Prototypes of Panaxadiol and Panaxatriol Saponins Suppress LPS-mediated iNOS/NO Production in RAW264.7 Murine Macrophage Cells (RAW264.7 대식세포에서 LPS 매개 iNOS/NO 생성에 대한 protopanaxadiol saponin 및 protopanaxatriol saponin의 억제효과)

  • This study was performed to investigate the modulatory effects of two prototypes of Panax ginseng saponin fractions, 20(S)-protopanaxadiol saponins (PDS) and 20(S)-protopanaxatriol saponins (PTS), on the induction of inflammatory mediators in lipopolysaccharide (LPS)-treated RAW264.7 murine macrophage cells. For this purpose, RAW264.7 cells were treated with LPS ($10{\mu}g/ml$) before, after, or simultaneously with PDS or PTS ($150{\mu}g/ml$), and the released level of nitric oxide (NO) and expression levels of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) were evaluated. When RAW264.7 cells were treated with LPS and ginseng saponin fractions simultaneously for 24 hr, PTS, compared to PDS, more strongly attenuated the NO production induced by LPS treatment. When the cells were pretreated with LPS for 2 hr followed by PDS or PTS treatment for 24 hr, both ginseng saponins strongly reduced NO release. The pretreatment of RAW264.7 cells with PDS or PTS for 2 hr followed by LPS treatment for 24 hr significantly attenuated the LPS-induced production of NO. PTS showed stronger inhibitory potency to NO generation than PDS. Our western blot experiment showed that both PDS and PTS ($150{\mu}g/ml$) also significantly down-regulated the expressions of iNOS and COX-2 induced by LPS treatment. Our results suggest that both PDS and PTS possess strong protective effects against LPS-stimulated inflammation and that their protective effects are mediated by the suppression of NO synthesis via down-regulation of pro-inflammatory enzymes, iNOS, and COX-2 in the RAW264.7 cells.

Adherence Rate of Listeria monocytogenes Contaminated from Pork Meat During Pork Meat Processing (포장돈육 가공공정 중 Listeria monocytogenes에 오염된 돈육으로부터 오염 부착율)

  • This study was done to determine the adherence rate of Listeria monocytogenes(LM) from pork meat to pork meat, conveyer belt and stainless steel, and cutting board at $10^{\circ}C$ for given times (2, 4, 24 hours). The adherence rates of ATCC and wild type LM from pork meat to red meat or fat meat after 2 hour contact time were approximately 65-79%, but 100% adherence rates of both LM after 6 hours were observed. The adherence rate of ATCC LM from pork meat to conveyer belt and stainless steel after 24 hours was 0.15%, while the adherence rate of wild type LM was rapidly increased from 4.34 after 2 hours to 28.6% after 24 hours. Also, The adherence rate of ATCC LM from pork meat to stainless steel after 24 hours ranged from 2.63% after 2 hours to 0.39% after 24 hours, while the adherence rate of wild type LM ranged from 0% after 2 hours to 24.4% after 24 hours. In the meantime, no adherence rate of both LM from pork meat to cutting board was observed. This results suggest that the adherence rate of LM from pork meat to pork meat or food contact surfaces was significantly affected by contact time and wild type LM showed much higher adherence rate to pork meat or food contact surfaces than ATCC LM.

Lack of any Prognostic Value of Body Mass Index for Patients Undergoing Chemoradiotherapy for Esophageal Squamous Cell Carcinoma

