Effects of Carbofuran and Compensation of Phenobarbital sodium in the NIH 3T3 Fibroblast and Rat Kidney (Carbofuran이 NIH 3T3섬유모세포와 흰쥐의 신장에 미치는 영향과 Phenobarbital sodium의 보상효과에 관한 연구)

  • This study was carried out to investigate toxicity of insecticide carbofuran and compensatory effects of phenobarbital sodium (PB) in vivo and in vitro. Sprague Dawley male rats were used as experimental animals and divided into carbofuran only administered group and simultaneous application group of carbofuran and PB. At 30 rain and 1, 3, 6, 12, 24, 48 and 96 hrs after each treatment, the animals were sacrificed by decapitation. Kidney were immediately removed, immersed in fixatives, and processed with routine method for light microscopic study. Paraffin sections were stained with H-E, PAM and PAS. $5.0\times 10^4$ cell/ml of NIH 3T3 fibroblast in each well of 24 multidish were cultured: After 24 hours, the cells were treated with solution of six groups; control group cultured in media only, carbofuran $MTT_50$ or $NR_50$ group cultured in the media containing carbofuran $MTT_50$ or $NR_50$ and four experimental groups cultured in the media containing carbofuran $NR_50$ plus various concentratins of PB. After the NIH 3T3 fibroblast of all groups were cultured in same condition for 48 hours, Tetrazolium MTT (MTT) and NR (neutral red) assay were performed to evaluate the cytotoxicity of cell organelles. Under the light microscope, atrophic change of renal corpuscles were frequently observed in 1 and 2 days after carbofuran treatment. The increase of the mesangium was apparent in 1 and 2 days after carbofuran treatment. Necrotic changes of the epithelium and loss of brush border of proximal tubules were most severe at 2 and 3 days after carbofuran treatment, respectively. In contrast, there were no evidences of the toxic effects on renal tissues at 48hrs in carbofuran-PB treated groups. Carbofuran $MTT_50$ and $NR_50$ were 78$\mu M$, 82.5$\mu M$ respectively. MTT and NR quantities were significantly increased in carbofuran-PB 100$\mu M$ treatment group and carbofuran-PB 100$\mu M$ treatment group. On the basis of these results, it is obvious that PB has compensatory effects against carbofuran toxicity.

  • Recently, photodynamic and photothermal therapies have received increasing attention as an effective cancer treatment. In this study, a gold nanorod (AuNR) colloidal solution was synthesized as a hyperthermia agent for photothermal therapy and also modified with photosensitizer (PS) for photodynamic therapy. PEG (polyethylene glycol) and FA (folic acid) ligand were also introduced into AuNR for the long circulation in human body and efficient targeting of cancer cells, respectively and AuNRs were modified with FA-PEG and poly-${\beta}$-benzyl-L-aspartate (PBLA) block copolymers through a 3,4-dihydroxy hydrocinnamic acid (HCA) linker. A series of AuNRs with various aspect ratios were synthesized by controlling the feeding ratio of $AgNO_3$. The physicochemical property and morphology of synthesized AuNR100 and FA-PEG-$P(Asp)_{50}$-HCA-AuNR100 were analyzed by UV-visible spectrophotometer, $^1H$ NMR, XPS measurements, TEM. The surface modified AuNR carrier with biocompatibility could be applied for the effective diagnosis as well tumor phototherapy.

A Study on the Elution Mechanism of Ni(II)-Isonitrosoethylacetoacetate Imine Chelates by Reversed Phase High Performance Liquid Chromatography (역상 액체 크로마토그래피에 의한 Ni(II)-Isonitrosoethylacetoacetate Imine 유도체 킬레이트의 용리 메카니즘에 관한 연구)

