2,4-Dichlorophenoxyacetic acid (2,4-D) as a widely used herbicide has caused serious environmental problems because of its difficult decomposition in nature. We isolated the strain capable of metabolizing 2,4-D as sole carbon and energy source by an enrichment culture technique from the 2,4-D contaminated soil collected at orchard in Gwangju, Korea. This strain was identified tentatively as Aeromonas sp. NOH2. With this strain, we established the response surface methodology (Box-Behnken Design) to optimize the principle parameters for maximizing biodegradation of 2,4-D such as culture pH, temperature, and nutrient concentration in liquid batch culture. The ranges of parameters were obtained from preliminary works done at our laboratory and chosen as 5.5, 6.5, and 7.5 for pH, 25, 30, and $35^{\circ}C$ for temperature, and 5, 20, and 35 g/1 nutrient concentration. Initial concentration of 2,4-D was 500 ppm and nutrient source was tryptic soy broth. The experimental data were significantly fitted to a second order polynomial equation using multiple regression. The most important parameter influencing 2,4-D degradation and biomass production was nutrient concentration. For 2,4-D degradation, the optimum values of pH and temperature, and nutrient concentration were obtained at pH (6.5), temperature (31.8 to $32.1^{\circ}C$), and nutrient concentration (29.6 to 30.1.0 g/1).
Objective: An experiment was conducted to investigate the effects of a specific mixture of essential oils (MEO), containing thyme, clove and cinnamon EO, on rumen microbial fermentation, nutrient apparent digestibility and blood metabolites in fistulated sheep. Methods: Six sheep fitted with ruminal fistulas were used in a repeated measurement design with two 24-d periods to investigate the effect of adding MEO at 0 (control), 0.8, and 1.6 mL/d on apparent nutrient digestibility, rumen fermentation characteristics, rumen microbial population and blood chemical metabolites. Animals were fed with a 50:50 alfalfa hay:concentrate diet. Results: Ruminal pH, total volatile fatty acids (VFA) concentration, molar proportion of individual VFA, acetate: propionate ratio and methane production were not affected with MEO. Relative to the control, Small peptides plus amino acid nitrogen and large peptides nitrogen concentration in rumen fluid were not affected with MEO supplementation; while, rumen fluid ammonia nitrogen concentration at 0 and 6 h after morning feeding in sheep fed with 1.6 mL/d of MEO was lower (p<0.05) compared to the control and 0.8 mL/d of MEO. At 0 h after morning feeding, ammonia nitrogen concentration was higher (p<0.05) in sheep fed 0.8 mL/d of MEO relative to 1.6 mL/d and control diet. Ruminal protozoa and hyper ammonia producing (HAP) bacteria counts were not affected by addition of MEO in the diet. Relative to the control, no changes were observed in the red and white blood cells, hemoglobin, hematocrit, glucose, beta-hydroxybutyric acid, cholesterol, total protein, albumin, blood urea nitrogen and aspartate aminotransferase and alanine aminotransferase concentration. Apparent total tract digestibility of dry matter, crude proten, organic matter, and neutral detergent fiber were not influenced by MEO supplementation. Conclusion:The results of the present study suggested that supplementation of MEO may have limited effects on apparent nutrient digestibility, ruminal fermentation and protozoa and HAP bacteria count, blood cells and metabolites.
Uptakes of N and P by Phragmites japonica Steudel. are determined in continuous flow tanks as a function of hydraulic retention time (HRT, days) and nutrient concentration (NC). Results show that the uptake was higher at shorter HRT and higher nutrient concentration, and the regression equations were estimated. Mean above ground biomass of Phragmites japonica in the middle reaches of Sagimak stream was 335.92 g/$m^2$, and estimated uptake by this vegetation were 7.31~20.15 NH$_4$-N, 31.15~95.84 NO$_3$-N, and 4.09~11.48 PO$_4$-P mg/$m^2$/day in Sagimak stream, respectively when HRT was 1~5 days. According to this study, it is concluded that Phragmites japonica Steudel. are suitable for water quality improvement of stream through nutrient uptake at short HRT and high concentration.
Field and nutrient cultures of American ginseng (Panax quinquefolius L.) were used to establish foliar symptoms related to boron (B) concentration in leaves and soils, and to evaluate radish as a time-saving model system for B nutrition. Application of excess B, 8 kg/ha versus the recommended 1.5 kg/ha, to field plantings of 2-, 3-, and 4-yr-old American ginseng plants just prior to crop emergence caused, within 4 wk after crop emergence, leaf symptoms of chlorosis followed by necrosis starting at the tips and progressing along the margins. The B concentration in leaves of 2-4-yr-old plants receiving 1.5 kg/ha Bwas $30{\mu}g/g$ dry mass compared to $460{\mu}g/g$ dry mass where 8 kg/ha B was applied. Similarly, B concentration in soils receiving the lower B concentration was 1.8 mg/g dry mass and $2.2-2.8{\mu}g/g$ dry mass where the higher B concentration was applied. Application of 8 kg/ha B reduced the dry yield of 3rd-yr roots by 20% from 2745 kg/ha to 2196 kg/ha and 4th-yr roots by 26% from 4130 kg/ha to 3071 kg/ha. Ginseng seedlings and radish were grown under greenhouse conditions in nutrient culture with four B concentrations ranging from 0 mg/L to 10 mg/L. At 5 mg/L and 10 mg/L ginseng and radish developed typical leaf B toxicity symptoms similar to those described above for field-grown plants. Increasing B in the nutrient solution from 0.5 mg/L to 10 mg/L decreased, in a linear fashion, the root and leaf dry mass of ginseng, but not radish. Given the many similarities of ginseng and radish to B utilization, radish might be used as a timesaving model system for the study of B, and other micronutrients, in the slow-growing perennial ginseng.
