  • This study was carried out in order to develop a compound fertilizer for raising rice seedling in trays. A compound fertilizer, a trail product, was manufactured using the major fertilizer sources of ammonium sulfate, diammonium phosphate and potassium chloride in combination with zeolite and glutamic acid fermentation waste. Besides, polyacrylamide for slow release control of the fertilzer and Tachigaren and sulfuric acid for reducing the occurrence of seedling rot were used. The component ratios of N, $P_2O_5$ and $K_2O$ of trial product were 4.19, 5.41 and 4.05 percent respectively. The dissolution rate of nitrogen component in water for the trial product with polyacrylamide was lower about fifteen percents than the product without polyacylamide in six hours. Hymexazole, main component of Thachigaren, from the product was released about 86.2 percents in forty eight hours. When the product with polyacylamide applied on red earth soil and paddy soil, the pH of soil ranged from 4.6 to 5.4 for 25 days experiment.

Natural killer cell activity in Naegzeria fowleri infected mice (Naegleria fowleri 자연살세포의 활성에 미치는 영향)

  • The natural killer (NK) cell activity of splenocytes and recycling capacity of NK cells were observed by combining the $^{51}Cr-release$ cytotoxicity assay and single cell cytotoxicity assay against YAC-1. The ICR mice were infected intranasally with Naegleria fewleri, that is a pathogenic free-living amoeba. The mice infected with $1{\times}10^5$ trophosoites showed mortality rate of 76.7% and mean survival time of 12. 9 days. The cytotoxic activity of NK cells in infected mice was significantly higher than that of non-infected mice during the period between 12 hours and day 3 after infection, and highest on day 1. The target-binding capacity of NK cells in infected mice was not different from that of non-infected ones. Maximal killing potential and maximal recycling capacity were remarkably increased in infected mice as compared with the control. The results obtained in this observation indicated that elevated NK cell activity in mice infected with N. fowieri was not due to target-binding capacity of NK cells but due to the increased activity of NK cells and increased recycling capacity of individual NK cells.

  • In this work, the effect of PMR-15 content on the variation of surface free energy of the DGEBA/PMR-15 blend system was investigated in terms of contact angles and mechanical interfacial tests. Based on FT-IR result of the blend system. C=O (1,772, $1,778cm^{-1}$) and C-N ($1,372cm^{-1}$) peaks appeared with imidization of PMR-15 and -OH ($3,500cm^{-1}$) peak showed broadly at 10 phr of PMR-15 by ring-opening of epoxy. Contact angle measurements were performed by using deionized water and diiodomethane as testing liquids. As a result, the surface free energy of the blends gave a maximum value at 10 phr of PMR-15, due to the significant increasing of specific component. The mechanical interfacial properties measured from the critical stress intensity factor ($K_{IC}$) and the critical strain energy release rate ($G_{IC}$) showed a similar behavior with the results of surface energetics. This behavior was probably attributed to The improving of the interfacial adhesion between intermolecules, resulting from increasing the hydrogen bondings of the blends.

Effects of Non-ionic Surfactants on Enzyme Distributions of Rumen Contents, Anaerobic Growth of Rumen Microbes, Rumen Fermentation Characteristics and Performances of Lactating Cows