  • Liquid Chromatographic behavior of Ni(II) in Isonitrosoethylacetoacetate Imine(IEAA-NR), Ni(IEAA-NH)(IEAA-NR)(R = H, CH_3, C-2H_5, n-C_4H_9, C_6H_5-CH_2) chelates were investigated by reversed-phase HPLC on Micropak MCH-5 column using methanol/water as mobile phase. The optimum conditions for the separation of Ni(IEAA-NH)(IEAA-NR) chelates were examined with respect to the effect of the flow rate, sample solvent, mobile phase strength and column temperature. It was fo$und that metal chelates were properly eluted in an acceptable range of capacity factor value(0{\le}logk'{\le}1). The dependence of the logarithm of capacity factor (k') on the volume fraction of water in the binary mobile phase as well as on the liquid-liquid extraction distribution ratio (Dc) in methanol-water/n-alkane extraction system showed the good linearties, and the dependence of the logarithm of capacity factor (k') on the column temperature and on the enthalpy exhibited the good linearties, and the compensation temperature ({\beta}) from the slope was 773.47{\circ}K. It was suggested that the retention of metal chelates was largely affected by the hydrophobic effect.

A phylogenetic analysis of the genus Pilea (Urticaceae) using nrDNA and cpDNA sequences (한국산 물통이속(Pilea) 식물의 nrDNA, cpDNA를 통한 계통분석)

  • A study of the genus Pilea in Korea including five taxa was carried out using molecular phylogenetic methods. The majority of members of the genus Pilea in Korea are annual herbs, and they live in moist habitats, flowering in summer and fruiting in autumn. The results of a phylogenetic analysis using nrDNA and cpDNA supported the recognition of P. japonica, P. peploides, and P. taquetii. Pilea taquetii from Mt. Sanbangsan in Jeju was nested within P. hamaoi and P. mongolica clade instead of the P. taquetii clade, with P. taquetii from Mt. Jirisan also separated from the P. taquetii clade. This indicates that the separation is not geographical isolation, but is instead related to taxonomic problems. Therefore, further study of the P. taquetii group is necessary.

Also, the dependence of k' on the liquid-liquid extration distribution ratio($D_c$) in methanol-water/n-alkane extration system was investigated. Both kinds of dependence are linear, which susggests that the retention of the electroneutral metal chelate is largely due to the solvophobic effect. Standard adsorption enthalpy changes (${\Delta}H^{\circ}$) and standard adsorption entropy changes (${\Delta}S^{\circ}$) of Pd(II) Isonitrosoethylacetoacetate imine chelates on Micropak MCH-5 column were calculated by measuring capacity factor with changing temperature of the column.

  • Epigallocatechin gallate (EGCG) was reported to exert weak cytotoxicity against normal healthy cells such as C3H10T1/2 cells, but profound inhibitory effects on the initiation or promotion stage of chemical carcinogenesis in mammary gland, blood and mouse skin. This study was carried out to develop antitumor agents with weak side effects and strong antitumor activity. Human skin melanoma cells (HBT 69) and human oral epitheloid carcinoma cells (OCL 17) were cultured in RPMI-1640 media containing 10% fetal bovine serum, antibiotic, and fungizone. After incubation for 24 hrs, the cells were treated with various amounts of (EGCG) for 48 hrs. The growth inhibitory effects of EGCG in human oral epitheloid carcinoma cells were evaluated by the 3- (4,5-djmethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT), neutral red (NR), and sulforhodamine B protein (SRB) assays of colorimetric methods. The light microscopic study was also carried out to observe morphological changes of the treated cells. These results obtained were as follows; 1. Significantly inhibitory effects of EGCG against cultured human oral epithelioid carcinoma cells. 2. Significantly inhibitory effects against cultured human skin melanoma cells treated with 50 $\mu$M EGCG, but decreased inhibitory effects in 100 $\mu$M EGCG. 3. Degenerative changes against cultured human oral epitheloid carcinoma cells. 4. Degenerative changes against human skin melanoma cells treated with 50 UM EGCG, but recovered degenerative changes in 100 $\mu$M EGCG.

