Anti-Inflammatory Effect of Erigeron annuus L. Flower Extract through Heme Oxygenase-1 Induction in RAW264.7 Macrophages (RAW264.7 대식세포에서 Heme Oxygenase-1의 유도에 의한 개망초 (Erigeron annuus L.) 꽃 Methanol 추출물의 항염증 효과)

  • This study investigated the anti-inflammatory effect of Erigeron annuus L. flower (EAF) methanol extract. We examined the involvement of heme oxygenase-1 (HO-1) in the inhibitory activities of EAF methanol extract on nitric oxide (NO) production in lipopolysaccharide (LPS)-stimulated RAW264.7 macrophages. Cell viability and NO assays were performed. In addition, inducible nitric oxide synthase (iNOS) and HO-1 expressions were detected by Western blotting and blocking HO-1 activity on NO production. EAF methanol extract (25, 50, 100, 200 ${\mu}g$/mL) significantly inhibited LPS-stimulated NO production (p<0.05; 12.82, 9.61, 6.83, 2.52 ${\mu}m$) in a concentration-dependent manner. EAF methanol extract also reduced the expression of iNOS protein. The EAF methanol extract induced the expression of HO-1 in a dose-dependent manner. Blockage of HO-1 activity by zinc protoporphyrin suppressed EAF methanol extract-induced reductions in the production of NO. The present results suggest that EAF methanol extract has a potent anti-inflammatory effect in RAW264.7 macrophages through the induction of HO-1.

Antioxidant and Anti-inflammatory Effects of Extracts from the Flowers of Weigela subsessilis on RAW 264.7 Macrophages (RAW 264.7 대식세포에 미치는 병꽃나무 꽃 추출물의 항산화 및 항염증 효과)

  • This study investigated the antioxidant and anti-inflammatory activity of ethanol extract from the flowers of Weigela subsessilis (WS-E) in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages. The total polyphenol and flavonoid content was 719.19±0.04 μg tannic acid equivalents/ml and 644.87±0.02 μg quercetin equivalents/ml, respectively. The antioxidant activities of WS-E were measured by 1,1-diphenyl-2-picrylhydrazyl (DPPH) and superoxide anion radical scavenging activity. The antioxidant activities of WS-E increased markedly, in a dose-dependent manner. To screen for anti-inflammatory agents, the inhibitory effects of WS-E on the production of proinflammatory cytokines in the LPS-stimulated RAW 264.7 macrophages was examined. WS-E had no effect on cell viability at a concentration of 100 μg/ml. Nitric oxide (NO) and interleukin (IL)-6 production were inhibited in a dose-dependent manner (p<0.05). WS-E had no effect on the production of tumor necrosis factor (TNF)-α at a concentration of 0.16–20 μg/ml but induced TNF-α at a concentration of 100 μg/ml. Inducible nitric oxide synthase (iNOS) expression was also inhibited at lower concentrations (p<0.05). In addition, WS-E reduced the activation of nuclear factor (NF)-κB by inhibition of inhibitoy (I) κB phosphorylation in RAW 264.7 macrophages upon stimulation with LPS (100 ng/ml) for 24 h but not that of mitogen-activated protein kinase (MAPK). These results suggest that WS-E may be a useful antioxidant and anti-inflammatory agent in functional cosmetics.

Inflammatory mediator regulation of the Zizyphus jujube leaf fractions in the LPS-stimulated Raw264.7 mouse machrophage (LPS로 염증이 유도된 Raw 264.7 대식세포에서 대추(Zizyphus jujube) 잎 분획물의 염증매개물질 억제)

  • Zizyphus jujube leaf fractions (ZLFs) showed no cytotoxic effects of up to $100{\mu}g/mL$, while the anti-inflammatory effects of ZLFs were analyzed by checking the productions of nitric oxide (NO), prostaglandin $E_2$ ($PGE_2$), cyclooxygenase-2 (COX-2), and inflammatory cytokines TNF-${\alpha}$, IL-$1{\beta}$, and IL-6 in the lipopolysaccharide (LPS)-stimulated Raw264.7 macrophage up to the concentration of $100{\mu}g/mL$. ZLFs ($100{\mu}g/mL$) demonstrated a strong anti-inflammatory activity that reduced 61~85% of NO and 71~100% of $PGE_2$ production in the LPS-stimulated Raw264.7 macrophage. Even the low ZLFs concentration of $1{\mu}g/mL$ have reduced NO and $PGE_2$ production by 34~64%. Expressions of COX-2 protein were also effectively inhibited by the ZLFs. Furthermore, the TNF-${\alpha}$, IL-$1{\beta}$, and IL-6 production were significantly suppressed through the treatment of ZLFs at concentrations of 1, 10, and $100{\mu}g/mL$. In the order of the Zizyphus jujube leaf water fraction (ZLWF) < buthanol fraction (ZLBF) < ethyl acetate fraction (ZLEF) showed anti-inflammatory activity. In particular, the ethyl acetate fraction ZLEF at $100{\mu}g/mL$ showed an excellent anti-inflammatory activity by reducing the production of NO, $PGE_2$, COX-2, and inflammatory cytokines (TNF-${\alpha}$, IL-$1{\beta}$, IL-6) in the level of Raw264.7 macrophage without LPS-stimulation or even better. The results of our study suggest the potential of ZLEF for use as an excellent ant-inflammatory inhibiting mediator and may be used as a therapeutic approach to various inflammatory diseases.

