Expression Analysis of OsCPK11 by ND0001 oscpk11 Mutants of Oryza sativa L. under Salt, Cold and Drought Stress Conditions (염분, 저온 및 가뭄 스트레스 조건에서 벼 ND0001 oscpk11 돌연변이체의 OsCPK11 발현 분석)

  • Calcium-dependent protein kinases (CDPKs) are known to be involved in regulating plant responses to abiotic stresses such as salinity, cold temperature and dehydration,. Although CDPKs constitute a large multigene family consisting of 31 genes in rice, only a few rice CDPKs' functions have been identified. Therefore, in order to elucidate the functions of OsCPK11 in rice, this study was intended to focus on the expression pattern analysis of OsCPK11 in wild type and ND0001 oscpk11 mutant plants under these abiotic stresses. For the salt, cold and drought stress treatment, seedlings were exposed to 200 mM NaCl, 4℃ and 20% PEG 6,000, respectively. RT-PCR and quantitative real-time PCR were performed to determine the expression patterns of OsCPK11 in wild type and ND0001 mutant plants. RT-PCR results showed that OsCPK11 transcripts in the wild type and heterozygous mutant were detected, but not in the homozygous mutant. Real-time PCR results showed that relative expression of OsCPK11 of wild type plants was increased and reached to the highest level at 24 hr, at 6 hr and at 24 hr under salt, cold and drought stress conditions, respectively. Relative expression of OsCPK11 of ND0001 homozygous plant was significantly reduced compared to that of wild type. These results suggested that oscpk11 homozygous mutant knocks out OsCPK11 and OsCPK11 might be involved in salt, cold and drought stress signaling by regulating its gene expression.

  • The crystallographic grain orientation of {{{{ { ZrB}_{2 } }}}}-ZrC composite sintered by pressureless a sintering(PLS) and spark plasma sintering (SPS) was analysed by the SEM-EBSP technique. In the case of PLS, (160) plane of {{{{ { ZrB}_{2 } }}}} was oriented to ND direction, (101) and (111) plane of ZrC were oriented to ND direction. In the case of SPS, (0001) plane of {{{{ { ZrB}_{2 } }}}} was strongly oriented to ND direction. Only (001) plane of ZrC was oriented to ND direction. The PLS specimen had weakly oriented grain structure and interface between {{{{ { ZrB}_{2 } }}}} and ZrC was found to be more stable than that of SPS but the SPS specimen had a preferentially oriented grain structure.

Effect of Obesity and Diabetes on Alzheimer's APP Gene Expression in Mouse Adipose Tissues (비만 및 당뇨가 생쥐 지방조직에서의 Alzheimer's APP 유전자 발현에 미치는 영향)

  • The aim of this study was to determine whether Alzheimer's amyloid precursor protein (APP) is dysregulated in adipose tissues of C57BL/6 male mice by high-fat diet (HFD) induced obesity, aging, or streptozotocin (STZ)-induced diabetes. APP mRNA expression was examined by quantitative real-time PCR (QPCR) in subcutaneous (SAT) and epididymal adipose tissues (EAT) from mice in 8 different condition groups. By combining conditions of age (16 weeks/26 weeks of age), diet (normal diet (ND)/high-fat diet), and induction of diabetes (non-diabetic/diabetic), 88 mice were divided into 8 different groups. QPCR demonstrated that APP expression in SAT was significantly increased by about two-fold in HFD-induced obese mice compared to both 16 week-old and 26 week-old mice in the ND group (16 weeks p=0.001; 26 weeks p<0.0001), but no changes in EAT was found. Particular effects of aging on APP gene expression were not observed in either adipose tissue depots. Significantly decreased APP expression was found in SAT in STZ-induced diabetic mice fed on ND or HFD at 16 weeks of age (ND p<0.05; HFD p<0.01). Linear regression analysis demonstrated that APP expression levels correlated with body weight in both the non-diabetic group (R=0.657, p<0.0001, n=39) and the diabetic group (R=0.508, p=<0.0001, n=49), but did not correlate with plasma glucose levels, which suggests that decreased APP expression in STZ-induced diabetic mice is most likely due to weight loss rather than hyperglycemia. These data confirm APP dysregulation by weight changes in humans and suggest a possible role linking midlife obesity with the later development of amyloidogenesis in the brain of older patients with Alzheimer's disease.

