Effect of NaNO3 and NaHCO3 Concentration on Microglae Arthrospira platensis Growth (NaNO3, NaHCO3 농도가 Arthrospira platensis 생장에 미치는 영향)

  • Arthrospira platensis (A. platensis) is one of the most explored cyanobacteria and has been studied for proteins, vitamins, pigment (chlorophyll and carotenoids) and fatty acid. In this study, we tested the effect of $NaHCO_3$ and $NaNO_3$ on the microalgae growth under photoautothrophic culture in A. platensis. As a result, cell growth and dry cell weight were increased in proportion to the $NaHCO_3$ and $NaNO_3$ concentration. Pigment (chlorophyll and carotenoids) contents of A. platensis were increased with proportion to $NaHCO_3$ concentration. But, the content of pigment (chlorophyll and carotenoids) in 100% $NaNO_3$ medium of A. platensis was the highest, 40%, 140% and 200% $NaNO_3$ medium with pigment content of A. platensis was reduced. In conditions of $NaHCO_3$ (50%) or $NaNO_3$ (40%) limitation, A. platensis could accumulate lipids to high as 1.7-fold and 1.3-fold.

Effect of Dietary Ca and Na Levels on Blood Pressure and Mineral Metabolism in Spontaneously Hypertensive Rats Fed High Fat Diet (고지방식을 섭취한 본태성 고혈압쥐에서 칼슘과 소디움 섭취가 혈압과 무기질 대사에 미치는 영향)

  • This study explored the effect of dietary levels of Na and Ca on spontaneously hypertensive rats (SHR). SHR were randomly divided into 5 groups and fed a high fat/cholesterol diet containing three levels of Na (0.05, 0.1, 1.5%) and Ca (0.1, 0.5, 1.5%) for 9 weeks. Body weight gain was not influenced by dietary intake but water intake significantly increased in high Na supplementation. Systolic blood pressure was not influenced by dietary Na and Ca levels but was decreased by dietary low Na/high Ca levels at 9 weeks. Angiotensin-II level was affected by dietary Na level but not by Ca levels. Plasma Ca, Mg, K and Na levels were in the normal range regardless of dietary Na and Ca levels. Weight, and K and Na contents of the heart and kidney were not significantly different among those with different dietary Na and Ca levels. Ca and Mg contents of the heart and kidney were significantly higher in the normal Na/normal Ca group. Ca and Mg in the feces were higher in those with high Ca intake. Na in the feces was higher in those with high Na intake. Therefore, Na and Ca had different mechanisms in the hypertension/hyperlipidemia models, respectively. And we suggested that Mg must be supplemented when Ca intake was high because Mg excretion was increased by Ca supplementation.

Growth Characteristics of Listeria monocytogenes Scott A under High Osmotic Condition and Antibacterial Effect by Morus alba L. Leaf Extract (고삼투압조건하에서 Listeria monocytogenes Scott A의 생육특성과 상엽(桑葉)추출물에 의한 증식억제효과)

  • Growth rate and osmolyte accumulation of L. monocytogenes were measured at the varying concentrations of NaCl. L. monocytogenes accumulated glycine betaine and glutamate intracellularly when grown under osmotic stress by NaCl, and the amounts of them increased as the concentration of NaCl was increased. They were 685 and 345 nmol/mg protein, respectively, when grown in the BHI supplemented with 4% NaCl. In order to inhibit L. monocytogenes effectively, both NaCl and Morus alba L. leaf extract were supplemented in TSB, and antibacterial effect of those supplements on L. monocytogenes was tested. Growth of L. monocytogenes grown in TSB supplemented with 2% NaCl and 100 ppm M. alba leaf extract decreased by 10 times in CFU/ml unit comparing to the growth of control. When grown in TSB, supplemented with 2% NaCl plus 500 ppm M. alba leaf extract and 2% NaCl plus 1,000 ppm M. alba leaf extract, growth of L. monocytogenes decreased by $10^5\;and\;10^8$ times in CFU/ml unit, respectively.

