Role of Nitric Oxide in Ischemia-evoked Release of Norepinephrine from Rat Cortex Slices (흰쥐 대뇌피질 절편에서 허혈에 의한 Norepinephrine 유리에 있어서 Nitric Oxide의 영향)

  • It has been generally accepted that glutamate mediates the ischemic brain damage, excitotoxicity, and induces release of neurotransmitters, including norepinephrine(NE), in ischemic milieu. In the present study, the role of nitric oxide(NO) in the ischemia-induced $[^3H]norepinephrine([^3H]NE)$ release from cortex slices of the rat was examined. Ischemia, deprivation of oxygen and glucose from $Mg^{2+}-free$ artificial cerebrospinal fluid, induced significant release of $[^3H]NE$ from cortex slices. This ischemia-induced $[^3H]NE$ release was significantly attenuated by glutamatergic neurotransmission modifiers. $N^G-nitro-L-arginine$ methyl ester(L-NAME), $N^G-monomethyl-L-arginine$ (L-NMMA) or 7-nitroindazole, nitric oxide synthase inhibitors attenuated the ischemia-evoked $[^3H]NE$ release. Hemoglobin, a NO chelator, and 5, 5- dimethyl-L-pyrroline-N-oxide(DMPO), an electron spin trap, inhibited $[^3H]NE$ release dose-dependently. Ischemia-evoked $[^3H]NE$ release was inhibited by methylene blue, a soluble guanylate cyclase inhibitor, and potentiated by 8-bromo-cGMP, a cell permeable cGMP analog, zaprinast, a cGMP phosphodiesterase inhibitor, and S-nitroso-N-acetylpenicillamine (SNAP), a nitric oxide generator. These results suggest that the ischemia-evoked $[^3H]NE$ release is mediated by NMDA receptors, and activation of NO system is involved.

Usefulness of four commonly used neuropathic pain screening questionnaires in patients with chronic low back pain: a cross-sectional study

  • Background: Recently symptoms-based screening questionnaires have gained attention for screening for a neuropathic pain component (NePC) in various chronic pain conditions. The present study assessed the usefulness of four commonly used NePC screening questionnaires including the Self-completed douleur neuropathique 4 (S-DN4), the ID Pain, the painDETECT questionnaire (PDQ), and the Self-completed Leeds Assessment of neuropathic Symptoms and Signs (S-LANSS) questionnaire in patients with chronic low back pain (CLBP) to assess the presence of NePC. Methods: This is a single-center cross-sectional study where patients with CLBP, with or without leg pain, were included. Participants were initially screened for NePC presence by a physician according to the regular practice, and later assessed using screening questionnaires. The diagnostic accuracy of these questionnaires was compared assuming the physician-made diagnosis as the gold standard. Results: A total of 215 patients with CLBP of which 164 (76.3%, 95% CI, 70.2-81.5) had a NePC were included. S-DN4, ID Pain, and PDQ have an area under the curve (AUC) > 0.8 indicating excellent discrimination. However, S-LANSS has an AUC of 0.69 (0.62-0.75), indicating low discrimination. S-DN4 has a significantly higher AUC as compared to ID Pain (d(AUC) = 0.063, P < 0.01) and S-LANSS (d(AUC) = 0.197, P < 0.01). But the AUC of S-DN4 does not significantly differ from that of PDQ (d(AUC) = 0.013, P = 0.62). Conclusions: S-DN4, ID Pain, and PDQ, but not S-LANSS, have good discriminant validity to screen for NePCs in patients with CLBP. Despite using all the tests, 20-30% of patients with an NePC were missed. Thus, these questionnaires can only be used as an initial clue in screening for NePCs, but do not replace clinical judgment.