This study was carried out to evaluate soil carbon and nutrient storage of three adjacent coniferous plantations (Larix leptolepis, Pinus densiflora and Pinus rigitaeda) growing on a similar site with a same planting age (42-year old) in the Sambong Exhibition Forests, Hamyang-gun, Gyungsangnam-do. The soil carbon concentration among three plantations was not significantly different in 0∼10cm soil depth, but other two depths (10∼20cm and 20∼30cm) showed higher carbon concentration in P. densiflora plantation than the other two plantations. The exchangeable cation concentrations (Ca and Mg) in 0∼10cm depth were significantly lower in L. leptolepis plantation than in the other two plantations, while nitrogen and phosphorus concentrations were not significantly different among three plantations except for nitrogen at 10∼20cm depth in P. rigitaeda plantation. Soil carbon storage in 0∼20cm depth of three plantations was unaffected by the stand types. Soil nutrient storage was not significantly different at each depth except for nitrogen storage at 10∼20cm depth in P. rigitaeda plantation because of the variation of bulk density and coarse fragment. This result demonstrates that soil carbon and nutrient concentrations among the plantations on a similar soil condition can be altered significantly by tree species effects over 40 years after plantation establishment.
Controlling the light energy and major nutrients is important for high cell density culture of cyanobacterial cells. The growth phase of Anabaena variabilis can be divided into an exponential growth phase and a deceleration phase. In this study, the cell growth in the deceleration phase showed a linear growth pattern. Both the period of the exponential growth phase and the average cell growth rate in the deceleration phase increased by controlling the light intensity. To control the light intensity, the specific irradiation rate was maintained above $10\;{\mu}mol/s/g$ dry cell by increasing the incident light intensity stepwise. The final cell density increased by controlling the nutrient supply. For the control of the nutrient supply, nitrate, phosphate, and sulfate were intermittently added based on the growth yield, along with the combined control of light intensity and nutrient concentration. Under these control conditions, both final cell concentration and cell productivity increased, to 8.2 g/l and 1.9 g/l/day, respectively.
This study was conducted to evaluate appropriate selenate ion concentration for the production of high functional vegetables. Sodium selenate was treated 0, 2, 4, 6 and 8mg/$\ell$ using herb nutrient solution developed by European R & D Vegetable Center in Belgium. Low level of Na2se04 concentration increased the growth of wormwood, but high selenate concentration decreased the growth. Total chlorophyll content was increased by sodium selenate. The higher selenate ion concentration in the nutrient solution, the more total chlorophyll content was. The vitamin C content in wormwood was high at 2 and 4mg/$\ell$ treatment, showing good growth, at higher concentration, however, the vitamin C content decreased. At 4mg/$\ell$ selenate ion concentration, essential oil content of wormwood was best. But higher selenate ion concentration decreased essential oil content. The uptake of Se by the plant increased with the increase of selenate ion concentration.
This study was carried out to investigate the effects of nutrient solution concentration, irrigation frequency on growth, flowering, and cut flower longevity of Dendranthema grandiflorum (Ramat.) Kitamura 'Shuhouno-chikara' hydroponically grown in perlite media. Not only stem length but also mineral contents of each plant organ in nutrient solution culture were much higher than those in soil culture. 1/2S of nutrient solution was good at early stage, but 1S of nutrient solution was better as chrysanthemum growth progressed. Among different concentrations of nutrient solution, mineral contents of each plant part showed no significant difference. 1S treatment of nutrient solution increased the vase life by 3 days than 2S treatment. The growth and mineral contents of each plant organ were great in plants irrigated 8 times a day, because of high moisture contents of medium and high water availability, followed by more frequent nutrient replenishment near roots. There was no correlation between nutrient solution concentration and vase life of cut flower grown in nutrient solution culture. Cut flowers irrigated twice per day had the longer vase life than other treatments (4, 8 times).
In this study, effects of nutrient and inorganic carbon on single cell emergence during the cultivation of microalgae were observed using colonial green algae, Pediastrum duplex. The concentration of inorganic carbon had significant effect on single cell emergence and its growth, but nitrogen and phosphorus concentration showed minor effects. According to P. duplex cultivation experiment, single cell started to be emerged around 500~750 mg-C/L of inorganic carbon concentration and it was bloomed dramatically at the higher values. And growth of P. duplex was started to be surpressed at the single cell formation concentration. From the results, it could be said that when we operate the microalgae systems for cultivation/harvesting or wastewater treatment, in order to avoid single cell formation, inorganic carbon should be maintained to the proper level.
This study was conducted to evaluate the optimum hydroponic system and nutrient solution for shortening the early growth period and quality improvement of dendrobium ( Dendrobium Phalaenopsis) seedlings. Dendrobium seedlings with 3 to 4 leaves were transplanted in the deep flow technique(DFT) system, aeroponic system, and ebb and flow system with different concentration of balanced nutrient solutions recommended by the Japanese Horticultural Experiment Station. Growth characteristics of shoot and root were recorded and evaulated among treatments. For autumn cultivation, plant height was the longest at the DFT system with quarter concentration of nutrient solution, where aeroponic system with half concentration of nutrient solution. Aeroponic system stimulated the root growth but fresh weight was observed in the plots of DFT system. For spring cultivation, pH values increased up 7.5 at the DFT and aeroponic system, where EC values did not fluctuate regardless of cultural system. Ebb and flow system showed the best result in the growth of plant followed by BFT system and aeroponic system. Higher concentration of nutrient solution within this range of treatment was recommended for the growth promotion of leaf length and width in DFT system. In conclusion, growth responses differed depending on the cultural system, concentrations of nutrient solutions and duration of cltivation.