  • 환경오염물질로부터 수생태계 보호를 위한 표준적인 평가 및 관리 수단인 수질환경기준은 오염물질의 독성작용이 일어나는 표적기관에서의 오염물질의 농도에 대한 대체측정치로서 환경 내 오염물질의 농도를 이용해 왔다. 이러한 '외부환경농도에 기반한 접근방법'은 표적기관에서의 독성물질의 농도가 생물체내 농도에 비례하고, 결국 외부환경농도에도 비례할 것이라고 가정한다. 따라서 환경오염물질의 생물이용도나 생물축적 양상의 차이 때문에 고유 독성치를 비교 평가하는데 한계가 있다. 이와 달리 '생물체내 농도에 기반한 접근방법(이하 생체잔류량 접근법)'은 환경오염물질의 생물이용도나 종 특이적 생물축적 양상과 관련된 불확실성을 제거하고, 환경오염물질 고유의 독성을 비교 평가할 수 있게 해준다. 특히 생체잔류량 접근법을 독성동태학 및 독성역학 모델과 함께 사용하는 경우는 실제 현장에서 일어나는 복잡한 노출조건에서의 독성영향을 예측하는데 활용할 수 있다. '생체잔류량 접근법'은 독성기작별 임계잔류량(Critical Body Residue)을 결정함으로써 생물모니터링의 결과를 해석하는데 적용되고 있다. 또한 생태위해성평가를 위해서 필요한 '무영향예측농도(Predicted No-effect Concentration, PNEC)를 예측하기 위한 방법으로 생체 내 잔류량에 기반해서 농도-시간-반응관계를 기술하고, 예측할 수 있는 새로운 유형의 독성역학 및 독성동태학 모델을 제시하고, 생체내 '무영향농도(No Effect Concentration, NEC)'를 추정하게 해 준다. 특히 생체내 NEC는 '무영향관찰농도(No Observed Effect Concentration, NOEC)'와 '영향농도(Effect Concentration, EC)'처럼 분산분석이나 회귀분석모델과 같은 통계적 모델에 기반해서, 농도-반응관계만을 기술할 뿐인 기존 독성모델을 대체할 대안으로 최근에 OECD와 ISO에 의해서 추천되었다.분석을 시행한 결과 인지기능 장애정도 및 MMSEK 점수 증가에 따른 사망위험도는 어느 모형에서도 인지기능 장애정도가 사망에 미치는 위험도는 통계적으로 유의하지 않았다(표 6, 표 7). 이상 본 연구는 농촌지역 노인들에서 인지기능 장애정도가 사망에 미치는 영향을 알아보고자 하였지만, 인지기능 장애정도가 사망에 미치는 영향을 통계적으로 유의하게 고찰하지 못하였다.의한 차이를 보였다. Effects of solvents on the anti-aging activity of Salvia miltiorrhiza extract (추출용매에 따른 단삼 추출물의 항노화 활성)

  • This study was conducted to determine the cell cytotoxicity, astringency, nitrite oxide scavenging, iNOS protein expression level, pro-inflammatory cytokine, elastase inhibition, and type I pro-collagen synthesis as a functional cosmetics material of Salvia miltiorrhiza root. We prepared the 80% ethanol(SE) and hot-distilled water(SW), respectively. Both SE and SW showed no toxicity from 0.05 to 0.5 mg/mL concentration as a result of MTT assay in NHDF or RAW264.7 cells. In the measurement of astringent effect, SE reveled 74.6% of astringent activity in 10 mg/mL. SE showed that LPS-induced nitric oxide production, iNOS protein expression, and cytokines were inhibited in a dose-dependent manner. Furthermore, two extracts significantly inhibited elastase activity and increased the type I pro-collagen production. Therefore, it is expected that Salvia miltiorrhiza extract is used as a natural material for functional cosmetics that can effectively prevent skin-related inflammation and wrinkles, and aging.