  • Pigs require energy for maintenance and productive purposes, and an accurate amount of available energy in feeds should be provided according to their energy requirement. Available energy in feeds for pigs has been characterized as DE, ME, or NE by considering sequential energy losses during digestion and metabolism from GE in feeds. Among these energy values, the NE system has been recognized as providing energy values of ingredients and diets that most closely describes the available energy to animals because it takes the heat increment from digestive utilization and metabolism of feeds into account. However, NE values for diets and individual ingredients are moving targets, and therefore, none of the NE systems are able to accurately predict truly available energy in feeds. The DE or ME values for feeds are important for predicting NE values, but depend on the growth stage of pigs (i.e., BW) due to the different abilities of nutrient digestion, especially for dietary fiber. The NE values are also influenced by both environment that affects NE requirement for maintenance ($NE_m$) and the growth stage of pigs that differs in nutrient utilization (i.e., protein vs. lipid synthesis) in the body. Therefore, the interaction among animals, environment, and feed characteristics should be taken into consideration for advancing feed energy evaluation. A more mechanistic approach has been adopted in Denmark as potential physiological energy (PPE) for feeds, which is based on the theoretical biochemical utilization of energy in feeds for pigs. The PPE values are, therefore, believed to be independent of animals and environment. This review provides an overview over current knowledge on energy utilization and energy evaluation systems in feeds for growing pigs.

  • This paper is to execute a study for characteristic analysis of synchronization clock and maximum network node number with various clock states, normal, SPT, LPT, in NG-SDH networks. Through the simulations, maximum network node numbers showed from 42 to 38 nodes in normal state. In SPT state, maximum network node numbers, when the last NE network applied to only SPT state, presented from 19 to 4 nodes, much less than normal state. Node numbers to meet specification in case of occurrence of SPT state in all NE networks decreased greatly. In LPT state, all maximum node numbers, when the last NE network applied to only LPT state, presented more than 50 nodes, and the results in case of occurrence of LPT state in all NE networks were also identified. However, node numbers to meet specification in case of LPT state in all DOTS networks were few large with difference between LPT and normal or SPT state.

Role of Nitric Oxide Produced During Endotoxic Shock in Sympathetic Nervous Function (Endotoxin에 의해 생성된 혈관의 nitric oxide가 교감신경계에 미치는 영향)

  • Endotoxic shock causes death in humans and animals via extreme hypoperfusion of peripheral organs. A massive production of nitric oxide (NO) both from the endothelical cells and smooth muscle cells has been proposed as a possible mechanism in this process. Since NO attenuated the contractility to vasoconstricting agents such as norepinephrine (NE) by directly acting on the smooth muscle cells, this mechanism was considered mainly as a postsynaptic mechanism. In this research it was investigated whether NO, thus released, also participates in the presynaptic events for the regulation of vascular tone in endotoxic shock. The role of NO was studied by adding NO donors or NO synthase inhibitor $N^\omega $methyl-L-arginine (NMA) in stimulated sympathetic nerves of the mesenteric vascular bed and the Langendorff heart of rats. Sodium nitroprusside (SNP), an NO donor, reduced the pressor responses of isolated mesenteric artery either to electrical stimulation or exogenously administered phenylephrine (PE). In this mesentery, although neither agent influenced NE release, in the presence of the adrenergic $\alpha_2$-receptor antagonist yohimbine, elecrical stimulation-evoked NE release was augumented by SNP. In the heart SNP facilitated the NE release induced by electrical stimulation, while NMA had no effect. From these results it is proposed that there exists a local reflex phenomenon in the junction between the sympathetic nerve terminals and the smooth muscle of resistance blood vessels; by which sympathetic responses are reduced by NO at the postjunctional level while NO facilitates NE release contributing to augumentation of sympathetic tone. All these facts suggest that NO produced during endotoxic shock has dual effects: whereas NO blunts the vasoconstrictive activity of NE at the postsynaptic level, NO presynaptically facilitates the release of NE from sympathetic nerve terminals.

Anti-migration Effects of the Daesiho-tang (Da Chai Hu-Tang) Water Extract in Cancer Cells by Regulating Macrophage Polarization (대식세포 분화 조절을 통한 대시호탕의 암세포 전이 억제 효과)

