl solution for 24 hrs was markedly reduced in total polyphenols as 73% for leaves and 33% for roots. Remarkable reduction in total polyphenols was not checked during fermentation followed after soaking. Trypsin inhibitor content correlated well(r=0.8873) with total polyphenols in all of Godulbaegi samples.

$WC-Ti_{1}$ -xA $l_{x}$ N multilayered coatings are performed by their periodically repeated structures of lamellae of WC-Ti/$WC-Ti_{1}$ -xA $l_{x}$ Nmaterials. The $WC-Ti_{1}$ -xA $l_{x}$ N coatings with variable Al content were deposited onto AISI D2 steel by cathodic arc deposition (CAD) method. The electrochemical behavior of multilayered $WC-Ti_{1}$ -xA $l_{x}$ N coatings with different phases (WC- Ti$0.6/Al_{0.4}$ N, $WC-Ti_{0.53}$$Al_{0.47}$N, $WC-Ti_{0.5}$$Al_{0.5}$ N and $WC-Ti_{ 0.43}$$Al_{0.57}$ N) was investigated in deaerated 3.5% NaCl solution at room temperature. The corrosion behaviors for the multilayered coatings were investigated by electrochemical techniques (potentiodynamic polarization) and surface analyses (X-ray diffraction (XRD), scanning electron microscopy (SEM), and glow discharge optical emission spectroscopy (GDOES)). In the petentiodynamic polarization test, the corrosion current density of $WC-Ti_{0.5}$$Al_{0.5}$N was lower than others.
The effect of different C and Cr/Mo contents, which were varied within the range allowed in the relevant specification, on the mechanical and corrosion properties of A487 alloy cast steel were examined. The increase in strength level with increasing C content and the decrease in strength along with the increase in tensile elongation with decreasing Cr/Mo contents were observed in A487 alloys with different C and Cr/Mo contents. At room temperature, the decrease in Cr/Mo content was beneficial in improving the impact properties of A487 alloy, with no notable difference in absorbed energy with varying C content. The impact properties of A487 alloy at $-60^{\circ}C$, on the other hand, greatly improved with the increase in C and the decrease in Cr/Mo contents. The corrosion behavior of A487 alloys in 3.5% NaCl solution did not show any meaningful change with different C and Cr/Mo contents.

Boride coating applied on steam turbine parts of power plants has provided good particle erosion resistance under temperature of $550^{\circ}C$, but it isn't able to protect the parts effectively any more in ultra super critical (USC) steam turbine which is being operated up to temperature of $650^{\circ}C$. To ensure stable durability for USC steam turbine parts, an alternative coating replacing boride coating should be developed. In this study, multi-component boride coatings containing elements such as chromium (Cr) and vanadium (V) were formed on base metal (B50A365B) using thermochemical treatment method called by pack cementation. The thermochemical treatments involve consecutive diffusion of boron(B) and Cr or/and V using pack powders containing diffusion element sources, activators and diluents. The top layer of Cr-boride coating is primarily consisted of $Cr_2B_3$ and $Cr_5B_3$, while that of V-boride coating is mostly consisted of $VB_2$ and $V_2B_3$. The (Cr,V)-boride coating is consisted of $Cr_2B_3$, $Cr_5B_3$ and $V_2B_3$ mostly. The top surfaces of 3 multi-component boride coatings show hardness of $3200-3400H_v$, which is much higher than that of boride, about $1600-2000H_v$. In 5 wt.% NaCl solution immersion tests, the multi-component boride coatings show much better corrosion resistance than boride coating.
Plasma nitrocarburising and plasma post oxidation were performed to improve the wear and corrosion resistance of S45C and SCM440 steel by a plasma ion nitriding system. Plasma nitrocarburizing was conducted for 3h at $570^{\circ}C$ in the nitrogen, hydrogen and methane atmosphere to produce the ${\varepsilon}-Fe_{2-3}$(N, C) phase. Plasma post oxidation was performed on the nitrocarburized samples with various oxygen/hydrogen ratio at constant temperature of $500^{\circ}C$ for 1 hour. The very thin magnetite ($Fe_3O_4$) layer $1-2{\mu}m$ in thickness on top of the $15{\sim}25{\mu}m$${\varepsilon}-Fe_{2-3}$(N, C) compound layer was obtained by plasma post oxidation. A salt spray test and electrochemical testing revealed that in the tested 5% NaCl solution, the corrosion characteristics of the nitrocarburized compound layer could be further improved by the application of the superficial magnetite layer. Throttle valve shafts were treated under optimum plasma processing conditions. Accelerated life time test results, using throttle body assembled with shaft treated by plasma nitrocarburising and post oxidation, showed that plasma nitrocarburizing and plasma post oxidation processes could be a viable technology in the very near future which can replace $Cr^{6+}$ plating.
