약용 식물 추출물의 항산화 활성 검색 (Screening for Antioxidant Activity of Plant Medicinal Extracts)

  • 현재 시판되고 있는 한약재 및 약용식물 118종에 대해서 80% 메탄올 추출물을 제조하였고 , 이들의 항산화 활성을 DPPH 와 superoxide anion radical 소거능 측정 방법을 이용하여 조사하였다. DPPH radical 소거 활성은 괴화 (Sophora japonica: 줄기부)의 메탄올 추출물에서 76.9%의 활성을 보였고 회수(Camptotheca acuminata Dence: 지상부)의 추출물에서는 50.9%의 활성을 보였으며 박하(Mentha arvensis: 지상부), 비파엽 (Eriobotryajaponica: 지상부), 소엽(Perilla frutescens: 줄기부), 애엽(Artemisia asiatica: 줄기부 ), 초과(Amomum. costatum: 지상부) 의 추출물과 산비장이(Serratula koreana: 잎), 살구나무(Prunus ansu: 잎)는 30% 이상의 활성을 보였다. Superoxide anion radical 소거능은 박하(Mentha arvensis), 비파엽(Eriobotrya jαponica), 초과(Amomum costatum), 희수(Camptotheca acuminata Dence)의 지상부에서 80% 이상의 활성을 보였으며 괴화(Sophora jαponica), 소엽(Perilla frutescens), 애엽(Artemisia asiatica), 순비기나무 (Vitex rotundifolia)의 줄기부 , 짚신나물(Agrimonia pilosa)의 잎, 창질경이(Plantago lanceolata) 지상부는 50% 이상의 활성을 나타내었다. 박하(Mentha arvensis) 와 비파엽(Eriobotrya jαponica), 초과(Amomum costatum), 희수(Camptotheca acuminata Dence)의 지상 부는 DPPH뿐만 아니라 superoxide anion radical 소거능에서도 높은 활성을 보였다.

느릅나무와 시무나무의 항산화 활성 (Antioxidant Activity of Ulmus davidiana var. japonica N. and Hemipteleae davidii P.)

  • 한약재로서 혼용되고 있는 느릅나무 및 시무나무의 수피와 근피의 항산화활성 및 총페놀 함량의 차이를 구명하고자 본 연구를 수행하였으며 그 결과는 다음과 같다. DPPH 라디칼에 대해 느릅나무 근피와 수피의 80% 에탄올 추출물은 $10\;{\mu}g/ml$의 농도에서도 각각 41.4% 및 35.6%의 활성을 보여 다른 추출물의 활성 $(2.3{\sim}24.0%)$에 비해 우수한 효과를 가지는 것을 알 수 있었고, superoxide 라디칼에 대해서는 $200{\sim}5\;{\mu}g/ml$의 농도에서 느릅나무 및 시무나무의 수피와 근피로부터 얻어진 모든 추출물이 항산화제인 ascorbic acid보다 우수한 활성을 나타내었으며 특히 느릅나무 근피의 80% 에탄올 추출물과 물 추출물이 좀 더 높은 활성을 나타내었다. 사람의 저밀도지단백 (LDL) 산화에 대한 실험 결과 느릅나무 수피 및 근피의 80% 에탄올 추출물이 $10\;{\mu}g/ml$의 농도에서 84.3% 및 81.7%로 다른 추출물들의 활성 $(31.3%{\sim}78.2%)$보다 비교적 높은 것을 확인할 수 있었다. 실험된 모든 추출물은 linoleic acid 과산화에 대해서 ${\alpha}-tocopherol$보다 낮은 저해활성을 나타내었다. 한편, 총페놀 함량은 느릅나무 근피의 80% 에탄올 추출물이 50.8%로 가장 높았으며 시무나무 근피의 물 추출물이 5.6%로 가장 낮았다. 이상의 실험결과, 느릅나무의 근피와 수피가 시무나무의 근피와 수피보다 더 효과적인 항산화 활성을 보유하며 특히, 느릅나무 근피의 80% 에탄올 추출물은 가장 항산화 활성이 우수한 것으로 확인되었다.