  • A series of experiments was carried out to determine the possibility for the non-ionic surfactant (NIS) as a feed additive for ruminant animals. The effect of the NIS on (1) the enzyme distribution in the rumen fluids of Hereford bulls, (2) the growth of pure culture of rumen bacteria and (3) rumen anaerobic fungi, (4) the ruminal fermentation characteristics of Korean native cattle (Hanwoo), and (5) the performances of Holstein dairy cows were investigated. When NIS was added to rumen fluid at the level of 0.05 and 0.1% (v/v), the total and specific activities of cell-free enzymes were significantly (p<0.01) increased, but those of cell-bound enzymes were slightly decreased, but not statistically significant. The growth rates of ruminal noncellulolytic species (Ruminobacter amylophilus, Megasphaera elsdenii, Prevotella ruminicola and Selenomonas ruminantium) were significantly (p<0.01) increased by the addition of NIS at both concentrations tested. However, the growth rate of ruminal cellulolytic bacteria (Fibrobacter succinogenes, Ruminococcus albus, Ruminococcus flavefaciens and Butyrivibrio fibrisolvens) were slightly increased or not affected by the NIS. In general, NIS appears to effect Gram-negative bacteria more than Gram-positive bacteria; and non-cellulolytic bacteria more than cellulolytic bacteria. The growth rates of ruminal monocentric fungi (Neocallimastix patriciarum and Piromyces communis) and polycentric fungi (Orpinomyces joyonii and Anaeromyces mucronatus) were also significantly (p<0.01) increased by the addition of NIS at all concentrations tested. When NIS was administrated to the rumen of Hanwoo, Total VFA and ammonia-N concentrations, the microbial cell growth rate, CMCase and xylanase activities in the rumen increased with statistical difference (p<0.01), but NIS administration did not affect at the time of 0 and 9 h post-feeding. Addition of NIS to TMR resulted in increased TMR intake and increased milk production by Holstein cows and decreased body condition scores. The NEFA and corticoid concentrations in the blood were lowered by the addition of NIS. These results indicated that the addition of NIS may greatly stimulate the release of some kinds of enzymes from microbial cells, and stimulate the growth rates of a range of anaerobic ruminal microorganisms, and also stimulate the rumen fermentation characteristics and animal performances. Our data indicates potential uses of the NIS as a feed additive for ruminant animals.

Evaluation of the Burning Properties of Various Carpet Samples by using the Cone Calorimeter and Gas Toxicity Test (콘칼로리미터와 가스유해성 시험법을 이용한 카페트류의 연소특성 평가)

  • In this study, the burning behaviours of five different kinds of carpet samples covered with nylon, PP (polypropylene), PTT (poly(trimethylene terephthalate)), wool fabric and NW (nylon and wool) were evaluated by using the cone calorimeter having a radiant flux of 50kW/$m^2$. And the combustion gas toxicity was evaluated according to KS F 2271 test method. As a result of the cone calorimeter test (KS F ISO 5660-1), nylon carpet samples were ignited most easily. In ignition ability or initial flammability, NW carpet samples showed the highest value. In heat release rate (HRR), fire intensity, PP carpet samples were larger than any other samples. Nylon carpet samples were the highest smoke production rate, while N/W carpet samples the lowest. The following were in mass loss rates: NW > wool > nylon > PP > PTT. CO (carbon monoxide) was one of the most toxic gases released from the combustion. PTT carpet samples gave rise to the highest CO concentration, while NW carpet samples the lowest. In addition, PP carpet samples caused the highest $CO_2$ (carbon dioxide) concentration, while NW carpet samples the lowest. Toxicity of the gas produced from carpet samples was determined by the mouse stop motion, and it resulted in the fact that the combustion gas of PTT carpet samples was more toxic than that of any other samples.

Cyanobacterial Development and Succession and Affecting Factors in a Eutrophic Reservoir (부영양 저수지에서 남조류의 발달과 천이 및 영향 요인)

  • This study was conducted to evaluate the causes and effects of cyanobacterial development and succession in a shallow eutrophic reservoir from March 2003 to February 2004. Phytoplankton succession, sedimentation rate, and sediment composition were analyzed. Algal bioassay also was conducted with the consideration of light, water temperature and nutrients. Cyanobacteria dominated throughout the year, except for spring season (March${\sim}$April) in which diatoms and flagellates dominated. Total cell density increased in July and November when P loading through inflows was high. Oscillatoria spp. and Aphanizomenon sp. were dominant in May and June, respectively, but replaced with Microcystis spp. in July. Thereafter, Microcystis spp. sustained until December, and again shifted to Oscillatoria spp. and Aphanizomenon sp. The dominance of Oscillatoria spp. in May was accompanied with high TN/TP ratio and the increase of water temperature and light intensity. While the dominance of Microcystis spp. was related with relatively low TN/TP ratio, ranging from 46 to 13 (average: 27). The sedimentation rate was highest in March (0.6 m $day^{-1}$) when diatoms dominated. During the period of cyanobacterial dominance, relatively high sedimentation rate was observed in May (0.4 m $day^{-1}$) and October (0.36m $day^{-1}$). C/N ratio of the sediment ranged $6{\sim}8$. Inorganic P concentration in the pore water was low when DO concentration was < 2 mg $O_2$ $L^{-1}$ in the hypolimnion, reflecting the P release from the sediment. Cyanobacterial growth rate depended on phosphorus concentration and water temperature, and high P concentration compensated for the low temperature in the growth rate. Our results suggest that the potential of cyanobacterial development and substantiality in eutrophic reservoirs be high throughout the year, as being supplied with enough P, and emphasize the consideration of sediment man. agement for the water quality improvement and algal bloom control.