  • As a part of continuing monitoring program since 1983, a study on pesticide residues on Korean agricultural products has been conducted to offer the tolerance of pesticide residues suiting Korean reality. The samples used in this experiment were collected from four areas in five provinces of Korea. The residue, levels of organochlorine pesticide (two kinds), organophosphoric pesticide (eight kinds) and carbamate pesticide (three kinds) on lettuce, garland chrysanthemum, radish Korean, potato, egg plant, green onion, persimmon and sweet potato, and the residue levels of captan and captafol on tomato, green pepper fresh, apple, peach, soybean, cucumber and cabbage Korean were determined by GC-NPD and ECD.

A Study on the Relationship between Discomfort and Depression of the Patients with Cast or Traction (석고붕대.견인환자의 불편감과 우울과의 관계연구)

  • This study was performed to explore relationships between depression and diacomfort. of patients with cast or traction. 112 parients were selected from several hospitals in Jeon-buk province. Data were collected from Feb. 26th. to March, 2nd. of 1996 using survey interview designed by the investigator. Data analysis included percentage, mean, and standard deviation, t-test, ANOVA, Pearson Correlation Coefficient. Results were as follows : 1. The depression score and the discomfort score were positively correlated(r=.3654, P=.0001). 2. The result shows that the score of ADLs(activites of daily living) and the score of discomfort were negatively correlated and they were statistically significant(activity r=-.1980, p=.0363 ; eating r=-3483, p=0002 ; bathroom r=-.4207, p=.0001 ; wearing r=-.4602 p=.0001 ; bathing r=-.3489, p=.0002).

Heavy Metal Contents in Beverages Consumed in Korea (국내 유통 음료류 중 유해중금속 함량)

  • This study was conducted to examine the contents of heavy metals in beverages and to estimate their intakes. The contents of lead (Pb), cadmium (Cd), arsenic (As), and tin (Sn) were determined in a total of 207 samples of beverages using atomic absorption spectrophotometer(AAS) and inductively coupled plasma spectrometer(ICP). Also a mercury analyzer was utilized for analysis of mercury. The values of heavy metals in beverages were as follows [min-max (mean), mg/kg]; Hg 0.0001-0.0025 (0.0004), As ND-0.0245 (0.0003), Pb ND-0.089 (0.004), Cd ND-0.006 (0.001), Sn ND-45.36 (1.97). Our results were similar to those reported by other countries. Our weekly intakes of heavy metals from beverages take $0.01%{\sim}0.06%$ of PTWI (Provisional Tolerable Weekly Intake) established by FAO/WHO.

Alteration of Thyroid Function in Indian HER 2-Negative Breast Cancer Patients Undergoing Chemotherapy

  • Background: Thyroid hormones (TH) are regulated by the hypothalamic-pituitary axis, which plays an important role in cell growth, differentiation, development and other aspects of metabolism. It is believed that an active hypothalamic-pituitary axis increases the susceptibility of thyroid dysfunction during systemic chemotherapy. In order to investigate the relation between thyroid function and chemotherapy the present study was designed to investigate TH in breast cancer patients receiving at least three cycles of chemotherapy. The levels of TH were measured at the baseline and before each cycle of chemotherapy. Materials and Methods: Blood samples for estimation of TH levels were collected from 80 (pre-menopausal-40; post-menopausal-40) breast cancer patients just before they were undergoing - $1^{st}$, $2^{nd}$, $3^{rd}$ and $4^{th}$ cycle of chemotherapy. The serum was separated and $T_3$, $T_4$ and TSH levels were determined by chemiluminescence method. Results: $T_3$ and $T_4$ were found significantly decreased and TSH was found significantly increased after $1^{st}$ (p<0.001), $2^{nd}$ (p<0.0001) and $3^{rd}$ cycle of chemotherapy (p<0.0001). The variation of $T_3$ levels (decreased) and TSH levels (increased) was found more in post-menopausal (p<0.0001) women then in pre-menopausal women after $3^{rd}$ cycle of chemotherapy as compared to baseline (p<0.001). Conclusions: TH were remarkably altered after each cycle of chemotherapy leading to decline in thyroid function of breast cancer patients. Further, the results also indicated that post-menopausal women were more prone towards decline in thyroid function then pre-menopausal women. The present study proposes the monitoring of TH after each cycle of chemotherapy in breast cancer patients.

