Studies on Immuno-Modulatory and Antitumor Effects of Crude Polysaccharides Extracted from Paecilomyces sinclairii (매미눈꽃동충하초(Paecilomyces sinclairii)로부터 추출한 조다당류의 면역 활성과 항암 효과에 관한 연구)

  • Neutral salt soluble [0.9% NaCl (Fr. NaCl)], hot water soluble (Fr. HW) and methanol soluble (Fr. MeOH) materials were extracted from Paecilomyces sinclairii. In vitro cytotoxicity tests. Fr. HW was not cytotoxic against MH3T3, Sarcoma 180 and HepG2 cancer cell lines at the concentration of $0{\sim}2,000\;{\mu}g/ml$, while HT-29 cell line was cytotoxic at the concentration of $100\;{\mu}g/ml$. Fr. NaCl and Fr. MeOH were slightly cytotoxic to the cell lines. Intraperitoneal injection with Fr. NaCl and Fr. MeOH exhibited antitumor activity with life prolongation effect of 32.3% in mice inoculated with Sarcoma 180. Fr. NaCl improved proliferation of spleen cells and the immunopotentiation activity of B lymphocyte by increasing spleen cell numbers and the alkaline phosphatase activity by $2.4{\sim}2.6\;and\;2.7{\sim}3.9$ folds, respectively. Fr. NaCl generated $89\;{\mu}M$ of nitric oxide (NO) when cultured with RAW 264.7, a mouse macrophage cell line, at the concentration of $50\;{\mu}g/ml$, while iipopolysaccharide, a positive control, produced $79\;{\mu}M$. The Fr. NaCl showed the hightest antitumor effect and activation of B lymphocytes and macrophage. From these results, antitumor effect of P. sinclaitii was likely due to immunopotentiation activity.

Effect of Sodium Chloride (NaCl) on the Mycelial Growth and Ascospore Germination of Rhizina undulata, the Root Rot Fungus of Coniferous Trees

  • Rhizina undulata, the causal agent of Rhizina root rot, is a soil-borne fungus occurring on coniferous trees. The destruction of coastal forests caused by R. undulata infection has been mainly associated with bonfires at camping sites. However, Rhizina root rot was observed in the western coastal forests without fire. It was hypothesized that Rhizina root rot in this area might be closely related to the soil salinity, which can facilitate the growth and survival of R. undulata. So, the variation in sodium chloride (NaCl) resistance among isolates of Rhizina undulata was compared using liquid media containing different concentrations of NaCl ranging from 0 mM to 300 mM. Our results showed that, albeit of no growth at a higher concentration of NaCl (300 mM), most of R. undulata isolates were capable of germinating and grew at up to 100 mM, indicating that NaCl resistance varies among R. undulata isolates. It was further found that isolates from coastal areas seemed to be more tolerant to NaCl than those further away the coast. We demonstrated that R. undulata could be possible to survive in coastal areas, but was lower NaCl tolerance than other fungi.

Effects of Lifestyles, Dietary Habits, Food Preferences and Nutrient Intakes on Sensitivity to and Preference for Salty Tastes of Korean Women

  • This study was performed to investigate factors affecting sensitivity and preference for salty tastes of Korean adult females. Sensitivity and preference for salty tastes were determined as detection threshold concentration of NaCl solution and the optimally-preferred NaCl concentration of the bean sprout soup, respectively. A self-administered questionnaire was used to obtain the information regarding general characteristics, self diagnosis of stress, health-related lifestyle practices, dietary habits and food preferences. Dietary intake using 24-hours recall and blood pressure were measured. Salty taste detection thresholds and optimally-preferred NaCl concentrations were 0.0197% and 0.357%, respectively. There was a significant positive correlation between the optimally-preferred salt concentration and age, despite no significant correlation between either sensitivity or preference for salty taste and sodium intake, which was 3,605mg/day. Those who had bread or cereal with milk as breakfast instead of a traditional Korean meal and those who preferred jjigae to soup had significantly higher NaCl preferences for bean sprout soup. Going to bed after midnight and skipping meals (${\geq}3/week$) decreased salty taste sensitivity without reaching statistical significance. Self awareness of one's own health, recent weight changes, family history of hypertension, sleep quality, getting-up time, rate of eating and other food preferences did not affect either perception. Stress level, TV watching, BMI and sodium intake did not have significant correlation to sensitivity or preference. Further research including a large number of well-controlled subjects and more accurate measurement of sodium intake should be directed to find other factors affecting salt preference and sensitivity in order to decrease Na intake and related diseases.