  • The aim of this study was to investigate the role of Daesiho-tang (Da Chai Hu-Tang) water extract (DSTE) in regulating chronic stress-induced cancer progression, focusing on its activity in modulating tumor-associated macrophages (TAMs). Different stimuli can polarize TAMs into immune-stimulating M1 macrophages or immunosuppressive M2 macrophages. During cancer progression, M2 phenotype increases and supports tumor growth, angiogenesis and metastasis. Notably, chronic stress-induced catecholamines promote M2 macrophage polarization. In this study, we investigated whether DSTE regulates norepinephrine (NE)-induced M2 macrophage polarization in RAW 264.7 mouse macrophage cells. Even though NE itself did not increase the expression of M2 markers, the conditioned media of NE-treated 4T1 mouse breast cancer cells (NE CM) significantly up-regulated M2 markers in RAW 264.7 cells, suggesting that NE-regulated cancer cell secretome stimulated M2 polarization. However, such increase was abrogated by DSTE. NE CM also induced phosphorylation of signal transducer and activator of transcription 6 (STAT6) in RAW 264.7 cells, which was clearly reversed by pretreatment with DSTE, demonstrating that DSTE inhibited M2 polarization by inactivating STAT6. Finally, M2-polarized RAW264.7 cells by NE CM markedly increased the migration of 4T1 cells. However, such increase was completely reversed by co-treating RAW264.7 cells with NE CM and DSTE, indicating that DSTE attenuated cancer cell migration by blocking M2 polarization. Taken together, our results suggest a probable use of DSTE for cancer patients under chronic stress by regulating M2 macrophage polarization.

  • 21세기는 정보화 시대로써 인터넷을 기반이 산업과 가정기기의 시설이 늘어나고 있는 추세이다. 아직까지 인터넷 기반의 시설이 많지는 않지만 곧 '유비쿼터스 네트워크 사회'로 접어들기 시작 하면 가정과 산업현장에서는 언제, 어디서라도 기기와 연결되는 것으로, 시간의 제약에서 벗어나 인간은 보다 풍요로운 생활의 실현을 가능하게 할 것이다. 또한 산업 현장에서는 실시간으로 관리기기의 상태를 파악하고, 데이터를 수집을 현장에 없어도 인터넷으로 할 수 있다. 장시간 산업 현장을 비우더라도 인터넷을 이용하여 산업 현장에서 일하는것과 같은 현상을 줄 수 있다. 따라서 본 논문에서는 MC9S12NE64를 이용하여서 소형의 웹서버를 구축하였다. 인터넷 웹 브라우저를 이용한 원격제어를 위해서는 MC9S12NE64를 서버로 이용하고 PC의 웹서버를 클라이언트로 사용하였다. 클라이언트인 PC에서 인터넷 웹브라우저를 이용하여 서버인 MC9S12NE64로 접근을 하여 I/O 포트를 제어하였다.

He-Ne Laser을 이용한 혼합용액의 Cloud-point curves 측정

  • 국내 및 해외에서 생산 되는 윤활유에 Cloud-point curves 및 상용성 개념을 도입하여 이러한 데이터를 측정할수 있는 장치를 기존의 Light scatering, Neutron scatering, Visible Method등이 아닌 He-Ne Laser를 이용하여 측정하는 장치이다. He-Ne Laser을 이용하여 공중합체 용액의 Cloud-point curves를 측정하였고, 측정 Cloud-point curves 데\ulcorner를 연속열역학적 상평형관계식에 적용 coex istence curves를 추산하였으며 추산치와 측정치를 비교 검토하였다. 멀지 않은 해에는 이러한 분석장치가 상용화되어 윤활유 제품의 상용성을 평가하는 장치가 될 것으로 사료된다. 따라서, 기존 윤활유 제품 및 신제품의 상용성(compatibility)을 He-Ne Laser을 이용하여 측정하고 다른 일면으로는 윤활유 제품의 고품질화 및 제품의 경쟁력을 강화하는데 커다란 영향을 미치리라 사료된다.

  • The optimal mixing condition of four components gas(Ne,Xe,He,Ar) in PDPs was caculated by a numerical simulation method. The dominated reactions in which $Xe^*(^3P_1)$ is produced and decays were investigated in three components gas (Ne,Xe,He) and our new components gas (Ne,Xe,He,Ar). A peak point of $Xe^*$ density appears in the range of 0.1% to 2% of Ar mixture ratio. The results of simulation show that the direct exitation of Xe by electrons has the greatest influence on the inceasing $Xe^*$ density in both gas mixtures.