Relationship of Early Lactation and Bovine Somatotropin to Water Metabolism and Mammary Circulation of Crossbred Holstein Cattle

  • The study was carried out to evaluate the effect of exogenous bovine somatotropin on water metabolism in relation to mammary function in early lactation of crossbred Holstein cattle. Ten, 87.5% crossbred Holstein cattle were divided into two groups of 5 animals each. At day 60 of lactation, the control group was given placebo while animals in the experimental group were given recombinant bovine somatotropin (rbST) by subcutaneous injection with 500 mg of rbST (14-days prolonged-release rbST). In rbSTtreated animals, milk yield increased 19.8%, which coincided with a significant increase in water intake (p<0.01), while DM daily intake was not different when compared to the control animals. Water turnover rate as absolute values significantly increased (p<0.05), while the biological half-life of water did not change in rbST-treated animals. Total body water (TBW) and total body water space (TOH) as absolute values significantly increased (p<0.01) in rbST-treated animals, while it was decreased in the control animals. Absolute values of empty body water (EBW) markedly increased (p<0.05), which was associated with an increase in the extracellular fluid (ECF) volume. Absolute values of plasma volume and blood volume were also significantly increased (p<0.05) in rbST-treated animals. The increase in mammary blood flow in rbST-treated animals was proportionally higher than an increase in milk production. The plasma IGF-1 concentration was significantly increased (p<0.01) in rbST-treated animals when compared with those of control animals during the treatment period. Milk fat concentration increased during rbST treatment, while the concentrations of both protein and lactose in milk were not affected. The present results indicate that rbST exerts its effect on an increase in both TBW and EBW. An increased ECF compartment in rbST-treated animals might partly result from the decrease in fat mass during early lactation. The action of rbST on mammary blood flow might not be mediated solely by the action of IGF-1 for increase in blood flow to mammary gland. An elevation of body fluid during rbST treatment in early lactation may be partly a result of an increase in mammary blood flow in distribution of milk precursors to the gland.

In Sacco Evaluation of Rumen Protein Degradation Characteristics and In vitro Enzyme Digestibility of Dry Roasted Whole Lupin Seeds (Lupinus albus)

  • The effects of dry roasting whole lupin seeds (lupinus albus, WLS) at 110, 130 or $150{^{\circ}C}$ for 15, 30 or 45 minutes on the in sacco rumen degradation characteristics, optimal heating conditions of time and temperature and in vitro enzyme digestibility were determined. Ruminant degradation characteristics (RDC) of crude protein (CP) of WLS were determined by in sacco technique in dairy cows. Measure ROC were soluble (S), undegradable (U), potentially degradable (D) fractions, lag time (TO) and rate of degradation (Kd) of insoluble but degradable fraction. Based on measured ROC, percentage bypass CP (%BCP) and bypass CP (BCP in g/kg, DM) were calculated. Degradability of CP was significantly reduced by dry roasting (p<0.001). The interaction of dry roasting temperature and time had significant effects on D (p<0.05), Kd (p<0.01), U (p<0.01), %BCP (p<0.001) and BCP (p<0.001) but not on S (p=0.923>0.05). With increasing time and temperature, S, D, Kd and U varied from 31.8%, 67.4%, 10.3%/h and 0.8% in the raw WLS (RWLS) to 27.1 %, 35.8%, 3.6%/h, 38.4% in $150{^{\circ}C}/45\;min$, respectively. All these effects resulted in increasing %BCP from 25.9 in RWLS to 61.0% in the $150{^{\circ}C}/45\;min$. Therefore BCP increased form 111.2 to 261.2 g/kg DM, respectively. Both %BCP and BCP at $150{^{\circ}C}/45\;min$ increased nearly 2.5 times over the RWLS. The effects of dry roasting on %BCP and BCP seemed to be linear up to the highest value tested. Although ROC had been altered by dry roasting, the In vitro perpsin-cellulase digestibility was generally unchanged. It was concluded that dry roasting was effective in shifting CP degradation from rumen to the lower gastrointestinal tract to potential reduce unnecessary N loss in the rumen. It might be of great value in successfully synchronizing the rhythms of release of nitrogen and energy in the rumen, thus achieving a more efficient fermentation of diets with high proportions of lignocellulosic resources. To determine the optimal dry roasting conditions, the digestibility of each treatment in the cows will be measured in the next trial using mobile bags technique.

