Recovery of TRIM25-Mediated RIG-I Ubiquitination through Suppression of NS1 by RNA Aptamers

  • Non-structural protein 1 (NS1) of influenza virus has been shown to inhibit the innate immune response by blocking the induction of interferon (IFN). In this study, we isolated two single-stranded RNA aptamers specific to NS1 with $K_d$ values of $1.62{\pm}0.30nM$ and $1.97{\pm}0.27nM$, respectively, using a systematic evolution of ligand by exponential enrichment (SELEX) procedure. The selected aptamers were able to inhibit the interaction of NS1 with tripartite motif-containing protein 25 (TRIM25), and suppression of NS1 enabled retinoic acid inducible gene I (RIG-I) to be ubiquitinated regularly by TRIM25. Additional luciferase reporter assay and quantitative real-time PCR (RT-PCR) experiments demonstrated that suppression of NS1 by the selected aptamers induced IFN production. It is noted that viral replication was also inhibited through IFN induction in the presence of the selected aptamers. These results suggest that the isolated aptamers are strongly expected to be new therapeutic agents against influenza infection.

Porcine parvovirus nonstructural protein NS1 activates NF-κB and it involves TLR2 signaling pathway

  • Background: Porcine parvovirus (PPV) is a single-stranded DNA virus that causes porcine reproductive failure. It is of critical importance to study PPV pathogenesis for the prevention and control of the disease. NS1, a PPV non-structural protein, is participated in viral DNA replication, transcriptional regulation, and cytotoxicity. Our previous research showed that PPV can activate nuclear factor kappa B (NF-κB) signaling pathway and then up-regulate the expression of interleukin (IL)-6. Objectives: Herein, the purpose of this study is to determine whether the non-structural protein NS1 of PPV also has the same function. Methods: Real-time quantitative reverse transcription polymerase chain reaction (RT-qPCR), enzyme-linked immunosorbent assay, western blot, immunofluorescence assay and small interfering RNA (siRNA) were used. Results: Our findings demonstrated that PPV NS1 protein can up-regulate the expression levels of IL-6 and tumor necrosis factor-alpha in a dose-dependent manner. Moreover, PPV NS1 protein was found to induce the phosphorylation of IκBα, then leading to the phosphorylation and nuclear translocation of NF-κB. In addition, the NS1 protein activated the upstream pathways of NF-κB. Meanwhile, TLR2-siRNA assay showed TLR2 plays an important role in the activation of NF-κB signaling pathway induced by PPV-NS1. Conclusions: These findings indicated that PPV NS1 protein induced the up-regulated of IL-6 expression through activating the TLR2 and NF-κB signaling pathways. In conclusion, these findings provide a new avenue to study the innate immune mechanism of PPV infection.

Immobilization Metallocene Inside Surface-functionalized Nanopore of Micelle-Templated Silica and its Ethylene Polymerization (표면 기능화된 Micelle-Templated Silica 나노세공 내 메탈로센 담지 및 에틸렌 중합)

  • A functionalization of mesoporous materials with organosilane was carried out via a post-synthesis grafting method and $(n-BuCp)_2ZrCl_2$/methylaluminoxane (MAO) as subsequently immobilized on the functionalized mesoporous materials for ethylene polymerization. Organosilanes having amine, cyano or imidazoline group such as $N$-[(3-trimethoxysilyl)propyl]ethylenediamine (2NS), 4-(triethoxysilyl)butyronitrile (1NCy), 1-(3-triethoxysilylpropyl)-2-imidazoline (2NIm) were used for the surface functionalization of mesoporous materials. In the SBA-15/2NS/$(n-BuCp)_2ZrCl_2$ catalyst preparation, the amount of MAO in feed increased with an decrease in the Zr content of the supported catalyst, and Al content in the supported catalyst increased. The ethylene homopolymerization activity of SBA-15/2NS/$(n-BuCp)_2ZrCl_2$ dramatically increased as the amount of MAO in feed increased. Furthermore, when the immobilization time was 6 hrs, SBA-15/2NS/$(n-BuCp)_2ZrCl_2$ showed the highest activity. The activities of supported 2NS-, 1NCy-, 2NIm-functionalized catalysts decreased in the following order, SBA-15/2NS/ > SBA-15/2NIm/ > SBA-15/1NCy/$(n-BuCp)_2ZrCl_2$. 2NS and 2NIm which have two amine groups per silane molecule were shown to interact with $(n-BuCp)_2ZrCl_2$ strongly compared to 1NCy which has one amine group. Thus, the activities increased with an increase in the nitrogen and the Zr content of the supported catalysts.