  • The salt concentration process in electrodialysis, which uses electrical energy to enhance ion concentrations in an aqueous electrolyte solution, has been studied on the transfer phenomenon of ions and water molecules over the ion exchange membrane. In this paper, we investigated various parameters for limiting concentration of electrolyte solution and the electroosmosis phenomenon in an electrodialysis system by varying salt concentration of electrolyte solution. The electroosmotic water transport was analyzed by measuring the ions and water fluxes in electrolyte solutions having two different NaCl concentrations (NaCl 2M/4M), and concentration change was observed for various volume ratios of the diluted reservoir to the concentration one As a result, it was found that the higher concentration of the aqueous electrolyte solution, the lower electroosmosis, and the higher volume ratio led to a higher concentration in the dilute reservoir, so the limiting concentration was enhanced and the specific energy consumption decreased.

The Effect of Concentration of Glucose and Salts on both the Growth and the production of Lipid and DHA of Thraustochytrium aureum ATCC 34304 (당농도 및 염농도가 해양미생물 Thraustochytrium aureum ATCC 34304의 성장 및 지질과 DHA 생성에 미치는 영향 규명)

  • The polyunsaturated fatty acids (PUFAs) are mostly synthesized by the marine original microorganisms. In this study, the effect of concentrations of glucose, NaCl, and $MgSO_4{\cdot}7H_2O$ on both the growth and the production of lipid and Docosahexaenoic acid (DHA) was investigated using Thraustochytrium aureum ATCC 34304. $Y_{Lipid/X}\;and\;Y_{DHA/Lipid}$ increased according to the increase of initial glucose concentration until 15 g/L of glucose. The maximum values of $Y_{Lipid/X}\;and\;Y_{DHA/Lipid}$ were 0.18 g/g and 0.41 g/g respectively at 15 g/L of glucose. The biomass yield, $Y_{x/s}$, however, was constant as 0.378 g/g regardless of initial glucose concentrations in the range of 5 g/L to 25 g/L. The specific growth rate of T. aureum increased also with initial glucose concentration from 5 g/L and 15 g/L. The maximum value was $0.79\;day^{-1}$ at 15 g/L of glucose, and the growth rate decreased to $0.57\;day^{-1}$ and remained constant according to initial sugar concentration in the range of higher than 15 g/L of glucose. The concentration of NaCl was changed from 0 g/L to 48 g/L, in order to study the characteristics of the growth and the production of lipid and DHA according to NaCl concentration. The growth and the production of lipid as well as DHA stopped all at 0 g/L of NaCl and the maximum values of all the three variables occurred at 24 g/L. The effect of $MgSO_4{\cdot}7H_2O$ concentration was also investigated not to find the growth and the production of lipid and DHA at 0 g/L. However, the growth and the Production with $MgSO_4{\cdot}7H_2O$ concentration increased to reach the maximum values at 18 g/L of $MgSO_4{\cdot}7H_2O$.

A Study on the Foaming Properties of Small red bean Protein Isolates at Various Conditions (분리 팥 단백질의 기포 특성에 영향을 주는 제 요인에 관한 연구)

  • The purpose of this study was to determine the foaming properties of two small red bean protein isolates at various conditions. Data concerning the effects of pH, temperature, MaCl concentration, sugar concentration and protein concentration on the properties such as solubility, foam expansion, foam stability were presented. The results were summarized as follows : 1. The crude protein contents of two small red beans were 26.14% and 22.71%. The percentage of nonpolar amino acid group was the highest and that of sulfur containing amino acid group was the lowest. 2. Protein solubility showed the minimum at pH 4.5 which is isoelectric point of small red bean protein isolate adn heat treatment lowered solubility(P<0.05). At pH 4.5, solubility increased sighificantly as 0.4M NaCl was added. However, the effect of sugar concentration in the solubility was not significant. 3. Foam expansion of two small red bean protein isolates was high at pH 4.5 and heat treatment at 10$0^{\circ}C$ lowered foam expansion(P<0.05). While addition of NaCl, sugar did not affect the foma expansion, gradual increment of the protein isolates concentration up to 9% decreased the foma expansion slightly. 4. Foam stability was significantly high at pH 4.5 and heat treatment at 10$0^{\circ}C$ lowered foam stability. Addition of sugar caused slight decrease in foam stability. From 1% to 9% suspension, foma stability increased significantly as protein concentration increased(P<0.05)

Effects of C3A Content on Chloride Concentration in Pore Solution (C3A함유량이 세공용액 Cl 농도에 미치는 영향)

  • 본 연구는 시멘트 경화체 중의 Cl 고정화 메카니즘을 규명하는 연구의 일환으로 시멘트의 C3A 함유량에 따른 Cl 고정화 효과를 세공용액 분석방법에 의해 조사한 것으로 C3A함유량 0.46 9.65%의 4가지 시멘트와 C3A를 함유하지 않은 초속경 시멘트 페이스트를 밀봉용기내에서 양생시켜 재령 28일에 세공용액을 추출하여 세공용액 중의 Cl 과 OH 농도를 측정, C3A함유량이 Cl 고정화에 미치는 영향을 검토한 것이다. 연구결과 세공용액 중의 Cl 농도는 NaCl 혼입량에 관계없이 시멘트 중의 C3A량이 증가함에 따라 낮아져 시멘트 경화체 내에서 Cl 의 고정화에 C3A가 매우 효과적임을 알 수 있었다. 세공용액의 Cl /OH 는 Cl 혼입량이 시멘트 중량의 0.3%인 경우 강재부동태막을 파괴하는 0.3보다 낮았으며 X선회절분석에 의해 C3A에 의한 고정화 메카니즘은 프리델씨염(Friedel's salt)의 생성에 의함을 확인하였다.

