Curative Radiation Therapy for T2N0M0 Non-small Cell Lung Cancer (T2N0M0 비소세포성 폐암의 근치적 방사선치료)

  • Purpose : Surgery is the treatment of choice for resectable non-small cell lung cancer. For patients who are medically unable to tolerate a surgical resection or who refuse surgery, radiation therapy is an acceptable alternative. A retrospective analysis of Patients with stage I non-samll cell lung cancer treated with curative radiation therapy was performed to determine the results of curative radiation therapy and patterns of failure, and to identify factors that may influence survival. Materials and Methods : From 1986 through 1993, 39 Patients with T2N0M0 non-small cell lung cancer were treated with curative radiation therapy at department of radiation oncology, Kyungpook national university hospital. All Patients were not candidates for surgical resection because of either Patient refusal (16 patients), poor pulmonary function (12 patients), old age (7 patients), Poor Performance (2 patients) or coexisting medical disease (2 patients). Median age of patients was 67 years. Histologic cell type was squamous cell carcinoma in 36, adenocarcinoma in 1, large cell carcinoma in 1 and mucoepidermoid carcinoma in 1. All patients were treated with megavoltage irradiation and radiation dose ranged from 5000cgy to 6150cGy with a median dose of 6000cGy. The median follow-up was 17 months with a range of 4 to 82 months, Survival was measured from the date therapy initiated. Results : The overall survival rate for entire Patients was $40.6\%$ at 2 years and $27.7\%$ at 3 years, with a median survival time of 21 months. The disease-free survival at 2 and 3 years was $51.7\%$ and $25.8\%$, respectively. Of evaluable 20 patients with complete response, 15 patients were considered to have failed. Of these, 13 patients showed local failure and 2 patients failed distantly. Response to treatment (p=0.0001), tumor size (p=0.0019) and age (p=0.0247) were favorably associated with overall survival. Only age was predictive for disease-free survival (p = 0.0452). Conclusion : Radiation therapy is an effective treatment for small (less than 3cm) tumors, and should be offered as an alternative to surgery in elderly or infirm patients. Since local failure is the prominent Patterns of relapse, potential methods to improve local control with radiation therapy are discussed.

Trichostatin A, a Histone Deacetylase Inhibitor, Potentiated Cytotoxic Effect of ionizing Radiation in Human Head and Neck Cancer Cell Lines (히스톤탈아세틸효소 억제제 Trichostatin A에 의한 인간 두경부암 셰포주의 방사선 감수성 증강)

  • Purpose : We have previously reported that human glioblastoma cells are sensitized to radiation-induced death after their exposure to trichostatin A (TSA), a histone deacetylase inhibitor (HDAC-1), prior to the irradiation. We aimed to measure the magnitude of the radiosensitizing effect of TSA in human head and neck cancer cell lines. Materials and Methods : Human head and neck cancer cell lines, HN-3 and HN-9, were exposed to 0, 50, 100, and 200 nM TSA for 18 hr prior to irradiation. Then, the TSA-treated cells were irradiated with 0, 2, 4, 6, and 8 Gy, and cell survival was measured by clonogenic assay. Results : Pre-irradiation exposure to TSA was found to radiosensitize HN-3 and HN-9 cell lines. In HN-9 cells, the fraction surviving after 2 Gy (SF2) was significantly reduced by treatment of TSA at concentration as low as 50 nM. However, a treatment with 200 nM TSA was required to significantly decrease SF2 in the HN-3 cell line. SER of pre-irradiation treatment with 200 nM TSA was 1.84 in HN-3 and 7.24 in HN-9, respectively. Conclusions : Our results clearly showed that human head and neck cancer cell lines can be sensitized to ionizing radiation by pre-irradiation inhibition of histone deacetylase (HDAC) using TSA, and that this potentiation might well be a general phenomenon.

  • The Debao pony ear marginal tissue fibroblast cell line (NDPEM 2/2) was uccessfully established using either primary explant technique or collagenase technique. The characterizations of the cell line were identified as following: the cells were adherent and of density limitation; population doubling time (PDT) of cells made with the two techniques were 35.9 h and 48 h, respectively; chromosome analysis showed that the frequency of cell chromosome number to be 2n=64 was 91.3%-92.8%. Confirmed by isoenzyme analysis, this cell line had no cross- contamination. Tests for microbial contamination from bacteria, fungi, virus or mycoplasma were negative. This newly established cell line meets all the standard quality controls of ATCC. It will provide a precious genetic resource for the conservation of the Debao pony breed, as well as effective experimental material for genetic studies on Debao ponies.

