Protective Effect of Fangchinoline on Cyanide-Induced Neuro-toxicity in Cultured Rat Cerebellar Granule Cells

  • The present study was performed to examine the effect of fangchinoline, a bis- benzylisoquinoline alkaloid, which exhibits the characteristics of a $Ca^{2+}$ channel blocker, on cyanide-induced neurotoxicity using cultured rat cerebellar granule neurons. NaCN produced a concentration-dependent reduction of cell viability, which was blocked by MK-801, an N-methyl-D-aspartate (NMDA) receptor antagonist, verapamil, L-type$Ca^{2+}$channel blocker, and L-NAME, a nitric oxide synthase inhibitor. Pretreatment with fangchinoline over a concentration range of 0.1 to 10 $\mu$M significantly decreased the NaCN-induced neuronal cell death, glutamate release into medium, and elevation of $[Ca^{2+}]_i$ and oxidants generation. These results suggest that fangchinoline may mitigate the harmful effects of cyanide-induced neuronal cell death by interfering with $[Ca^{2+}]_i$influx, due to its function as a $Ca^{2+}$ channel blocker, and then by inhibiting glutamate release and oxidants generation.

Effects of Stabilizing Additives on Electroless Copper Deposition (무전해 동 도금용액 속에서 안정제의 역할)

  • The effects of the stabilizing additives such as NaCN, 2-MBT and Thiourea on bath decom-position, plating rate and surface morphology have been studied. Bath stability was increased in the order of an additive-free bath, and NaCN-, 2-MBT-, and Thiourea-stabilized baths. The sta-bilizing effects may be attributed to the stability of Cu(II) -complexes. The plating rate is the re-verse order of the bath stability. Accelerative effect of 2-MBT in proper quantity(0.3mg/$\ell$) may be explained by visualizing it absorbed through benzene ring or sulfur atom on portions of the sub-strates. The strong bond of the complexing part of the molecule to nearby chelated copper ions would tend to accelerate plating by making it easier for the Cu2+ -ligand bond to be broken. Sur-face morphologies of copper deposits depend on the bath additives. Electroless copper deposits from the 2-MBT stabilized baths are finer than the deposits from the NaCN- and Thiourea- stabi-lized baths due to the strong adsorption on the substrates.

Wet SiO2 As a Suitable Media for Fast and Efficient Reduction of Carbonyl Compounds with NaBH3CN under Solvent-Free and Acid-Free Conditions

  • Reduction of carbonyl compounds such as aldehydes, ketones, $\alpha,\beta$-unsaturated enals and enones, $\alpha$-diketones and acyloins was carried out readily with $NaBH_3CN$ in the presence of wet $SiO_2$ as a neutral media. The reactions were performed at solvent-free conditions in oil bath (70 - $80^{\circ}C$) or under microwave irradiation (240 W) to give the product alcohols in high to excellent yields. Regioselective 1,2-reduction of conjugated carbonyl compounds took place in a perfect selectivity without any side product formation.

Characterization of Enzymes Against Oxygen Derivatives Produced by Rhodobacter sphaeroides D-230 (Rhodobacter sphaeroides D230이 생성하는 산소 유도체에 작용하는 효소의 특성)

  • The activities of enzymes that act on oxygen derivatives in Rhodobacter sphaeroides D-230 were investigated under various culture conditions. Intracellular SOD activity from the cells grown in aerobic or anaerobic culture conditions was highest at pH 7.0 and pH 8.0, respectively. On the other hand, extracellular SOD activity was highest at pH 6.0. Catalase activity was highest at neutral pH in both cases. Growth of R. sphaeroides D-230 in aerobic or anaerobic culture conditions was inhibited by methyl viologen. As R. sphaeroides D-230 was cul-tured aerobically, SOD activity was increased about 2-fold by addition of iron ion. But $Mn^+2$ had little effect on the SOD activity of R. sphaeroides D-230 grown in aerobically. NaCN, the inhibitor of Cu$.$Zn-SOD, did not inhibit SOD activity. But, $NaN_3$, the inhibitor of Mn-SOD, inhibited SOD activity in anaerobic cultures con-dition. Therefore, R. sphaeroides D-230 produce Mn-SOD in anaerobic condition, although Fe-Sod is produced in aerobic condition. The activity of catalase was induced by methyl viologen, however, extremely inhibited by NaCN and $NaN_3$.

Chemical Weathering Trend of Granitic Rock in Hwangtohyun, Korea (한반도 서부 황토현 일대 적색토의 화학적 풍화 경향)

  • In Hwangtohyun, meaning 'red soil pass', reddish surface mantles is widespread. Other granitic hills, exceptionally Naju and Youngam area, in Korean peninsula don't commonly have that color. This paper attempts to address this issue by CIA(chemical index of alteration). CIA data and A-CN-K diagram provide crucial insights into the changes in the relative contributions of chemical physical weathering in difference of grus regolith and saprolite. CaO and $Na_2O$ show strong depletion and $K_2O$ is progressive loss. In grus regolith, weathering trends are (sub)parallel to the CN-A join of the A-CN-K diagram, but the sample's composition plot ever closer to the A-K join in saprolite. The difference of weathering trend obtained using CIA data corresponds closely with the visual interpretation of soil color and texture.

