Immobilization and Characterization of a Liposome-Mediated Reconstituted Nicotinic Acetylcholine Receptor

  • A nicotinic acetylcholine receptor (nAchR) isolated from the electric tissues of Torpedo californica has been reconstituted into a vesicle comprising a bifunctional azo-ligand (Bae 1) compound, and a liposome containing phospholipids and cholesterol (1 : 1, w/w). The liposome-mediated reconstituted receptor showed a concentration-dependent response to cholinergic drugs in a lithium ion flux assay. This liposome-mediated reconstituted nAchR was immobilized onto an electrode using various synthetic polymers which were tested for their response to the cholinergic ligands. The immobilized nAchR not only exhibited a linear response to a wide range of cholinergic ligand concentrations but also retained an operational stability which lasted for longer than 6 days. Thus, this result provides a basis for application of the immobilized nAchR-based biosensor in detecting cholinergic ligands in vitro.

Overexpression of N -terminal lacking mutant HFR1 confers light-independence in a subset of photo-responses

  • Phytochrome controls diverse aspects of plant development in response to the ambient light conditions. HFRl, a basic helix-loop-helix protein, is required for a subset of phytochrome A (phy A)-mediated photo-responses in Arabidopsis. Here, we show that overexpression of HFR1-N105, but not the one of the full-length HFR1, confers exaggerated photo-responses. The transgenic plants overexpressing HFR1- N105 exhibited light-independence in a subset of photo-responses, including germination, de-etiolation, gravitropic hypocotyl growth, and blocking of greening. Overexpression of HFR1-N105 also caused constitutive light-responses in the expression of some light-regulated genes. In addition, the HFR1-N105 overexpressor showed hypersensitive responses under R and FR light, dependently on phyB and phyA, respectively. End-of-day far-red light response and petiole elongation were suppressed in the HFR1-N105 overexpressor plants. Together these results imply that overexpression of HFR1-N105 activated a branch of light signaling, supporting the hypothesis that transcriptional regulation in the nucleus would be the primary mechanism of light signaling in Arabidopsis. We discuss the biotechnological potential of the mutant bHLH protein, HFR1-N105 in regard to suppressed shade avoidance syndrome.

  • Polymeric electrodes for lead ion based on N,N'-bis-thiophen-2-ylmethylene-pyridine-2,6-diamine as an ion carrier were prepared. The membrane electrode (m-3) containing o-NPOE as a plasticizer and 50 mol% additive of ionophore gives an excellent Nernstian response (29.59 mV/decade) and the limit of detection of ?log a (M) = 5.74 to Pb2+ in Pb(NO3)2 solution at room temperature. The prepared electrode provided good sensitivity and outstanding selectivity and response for Pb2+ over a wide variety of other metal ions in pH 7.0 buffer solutions. The good sensitivity and selectivity towards lead ion are attributed to the strong complexation of lead ion to N,N'-bis-thiophen-2-ylmethylene-pyridine-2,6-diamine which has geometrically the proper cavity to coordinate to the ligand.

Immune-Enhancing Effect of Nanometric Lactobacillus plantarum nF1 (nLp-nF1) in a Mouse Model of Cyclophosphamide-Induced Immunosuppression

  • Nanometric Lactobacillus plantarum nF1 (nLp-nF1) is a biogenics consisting of dead L. plantarum cells pretreated with heat and a nanodispersion process. In this study, we investigated the immune-enhancing effects of nLp-nF1 in vivo and in vitro. To evaluate the immunostimulatory effects of nLp-nF1, mice immunosuppressed by cyclophosphamide (CPP) treatment were administered with nLp-nF1. As expected, CPP restricted the immune response of mice, whereas oral administration of nLp-nF1 significantly increased the total IgG in the serum, and cytokine production (interleukin-12 (IL-12) and tumor necrosis factor alpha (TNF-${\alpha}$)) in bone marrow cells. Furthermore, nLp-nF1 enhanced the production of splenic cytokines such as IL-12, TNF-${\alpha}$, and interferon gamma (IFN-${\gamma}$). In vitro, nLp-nF1 stimulated the immune response by enhancing the production of cytokines such as IL-12, TNF-${\alpha}$, and IFN-${\gamma}$. Moreover, nLp-nF1 given a food additive enhanced the immune responses when combined with various food materials in vitro. These results suggest that nLp-nF1 could be used to strengthen the immune system and recover normal immunity in people with a weak immune system, such as children, the elderly, and patients.

Evaluation of Design Response Spectrum in Sejong City Using Gyeongju and Pohang Type Seismic Waves (경주·포항형 지진파에 대한 세종시 지역의 설계응답스펙트럼 성능평가)

  • In the seismic design standard, input waves for different levels of seismic performance are proposed in the form of design response spectra. At the time of establishing these standards, measured records of significant earthquakes that occurred domestically, such as the 2016 Gyeongju earthquake and the 2017 Pohang earthquake, were not included. Additionally, for the ground response analysis, shear wave velocities representing ground amplification characteristics were derived from the results of standard penetration tests (N-values) and applied in empirical formulas. This approach may not adequately capture sufficient information about the characteristics of domestic ground properties. Therefore, in this study, seismic records from the Gyeongju and Pohang earthquakes were modified to adjust the bedrock standard design response spectra. Ground response analyses were conducted using shear wave velocity profiles obtained from borehole tests in the Sejong City area. The shape of the response spectrum and ground amplification coefficient obtained from the ground response analysis were then compared with those from existing studies and seismic design standard.

