New Records of Benthic Dinoflagellates of Four Genera (Bispidodinium, Cabra, Prorocentrum, Sinophysis) from the Coastal Beach of Korea

  • A research probing for the unrecorded and taxonomically undescribed indigenous species has been initiated since 2006. Samples were collected from an intertidal zone along the coasts of Korea as well as around the Jeju Island. We have found five unrecorded species of four genera belonging to the order Dinophysiales, Gymnodiniales, Peridiniales, and Prorocentrales. The species are as follows, Sinophysis canaliculata (2017) Bispidodinium angelaceum (2015), Cabra armorica (2016), Prorocentrum bimaculatum (2017), and P. tsawwassenense (2017) (note; The numbers in parenthesis refer to the year in which the species was reported as unrecorded indigenous species by National Institute of Biological Resources, NIBR hereafter).

A New Record of Parasquillid Species, Faughnia haani (Crustacea: Stomatopoda: Parasquillidae), from Korea

  • A parasquillid stomatopod, Faughnia haani(Holthuis, 1959), collected from Jeju Island, is reported for the first time in Korea. This species is characterized by the presence of obtuse anterolateral angles of carapace, median and three pairs of longitudinal carinae on telson, and crenulate inner margin of uropodal protopod. In this paper, a detailed description and illustrations are provided. As a result of this study, the geographical distribution of the present species is extended and 15 species of stomatopods including this species are now recorded in Korean fauna. The specimen examined herein was deposited in the National Institute of Biological Resources(NIBR).

Unrecorded species of Korean protozoans discovered through the project of 'Discovery of Korean Indigenous Species' III

  • In this paper, we present unrecorded protozoans of Korea that were discovered, through the 'Discovery of Korean Indigenous Species' project hosted by the 'National Institute of Biological Resources (NIBR)'. A taxonomic account is provided for each identified species, offering comprehensive information such as species name, Korean name, collection site, synonyms, specimen vouchers, diagnoses, and figures. This study introduces 13 previously unrecorded Korean protozoan species that are classified into three phyla: Amoebozoa, Cercozoa, and Ciliophora. Notably, the cercozoan family Chlamydophryidae was recorded in Korea for the first time, together with the discovery of three previously unreported genera: Diaphoropodon within Cercozoa, and Metauroleptus and Hemicycliostyla within Ciliophora.

홍해삼 유전체 분석에 의한 microsatellite의 분포도 연구 (Analysis of Microsatellite Patterns in the Genome of Red Sea Cucumber)

  • 본 연구는 홍해삼의 유전체를 분석하여 홍해삼의 유전자 마커 개발을 위한 기초 자료로 활용하기 위해 수행되었다. 울릉도_일반과 울릉도_토착으로 microsatellite marker 분석을 실시하였다. 그 결과 dinucleotide repeat 서열이 81.3~81.4%로 가장 많이 차지 되었으며, 반복서열 개수가 증가될수록 감소되는 경향을 보였다. 일반적으로 microsatellite는 5~10 반복수 사이에 집중적으로 존재하였으며, 반복 서열의 크기가 클수록 반복수가 적어지는 양상을 보였다. Di, tri, tetra-nucleotides 반복에서 각각 (AT)5, (AAT)5, (AAAT)5 등이 가장 높은 것들로 나타났다. (CG), (CCG) 등은 동일 반복 단위의 다른 반복 단위에 비교하여 매우 낮게 관찰되었다. Di-와 tri-nucleotide는 반복수가 각각 35와 32까지 지속적으로 나타난 다음에 비연속적으로 44와 43 반복까지 계수 되었다. Tetra-, penta- 및 hexa-nucleotide는 각각 25, 21 및 14까지 연속적으로 나타났다. 본 분석결과에 따르면 microsatellite는 특이서열반복에 대해 편중되는 경향성을 보이는 것으로 나타났다. 따라서 홍해삼의 microsatellite 분석에서 고유의 반복 서열과 반복수를 유지하는 것으로 추정되므로 향후 연구를 위한 기초 자료로 활용하는 것이 가능할 것으로 판단된다.

갈색거저리 유충에서 추출한 펩타이드의 항산화 특성 (Antioxidant Properties of Peptides Extracted from Tenebrio molitor Larvae)

  • 본 연구는 기능성 식품의 개발을 위해 갈색거저리 유충 유래 추출물에서 새로운 생리활성 펩타이드를 확인하는 것이다. 동결건조된 T. molitor 유충에서 다양한 용매로 시간에 따라 추출하였다. T. molitor 유충 추출 결과 물 추출물에서 5시간과 10시간에서 총 단백질과 고형분 함량당 각각 가장 높은 2,2,1-diphenyl-1-picrylhydrazyl (DPPH) 활성을 보였다. 물 추출물을 메탄올 농도에 따라 분획하였을 때, 높은 메탄올 농도는 상등액 중에 고분자량 단백질의 수준을 감소시키는 반면, 상등액의 DPPH 활성 수준을 증가시켜 저분자량 펩타이드가 항산화 활성을 매개하는 것으로 나타났다. 유기용매 분배에서 부탄올을 제외한 유기 용매 분배에서 물층에서 유사한 활성을 보였다. 유기용매분획물을 열처리 후 활성이 28~44% 감소하여 열에 대한 불안정한 성분이 존재하는 것으로 추정되었다. 물 추출물에 프로테아제 K를 첨가하면 DPPH 활성이 10~20% 증가하여 전체 단백질에서 방출되는 펩타이드가 항산화 활성을 부분적으로 증가시켰음을 알 수 있다. 그러므로, T. molitor 유충 추출물은 항산화제로써 기능을 수행할 수 있는 물질이 존재하는 것으로 제의할 수 있다.

