Alterations in Cerebrovascular Reactivity by Trigeminovascular System Injury in Rats

  • Trigeminovascular system plays an important role for the cerebral memodynamics. The aim of this study was to investigate the alterations in cerebrovascular reactivity by trigeminovascular system injury in rats. Trigeminovascular system of male Sprague-Dawley rats was injured by either denervation of nasocilliary nerve or neonatal capsaicin treatment. Trigeminovascular system was stimulated by controlled hemorrhagic hypotension or somatosensory (whisker) stimulation. Changes in regional cerebral blood flow (rCBF) and pial arterial diameter were continuously measured by laser-Doppler flowmetry and videomicroscopy, respectively. Nitric oxide synthase (NOS) activity in cerebral cortex was determined by measuring the conversion of $L-^3H-arginine\;to\;L-^3H-citrulline$. Cyclic GMP levels in cerebral cortex and pial artery were determined using the cyclic GMP $^{125}I$ scintillation proximity assay system. rCBF autoregulation was impaired or almost abolished by trigeminovascular system injury. rCBF response to whisker stimulation was significantly attenuated by trigeminovascular system injury. NOS activity as well as cyclic GMP level in cerebral cortex and pial artery were significantly reduced in the group of trigeminovascular system injury. These results suggest that trigeminovascular system injury causes prominent alterations in cerebrovascular reactivity, and that NO, which is generated by neuronal NOS in the trigeminovascular system, is implicated in the regulation of rCBF.

신이화(辛夷花, Magnoliae Flos)추출물의 화장품약리활성 및 항염증효과에 관한 연구 (Study on Cosmeceutical Activities and Anti-inflammatory Activities of Magnolia biondii Extracts)

  • 본 연구에서는 신이화에 함유된 물질을 열수와 에탄올을 이용하여 추출한 후 화장품약리활성 및 항염증효과를 확인하고, 화장품 및 바이오산업에 적용하기 위한 천연소재로써의 이용 가능성을 살펴보았다. DPPH 라디칼 소거능 실험결과 농도의존적으로 소거능 효과를 보이며, 열수추출물의 경우 5,000 ug/ml 농도에서 대조군인 BHA보다 우수한 효능을 보였다. SOD 유사활성에서는 농도가 증가할수록 유사활성이 증가하는 것을 확인할 수 있었고, 5,000 ug/ml에서 열수추출물의 경우 85% 유사활성을 나타내었고, 에탄올 추출물의 경우 43%의 유사활성을 나타내었다. Xanthine oxidase 저해능 실험에서도 5,000 ug/ml 농도에서 대조군 BHA 보다 높은 저해능을 효여 우수한 항산화효과를 보였으며, 합성항산화제를 대체할 수 있는 천연항산화제로 사용이 가능할 것으로 사료된다. 항염증효과를 확인하기 위하여 NO 저해 활성 및 iNOS 발현량을 측정하였는데, 열수와 에탄올 추출물 모두 농도가 증가할 수로 NO 저해활성과 iNOS 발현량이 감소하여 우수한 항염증효과를 보였다. 이상의 결과에서 신이화 열수추출물과 에탄올추출물은 우수한 화장품약리활성 및 항염증효과가 있음을 확인할 수 있었고, 안티에이징 및 트러블 피부 개선 화장품 소재로서의 가능성을 확인할 수 있었다.

Anti-inflammatory Effects of Ethanolic Extracts from Codium fragile on LPS-Stimulated RAW 264.7 Macrophages via Nuclear Factor kappaB Inactivation

  • Bacterial lipopolysaccharide (LPS) induces expression of pro-inflammatory cytokines and enzymes producing nitric oxide (NO) and prostaglandins (PGs) in immune cells. This process is mediated by the activation of nuclear factor kappaB (NF-${\kappa}B$). In this study, we investigated the anti-inflammatory characteristics of Codium fragile ethanolic extract (CFE) mediated by the regulation of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) using LPS-stimulated murine macrophage RAW 264.7 cells. CFE significantly inhibited LPS-induced NO and $PGE_2$ production in a dose-dependent manner and suppressed the expression of iNOS and COX-2 proteins in LPS-stimulated RAW 264.7 cells with no cytotoxicity. Pro-inflammatory cytokines, such as interleukin (IL)-$1{\beta}$, IL-6, and tumor necrosis factor-${\alpha}$, were significantly reduced by treatment of CFE in LPS-stimulated RAW 264.7 cells. CFE inhibited the promoter activity of (NF)-${\kappa}B$ in LPS-stimulated macrophages. Treatment with CFE suppressed translocation of the NF-${\kappa}B$ p65 subunit by preventing proteolytic degradation of inhibitor of ${\kappa}B-{\alpha}$. These results indicate that the CFE-mediated inhibition of NO and $PGE_2$ production in LPS-stimulated RAW 264.7 cells is mediated through the NF-${\kappa}B$-dependent transcriptional downregulation of iNOS and COX-2, suggesting the potential of CFE as a nutraceutical with anti-inflammatory activity.

