Influence of Nitrite and Ascorbic acid on N-Nitrosamine Formation during the Fermentation of Salt-fermented Small Shrimp (새우젓 숙성중 아질산염과 아스코르브산이 N-Nitrosamine의 생성에 미치는 영향)

  • The changes of contents of trimethylamine oxide nitrogen (TMAO-N), trimethylamine nitrogen (TMA-N), dimethylamine nitrogen (DMA-N), nitrite nitrogen (nitrite-N), nitrate nitrogen (nitrate-N) and N-nitrosamine (NA) of salt-fermented small shrimp were investigated during fermentation. The contents of TMAO-N decreased, while TMA-N and DMA-N increased during fermentation in all samples. Contents of nitrite-N decreased in the samples supplemented with sodium nitrite during fermentation, whereas the formation of N-nitrosodimethylamine (NDMA) increased. Treatment of ascorbic acid revealed inhibiting effort on NDMA formation compared with the control. The model system was used for the evaluation of ascorbic acid (inhibitor) or thiocyanate (promoter) on the formation of NDMA using salt-fermented small shrimp supplemented with sodium nitrite, The optimum pH for the formation of NDMA was 3.5, and ascorbic acid inhibited the formation of NDMA whereas thiocyanate promoted.

  • The changes of contents in trimethylamine oxide nitrogen(TMAO-N), trimethylamine nitrogen(TMA-N), dimethylamine nitrogen(DMA-N), nitrite nitrogen(nitrite-N), nitrate nitrogen(nitrate-N) and the effect on the formation of N-nitrosamine(NA) during fermentation were investigated with salted anchovy added different amounts of sodium nitrite, sodium nitrate and ascorbic acid, respectively. When the sodium nitrite was added in salted anchovy, the contents of nitrite-N was decreased during fermentation . Whereas the formation of N-nitrosodimethylamine(NDMA ) was increased . Contents of TMAO-N was decreased, while TMA-N and DMA-N were increased during fermentation in all samples. Addition of ascorbic acid inhibited the formation of NDMA significantly. The formation of NDMA was inhibited by 81.3% at the concentration of 130mM as compared with non-added the control group. The aqueous model system was used for the evaluation of ascorbic acid(inhibitor) or thiocyanate (promoter) on the formation of NDMA using salt-fermented anchovy added with sodium nitrite. The optimum pH on the formation of NDMA was shown to be 3.8, and ascorbic acid inhibited the formation of NDMA whereas thiocyanate promoted. NDMA was not detected in the salt-fermented anchovy (control sample). However it is a possibility to form carcinogenic NDMA in stomach if both saltfer-mented anchovy and the materials contained abundant nitrite or nitrate were took in.

Studies on Tip-burn of Chinese Cabbage by Ammonium Toxicity (암모니움 Toxicity에 의(依)한 배추의 Tip-burn에 관(關)한 연구(硏究))

  • The effect of N forms ($NO_3{^-}-N$, $NH_4{^+}-N$) and concentrations (4, 8, $16meq/{\ell}$) which were changed at head formation stage on the tip-burn incidence of chinese cabbage was studied under the three levels (0, 8, 16 meq/l) of Ca. All of the plants grown on $NH_4{^+}-N$ showed the symptoms of tip-burn and low yields regardless of Ca levels. Roots of plants grown on $NH_4{^+}-N$ were severely damaged. The pH of the leachate from $NH_4{^+}-N$ pot was decreased to below pH 5. Plants which had been grown on $NH_4{^+}-N$ before the head formation stage, but changed to $NO_3{^-}-N$ were recovered from abnormal growth. But, the reverse showed the tip-burn symptoms. $NH_4{^+}-N$ treatments increased the T-N contents, but lowered K and Ca contents of inner leafblades. Icreased applications of Ca did not affect the T-N and K contents of the inner leafblades. $NH_4{^+}-N$ suppressed Ca translocation into the inner leaves from outer leaves after the head formation stage, but $NO_3{^-}-N$ accelarated it. Ca contents were much lower in leaves showing tip-burn symptoms than in healthy leaves. Internal rot which is a tip-burn symptom occuring after head formation were noted in plants applied with high concentration of $NO_3{^-}-N$ both before and after the head formation stage. Ca contents correlated well with the rates of Ca application, but there was no correlation between ca conents and internal rot incidence. Chinese cabbage tip-burn is not caused by Ca deficiency, as is commonly believed, but rather by the water stress (95% water contents in $NO_3{^-}$-fed plants, 91% in $NH_4{^+}$-fed plants) resulting from root damage caused by ammonium toxicity. Internal rot is also caused by excess applications of nitrogen, and is unaffected by calcium levels.