Effect of Additives for Prevention of NaBO2 Precipitation on Hydrogen Generation Properties of NaBH4 Hydrolysis (NaBO2의 석출 방지를 위한 첨가제가 NaBH4 가수분해의 수소발생특성에 미치는 영향)

  • Additives such as glycerol, methanol, acetone, and ethanol were used to prevent $NaBO_2$ from precipitation, and their effects on hydrogen generation properties of $NaBH_4$ hydrolysis were investigated. When the concentration of additives was 5 wt%, the additives such as methanol, acetone, and ethanol could not prevent $NaBO_2$ precipitation. Although glycerol prevented $NaBO_2$ precipitation, conversion efficiency decreased to 78.0% due to its viscosity. Based on test results, hydrogen generation tests were also performed at various concentration of glycerol and methanol to investigate the concentration effects on hydrogen generation properties. As the concentration of glycerol increased from 1 wt% to 3 wt%, conversion efficiency increased owing to additive effect. When its concentration increased to 5 wt%, conversion efficiency decreased due to its viscosity. As the concentration of methanol increased from 5 wt% to 10 wt%, conversion efficiency increased owing to additive effect. When its concentration increased to 15 wt%, conversion efficiency decreased due to $NaB(OCH_3)_4$ precipitate. Although conversion efficiency decreased about 1% when 3 wt% glycerol was added, $NaBO_2$ precipitation was prevented. Consequently, addition of 3 wt% glycerol to $NaBH_4$ solution improves stability of hydrogen generation system.

Effect of $Na^+$ Removal on the Action of Ethanol in Cat Ileal Longitudinal Muscle (장관 평활근에서 Na-free용액이 Ethanol의 효과에 미치는 영향)

  • The effect of ethanol on the contraction of intestinal smooth muscle in Na-free solution was studied using cat ileal longitudinal muscle strips. Ethanol $(0.5{\pm}4%)$ inhibited both the spontaneous mechanical activity and base-line tension in normal physiological salt solution. However, in Na-free solution it induced a reversible contraction. The excitatory effect by ethanol in Na-free solution was increased with increasing the concentrations of ethanol and the time incubated in Na-free solution. The excitatory response by ethanol was reduced by increasing the concentrations of Na in incubated medium . Ethanol-induced contractile response was not affected by $Ca^{2+}$ removal in bathing medium. In Na-free solution, the contraction by ethanol was inhibited by Las. but was not affected by verapamil. The contraction induced by Na removal in solution was inhibited by the pretreatment of ethanol. These results suggest that ethanol may induce the contraction by increasing the release of superficially membrane-bound $Ca^{2+}$ and/or intracellular $Ca^{2+}$ in Na-free physiological salt solution.

Effect of Neutral Salts on the Reactive Dyeing of Silk(I) -Effect of Cations- (중성염이 견의 반응염색에 미치는 영향(I) - 양이온의 영향-)

  • Four kinds of neutral salts with different cations, LiCl, NaCl, KCl, and CsCl, were added to the dye bath to accurately understand the effect of cations on the reactive dyeing of silk with C. I. Reactive Black 5. The cations of salts added lowered the negative surface potential of the silk, improving equilibrium adsorption and the accessibility of the dyestuff to the fiber greatly and speeding up the dyeing rate in the order of $Li^+>Na^+>K^+>Cs^+$. The activation energy$(E_a)$ for the dyeing was in the order of$Li^+>Na^+>K^+>Cs^+$ but the activation free energy$(\Delta{G}^*)$, or the real energy barrier for the reaction, was in the order of $Li^+>Na^+>K^+>Cs^+$ because the degree of the contribution of E$^{a}$ to the activation entropy$(\Delta{S}^*)$ was $Li^+>Na^+>K^+>Cs^+$. It was found from this result that LiCl had the strongest lowering effect on the negative surface potential of silk. The decrease in $\Delta{S}^*$ should be attributed to the loosely bonded activated complex of dyestufffs, cations and fiber molecules at transition state. It was clarified from the Bronsted equation that salts had the ionic strength effect and the specific salt effect on the reactive dyeing.

  • This study is a preliminary investigation of the influence of surface characteristics of substrates on detergency of particulate soil. The surface of PET film was modified with NaOH and NaOH+ethylene diamine on different times. The surface morphology of the film was scanned by AFM and surface energies were calculated from measured contact angles between several solutions and film based on the geometric mean and a Lewis acid base method. The surface morphology of PET film treated with NaOH and NaOH+ethylene diamine became more etched, and the surface area, surface roughness and the coefficient of friction of film increased with treatment of NaOH and NaOH+ethylene diamine. The contact angle of film treated with NaOH and NaOH+ethylene diamine decreased in water and surfactant solution; in addition, the surface energy increased was largely attributed to the increased portion on the polar surface energy of film. However, the effect of the diamine addition to the NaOH treatment solution on surface characteristics of PET film was insignificant.