오크라(Abelmoschus esculentus) 추출물의 항염증 및 보습 효과 연구 (Study on Anti-Inflammatory and Moisturizing Effect of Extracts from Abelmoschus esculentus)

  • In this study, we studied the anti-inflammatory and physiological activities Abelmoschus esculentus extracts by hot water (AEW) and 70% ethanol (AEE) and confirm the possibility to use it as a natural ingredient. For this study measure the antioxidative activity, total polyphenol content was measured, and DPPH and ABTS scavenging activity assays were conducted. Total polyphenol content of AEW and AEE was measured, and the results showed that they were 126.76 mg TAE/100 g and 144.21 mg TAE/100 g, respectively. DPPH and ABTS radical scavenging activities were measured to determine the antioxidative activity, and the results indicated that DPPH and ABTS radical scavenging activities increased in both extracts concentration-dependently. The moisturizing effect was measured by measuring the amount of hyaluronic acid (HA) produced within HaCaT cell using the ELISA kit. AEW and AEE increased the amount of HA production in a concentration-dependent manner. In order to determine the anti-inflammatory activity of AEW and AEE, the NO assay was conducted, and the inhibitory effects were found to be 11.46% and 25.28%, respectively in 100 ㎍/ml. In order to measure the anti-inflammatory activity, nitric oxide (NO) inhibitory activity was measured, and the inhibition of expression of iNOS, COX-2 proteins was measured and shown. Furthermore, inhibition of expression was found in inflammatory inducing factors iNOS and COX-2 proteins showing concentration-dependent inhibition. This study found the excellent effects of antioxidative, moisturizing effect, anti-inflammatory activity in Abelmoschus esculentus extracts, which indicates that can be used as functional materials for aesthetics, food and functional cosmetics.

RAW 264.7 세포에서 분비나무(Abies nephrolepis MAX.) 추출물의 항염 효과에 대한 연구 (A Study of the Anti-inflammatory Effects of Abies nephrolepis MAX. Extract in RAW 264.7 Cells)

  • 본 연구에서는 분비나무(Abies nephrolepis MAX.)를 분비나무 줄기(Abies nephrolepis MAX. stem) 추출물과 분비나무 잎(Abies nephrolepis MAX. leaf) 추출물로 나누어 항염 관련 기능성을 연구하여 화장품 소재로서의 이용가능성을 알아보고자 하였다. MTT assay를 통해 세포 생존율을 확인한 결과, 대식세포(RAW 264.7)에서 분비나무 줄기와 잎 추출물의 500 ㎍/ml의 농도에서 각각 97.8%, 95.6%의 생존율을 보였다. 이에 따라 세포 관련 실험을 세포의 생존율이 100%에 가까운 25, 50, 100 ㎍/ml의 농도로 진행하였다. 염증을 유발하는 활성산소인 NO 생성에 대해 분비나무 추출물을 측정한 결과, 분비나무 추출물을 처리한 구에서 NO 발현이 억제되는 것을 확인할 수 있었으며, 분비나무 줄기와 잎은 100 ㎍/ml의 농도에서 71.1%, 42.9%의 저해율을 나타내었다. 분비나무 추출물을 RAW 264.7 세포에 처리하여 단백질 발현을 측정한 결과, COX-2, iNOS 및 pro-inflammatoty cytokine인 IL-1β, IL-6, TNF-α 인자에서 농도 의존적으로 단백질 발현이 억제되었으며, 분비나무 줄기의 iNOS와 분비나무 잎의 COX-2, iNOS, IL-6 및 TNF-α의 인자가 대조군인 Vit. C와 비교하였을 때 우수한 단백질 발현 억제를 보였다. 또한, RAW 264.7 세포에 분비나무 추출물을 처리하여 mRNA 발현을 측정한 결과, 분비나무 줄기의 iNOS, 분비나무 잎의 COX-2와 iNOS의 mRNA 발현양은 대조군인 Vit. C와 비교하였을 때 우수한 억제율을 확인할 수 있었으며, pro-inflammatoty cytokine인 IL-1β, IL-6 및 TNF-α인자에서는 분비나무 줄기 추출물의 IL-1β는 대조군과 유의한 결과를 나타냈지만, 나머지 인자에서는 대조군인 Vit. C와 비교하였을 때 mRNA 발현 억제율이 우수한 것을 확인할 수 있었다. 이에 따라 분비나무 추출물이 항염 관련 기능성 활성에 우수함을 나타내어 화장품 소재로써의 이용가능성을 확인할 수 있었다.