쇠비름의 무기물 함량 및 항산화 활성 (Mineral Content and Antioxidants Activity of Portulaca oleracea)

  • 쇠비름의 생리활성을 알아보기 위하여 무기물 함량 및 항산화 활성을 분석하였다. 쇠비름의 무기물은 총 9종 검출되었으며, 총량은 6025.80 mg/100 g으로 칼륨이 3846.99 mg/100 g으로 가장 높았다. 물 및 에탄올 추출물의 추출 수율은 각각 14.84% 및 24.93%였다. 쇠비름 에탄올 추출물의 총 페놀화합물 및 플라보노이드 함량은 각각 58.16 mg/g, 20.08 mg/g으로 물 추출물(49.09 mg/g, 14.98 mg/g)보다 유의적으로 높았다. 항산화 활성 및 아질산염 소거활성은 시료의 첨가량에 따라 유의적으로 상승하였으며, 에탄올 추출물이 물 추출물보다 활성이 더 높았다. ${\alpha}$-glucosidase 저해활성은 에탄올 추출물이 유의적으로 높은 활성을 보였다. 따라서, 쇠비름의 물 및 에탄올 추출물은 항산화 활성 및 혈당강하에 효과적일 것으로 판단되며, 시료 중의 무기물, 페놀 화합물 및 플라보노이드 함량과 관련성이 높을 것으로 사료된다.

SM520C-TMC 강재의 피로강도에 대한 두께효과 (Thickness Effects on the Fatigue Strength of Butt Welded Specimens using SM520C-TMC Steel)

  • 피로설계에 대한 기준, 시방서 또는 추천안 등은 일반적으로 두께 10~30mm의 강판으로 만든 실험체의 피로실험을 통해 얻은 피로강도곡선을 근거하고 있다. 그러나 강판의 두께가 증가함에 따라서 피로강도가 감소하게 될 가능성에 대해 관심을 갖게 되었고, 파괴역학적 방법에 의한 분석과 강재 용접에 따른 영향을 근거로 하여 강판의 두께에 따른 영향이 상당히 클 수도 있다고 지적되었다. 기존의 몇몇 피로실험 결과로부터 얻은 피로강도곡선으로부터 강판 두께에 대한 영향을 고려하여 피로강도를 수정하는 식이 제안되어왔다. 본 연구에서는, 두께 80mm까지의 국내생산 SM520C-TMC 강재에 대한 피로강도시험을 실시하고, 그 결과로부터 강재의 두께에 따른 영향을 평가하였다. 최종적으로, 강판 두께에 대한 영향을 고려하여 피로강도를 수정하는 제한적인 시편에 대한 수정식을 제공하고, 이를 기존의 연구결과와 비교 검토하였다. 결과로서, 두께에 따른 강도감소효과는 기존의 연구와 유사한 경향을 갖고 감소하나 그 정도는 적어지고 있는 결과를 얻을 수 있었다.

Hypolipidemic and hypoglycemic effects of Orostachys japonicus A. Berger extracts in streptozotocin-induced diabetic rats

  • The hypolipidemic and hypoglycemic effects of two dietary dosages (0.1% and 0.5%) of water and 80% ethanol extracts from hot-air dried Orostachys japonicus A. Berger were evaluated in the serum and organ tissues of streptozotocin-induced diabetic rats. The STZ-induced diabetic groups supplemented with the O. japonicus extracts showed significantly higher body weight compared to a diabetic control group at the end of experiment. The extracts exhibited substantial hypoglycemic effects by significant reductions of fasting blood glucose levels at all time points tested compared to the initial stage before treatment of the extracts. Declines of serum and hepatic triglyceride levels were greater than declines of total cholesterol in the groups treated with the 0.5% O. japonicus extract (DBW2 and DBE2) when compared to the DBC group. Hepatic glycogen content was higher in the groups treated with O. japonicus extract, while lipid peroxide content was decreased in these treated groups compared to the DBC group. Hepatic antioxidant activity was significantly increased in the groups supplemented with the O. japonicus ethanol extract The hypolipidemic and hypoglycemic effects of the O. japonicus ethanol extract were significantly greater than the effects of the water extract. Based on this study, it seems that O. japonicus ethanol extract, due to its higher phenolic and flavonoid components than the water extract, may control blood glucose and alleviate hyperlipidemia in diabetes.