Effect of sodium on transmembrane calcium movement in the cat ileal longitudinal muscle

  • To get a better insight into the exxistence and the role of a Na-Ca exchange mechanism in smooth muscle, the effect of Na substitution with sucrose on tension development, cellular Ca uptake and $^{45}Ca$ efflux was investigated using isolated cat ileal longitudinal muscle strips. Experimental results were summarized as follows;1) Exposure of the cat ileal longitudinal muscle to Na-free solution induced a contraction, and the magnitude of the contraction increased after incubation of the muscle strips with ouabain ($2{\times10^{-}5}$M) for 1hr. 2) Cellular Ca uptake in Na-free solution increased with an increase in Na content of the Na-loading media, and a linear relationship existed between tissue Na content and cellular Ca uptake for 10 min 3) After tissues were equilibrated in PSS containing $^{45}Ca$ for 2hr, cellular Ca uptake decreased with rising the external Na concentration. 4)Removal of medium Na or inhibition of the Na-K pump decreased the rate of $^{45}Ca$ efflux. These results strongly suggested that Na substitution increases cellular Ca uptake and decreases the rate of $^{45}Ca$ efflux via a Na-Ca exchange mechanism.

Antidoting Effect of 1,8-Naphthalic Anhydride on Butachlor Phytotoxicity in Rice (벼에 있어서 Butachlor 약해(藥害)에 미치는 1, 8-Naphthalic Anhydride 의 약해경감효과(藥害輕減效果))

  • Antidoting effect of 1,8-naphthalic anhydride (NA) on butachlor [2-chloro-2', 6'-diethyl-N-(butoxyethyl) acetanilide] phytotoxicity in rice was determined at different seeding depths and application rates of butachlor. The most sensitive response to NA was found in mesocotyl of rice (Oryza saliva L.). The mesocotyl length decreased with use of NA when seeded 2 to 4 cm deep, whereas no effect was obtained in plant height, root length and coleoptile length. Phytotoxic effect of butachlor to rice sown in vermiculite saturated with butachlor solutions decreased with use of NA at all seeding depths employed. However, depth protection was not observed when planted 3 to 4 cm deep without use of NA. Use of NA resulted in reduction in the phytotoxicity at concentrations of butachlor lower than 40 ppmw. The results indicated that the antidoting effect of NA was not due to reduction of mesocotyl elongation which would result in decrease in butachlor uptake through the mesocotyl.

Effect of Methanethiol Administration on the Erythrocyte Damage in Rats (흰쥐에 Methanethiol 투여가 적혈구막 손상에 미치는 영향)

  • To evaluate an effect of methanethiol on a cause of erythrocyte membrane damage in rats, methanethiol was given at 11.25 rag/100 g body weight, and after 4 hr, the animals were sacrifled, the activities of Na$^+$/K$^+$ ATPase, protein contents in partial purified erythrocyte membrane and erythrocyte indices were determined Concomitantly, in vitro, effect of methanethiol on the erythrocyte fragility, Na$^+$/K$^+$ ATPase activity and its kinetics in various concentration of substrate from the preincubated erythrocyte membrane with methanethiol were demonstrated. The spleen weight per body weight (%) and MCV of erythrocyte in methanethiol-treated rats were more increased than those in the control group. The Na$^+$/K$^+$ ATPase activities in erythrocyte membrane were more decreased in methanethiol-treated rats than those in the control group. The apply of 0.05 ng rat whole blood to the 0.24 mg/ng of methanethiol solution in isotonic condition showed the complete hemolysis. The Na$^+$/K$^+$ ATPase activity in preincubated erythrocyte membrane with methanethiol at 37$\circ$C showed the dual effect and the K$_m$ value of Na$^+$/K$^+$ ATPase was higher in the preincubated erythrocyte membrane with methanethiol than that in the preincubated erythrocyte membrane omitted the methanethiol. These results suggest that the methanethiol may induce the damage of rat's erythrocyte membrane due to a change in substrate binding affinity of Na$^+$/K$^+$ ATPase.