  • The new optical link receiver with data transfer rate higher than 10Mbps at the supply voltage of 1.8V was designed and fabricated using bipolar technology. The fabricated IC showed the dissipation current of 4.6mA under high level input voltage of 1.5V. The high level output voltage($V_{OH}$) and the low level output voltage($V_{OL}$) were 1.15V and 0V, respectively, for a given 10 Mbps signal which has duty ratio of 50%, $V_{IL}$(low level input voltage) of 0.5V, and $V_{IH}$(high level input voltage) of 1.5V, The duty ratio of output waveform was 52.6%. The rising time(t$_{r}$) and the falling time(t$_{f}$) were 9.5ns and 6.8ns, respectively. The propagation delay difference($t_{PHC}-t_{PLH}$) and the jitter($t_j$) were 11.7ns and 4.3ns, respectively.y.

  • Serpins are a group of proteins involved in the regulation of serine and other type of proteases, and have been identified in many kinds of organisms from invertebrates to vertebrates. Serpins are known to regulate the proteolytic cascades of the innate immune pathways in addition to their roles in blood coagulation, angiogenesis, fibrinolysis, inflammation and tumor suppression. In this study, we have isolated two partial serpin gene fragments from expressed sequence tags (ESTs) of Nesiohelix samarangae. Dotplot analysis indicates that they are of two different types, Ns-serpin type 1 and Ns-serpin type 2. Ns-serpin type 1 has 819 bp coding region (272 amino acids), whereas Ns-serpin type 2 has 555 bp coding region (185 amino acids). Molecular phylogenetic analysis shows that the identified serpins have high similarities to their counterparts in the California see slug, Aplysia californica. Yet, the precise biological and immunological roles of these Ns-serpins remain to be further investigated using RNA interference and other molecular techniques.

In vitro Study of Nucleostemin as a Potential Therapeutic Target in Human Breast Carcinoma SKBR-3 Cells

  • Although nucleolar protein nucleostemin (NS) is essential for cell proliferation and early embryogenesis and expression has been observed in some types of human cancer and stem cells, the molecular mechanisms involved in mediation of cell proliferation and cell cycling remains largely elusive. The aim of the present study was to evaluate NS as a potential target for gene therapy of human breast carcinoma by investigating NS gene expression and its effects on SKBR-3 cell proliferation and apoptosis. NS mRNA and protein were both found to be highly expressed in all detected cancer cell lines. The apoptotic rate of the pcDNA3.1-NS-Silencer group ($12.1-15.4{\pm}3.8%$) was significantly higher than those of pcDNA3.1-NS ($7.2-12.0{\pm}1.7%$) and non-transfection groups ($4.1-6.5{\pm}1.8%$, P<0.01). MTT assays showed the knockdown of NS expression reduced the proliferation rate of SKBR-3 cells significantly. Matrigel invasion and wound healing assays indicated that the number of invading cells was significantly decreased in the pcDNA3.1-NS-siRNA group (P<0.01), but there were no significant difference between non-transfected and over-expression groups (P>0.05). Moreover, RNAi-mediated NS down-regulation induced SKBR-3 cell G1 phase arrest, inhibited cell proliferation, and promoted p53 pathway-mediated cell apoptosis in SKBR-3 cells. NS might thus be an important regulator in the G2/M check point of cell cycle, blocking SKBR-3 cell progression through the G1/S phase. On the whole, these results suggest NS might be a tumor suppressor and important therapeutic target in human cancers.