반응표면분석을 이용한 고등어 절임용 흑마늘 추출액 절임조건의 최적화 (Optimization of Black Garlic Extract Curing Conditions for Mackerel Using Response Surface Methodology)

  • We examined the quality characteristics of obtaining the optimal curing conditions for black garlic extract (BGE) added mackerel by using response surface methodology (RSM). The condition of RSM was included in 13 experimental points using two dependent variables: concentration of BGE (2.6~12.5 brix, X1) and curing time of BGE (17.58~102.42 min, X2). An assessment was conducted with four replicates on three independent variables: volatile basic amine (VBN, Y1), thiobarbituric acid (TBA, Y2), and shear force (Y3). The minimum content of VBN was 1.625 mg% at 3.17 brix, for 80.5 min. This was obtained when the BGE concentration was lower and the curing time was longer. The content of the TBA was 3.13~12.24 MA mg/kg at experiment range and the stationary point was at saddle point. Therefore, by using ridge analysis, the minimum point of TBA was 2.414 MA mg/kg at 3.5 brix for 84.4 min. The truth coefficient of the BGE concentration and curing time were 2.9 brix and 75.1 min, respectively, with a shear force ($120.764cm/kg^2$) shown at maximum point. The optimum Hunter color range was estimated from the lowest TBA value. The optimum range of the L color was 39.16~40.26, a and b color were 4.40~4.88 and 12.35~13.20, respectively.

로테르담 규칙상 수량계약조항의 시사점에 관한 연구 (A Study on the Implication of Volume Contract Clause under Rotterdam Rules)

  • The purpose of this study aims to analyse the implications of volume contract clause with Rotterdam Rules. The Hague-Visby Rules have been in force this jurisdiction for over 30 years. In those three decades they have performed valiant service, both for the development of maritime law in this country and for the countless parties from around the world who have chosen courts and arbitral tribunals in London for the resolution of disputes arising under bills of lading or under charterparties incorporating the Hague-Visby Rules. While the Hague-Visby Rules apply only to bills of lading or any other similar documents of title and hence all other contracts of carriage are not subject to the current regime, this is not the case for the Rotterdam Rules which, broadly speaking, apply to contracts of carriage whether or not a shipping document or electronic transport record is issued. To preserve freedom of contract where necessary, however, a number of significant concessions were made and Article 80 represents one of the most controversial: that of volume contracts. However, the provision lends itself to abuse under each one of the elements as there is no minimum quantity, period of time or frequency and the minimum number of shipments is clearly just two. This means that important contracts of affreighment concluded pursuant to, for example, oil supply agreements have the same right to be excluded from the scope of application of the Rotterdam Rules. The fact that a volume contract may incorporate by reference the carrier's public schedule of services and the transport document or other similar documents as terms of the contract would make a carefully drafted booking note for consecutive shipments a potential volume contract as well.

이종재료 마찰용접에 의한 초내열합금 대형 배기밸브 스핀들 개발 (Development of Large Superalloy Exhaust Valve Spindle by Dissimilar Inertia Welding Process)