Effect of NaOCl and Gold Plating Additive on the Gold Recovery in Cyclone Electrolytic Cell from Solution for Stripping Gold of PCB by Cyanide (시안을 이용한 PCB 금 박리용액으로부터 사이클론 전해조에서의 금 회수거동에 미치는 차아염소산나트륨과 금 도금첨가제의 영향)

  • The recovery of gold in cyclone type electrolytic cell was conducted from solution for stripping gold of PCB by cyanide. The electrolytic recovery behaviors of gold was investigated by reaction time and addition of electrolytic sodium hypochlorite (NaOCl) and gold plating additive (KG-120). Because the electrolysis generated NaOCl reacted with the cyanide in the leachate by alkaline chlorination, more than 99% of the cyanide was removed at a $NaOCl(g)/CN^-(g)$ ratio of 1.0. When NaOCl was added during the recovery of the gold from cyanide leachate in the cyclone electrolytic cell, the recovery of gold was 98% at the ratio of $NaOCl(g)/CN^-(g)$ from 0.5 to 2.5 in 480 minutes and decreased rapidly over the ratio of 3.0. Gold was recovered more than 99% by adding 1.5 and 4.5%(v/v) of KG-120 with NaOCl in 480 minutes. In particular, when the concentration of KG-120 was 3.5 and 4.5%(v/v), more than 96% of gold was recovered within 240 minutes and the initial recovery rate was relatively faster. The optimum concentration of KG-120 is 3.5%(v/v) considering the economic feasibility and efficiency.

Effect of Transglutaminase Addition on the Physicochemical Properties of Sodium Caseinate and Whey Proteins

  • In this study, several factors were analyzed in an effort to determine the effects of transglutaminase (TGase) treatment on sodium caseinate (NaCN), ${\alpha}--lactalbumin$ (${\alpha}-La$), and ${\beta}-lactoglobulin$ (${\beta}-Lg$) polymerization reactions. The results of SDSPAGE showed that NaCN was slightly hydrolyzed to molecular weights of 50-400 kDa according to activation time. ${\alpha}-La$ formed high-molecular polymers of 30-300 kDa, whereas ${\beta}-Lg$ remained almost completely unhydrolyzed. Melting temperatures of NaCN, ${\alpha}-La$ with and without TGase were all in the range of $100{\pm}10^{\circ}C$ under the endothermic curve, and the melting temperature of ${\beta}-Lg$ with TGase was lower than that with TGase. When the proteins were incubated for 3 h with TGase, the micrographic structures showed a small quantity of sediment and broad layers. The final ${\alpha}-La$ residues remained at a level of 21.38%, and the TGase-treated ${\alpha}-La$ was confirmed to have undergone a profound loss of mass, to 18.25%. The DPPH-radical scavenging activity of NaCN and ${\beta}-Lg$ with TGase treatment was higher than that observed in the untreated sample, while those of ${\alpha}-La$ increased with concentration.

Treatment Characteristics of Plating Wastewater Containing Freecyanide, Cyanide Complexes and Heavy Metals (I) (도금폐수내 유리시안과 착염시안 및 중금속의 처리특성 (I))

  • The mean pH of wastewater discharged from the plating process is 2, so a less amount of alkali is required to raise pH 2 to 5. In addition, if sodium sulfite is used to raise pH 5 to 9 in the secondary treatment, caustic soda or slaked lime is not necessary or only a small amount is necessary because sodium sulfite is alkali. Thus, it is considered desirable to use only $FeSO_4{\cdot}7H_2O$ in the primary treatment. At that time, the free cyanide removal rate was highest as around 99.3%, and among heavy metals, Ni showed the highest removal rate as around 92%, but zinc and chrome showed a low removal rate. In addition, the optimal amount of $FeSO_4{\cdot}7H_2O$ was 0.3g/L, at which the cyanide removal rate was highest. Besides, the free cyanide removal rate was highest when pH value was 5. Of cyanide removed in the primary treatment, the largest part was removed through the precipitation of ferric ferrocyanide: $[Fe_4(Fe(CN)_6]_3$, and the rest was precipitated and removed through the production of $Cu_2[Fe(CN)_6]$, $Ni_2[Fe(CN)_6]$, CuCN, etc. Furthermore, it appeared more effective in removing residual cyanide in wastewater to mix $Na_2SO_3$ and $Na_2S_2O_5$ at an optimal ratio and put the mixture than to put them separately, and the optimal weight ratio of $Na_2SO_3$ to $Na_2S_2O_5$ was 1:2, at which the oxidative decomposition of residual cyanide was the most active. However, further research is required on the simultaneous removal of heavy metals such as chrome and zinc.

A Magnetization Study of Prussian-blue Analogue NaxMny[Fe(CN)6]

  • In this report, we present the results of a study on the effects of the particle size on the properties of the Prussian blue (PB) analog $Na_xMn_y[Fe(CN){_6}]$. A novel synthesis method of the $Na_xMn_y[Fe(CN){_6}]$ nano-particles using an organic solvent, formamide, is employed. The size of the PB particles is found to be 100-150 nm for the samples prepared in the formamide solvent, which is much smaller than that of the samples prepared using water only. The broadening of the X-ray diffraction peaks of the nano-sized PB samples is attributed to the lattice disorder and a dramatic reduction in the particle size. The compositions of the samples are confirmed by an energy-dispersive X-ray analysis (EDAX), and the result proves that the samples are actually $Na_xMn_y[Fe(CN){_6}]$ Prussian blue. The UV-vis spectra show a broad intervalence charge-transfer (CT) band in the visible region between 400 and 700 nm, and the absorption decreases abruptly in the green region for the nano-sized PB sample. A divergence between the field cooled (FC) and zero field cooled (ZFC) magnetization curves is observed for the nano-sized PB sample at 11 K, indicating that nanoparticles in the sample are single domain superparamagnets with a blocking temperature of 11 K. Our results reveal that the nano-sized PB samples show significantly different optical and magnetic properties than those of the bulk PB samples.