  • Objectives: This study aimed to investigate the cytotoxicity of Nexus RMGIC, an indirect restorative cement, on cell survival rate and reactive oxygen species (ROS) production in periodontal stem cells (PDSCs). Methods: PDSCs were incubated with serially diluted Nexus RMGIC eluates with and without the addition of N-acetyl-cysteine (NAC). Cell survival was determined using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. The ROS generation was determined by measuring the fluorescence intensity for 2',7'-dichlorofluorescin diacetate. Results: Nexus RMGIC exposure decreased cell proliferation and cell survival rate in a dose-dependent manner (1:8, 1:4, 1:2, 1:1) in PDSCs. The cytotoxicity of Nexus RMGIC was inhibited by treatment with 10-mM NAC. In addition, the production of ROS was detected by immunofluorescence after PDSCs were exposed to Nexus RMGIC. However, ROS generation was significantly suppressed in the NAC pretreatment compared with the Nexus RMGIC group. Conclusions: Nexus RMGIC increased the cytotoxicity and ROS generation. ROS was involved in Nexus RMGIC-induced cell toxicity.

  • The purpose of study were the effect of stress levels and duty type of guarder on 8-OHdG(hydroxydeoxyguanosin), Immunoglobin, T-cell, NK-cell and antioxidant enzyme. The subject of this study is people who have the guard's career over 2 years. A group of object sample consists of sixty guards carrying the guardbusiness. The group division and distribution of the number of persons a group divided into the guard group of international factor(N=23), private guard group(N=23),and the others(N=14) as a group following the guardbusiness. The group following a stress level divided into 3groups as their score of stress measure; low stress group(N=14), middle stress group(N=26),and high stress group(N=20). In the office type, the experiment sampling the blood, analysing result of following the office type and stress had a conclusion after measuring stress as the followings. The stress level in the office type of guard showed highly in the guard group of international factor(p<.05). Finally we knew that the guard have different stress levels in the office type, even the immune function and the activation of superoxide dismutase affected them in the stress level. Stress showing in the guard career can do them an injury so it is desirable for the guard themselves to participate in the activities settling their stresses and in the development of exercise programs.

Development of ELISA System for Screening of Specific Binding Inhibitors for Src Homology (SH)2 Domain and Phosphotyrosine Interactions

  • In the present study, an in vitro ELISA system to assess the interaction between Src homology (SH)2 domains and phosphotyrosine that contain peptides was established using purified GST-conjugated SH2 proteins and synthetic biotinylated phosphotyrosine that contain oligopeptides. The SH2 domains bound the relevant phosphopeptides that were immobilized in the streptavidin-coated microtiter plate in a highly specific and dose-dependent manner. The epidermal growth factor receptor (EGFR)-, T antigen (T Ag)-, and platelet-derived growth factor receptor (PDGFR)-derived phosphopeptides interacted with the growth factor receptor binding protein (Grb)2/SH2, Lck/SH2, and phosphatidyl inositol 3-kinase (PI3K) p85/SH2, respectively. No cross-reactions were observed. Competitive inhibition experiments showed that a short phosphopeptide of only four amino acids was long enough to determine the binding specificity. Optimal concentrations of the GST-SH2 fusion protein and phosphopeptide in this new ELISA system for screening the binding blockers were chosen at 2nM and 500nM, respectively. When two candidate compounds were tested in our ELISA system, they specifically inhibited the Lck/SH2 and/or p85/SH2 binding to the relevant phosphopeptides. Our results indicate that this ELISA system could be used as an easy screening method for the discovery of specific binding blockers of protein-protein interactions via SH2 domains.