하고초물추출물이 LPS로 활성화된 Raw 264.7 cell에서의 염증매개물질 억제효과 (Inhibitory Effects of Water Extracy of Prunellae Spica on the Production of Pro-inflammatory Mediator in LPS-activated Raw 264.7 Cells)

  • Prunellae Spica is the spike or whole plant of Prunella vulgaris Linne, which has been used for clearing heat from the liver, brightening the eyes and treating headache in traditional oriental medicines. This study was conducted to evaluate the inhibitory effects of the aqueous extract of Prunellae Spica (PSE; PS extract) on the production of NO and PGE2 in LPS-activated Raw 264.7 cells. Cell viability was determined by MTT assay, and all three doses of PS extract (0.03, 0.1 and 0.3 mg/ml) had no significant cytotoxicity during the entire experimental period. The cells were treated with 1 ${\mu}g/ml$ of LPS 1 h before adding PS extract, and increased NO and PGE2 production were detected in LPS-activated cells compared to control. However, these increases were dose-dependently attenuated by treatment with PS extract. The inhibition of NO by PS extract was due to the suppression of iNOS expression via inhibition of $NF{\kappa}B$ nuclear translocation and proteolytic degradation of $I{\kappa}B{\alpha}$. The decreased level of PGE2 was derived from inhibition of COX-2 activity, but expression of COX-2 protein was not affected by PS extract. Moreover, PS extract reduced the elevated production of IL-${\beta}$ and IL-6 by LPS. These results demonstrate that PS extract has inhibitory effects on the production of NO and PGE2 as a consequence of the reduction of proinflammatory cytokines, especially IL-${\beta}$ and IL-6 in LPS-activated Raw 264.7 cells.

  • Objectives Trachelospermi caulis, known as Nak-Suk-Deung in Korea, is the dried leafy stem of Trachelospermum asiaticum var. intermedium Nakai, and climbing stems and branches of Trachelospermum sdisyivum var, intermedium nakai or Apocyanaceae. Trachelospermi caulis has antipyretic and analgesic activity. It has traditionally been used as a folk remedy in Korea for the treatment of various infla mMatory diseases, including rheumatoid arthritis. The purpose of this study was to evaluate the Effects of Trachelospermum caulis extract on SNP-induced infla mMatory responses in rabbit HIG-82 synovial membrane cells. Methods Anti-infla mMatory effects of the extract of Trachelospermum caulis were investigated using rabbit HIG-82 synovial membrane cells. For this study, 3-(4,5-dimethylthiazol- 2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay, western blot analysis, PGE2 i mMunoassay, and NO detection were conducted. Results The aqueous extract of Trachelospermum caulis exerted cytotoxicity and suppressed PGE2 synthesis and NO production in rabbit HIG-82 synovial membrane cells. The aqueous extract of Trachelospermum caulis also inhibited the SNP-induced expressions of COX-2, iNOS, and TNF-$\alpha$ in rabbit HIG-82 synovial membrane cells. Conclusions These results showed that the extract of Trachelospermum caulis exerts the anti-infla mMatory effect by suppressing COX-2, iNOS, and TNF-$\alpha$ expressions in the synovial membrane cells.