Effects of Different NaCl Concentrations on the Growth of Suaedu asparagoides, Suaeda maritima, and Salicornia herbacea (염분농도에 따른 나문재, 해홍나물, 퉁퉁마디의 생장반응)

  • To investigate the effect of NaCl concentration on plant growth in three halophytes, Suaeda asparagoides, Suaeda maritima, and Salicornia herbacea, and to select afforestation species for reclaimed land along the shore, this experiment was conducted by ebb and flow system added fertilize of 1/2 Hoagland solution and halophytes were cultured for 4 month treated by 0, 50, 200, 400 mM NaCl, respectively. The shoot height was increased with 0 and 50 mM NaCl treatment of Suaeda asparagoides and Suaeda maritima and decreased with 200, 400 mM NaCl treatment, whereas them of Salicornia herbacea was increased by 200 mM NaCl treatment. The higher NaCl concentrations, the lower fresh and dry weight in Suaeda asparagoides. In case Suaeda maritima and Salicornia herbacea, fresh and dry weight was increased by 200 mM NaCl treatment. The higher NaCl concentrations, the lower chlorophyll contents in shoot of all treatment. The proline contents of 400 mM treatment in Suaeda maritima was higher than them of other treatments. The shoot water potential was generally lower in 50mM as compared to the control in Suaeda asparagoides, Suaeda maritima, significantly.

Hydrothermal Synthetic Study of Zeolites from Siliceous Mudstone (규질 이암으로부터 제올라이트의 수열 합성에 관한 연구)

  • Siliceous mudstones are embedded on a large scale in the Tertiary formations of Pohang area. Some useful zeolites such as NsP, (Na, TMA)P, analcime and hydroxysodalite were synthesized from the siliceous mudstones by treating with the variety of solution, i.e ., NaOH, NaOH+NaCl, NaOH, NaOH+$NaAlO_2$and NaOH+TMAOH at the low-temperature hydrothermal system ranging 60~12$0^{\circ}C$. Major precursor of zeolites is found as opal-CT in the zeolite-forming reaction. Smectite, which is included in considerable amounts in the mudstone, appears to play a major role of Al-source in the zeolite synthesis. In comparison, chalcedonic quartz and mica are rather insoluble in alkaline solution, and thus, these are observed as major impurities in the reaction products. An addition of $NaAlO_2$to NaOH solution is effective for eliminating these impurities in the reaction procedure, through these are partly dissolved when elevating the reaction temperature, concentration, and time. Phase change from NaP to hydroxysodalite takes place at the NaOH concentrations of 3.0~4.0 M, and the reaction is not sensitive to the temperature shift. NaP is more stable at lower NaOH concentration and higher activity of $Na_{+}$ whereas analcime is sensitive to the temperature change and stable at higher than $100^{\circ}C$ and 2.0~4.0 M in NaOH concentration. For the pure NaP synthesis without any other products, the treatment of mudstones with 1:1 solution of NaOH and $NaAlO _2$ turns out to be quite effective. NaP was successfully synthesized together with analcime at $100^{\circ}C$ as well as lower concentrations of NaOH+NaCl solution. In addition, the organic type, (Na, TMA)P was formed together with smectite in the 1:1 solution of NaOH and TMAOH.

  • To investigate the effect of NaCl concentration and nitrogen sources in the medium for the Streptococci on the intra- and extracellular proteinase(ICP and ECP) which have been known as one of the major causes for the bitter peptide formation, Streptococcus lactis $ML_8$ and Streptococcus cremoris $ML_4$ strains were incubated at 0-4% NaCl in the medium and Na-caseinate as a nitrogen source 0-100%, and the cell growth, ICP and ECP activities were analyzed. As the concentration of the NaCl in the medium increased, the growth and ECP activity decreased but the ICP $activity/10^{10}$ cells increased on the contrary. This implied that the NaCl in the medium affects only the ECP which is associated mainly to the cell wall and cell membrane but not the ICP activity. When the content of the caseinate instead of other low molecular nitrogen sources were increased in the medium, the cell growth was lowered while the ECP activities increased probably by induction of proteinase production.