Nitrogen Removal in Column Wetlands Packed with Synthetic Fiber Treating Piggery Stormwater (축산단지 강우 유출수 처리를 위한 합성섬유충진 습지의 질소제거에 관한 연구)

  • A set of lab-scale polymer synthetic fiber packed column wetlands composing three columns (CW1, CW2 and CW3) with different hydraulic regimes, recirculation frequencies and pollutant loading rates, were operated in 2012. Synthetic fiber tested as an alternative wetland medium for soil mixture or gravel which has been widely used, has very high pore size and volume, so that clogging opportunity can be greatly avoided. The inflow to the wetland was artificial stormwater. All the wetlands achieved effective removal of TSS (94%~96%), TCOD (68%~73%), TN (35%~58%), TKN (62%~73%) and NH4-N (85%~ 99%). Particularly, it was observed that COD was released from the fiber during one distinct period in all wetlands. This was probably due to the degradation of polymer fiber, and the released organic matters were found to serve as carbon source for denitrification. In addition, with longer retention time and frequent recirculation, lower effluent concentration was observed. With higher pollutant loading rate, higher nitrification and denitrification rates were achieved. However, although organic matters were released from the fiber, the lack of carbon source was still the limiting factor for the system since the release persisted only for 40 days.

Effects of Green Manure on Soil Properties and Grain Yield of Sorghum (Sorghum bicolor Moench) (수수 재배 시 풋거름작물 이용이 토양특성 변화와 수수의 수량에 미치는 영향)

  • Green manure has been used as alternative to chemical fertilizer. To evaluate the effect of green manure on the chemical properties of top-soil and sorghum yield, hairy vetch (Vicia villosa Roth, HV), manure barley (MB), and a mixture of hairy vetch and manure barley (HV+MB) were incorporated into the soil at a rate of $100kg-N\;N\;ha-1$ before the sorghum was transplanted. Total biomass of sorghum grown in the HV, MB, and HV+MB treatments was 13.1, 31.6, and $25.2t\;ha^{-1}$, respectively, and the nitrogen production of the treatments was 81, 74, and $145kg\;ha^{-1}$, respectively. The SPAD value of the uppermost leaf of sorghum plants grown in the soils with HV, MB, or HV+MB were very similar until heading stage; however, at maturity, the SPAD value of sorghum cultivated in the soils with HV was lower than that of sorghum in the soils with MB or HV+MB. This could be because the nitrogen release from HV was too rapid to supply nitrogen to sorghum during the later stage of grain filling. Compared with chemical fertilizers, the incorporation of green manure increased the pH, exchangeable cations ($K^+$, $Mg{^{+}^{+}}$, and $Ca{^{+}^{+}}$), and total nitrogen in soil postharvest, indicating an improvement in soil chemical properties. Total carbon content increased in soil with green manure incorporated, but decreased in the chemical fertilized soil, suggesting that sorghum cultivation using green manure may sequester carbon in soils. The yield of sorghum cultivated with green manure was not different from the yield of sorghum cultivated with chemical fertilizers. These results suggest that the mixture of hairy vetch and manure barley can be a useful chemical fertilizer alternative in sorghum cultivation.
