Generation of Antibodies Against Rice stripe virus Proteins Based on Recombinant Proteins and Synthetic Polypeptides

  • Rice stripe virus (RSV) is one of serious epidemic pathogens for rice species grown in many Asian countries. Therefore, it is necessary to produce a diagnostic detection kit applicable in fields for RSV detection. In this study, RSV proteins that were derived from recombinant proteins and synthetic polypeptides as antigens were generated and were raised in rabbits for antiserum production. Among seven proteins in RSV, genes that code for NCP and NS3 proteins were cloned and subcloned into vector carrying His-tag protein and were expressed in E. coli. Of two recombinant proteins, only anti-NCP displayed stable hybridization signals in western blot analysis. Alternately, synthetic RSV polypeptides for CP, NCP, NS3 and NSvc4 we also generated and only antibodies against CP and NCP were very effective to detect RSV in both RSV infected rice and weed plants. However, antibodies against NS3 and NSvc4 showed weak specific bands as well as strong non-specific background due to the difference of viral proteins produced in the infected leaves. In summary, the antibodies generated against RSV proteins produced in this study will be useful for various assays such as for RSV diagnostic detection, immunoprecipitation, protein purification, and western blot analysis.

Changes in the thyroid hormone profiles in children with nephrotic syndrome

  • Purpose: We compared thyroid hormone profiles in children with nephrotic syndrome (NS) during the nephrotic phase and after remission. Methods: This study included 31 pediatric NS patients. The thyroid hormone profiles included serum levels of triiodothyronine (T3), thyroxine (T4), thyroid-stimulating hormone (TSH), and free T4. Results: Of the 31 patients, 16 (51.6%) showed abnormal thyroid hormone profiles: 6 had overt hypothyroidism, 8 had subclinical hypothyroidism, and 2 had low T3 syndrome. The mean serum T3, T4, and free T4 levels in the nephrotic phase and after remission were $82.37{\pm}23.64$ and $117.88{\pm}29.49ng/dL$, $5.47{\pm}1.14$ and $7.91{\pm}1.56{\mu}g/dL$, and $1.02{\pm}0.26$ and $1.38{\pm}0.23ng/dL$, respectively; the levels were significantly lower in the NS nephrotic phase (P=0.0007, P<0.0001, and P=0.0002). The mean serum TSH levels during the nephrotic phase and after remission were $8.05{\pm}3.53$ and $4.08{\pm}2.05{\mu}IU/mL$, respectively; they were significantly higher in the nephrotic phase (P=0.0005). The urinary protein/creatinine ratio during the nephrotic phase was significantly correlated with serum T3, T4, and free T4 levels (r=-0.5995, P=0.0032; r=-0.5797, P=0.0047; r=-0.5513, P=0.0078) as well as with TSH levels (r=0.5022, P=0.0172). A significant correlation was found between serum albumin and serum T3 levels during the nephrotic phase (r=0.5385, P=0.0018) but not between serum albumin and T4, TSH, or free T4 levels. These significant correlations all disappeared after remission. Conclusion: Abnormal thyroid hormone profile findings were observed in 51.6% of pediatric patients with NS. Thyroid hormone levels normalized after remission, regardless of levothyroxine therapy.

소아 특발성 신증후군에서의 cyclosporine A(Cipol-$N^{(R)}$)의 치료 효과 (Cyclosporine A (Cipol-$N^{(R)}$) Therapy in Children with Idiopathic Nephrotic Syndrome)

  • 목 적 : 소아 특발성 신증후군에게 cyclosporine A(Cipol-$N^{(R)}$,종근당)를 투여하여 이의 효과와 부작용을 관찰하고 또한 기존의 cyclosporine A 제제를 사용한 치료 효과와 비교하고자 본 연구를 시행하였다. 방 법 : 대상 환아는 특발성 신증후군 환아로서 스테로이드 의존형(SD) 또는 빈발 재발형(FR) 39명과 스테로이드 저항형(SR) 3명으로 하였으며 이들에게 cyclosporine A를 1일 체중당 4-5mg을 경구 투여하고 스테로이드 제제 (prednisolone 혹은 같은 역가의 deflazacort)는 격일 요법으로 감량 투여하였다. 4개월의 조사 기간중 2, 4, 8, 12, 16주에 이들 환아에 대한 문진, 이학적 검사 및 각종 검사를 시행하였다. 결 과 : 39명의 SD/FR NS 환아중 35명 ($89.7\%$)에서 16주간 관해가 지속되었으며 혈청 단백, 알부민, 콜레스테롤 및 24시간 뇨 단백은 치료 개시후 4주째에 정상 수치를 보였고 BUN, creatinine 및 24시간 뇨를 이용한 사구체 여과율은 16주간 변동 사항이 없었다. 3명의 SR NS 환아에서는 2명에서 완전 관해가 유도되고 지속되었으며 1명에서는 반응이 없었다. Cyclosporine A에 대한 부작용은 다모증이 8명, 고뇨산혈증이 5명에서 관찰되었으나 이로 인해 투약을 중지할 필요가 없었으며 기타 CBC, 간기능 검사, 혈압 등은 16주간 정상 소견을 보였다. 결 론 : 스테로이드 의존형, 빈발 재발형 및 스테로이드 저항형의 특발성 신증후군 환아에게 cyclosporine A는 유용하게 사용할 수 있을 약제로 생각되며 본 연구의 다음 과제는 cyclosporine A의 장기 투여(1-2년)로서 얻을 수 있는 효과 및 약제의 안전성 (특히 신독성)여부를 확인하여야 할 것으로 생각한다.