  • Inertia welding is a solid-state welding process in which butt welds in materials are made in bar and in ring form at the joint race, and energy required lot welding is obtained from a rotating flywheel. The stored energy is converted to frictional heat at the interface under axial load. The quality of the welded joint depends on many parameters, including axial force, initial revolution speed and energy amount of upset. working time, and residual stresses in the joint. Inertia welding was conducted to make the large exhaust valve spindle for low speed marine diesel engine. superalloy Nimonic 80A for valve head of 540mm and high alloy SNCrW for valve stem of 115mm. Due to different material characteristics such as, thermal conductivity and flow stress. on the two sides of the weld interface, modeling is crucial in determining the optimal weld geometry and Parameters. FE simulation was performed by the commercial code DEFORM-2D. A good agreement between the Predicted and actual welded shape is observed. It is expected that modeling will significantly reduce the number of experimental trials needed to determine the weld parameters. especially for welds for which are very expensive materials or large shaft. Many kinds of tests, including macro and microstructure observation, chemical composition tensile , hardness and fatigue test , are conducted to evaluate the qualify of welded joints. Based on the results of the tests it can be concluded that the inertia welding joints of the superalloy exhaust valve spindle are better properties than the material specification of SNCrW.

수질(水蛭)이 비만세포의 알러지 염증 반응에 미치는 영향 (Effect of Whitmania pigra whitman on the Allergic Inflammatory Response)

  • Objectives: Whitmania pigra whitman (WP) has been used in herbal medicine to treat various conditions, such as eczema, skin burns and frostbites in herbal medicine. The purpose of this study is was to investigate the effect of WP on anti-allergy mechanism. Methods: To clarify the mechanism, the effect of WP on vascular permeability of rat cutaneous tissue and histamine and cytokines (IL-6, IL-8, $TNF-{\alpha}$) released from mast cells were observed. Results: The results were 1. the pretreatment with WP significantly decreased the compound 48/80-induced degranulation and histamine release from RPMC 2. WP did not inhibit the anti-DNP IgE-induced increment of vascular permeability of rat cutaneous tissue 3. WP significantly reduced the PMA plus A23187-induced increment of expression of IL-6, IL-8, and $TNF-{\alpha}$ in HMC-1 cells. Conclusions: The present study providesevidence that WP acid inhibits mast cell-derived inflammatory allergic reactions by blocking histamine release and pro-inflammatory cytokine expression, and suggest the mechanisms of action. Furthermore, in vivo and in vitro anti-allergic effect of WP suggests a possible therapeutic application of this agent in inflammatory allergic diseases.

담수어(淡水魚)의 식품학적(食品學的) 연구(硏究)(II)-붕어, 메기, 가물치 및 미꾸리의 맛성분(成分)- (Studios on the Food from Fresh Water Fish(II)-The Taste Compounds in Meat of Crucian Carp, Skate$\cdot$Fish, Snake Head and Loaches-)

  • Crucian carp Carassius carassius L., Skate-fish Parasilurus asolus, snake head Ophicephalus argus and loaches Misgrunus anguillicaudatus have been esteemed as the tasty fresh water fish in Korea. However, a little has been studied on their taste compounds. Amino acids, nucleotides and their related compounds as the taste giving substance in the dorsal muscle of crucian carp, skate-fish, snake head and loaches were analyzed. Hypoxanthine (4.6-30.3 ${\mu}mole/g$, on dry base) was dominant among the nucleotide materials in the dorsal muscle of crucian carp, skate-fish, and snake head while IMP (12.8 ${\mu}mole/g$) was detected with the highest amount in loaches meat. IMP was also high in the muscle of skate-fish (13.5 ${\mu}mole/g$) and snake head (5.6 ${\mu}mole/g$). The other nucleotide materials, CMP, UMP, GMP, AMP, and inosine were present less than 2.0 ${\mu}mole/g$ in all the fish meat. The amino acid composition showed that glutamic acid and aspartic acid were the most abundant in the dorsal muscle of crucian carp, skate-fish, snake head and loaches. The analysis of free amino acids showed that histidine, lysine, glycine, and serine were the leading amino acids in the dorsal muscle of crucian carp, alanine, serine, and lysine were of skatefish, glycine, lysine, alanine, and serine were of snake head, while alanine, serine, and glycine were of loaches. Isoleucine and leucine were detected in trace amount in the muscle of skate-fish, snake head, and loaches. It is believed that alanine, Iysine, serine, glycine and histidine along with IMP and hypoxanthine may play an important role as the taste compounds in fresh water fish.