  • Objectives : Scrophulariae Radix (SRE) is commonly used in combination with other herbs as a nutrient and health strengthening agent, and to remove 'heat' and replenish vital essence. The water-based extract of this herb can lower blood pressure in both anesthetized and concious animals, and exhibits an anti-inflammatory activity. But, there is lack of studies regarding the effects of SRE on the immunological activities in molecular levels. The present study was conducted to evaluate the effect of SRE on the regulatory mechanism of cytokines and nitric oxide (NO) in Raw 264.7 cells. Method : After the treatment of Scrophulariae Radix methanol extract, cell viability was measured by MTT assay, NO production was monitored by measuring the nitrite content in culture medium. COX-2 and iNOS were determined by Immunoblot analysis, and levels of cytokine were analyzed by sandwich immunoassays. Results : Results provided evidence that SRE inhibited the production of nitrite and nitrate (NO), inducible nitric oxide synthase (iNOS), $interleukin-1{\beta}\;(IL-1{\beta})$ and interleukin-6 (IL-6), and the activation of phospholylation of inhibitor ${\kappa}B{\alpha}\;(p-I{\kappa}B{\alpha})$ in Raw 264.7 cells activated with lipopolysaccharide (LPS). Conclusion : These findings suggest that Scrophulariae Radix can produce anti-inflammatory effect, which may playa role in adjunctive therapy in Gram-negative bacterial infections.

마우스대식세포주인 RAW 264.7에서 금령자산(金鈴子散)(金鈴子散)의 항염증 활성 연구 (Anti-inflammatory effect of methanol extract of Keum-Ryung-Ja-San in mouse macrophages)

  • Objective : The aim of this study was to determine whether methanol extract of Keum-Ryung-Ja-San (KRJS) inhibit production of NO, $PGE_2$, iNOS, COX-2 and pro-inflammatory cytokines in lipopolysaccharide (LPS)-treated RAW 264.7 macrophages. Methods : Cytotoxic activity of extracts on RAW 264.7 cells was measured using 5-(3-caroboxymeth-oxyphenyl)-2H-tetra-zolium inner salt (MTS) assay. The nitric oxide (NO) production was measured by Griess reagent system. And proinflammatory cytokines and PGE2 were measured by ELISA kit. The levels of inducible nitric oxide synthase (iNOS), cyclooxygenase-2(COX-2), $I{\kappa}-B-\alpha$ and nuclear NF-${\kappa}B$ p65 expression were detected by western blot. Results : Our results indicated that methanol extract of KRJS significantly inhibited the LPS-induced NO, $PGE_2$ production and iNOS, COX-2 expression accompanied by an attenuation of TNF-$\alpha$, IL-$1{\beta}$ and IL-6 production in RAW 264.7 cells. Moreover, methanol extract of KRJS treatment also blocked LPS-induced NF-${\kappa}B$ activation. Conclusion : These findings indicate that methanol extract of KRJS inhibits the production of pro-inflammatory mediators and cytokines via suppression of NF-${\kappa}B$ activation. Take together, these results indicate that methanol extract of KRJS has the potential for use as an agent of anti-chronic inflammatory diseases.

곰취(Ligularia fischeri), 미역취(Solidago virga-aurea), 삼나물(Aruncus dioicus) 복합 추출물의 항염증 효과 (Anti-Inflammatory Effect of Ligularia fischeri, Solidago virga-aurea and Aruncus dioicus Complex Extracts in Raw 264.7 Cells)

  • 본 연구에서는 울릉도 특산 약용 작물 추출물이 항염증에 대한 실험연구가 이루어져 있지 않은 것에 착안하여 LPS에 의해 활성화된 대식세포로부터 유도되는 염증반응에 대한 억제효과를 조사하였다. 울릉도 자생 식물인 곰취, 미역취, 삼나물의 세가지 식물 추출물을 이용하여 피부 염증에 대하여 연구를 하였다. 산화질소와 cytokine의 생산은 면역세포의 대표적인 염증인자이다. 세포는 LPS 처리 후 한 시간 뒤에 곰취, 미역취, 삼나물 70% 아세톤 추출물을 처리를 하였다. 세포 독성이 나타나지 않는 농도인 1, 10, 100 ug/ml를 사용하였다. 곰취, 미역취, 삼나물 70% 아세톤 추출물은 NO, $PGE_2$, TNF-${\alpha}$, IL-1${\beta}$, IL-6, iNOS, COX-2의 생성을 저해 시켰다. $PGE_2$는 100 ug/ml의 농도에서 60%에 가까운 저해율을 나타내었다. iNOS와 COX-2 역시 100 ug/ml의 농도에서 각각 54%, 65%가 저해가 되었다. 게다가 곰취, 미역취, 삼나물 70% 아세톤 추출물은 염증성 사이토 카인인 TNF-${\alpha}$, IL-1${\beta}$, IL-6의 생성을 감소 시켰다. 이러한 결과로 곰취, 미역취, 삼나물 추출물은 염증 예방과 치료에 효과적임을 확인할 수 있었다.