청폐사간탕(淸肺瀉肝湯)이 $CCl_4$로 유발(誘發)된 휜쥐의 간손상(肝損傷)에 마치는 영향(影響) (The Effects of Chungpyesagan-tang on the Recovery of Liver Function in Rat Injured by $CCl_4$)

  • 1. Objectives The present study was carried out to investigate the effects of Chungpyesagan-tang on the $CCI_4-induced$ Liver Damage in Rats. 2. Methods Sprague-Dawley rats were devided into 5 experimental groups : Normal, $NS+CCI_4(Solid$ extract of $CCI_4$ injection group after Normal Saline feed), $CP+CCI_4(Solid$ extract of $CCI_4$ injection group after Chungpyesagantang feed), $CCI_4+NS(Nomal$ Saline feed group after $CCI_4$ injection), $CCI_4+CP(Solid$ extract of Chungpyesagantang feed group after $CCI_4$ injection). Biochemical assays for serum enzyme activities such as AST, ALT, ALP, BUN, Creatinine, Uric Acid, Total Protein, Albumin, Total Cholesterol, Triglyceride, Glucose, and mRNA Revelation of Cytochrome p450 and activities such as LPO(Lipid Peroxidation), GSH(Glutathione), GST(Glutathione-S- Transferase), Glutathione Reductase, Glutathione Peroxidase, SOD(Superoxide Dismutase), Catalase, Hydroxyproline, and ${\beta}-Glucuronidase$ were performed. 3. Results 1) $CP+CCI_4$ showed significantly lower relation of Cytochrome p450 than $NS+CCI_4$. 2) As to LPO Hydroxyproline, $CCI_4+CP$ showed significantly lower activity than $CCI_4+NS$. 3) As to GSH GST Glutathione Peroxidase Catalase, $CP+CCI_4$ showed higher activity than $NS+CCI_4$, $CCI_4+CP$ showed significantly higher activity than $CCI_4+NS$. 4) As to Glutathione Reductase SOD, $CCI_4+CP$ showed significantly higher activity than $CCI_4+NS$. 5) As to ${\beta}-Glucuronidase$, $CP+CCI_4$ showed signficantly lower activity than $NS+CCI_4$. 4. Conclusions Chungpyesagantang has the recovering effects on the $CCI_4-induced$ Liver Damage.

Antiviral effect of fucoxanthin obtained from Sargassum siliquastrum (Fucales, Phaeophyceae) against severe acute respiratory syndrome coronavirus 2

  • Human coronavirus diseases, particularly severe acute respiratory syndrome coronavirus 2, still remain a persistent public health issue, and many recent studies are focusing on the quest for new leads against coronaviruses. To contribute to this growing pool of knowledge and explore the available marine natural products against coronaviruses, this study investigated the antiviral effects of fucoxanthin isolated from Sargassum siliquastrum-a brown alga found on Jeju Island, South Korea. The antiviral effects of fucoxanthin were confirmed in severe acute respiratory syndrome coronavirus 2-infected Vero cells, and its structural characteristics were verified in silico using molecular docking and molecular dynamic simulations and in vitro colorimetric method. Fucoxanthin inhibited the infection in a concentration-dependent manner, without showing cytotoxicity. Molecular docking simulations revealed that fucoxanthin binds to the angiotensinconverting enzyme 2-spike protein (binding energy -318.306 kcal mol-1) and main protease (binding energy -205.118 kcal mol-1). Moreover, molecular dynamic simulations showed that fucoxanthin remains docked to angiotensin-converting enzyme 2-spike protein for 20 ns, whereas it breaks away from main protease after 3 ns. Also, the in silico prediction of the fucoxanthin was verified through the in vitro colorimetric method by inhibiting the binding between angiotensinconverting enzyme 2 and spike protein in a concentration-dependent manner. These results indicate that fucoxanthin exhibits antiviral effects against severe acute respiratory syndrome coronavirus 2 by blocking the entry of the virus. Therefore, fucoxanthin from S. siliquastrum can be a potential candidate for treating coronavirus infection.