  • Background: The relationship between body mass index(BMI) and outcomes after chemoradiotherapy(CRT) has not been systematically addressed. The purpose of this study was to evaluate the effect of BMI on survival in patients with esophageal squamous cell carcinoma (ESCC). Materials and Methods: Sixty ESCC cases were retrospectively reviewed in this study. Patient overall survival(OS) and disease-free survival (DFS) were compared between two groups (BMI< $24.00kg/m^2$ and $BMI{\geq}24.00kg/m^2$). Results: There were 41 patients in the low/normal BMI group (BMI< $24.00kg/m^2$) and 19 in the high BMI group ($BMI{\geq}24.00kg/m^2$). No significant differences were observed in patient characteristics between these. We found no difference in 2-year OS and DFS associated with BMI (p=0.763 for OS; p=0.818 for DFS) using the Kaplan-Meier method. Univariate analysis revealed that higher clinical stage was prognostic for worse 2-year OS and DFS, metastasis for 2-year OS, lymph node status for 2-year DFS, while age, gender, smoking, drinking, tumor location and BMI were not prognostic. There were no differences in the 2-year OS (hazard ratio=1.117; p=0.789) and DFS(hazard ratio=1.161; p=0.708) between BMI groups in multivariate analysis, whereas we found statistical differences in the 2-year OS and DFS associated with clinical stage, gender and tumor infiltration (p<0.04), independent of age, smoking, drinking, tumor location, the status of lymph node metastases and BMI. Conclusions: BMI was not associated with survival in patients with ESCC treated with CRT as primary therapy. BMI should not be considered a prognostic factor for patients undergoing CRT for ESCC.

Anti-inflammatory Activity of Wild Yeasts, Meyerozyma guilliermondii YJ34-2 and Rhodotorula graminis YJ36-1, on Lipopolysaccharide-induced Nitric Oxide in RAW 264.7 Cells Through the Inhibition of Nitric Oxide and Cytotoxic Effects (Lipopolysaccharide로 유도한 RAW 264.7 세포에 대한 Meyerozyma guilliermondii YJ34-2와 Rhodotorula graminis YJ36-1의 항염활성과 Nitric Oxide 생성 저해물질의 생산)

  • The anti-inflammatory effects of cell-free extracts from wild yeasts, Meyerozyma guilliermondii YJ34-2 and Rhodotorula graminis YJ36-1, caused by the inhibition of nitric oxide (NO) activity and cytotoxic effects were determined. Cell-free extracts from these two yeast strains had dose-dependent inhibitory effects on the production of lipopolysaccharide-induced NO and there were no cytotoxic effects on the treated cells or negative effects on their proliferation. Their cell-free extracts were also shown to have inhibitory effects on pro-inflammatory cytokines, such as tumor necrosis factor $(TNF)-{\alpha}$ and $prostaglandin-E_2$, in a dose-dependent manner. Maximal inhibitory activity on NO production occurred in cell-free extracts of Meyerozyma guilliermondii YJ34-2 cultivated at $30^{\circ}C$ for 24 hr and Rhodotorula graminis YJ36-1 cultivated at $25^{\circ}C$ for 24 hr in the yeast extract-peptone-dextrose (YPD) media.

DENTINAL MICROLEAKAGE STUDY ON THE LIGHT CURABLE RESTORATIVE GLASS IONOMER CEMENT (광중합형 충전용 글라스 아이오노머 시멘트의 상아질 변연누출에 관한 정량분석)

  • The purpose of this study was to evaluate the amount of marginal microleakage of 2 light curable GI cements(Fuji II LC & VariGlass), which contain some resin components. 4 volunteers kept on acrylic resin plates, which contained dentin disks with cavities filled with test materials for 2 weeks. The time when polishing was done(5 minutes and 24 hours after filling) and the use of protective agents were varied, so 8 groups with each 6 specimens were tested. After having specimens(disks with cavities filled with materials) penetrated with 1% Methylene Blue solution, specimens were stored in 40% nitric acid solution for 4 days to extract adsorbed dye material. Supernatants of centrifuged samples were diluted 5 times and Spectrophotometer was used to determine the degree of absorption. Dye concentration was calculated through the pre-obtained Linear Regression Curve. The results were as follows. 1. The best result was seen in groups (PF24, PV24) which were protected and polished 24 hours later and the opposite phenomenon was seen in groups(NF24, NV24) which were held without protection and polished 24 hours later. Groups polished S minutes later showed moderate leakage pattern. 2. Groups polished 5 minutes later showed similar leakage amount irrespective of using of protective agent. But statistically insignificant lower values were seen in VariGlass than in Fuji II LC groups, So It was considered that VariGlass may be more resistant to early moisture attack than Fuji II LC. 3. In groups polished 24 hours later, there was no significant difference between materials but was definitely significant difference according to the use of protective agent. If the cement in which polishing will be done 24 hours later, Protective agent should be used to cover the surface.