A Study on Fracture Behavior and Impact Stability of Sintered Rare-earth Permanent Magnets

  • The fracture behavior and mechanical characteristics of sintered rare-earth magnets were investigated. It shows that the fracture behavior and bending strength of the magnets obviously exhibit anisotropy. Sm-Co magnets tend to cleavage fracture in the close-packed (0001) plane or in the ($10\bar{1}1$) plane. The fracture mechanism of $Nd_2Fe_{14}B$ magnet mainly appears to be intergranular fracture. The anisotropy of fracture behavior and mechanical strength of sintered rare-earth magnets is caused mainly by the strong crystal-structure anisotropy and the grain alignment texture. The effects of Nd content, and Pr, Dy substitution on the impact stability of $Nd_2Fe_{14}B$ magnets were also reported.

Correlation of Gene Expression between Adiponectin and Glucose Transporter 4 in Mouse Adipose Tissue (생쥐 지방조직에서의 아디포넥틴과 포도당수송체-4 유전자 발현의 상관관계)

  • Adiponectin has been known to improve insulin sensitivity and elicit glucose uptake via increased glucose transporter 4 (GLUT4) translocation. In the current study, mRNA expression levels of adiponectin and GLUT4 were measured in subcutaneous adipose tissue from C57BL/6 mice fed normal (ND) or high-fat diet (HFD) until 16, 26, 36, 47, or 77 weeks of age starting from 6 weeks of age. Expression levels were also measured in mice with calorie restriction (CR) and in thiazolidinedione (TZD) treated mice. Using quantitative real-time PCR, we demonstrated that GLUT4 expression in adipose tissue significantly decreased in HFD mice groups and increased in CR (p<0.05) and TZD (p=0.007) groups while there was no difference in adiponectin mRNA expression levels between experimental and control groups. General linear regression models were used to assess the association of gene expression levels between adiponectin and GLUT4 and to determine whether adiponectin affects GLUT4 transcription. mRNA expression levels of adiponectin and GLUT4 are significantly associated each other in mice fed a ND (p<0.0001) or HFD (p<0.0001), in groups separated into each age and diet, and CR group (p=0.002), but not in TZD group (p=0.73). These results demonstrated that gene expression of adiponectin and GLUT4 is strongly associated, suggesting that there is a common regulatory mechanism for adiponectin and GLUT4 gene expression and/or adiponectin has a direct role in GLUT4 gene expression in adipose tissue.

Heavy Metal Contents in Ginseng and Ginseng Products (인삼 및 인삼제품류의 중금속 함량)

  • Contents of heavy metals, mercury (Hg), lead (Pb), cadmium (Cd), arsenic (As), manganese (Mn), zinc (Zn), and copper (Cu), In ginsengs and ginsengs products their safety were evaluated using a mercury analyzer, atomic absorption spectrophotometer and Inductively coupled plasma spectrometer. Contents of heavy metals in ginsengs were [min-max(mean), mg/kg]; Hg 0.002-0.02(0.007), Pb 0.018-0.39(0.106), Cd 0.004-0.413(0.106), As ND-0.094(0.013), Cu 0.24-5.5(3.2), Mn 3.18-50.4(20.09), Zn 0.76-44.27(13.42). Ginseng products values were; Hg 0.0001-0.002(0.001), Pb 0.001-0.133(0.017), Cd ND-0.07(0.004) As ND-0.181(0.008), Cu ND-1.1(0.13), Mn 0.73-30.15(1.61). Zn 0.02-13.42(1.02), similar to those reported by other countries. Average weekly Intakes of Hg, Pb and Cd from ginseng and ginseng products were 0,003, 0.01 and 0,02% of provisional tolerable weekly intake established by FAO/WHO, respectively. Our result could be utilized as important references to establish the standard of lead in ginseng and ginseng products.