Note of Five Unrecorded Mushrooms Including Three Rare Species on Mount Juwang in Korea

  • The Mount Juwang was designated as a national park in 1976 because of its unique bedrock geology. Although Juwang National Park has maintained its natural ecosystem well, few macrofungal surveys have been carried out. As a part of a project supported by the National Institute of Biological Resources (NIBR) for the discovery of indigenous fungal species, we surveyed the mushrooms in Juwang National Park from 2018 to 2019. The macrofungi were identified based on morphological and molecular analyses. Among these fungi, five specimens were identified as species previously unrecorded in Korea: Calocybe decolorata, Crepidotus brunnescens, Mycena pearsoniana, Psathyrella phegophila, and P. sulcatotuberculosa. Three of these species are known to be rare in the world: Crepidotus brunnescens, P. phegophila, and P. sulcatotuberculosa. In this study, we provide detailed morphological descriptions of the five unrecorded species from Mt. Juwang in Korea.

Account of montane and insular speciation in some Korean megadriles (Annelida: Oligochaeta)

  • Surveys of easily accessible or moderately remote South Korean mountains has revealed several common exotic and early species-complexes [Amynthas corticis (Kinberg, 1867) with A. diffringens (Baird, 1869), A. gracilis (Kinberg, 1867) and Metaphire californica (Kinberg, 1867)], plus an unexpected number of new native taxa. Megascolecid Metaphire muuido sp. nov. and lumbricid Eisenia muuido sp. nov. are newly described from Muuido Island, Incheon. Montane taxa are parthenogenetic Amynthas tokioensis oculo sub-sp. nov. that lacks male pores but is yet comparable to both Amynthas tokioensis (Beddard, 1892) and Metaphire soulensis (Kobayashi, 1938) with its possible new synonym A. chiakensis Hong & James, 2013. Apparently unique sympatric taxa are Amynthas bangtaesan bangtaesan and A. b. confinius sup-spp. nov., Amynthas centurio sp. nov., Amynthas punicans sp. nov., Amynthas seoraksan and A. seoraksan iti sub-spp. nov. These are newly described and their DNA COI gene barcodes, where obtainable, are presented in a phylogram with outgroup Acanthodrilidae Microscolex dubius (Fletcher, 1887) from Lake Biwa Japan being a new exotic record for Asia.

A report of 22 unrecorded bacterial species in Korea in the phyla Bacteroidetes and Rhodothermaeota

  • A total of 22 bacterial strains belonging to the phylum Bacteroidetes were isolated primarily from aquatic environments such as seawater, freshwater, lagoon and tidal flat. One of these 22 strains was isolated from ginseng soil. Phylogenetic analyses based on 16S rRNA gene sequences revealed that 21 strains showed the high sequence similarities(${\geq}98.7%$) to the closest type strains and formed robust phylogenetic clades with closely related species in the phylum Bacteroidetes. One strain, which had been previously classified as Balneola vulgaris in the phylum Bacteroidetes, was identified as a member of the newly described phylum Rhodothermaeota. These strains had not been previously reported in Korea. Here, we report 21 species of 13 genera in the phylum Bacteroidetes and one species in the phylum Rhodothermaeota which were not reported in Korea. Morphological, biochemical, and physiological characteristics, isolation sources, and NIBR deposit numbers are described in the species descriptions.

A report of 18 unrecorded prokaryotic species isolated from the feces of an Oriental stork (Ciconia boyciana), and from the intestinal tracts of a cobitid fish (Kichulchoia multifasciata) and a Korean splendid dace (Coreoleuciscus splendidus)

  • The animal gut is filled with highly diverse microbes associated with host metabolism, physiology, and pathology. However, numerous animal gut microbes have not been cultured or reported. We isolated various bacterial species using culture-dependent approaches during a comprehensive investigation of endangered endemic vertebrate species in the Republic of Korea. A total of 18 unrecorded bacterial species were isolated from the feces of an Oriental stork (Ciconia boyciana), and from the intestinal tracts of a cobitid fish (Kichulchoia multifasciata) and a Korean splendid dace (Coreoleuciscus splendidus). Based on a phylogenetic analysis of 16S rRNA gene sequences, we discovered species belonging to the phyla Actinobacteria (eight species), Firmicutes (seven species), Proteobacteria (two species), and Bacteroidetes (one species). Based on their high 16S rRNA gene sequence similarities (>98.7%) and formation of monophyletic clades with type species, each species was classified into an independent and predefined bacterial species. Gram-stain reaction, colony and cell morphology, basic biochemical characteristics, isolation source, and NIBR IDs for each species are described in the species description section.

Eight unrecorded bacterial species isolated from soil and marine sediment in Korea

  • The Earth contains billions of microbial species, although the vast majority cannot be cultured in laboratories and are thus considered unidentified and uncharacterized. Extremophiles are microorganisms that thrive in extreme conditions, including temperature, salinity, and pH. Extremophilic microorganisms have provided important insights for biological, metabolic, and evolutionary studies. Between 2017 and 2019, as part of a comprehensive investigation to identify bacterial species in Korea, eight bacterial strains were isolated from marine and non-marine environments in Jeju Island. These strains were cultured under extreme salinity or pH conditions. Phylogenetic analysis using 16S ribosomal RNA(rRNA) gene sequencing indicated that all eight strains belonged to the phyla Gammaproteobacteria, Bacilli, and Alphaproteobacteria. Based on their high 16S rRNA gene sequence similarities(>98.7%) and the formation of strong monophyletic clades with their closest related species, all isolated strains were considered as an unrecorded strain, previously unidentified species. Gram stain reaction, culture conditions, colony and cell morphology, biochemical characteristics, isolation source, and National Institute of Biological Resources(NIBR) IDs are described in this article. The characterization of these unrecorded strains provides information on microorganisms living in Korea.