  • Nephrotic syndrome (NS) is the most common glomerular disorder in childhood, and a vast majority of cases are idiopathic. The precise cause of this common childhood disease is not fully elucidated despite significant advancements in our understanding of podocyte biology. Idiopathic NS has been considered "a disorder of T-cell function" mediated by a circulating factor that alters podocyte function resulting in massive proteinuria since the last four decades. Several circulatory factors released from T-cells are considered to be involved in pathophysiology of NS; however, a single presumptive factor has not been defined yet. Extended evidence obtained by advances in the pathobiology of podocytes has implicated podocytes as critical regulator of glomerular protein filtration and podocytopathy. The candidate molecules as pathological mediators of steroid-dependent NS are CD80 (also known as B7-1), hemopexin, and angiopoietin-like 4. The "two-hit" hypothesis proposes that the expression of CD80 on podocytes and ineffective inhibition of podocyte CD80 due to regulatory T-cell dysfunction or impaired autoregulation by podocytes results in NS. Recent studies suggest that not only T cells but also other immune cells and podocytes are involved in the pathogenesis of MCNS.

  • We have proposed and analysed a novel Lookup Algorithm which had a short switching speed and tiny memory size for IP router. This algorithm could simply be implemeted by a hardware with SRAM because of simple structure. This Lookup scheme needs 1${\sim}$3 memory access times. When we simulated with 40,000 routing record obtained from IPMA Website, the maximum memory size of this algorithm was 316KB(the offset threshold for compression algorithm was 8). When we simulated by HDL using ALTERA EPM7256 series and 100MHz clock and SRAM of 10ns access time, the total lookup time was 45ns for two memory access, 175ns for three memory access.

Effect of Surface Treatment Condition of Aminosilane on Ethylene Polymerization of Supported Metallocene (아미노실란 표면 처리 조건이 담지메탈로센 촉매의 에틸렌 중합에 미치는 영향)

  • The effects of surface treatment method of unreacted N-[3-(trimethoxysilyl)propyl]ethylenediamine (2NS), $N^1$-(3-trimethoxysilylpropyl)diethylenetriamine (3NS), and 3-cyanopropyltriethoxysilane (1NCy) after grafting on the surface of silica and of the surface treatment temperature on ethylene polymerization were investigated. The Zr content of supported catalyst employing filtering method was higher than that of washing method, and the activities of supported catalysts prepared by washing method were higher than those of filtering methods significantly. Regardless of surface treatment methods the activities were in order by $SiO_2/2NS/(n-BuCp)_2ZrCl_2>SiO_2/1NCy/(n-BuCp)_2ZrCl_2>SiO_2/3NS/(n-BuCp)_2ZrCl_2$. The ethylene polymerization activity was increased as the surface treatment temperature of aminosilane on silica increased.

Characteristics of Partial Discharge Under HVDC in SF6 Gas (SF6 가스 중 직류 고전압 하에서 부분방전 특성)

  • This paper dealt with the measurement and analysis of partial discharge (PD) under high voltage direct current (HVDC) in SF6 gas. Electrode systems such as a protrusion on conductor (POC), a protrusion on enclosure (POE), a crack on epoxy plate and a free particle (FP) were fabricated to simulate the insulation defects. The analysis system was designed with a Time-Frequency (T-F) map algorithm programed based on LabVIEW. This can arrange the acquired PD pulses into frequency and time domain. A HVDC power source is composed of a transformer (220 V/50 kV), a diode (100 kV) and a capacitor (50 kV, 0.5 ${\mu}F$). The gap between the electrodes is 3 mm, and the $SF_6$ gas was set at 5 bar. PD pulses were detected by a 50 ${\Omega}$ non-inductive resistor. In the analysis, PD pulses were distributed below 0.5 MHz and 20 ns ~ 35 ns for the POC, 0.7 MHz ~ 1.7 MHz, below 0.6 MHz and 10 ns ~ 40 ns and 60 ns ~125 ns for the POE, below 0.1 MHz and 135 ns ~ 215 ns for the crack, and below 1.6 MHz and 250 ns for the FP.