  • The response displacement method is the most frequently used method for seismic design of buried structures. This method is pseudo-static method, and the evaluations of velocity response spectrum of seismic base and response displacement of surrounding soil are the most important steps. In this study, the evaluation of velocity response spectrum of seismic base according to the Korean seismic design guide and the simple method of calculating the response displacement were studied. It was found that velocity response spectrum of seismic base can be estimated by direct integrating the ground-surface acceleration response spectrum of soil type $S_{A}$, and the evaluation of the response displacement using double cosine method assuming two layers of soil profile shows the advantages in the seismic design.n.

Growth and community response of phytoplankton by N, P and Fe nutrient addition in around water of Ulleungdo and Dokdo in East Sea (동해 울릉도-독도해역에서 질소, 인, 철 첨가실험에 따른 식물플랑크톤의 성장 및 군집반응)

  • We investigated phytoplankton and vertical inorganic nutrients at two stations around water of Ulleungdo and Dokdo in the East Sea, Korea. Nutrient addition experiments (+N, +P, +NP and +Fe) were also conducted to determine the growth response and nutrient consumption of the phytoplankton assemblage using the surface water of St. UD3 and St.50. In the field, although inorganic nutrients were low in the euphotic layer, these nutrients were increased at depths below 100 m. The total phytoplankton abundances in St. UD3 and St.50 were $4.9{\times}10^5cells\;L^{-1}$ and $1.9{\times}10^5cells\;L^{-1}$, respectively. The dominant species at St. UD3 was observed to be Raphidophyta Heterosigma akashiwo, Cryptophyta Crytomonas spp., and diatom Leptocylindrus danicus, while L. danicus and H. akashiwo including small diatom species Chaetoceros socialis were dominant at St. 50. In the nutrient addition experiments, phytoplankton growth (in vivo flourescence) in the +N and +NP treatment was 2-3 times higher than that in the +P treatment, indicating that the natural phytoplankton can respond to pulsed nutrient loading events. In addition, in vivo fluorescence in +Fe treatment was not statistically (p>0.05) different from that of the non-Fe treatments, indicating that the phytoplankton growth response in +Fe treatment was not significant. Dominant H akashiwo and L. danicus in the field showed a rapid response in nutrient additional bio-algal assay, particularly L. danicus in the +Fe treatments.

Preparation and characterization of Mn doped copper nitride films with high photocurrent response

  • The Mn-doped copper nitride ($Cu_3N$) films with Mn concentration of 2.0 at. % have high crystallinity and uniform surface morphology. We found that the as-synthesized Mn-doped $Cu_3N$ films show suitable optical absorption in the visible region and the band gap is ~1.48 eV. A simple photodetector based on Mn doped $Cu_3N$ films was firstly fabricated via magnetron sputtering method. The fabricated device with doping of Mn demonstrated high photocurrent response and fast response shorter than 0.1 s both for rise and decay time superior to the pure $Cu_3N$. Furthermore, the energy levels of Mn-doped Cu3N matched well with ITO and Ag electrode. The excellent photoelectric properties reflect a good balance between sensitivities and response rate. Our investigation reveals the excellent potential of Mn-doped $Cu_3N$ films for application of photodetectors.

Quantification of Serum Free RNA as a Predictive Biomarker for the Response to Chemotherapy in Patients with Lung Cancer: A Pilot Study

  • Background: It is well-known that cell-free nucleic acids rise in patients with many types of malignancies. Several recent experimental studies using cancer cell lines have shown that changes in cell-free RNA are predictive of the response to chemotherapy. The objective of this study was to determine whether quantification of free RNA can be used as a biomarker for clinical responses to chemotherapy in patients with lung cancer. Methods: Thirty-two patients with lung cancer (non-small cell lung cancer, n=24; small cell lung cancer, n=8) were divided into 2 groups according to their responses to chemotherapy (response group, n=19; non-response group, n=13). Blood samples were collected before and after two cycles of chemotherapy. Real-time quantitative RT-PCR was used for transcript quantification of the glyceraldehyde-3-phosphate dehydrogenase gene. Results: The pre chemotherapy values (Response group $41.36{\pm}1.72$ vs. Non-response group $41.33{\pm}1.54$, p=0.78) and post chemotherapy values (Response group $39.92{\pm}1.81$ vs. Non-response group $40.41{\pm}1.47$, p=0.40) for cell free RNA concentrations, expressed as Ct GAPDH (threshold cycle glyceraldehyde-3-phosphate dehydrogenase gene) levels, was not different between the two groups. There was no significant relationship between changes in the cell free RNA level clinical responses after chemotherapy (p=0.43). Conclusion: We did not find a correlation between quantification of serum cell free RNA levels and clinical responses to chemotherapy in patients with lung cancer. Further investigations are needed to determine whether the cell free RNA level is a useful predictor of responses to chemotherapy in patients with lung cancer.

Optoelectronics based on 2D semiconductor heterostructures

  • Van der Waals (vdW) heterostructures built from two-dimensional layered materials provide an unprecedented opportunity in designing new material systems because the lack of dangling bonds on the vdW surfaces enables the creation of high-quality heterointerfaces without the constraint of atomically precise commensurability. In particular, the ability to build artificial heterostructures, combined with the recent advent of transition metal dichalcogenides, allows the fabrication of unique semiconductor heterostructures in an ultimate thickness limit for fundamental studies as well as novel device applications. In this talk, we will present the characterization of the electronic and optoelectronic properties of atomically thin p-n junctions consisting of vertically stacked WSe2 and MoS2 monolayers. We observed gate-tunable diode-like current rectification and a photovoltaic response across the p-n interface. Unlike conventional bulk p-n junctions, the tunneling-assisted interlayer recombination of the majority carriers is responsible for the tenability of the charge transport and the photovoltaic response. Furthermore, we will discuss the enhanced optoelectronic characteristics in graphene-sandwiched vdW p-n junctions.