Sulfur Deficiency Effects on Nitrate Uptake and Assimilatory Enzyme Activities in Rape Plants (유채에서 황 결핍이 질산염의 흡수 및 동화관련 효소활력에 미치는 영향)

  • Rape plants, especially forage type, are known as one of high S-demanding plants. Their productivity and quality have often limited under S-deficient condition. To investigate the effect of S-deficiency on N uptake and its assimilation, $NO_3^-$ absorption, nitrate reducatse (NR) and glutamine synthetase (GS) activity in leaf and root tissues as affected by different S-supplied level was determined. $NO_3^-$ uptake was not significant between control and S-deficient treated plants, while significantly depressed in S-deprived plants for the early 8 h. NR activity decreased as S-availability decreased, especially in young and middle leaves, representing more than 35% of decrease in S-deficient and 70% in S-deprived plants when compared with control. In roots, a significant decrease (-29%) in NR was observed only in S-deprived plants. Relatively higher GS activity was found in young leaves for three all treatments. As a whole leaf tissue, S-limited conditions resulted in a reduction of GS activity. In root which showed the lowest activity, a significant decrease (-30%) was observed only in S-deprived plants.

Cytotoxic Effects of Extracts from Hairy Roots of Rheum undulatum L. (대황의 모상근 배양조직 추출물의 세포독성)

  • The purpose of this research was to investigate the effects of extracts from cultured hairy roots of R. undulatum on human kidney epithelial cells. The cytotoxicity was measured by colorimetric assay using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT), neutral red (NR) and sulforhodamine protein B (SRB) with human kidney epithelial cell lines A498. MTT, NR and SRB quantities decreased propotionally in cultured A498 cells treated with the water or chloroform extracts of cultured hairy roots at increasing concentrations. These results suggest that extracts of cultured hairy roots are cytotoxic on human epithelial cells. The cytotoxicity of chloroform fraction was stronger than that of water fraction. The values of $MTT_{50},\;NR_{50}\;SRB_{50}$ of the extracts of chloroform fraction and those of water fraction were measured to be $289.3{\mu}g/ml,\;302.7{\mu}g/ml,\;433.8{\mu}g/ml\;and\;475.8{\mu}g/ml,\;428.3{\mu}g/ml,\;549.5{\mu}g/ml$, in A498 cell line.

Effects of Repeated Nicotine Treatment on the Changes in Glutamate Receptor Subunits Levels in Mesocorticolimbic Dopamine Areas

  • Recent studies suggest that alterations in glutamate receptor subunit levels in mesocorticolimbic dopamine areas could account for neural adaptations in response to psychostimulant drugs. Although many drugs of abuse induce changes in ionotropic glutamate receptor subunits in mesocorticolimbic dopamine areas, the changes of ionotropic glutamate receptor subunits by repeated nicotine treatment in these areas are not known. To answer this question, we injected male Sprague-Dawley rats twice daily with nicotine (0.4 mg/kg) or saline (1 ml/kg) for 10 days. The immunoreactivity of NR1, GluR1, and GluR2 glutamate receptor subunits was examined $16{\sim}18 h$ after the last injection of saline or nicotine. Repeated nicotine treatment significantly increased NR1 levels in the ventral tegmental area (VTA). In addition, repeated nicotine treatment showed a tendency towards an increase in GluR1 levels in the VTA as well as in striatum. However, there was no significant change in glutamate receptor subunits in other areas including nucleus accumbens (NAc). These results demonstrate that repeated nicotine treatment increases NR1 levels in VTA similarly to other drugs of abuse, suggesting that elevated glutamate receptor subunits in the VTA, but not NAc may be involved in the excitation of mesocorticolimbic dopamine neurons by nicotine.

Selective Reduction of Carbonyl Compounds with Dilsobutyldialkylaminoalanes

  • Details are described of reaction of carbonyl compounds with diisobutyldialkylaminoalane (DIBAL-NR2; R=Et, i-Bu, Ph) in order to establish their reduction characteristics. The reagents were extremely mild and reduced only aldehydes and ketones effectively in ethyl ether at 25 ℃. The stereoselectivity in the reduction of representative cyclic ketones appeared not so high but quite different from that obtained by DIBAL-H itself. The reagents reduced α,β-unsaturated aldehydes and ketones to the corresponding allylic alcohols without any detectable 1,4-reduction products. DIBAL-NR2 also achieved the selective reduction of aldehydes or ketones in the presence of keto or other readily reducible functional group, however the chemoselectivity was less satisfactory than that achieved by diisobutylethoxyalane (DI-BAL-OEt).