Antioxidant, Antibacterial and Anti-inflammatory Effects of Stachys sieboldii Extract (초석잠 추출물의 항산화, 항균 및 항염 활성)

  • This study was conducted to evaluate antioxidant, antibacterial, anti-inflammatory, and digestive enzyme activity in water extract (SAW) and 60% ethanol extract (SAE) from Stachys sieboldii. As the treatment concentration of each extract S. sieboldii extract increased, antibacterial and antioxidant activity increased. The total polyphenol and total flavonoid contents of SAW were 106.25 ± 0.94 mgGAE/g, 24.4 ± 0.24 mgQE/g and SAE were 124.61 ± 1.11 mgGAE/g, 45.2 ± 3.52 mgQE/g, respectively. The 400 ㎍/mL of SAW and SAE performed more than 53% protective effects against oxidative stress in HepG2 cell lines. All extracts were not showed cytotoxicity to RAW 264.7 cell line at 100 ㎍/mL. NO production was reduced to 44.3 ± 1.4% for SAW and 45.1 ± 1.0% for SAE at a concentration of 100 ㎍/mL. The production of inflammatory cytokines each TNF-α, IL-1β, and IL-6 was inhibited in a concentration-dependent. S. sieboldii extract did not showed Caco-2 cells cytotoxicity and inhibited NO production in concentration-dependent. As the concentration of the S. sieboldii extract increased of α-amylase and protease enzymes activity, which are digestive enzyme. As a result of the experiment, it is judged that it can be used as basic data for the development of health food using S. sieboldii.

Immunological Activity of Bovine Colostral Whey Protein Containing TGF-β from Imsil Province (임실지역 젖소 초유로부터 분리한 TGF-β 함유 유청 단백질의 면역활성)

  • This experiment was carried out in order to separate bovine colostral whey protein from Imsil province and to test the effect of immunological activity on RAW 264.7 cells. The colostral whey protein contained TGF-${\beta}$ 7, 475 pg/g in total. We first tested the effect of the colostral whey protein on the proliferation of RAW 264.7 cells and it demonstrated cytotoxicity at concentrations greater than 20 mg/mL. Therefore, the immunological activities of colostral whey protein were investigated in maximum concentration of 10 mg/mL on LPS-induced RAW 264.7 cells. Results indicated that colostral whey protein inhibited the LPS-induced nitric oxide (NO) production in a dose-dependent manner. The colostral whey protein also suppressed the productions of proinflammatory cytokines (TNF-${\alpha}$, IL-$1{\beta}$, IL-6) in a dose-dependent manner. In addition to the immunological activity, colostral whey protein led to the expression of heme oxygenase-1 (HO-1) in RAW 264.7 cells. In conclusion, colostral whey protein containing TGF-${\beta}$ inhibited the production of NO, TNF-${\alpha}$, IL-$1{\beta}$, and IL-6 via expression of HO-1.

Anti-Inflammatory Effects of Extracts from Ligustrum ovalifolium H. Leaves on RAW264.7 Macrophages (RAW264.7 대식세포에서 왕쥐똥나무잎 추출물의 항염증 효과)

  • This study investigated the anti-inflammatory effects of Ligustrum ovalifolium H. (LOH) leaf extracts on RAW264.7 macrophages. Cell toxicity was determined by MTT assay. We evaluated the anti-inflammatory effects of LOH extracts by measuring nitric oxide (NO), reactive oxygen species (ROS), inducible NOS (iNOS) production, and cyclooxygenase-2 (COX-2) expression by Western blotting. LOH ethanolic extracts (0.05, 0.1, and 0.2 mg/mL) significantly suppressed LPS-stimulated production of NO. The intracellular ROS level also significantly decreased. LOH ethanolic extracts reduced the expression of iNOS and COX-2 proteins. The present results show that LOH ethanol extract has potent anti-inflammatory effects on RAW264.7 macrophages. These results also suggest that the anti-inflammatory effects of LOH extracts may be related to the inhibition of LPS-stimulated ROS and NO production. Therefore, ethanolic extracts of LOH leaves may be utilized as a good source of functional foods for protection against inflammatory diseases.

Study on Applicability of Pinus koraiensis Siebold et Zucc Leaf Extract as a Cosmetic Ingredient (잣나무(Pinus koraiensis Siebold et Zucc)잎 추출물의 화장품 소재로서의 활용 가능성에 관한 연구)

  • This study was performed to verify various effects of 70% ethanol extract and distilled water extract of pine leaf such as antioxidant, anti-inflammatory, skin-lightening effects analysis in cells. The result of antioxidant experiment showed that polyphenol content increased concentration dependently in polyphenol and flavonoid effects. The total content of polyphenols and flavonoids was much higher in ethanol extract than water extract. In all the cells of B16F10, RAW264.7 cytotoxicity at each concentration level didn't appear. The result of measuring NO production inhibition showed that it inhibited NO production led to LPS effectively, so anti-inflammatory activity was confirmed. The result of measuring melanin biosynthesis inhibition showed that there was considerably reduction effect, but it performed western blot, and as a result of MITF, Tyrosinase protein revelation, the inhibition of MITF, Tyrosinase revelation was confirmed concentration dependently, Therefore, in this study pine leaf extract was expected to be used as a cosmetic ingredient.