3D-QSAR, Docking and Molecular Dynamics Simulation Study of C-Glycosylflavones as GSK-3β Inhibitors

  • Abnormal regulation, hyperphosphorylation, and aggregation of the tau protein are the hallmark of several types of dementia, including Alzheimer's Disease. Increased activity of Glycogen Synthase Kinase-3β (GSK-3β) in the Central Nervous System (CNS), increased the tau hyperphosphorylation and caused the neurofibrillary tangles (NFTs) formation in the brain cells. Over the last two decades, numerous adenosine triphosphate (ATP) competitive inhibitors have been discovered that show inhibitory activity against GSK-3β. But these compounds exhibited off-target effects which motivated researchers to find new GSK-3β inhibitors. In the present study, we have collected the dataset of 31 C-Glycosylflavones derivatives that showed inhibitory activity against GSK-3β. Among the dataset, the most active compound was docked with the GSK-3β and molecular dynamics (MD) simulation was performed for 50 ns. Based on the 50 ns MD pose of the most active compound, the other dataset compounds were sketched, minimized, and aligned. The 3D-QSAR based Comparative Molecular Field Analysis (CoMFA) model was developed, which showed a reasonable value of q2=0.664 and r2=0.920. The contour maps generated based on the CoMFA model elaborated on the favorable substitutions at the R2 position. This study could assist in the future development of new GSK-3β inhibitors.

사람 선유아세포 인터페론(Hu IFN-$\beta$)에 대한 단 Clone성 항체생산세포의 조작과 그 성질에 관한 연구 (Preparation and Characterization of Cell Hybrids Producing a Monoclonal Antibody to Human Fibroblast Interferon (Hu IFN-$\beta$))

  • 사람 선유아세포 인터페론의 정제에 사용되는 단 clone성 항체생산 세포주를 조작하기 위하여 BA-LB/C mouse의 복강과 꼬리정맥을 통하여 HuIFN-$\beta$를 면역화시키고 그 비장세포(spleen cells) 와 NS-O 세포주를 세포융합 시켰다. 융합된 1300 hybrids를 ELISA방법으로 선별하고 soft agarose 방법과 limiting dilution방법으로 subcloning하여 높은 항체를 생성하는 것으로 판명된 11 hybrids를 재선별 하였다. 재선별된 11 hybrids 각각의 항체형 (Ig type)을 조사하고 최종 Protein A-sepharose와 친화성이 높은 IgG 2a/형의 clone # 4-1-19와 clone # 551-4-1을 선별하여 배양된 세포를 각각 nude(nu/nu) mouse 및 BALB/c mouse 복강에 접종배양 하였다. 이들 mouse복강액으로 부터 얻은 ascites fluid를 protein A-sepharose를 이용한 affinity column분획으로 항체를 정제하였으며 ascites fluid $m{\ell}$당 약 4mg의 정제된 항체를 얻을 수 있었고 SDS-polyacrylamide gel상에서 전기영동 시킨 결과 분자량 14-16만 dalton으로 추정되는 항체를 확인할 수 있었다.

인간 A549 폐암세포에서 비스테로이드성 항염증제와 genistein의 복합처리에 의한 NAG-1 의존적 세포사멸 증진 효과 (Combined Treatment of Nonsteroidal Anti-inflammatory Drugs and Genistein Synergistically Induces Apoptosis via Induction of NAG-1 in Human Lung Adenocarcinoma A549 Cells)

  • 비스테로이드성 항염증약(nonsteroidal anti-inflammatory drugs; NSAIDs)은 항염 및 진통효과를 나타내며, 염증억제 외에 다양한 신호전달 분자를 통해 여러 가지 세포생리활성을 조절하며, 암세포에서는 세포사멸 유도를 통한 항암제 효과를 보이고 있다. 본 연구에서는 NSAIDs가 암세포사멸프로그램을 작동시키는데 있어 phosphatidyl inositol 3-kinase (PI3K)-Akt/protein kinase B (PKB) 그리고 MEK1/2-ERK1/2 신호 전달계과 같은 anti-apoptotic program이 NSAIDs의 효과를 경감시키는 것으로 예상하고, 이들 항세포사멸 프로그램을 억제하였을 경우, NSAIDs의 세포사멸 유도작용이 증가되는지 그 가능성을 조사하였다. 세포사멸은 Hoeschst 33342으로 핵응축과 핵 쪼개짐을 염색하여 확인하였다. Western blotting을 통해 단백질 발현과 역전사중합효소연쇄반응을 통해 mRNA 발현을 확인하였다. NSAIDs 처리와 동시에 PI3K-Akt/PKB와 MEK-ERK1/2 신호전달계의 억제제를 함께 처리했을 때, NSAIDs의 세포사멸유도작용이 증가함을 확인하였다. 또한 PI3K와 MEKl/2 신호전달계의 상위에 존재하는 receptor tyrosine kinases (RTKs)의 억제제인 genistein을 함께 처리하였을 때에도 유사한 효과가 나타남을 확인하였다. 그리고 이들 복합처리에 의해 NAG-1 발현이 증가하며 NAG-1 interference 하였을 경우 복합처리에 의한 세포사멸증진 효과가 사라짐을 확인하였다. 본 연구결과는 암세포에 활성화 되어 있는 세포생존프로그램을 제어하는 물질(genistein 혹은 LY294002+U0126)을 복합처방함으로써 NSAIDs의 항암작용을 증진시킬 수 있음을 보여준다.