Hypoxia Induces Paclitaxel-Resistance through ROS Production

  • Oxygen supply into inside solid tumor is often diminished, which is called hypoxia. Many gene transcriptions were activated by hypoxia-inducible factor (HIF)-$1{\alpha}$. Here, we investigated the effect of hypoxia on paclitaxel-resistance induction in HeLa cervical tumor cells. When HeLa cells were incubated under hypoxia condition, HIF-$1{\alpha}$ level was increased. In contrast, paclitaxel-mediated tumor cell death was reduced by the incubation under hypoxia condition. Paclitaxel-mediated tumor cell death was also inhibited by treatment with DMOG, chemical HIF-$1{\alpha}$ stabilizer, in a dose-dependent manner. A significant increase in intracellular ROS level was detected by the incubation under hypoxia condition. A basal level of cell density was increased in response to 10 nM $H_2O_2$. HIF-$1{\alpha}$ level was increased by treatment with various concentration of $H_2O_2$. The increased level of HIF-$1{\alpha}$ by hypoxia was reduced by the treatment with N-acetylcysteine (NAC), a well-known ROS scavenger. Paclitaxel-mediated tumor cell death was increased by treatment with NAC. Taken together, these findings demonstrate that hypoxia could play a role in paclitaxel-resistance induction through ROS-mediated HIF-$1{\alpha}$ stabilization. These results suggest that hypoxia-induced ROS could, in part, control tumor cell death through an increase in HIF-$1{\alpha}$ level.

  • Research on mutant generation and isolation for microalgae yielding enhanced lipid accumulation is an important issue for the production of economic biodiesel. In the present study, ultraviolet (UV-B type) ray induced mutant generation was tried using a photosynthetic microalgae, Nannochloropsis oculata (N. oculata), for the production of biodiesel. The resulting colonies were isolated and further cultured with both liquid and solid state f/2 media. After a few week cultivation, changes of cell growth rate, dry cell weight, and several important intracellular components (chlorophyll, carotenoid, and lipid) were investigated. Two mutants among thousands colonies showed an increased cell growth and high lipid accumulation as compared to those of wild type. It was also observed that the increased cell growth rate is associated with the overexpressed intracellular proteins. However, the mutants showed a decrease in the chlorophyll biosynthesis.

  • Purpose : In our Previous study, we have shown the main cel1 death pattern Induced by irradiation or protein tyrosine kinase (PTK) inhibitors in K562 human myeiogenous leukemic cell line. Death of the cells treated with irradiation alone was characterized by mitotic catastrophe and typical radiation-induced apoptosis was accelerated by herblmycin A (HMA). Both types of cell death were inhibited by genistein. In this study, we investigated the effects of HMA and genistein on cell cycle regulation and its correlation with the alterations of radiation-induced cell death. Materials and Methods: K562 cells In exponential growth phase were used for this study. The cells were Irradiated with 10 Gy using 6 MeV Linac (200-300 cGy/min). Immediately after irradiation, cells were treated with 250 nM of HMA or 25 $\mu$N of genistein. The distributions of cell cycle, the expressions of cell cycle-related protein, the activities of cyclin-dependent kinase, and the yield of senescence and differentiation were analyzed. Results: X-irradiated cells were arrested In the G2 phase of the cell cycle but unlike the p53-positive cells, they were not able to sustain the cell cycle arrest. An accumulation of cells in G2 phase of first ceil-cycle post-treatment and an increase of cyclin Bl were correlated with spontaneous, premature, chromosome condensation and mitotic catastrophe. HMA induced rapid G2 checkpoint abrogation and concomitant p53-independent Gl accumulation. HMA-induced cell cycle modifications correlated with the increase of CDK2 kinase activity, the decrease of the expressions of cyclins I and A and of CDK2 kinase activity, and the enhancement of radiation-induced apoptosis. Genistein maintained cells that were arrested in the G2-phase, decreased the expressions of cyclin Bl and cdc25c and cdc25C kinase activity, increased the expression of pl6, and sustained senescence and megakaryocytic differentiation. Conclusion: The effects of HMA and genistein on the radiation-induced cell death of KS62 cells were closely related to the cell cycle regulatory activities. In this study, we present a unique and reproducible model in which for investigating the mechanisms of various, radiation-induced, cancer cell death patterns. Further evaluation by using this model will provide a potent target for a new strategy of radiotherapy.

  • In order to test the function of Iymphocytes in Naegleria fowleri-nniected mice, the in nitro blastogenic response of splenocyte cultures to non-specific mitogens was studied. Concanavalin A and lipopolysaccharide stimulation were used as tests of T cell and B cell function. For the first 14 days following N. fowleri infection, Iymphoblastic transformation induced by T-cell mitogen was markedly reduced in comparison to the uninfected control mice. The blastogenic response to B-cell mitogen remained depressed in the infected mice up to 14 days after infection. The fluorescent antibody titers of sera of N. fowleri infected mice were between 1 : 4 and 1 : 32. The results suggest that there is a suppression of cell mediated immunity during the acute course of experimental Naegleria meningoencephalitis in mice.