  • 인플루엔자 A 바이러스의 아형인 H5N1은 고병원성으로 조류 독감을 일으킨다. H5N1 바이러스는 원래 조류끼리만 감염되는 독감이고, 사람에게는 전염되지 않는다고 알려져 있었으나, 2003년에 베트남과 중국을 시작으로 현재까지 168명의 사망자가 기록되고 있다. 그러나 현재 시판되고 있는 진단키트(Rapid diagnostic kits)들은 H5N1 에 특이적인 것이 아니라 influenza A virus 모두를 진단한다. 따라서 influenza 감염여부는 확인 할 수 있지만, 이것이 H5N1 인지는 확인 할 수가 없다. H5N1은 전염성이 강하기 때문에 빠르게 진단하여 감염조류를 살 처분 하여야 더 많은 경제적 손실을 줄일 수 있다. 따라서 H5N1 에만 특이적인 epitope를 네트워크 기반으로 예측하여 진단제에 응용할 수 있도록 하고자 한다. 각 서열 정보는 Openflu (http://openflu. vital-it.ch/browse.php)에서 얻었다. H5N1은 H1N1에서 유래되었기 때문에 두 subtype의 차이점을 알아보고자 TCOFFEE에서 multiple sequence alignment를 수행한 결과 N-terminal 부분이 상이하였다. 상이한 H5N1의 N-terminal 부분이 H5N1 virus에 감염된 모든 host에서 존재하는지 알아보기 위해 host가 사람인 경우와 조류인 경우를 TCOFFEE에서 alignment 하였다. 그 결과 H5N1의 N-terminal 부분은 사람과 조류에서 보존적이었다. 따라서 H5N1의 N-terminal이 다른 subtype과 유사하지 않고 H5에만 특이적이기 때문에 진단키트 제작을 위한 epitope로 사용할 수 있을 것으로 기대된다.

Studies on the Denitrification in the Submerged Paddy Soil -IV.   • A series of laboratory experiments were carried out to find the effects of soil organic matter contents, soil temperature, pH values, kinds and amount of nitrogen fertilizers on the denitrification-$N_2O$ gas evolution-. The results obtained were summarized as follows: 1. Denitrification rate, amount of $N_2O$ gas evolution, was influenced the order of organic matter contents>soil temperature>pH values>kinds of N-fertilizer>levels of N-fertilizer. 2. The highest dentrification rate was observed in organic matter content of 3.0%, pH values at 6.0 with application of $KNO_3$ at levels of 20 mgN/100g soil. 3. For the evolution of I mole $N_2O$ gas, averaged carbon consumption was obtained as 0.5 mole in all these experiment condition. However, the highest carbon consumption rate was obtained in organic matter contents for 1.0% with application of $(NH_4)_2SO_4$ at levels of 10 mgN/100g soil (1.06 mole) while lowest carbon consumption rate was obtained in organic matter contents for 3.0% with application of $KNO_3$ at levels of 20 mgN/100g soil (0.13 mole). 4. According to Michaelis-Menten's equation, the V/2 values for evolution of $N_2O$ gas was estimated by progress curve. The results obtained was as 550 ug for $(NH_2)_2CO$ and 1100 ug $N_2O/100g$ soil by application of $KNO_3$ in organic matter contents of 1.0% soil. On the other hand, when the application $(NH_4)_2SO_4$ the V/2 values of $N_2O$ gas was obtained as the amount of 490 ug/100g soil while V/2 values of $N_2O$ gas by application of $KNO_3$ was on the linear line in soil organic matter contents of 3.0%.

Selenium Content of Infant Formulas and Estimated Intake of Infants in Japan and Korea

  • Selenium contents of infant formulas that were commerically available in Japan and Korean were determined by the method of atomic absorption spectrometry with hydrode generation. Total means of selenium contents were 45.2$\pm$22.1ng/g(n=34) in all the Japaneses formulas and 58.3$\pm$18.9ng/g(n=22) in all the Korean formulas. selecnium contents of milk-based formulas (54.7$\pm$23.2ng/g in Japan, n=20 and 64.8$\pm$12.6ng/g in Korean , n=18) were significantly higher than those of soy-based formulas (36.8$\pm$11.0ng/g in Japanes brands, n=11 and 36.4ng/g in Korean one , N=1). Based on selenium data of these formulas the selenium intakes of infants have been estimated to be about 6.6$\mu\textrm{g}$/day and 8.1$\mu\textrm{g}$/day in Korean by milk-based formula feeding.

Changes of Nitric Oxide Synthase Activity and Free Methylarginines Contents in Regenerating Rat Liver after Partial Hepatectomy

  • In the present study, liver regeneration rate (%) was increased up to 70% 3 days after partial hepatectomy (PH). Nitric oxide synthase (NOS) activity in liver tissue as well as serum nitrite/ nitrate content had no timed response, revealing no significant difference between shamoperated and partially hepatectomized rat liver. Contents of free methylarginines in liver tissue were increased biphasically in a time-dependent manner after PH. However, those in serum did not exhibit the same patterns as in liver. Taken together, the results suggest that $N^{G}$-monomethyl-L-arginine (MMA) and $ N^{G}, N^{G}$-dimethylarginine (DMA) play a role in inhibiting nitric oxide (NO) synthesis in regenerating rat liver because the increase of their contents was synchronized with NOS expression.

Evaluation of Micro End-Milling Characteristics of AlN-hBN Composites Sintered by Hot-Pressing (열간가압소결에 의해 제조된 AlN-hBN 복합재료의 마이크로 엔드밀링 가공특성 평가)

  • The objective of this study is to evaluate various machining characteristics of AlN-hBN machinable ceramics in micro end-milling process for its further application. First, AlN based machinable ceramics with hBN contents in the range of 10 to 20vol% were prepared by hot-pressing. Material properties of the composites, such as relative density, Vickers hardness, flexural strength, Young's modulus and fracture toughness were measured and compared. Then, micro end-milling experiments were performed to fabricate micro channels using prepared system. During the process, cutting forces, vibrations and AE signals were measured and analyzed using applied sensor system. Machined micro channel shapes and surface roughness were measured using 3D non-contact type surface profiler. From the experimental results, it can be observed that the cutting forces, vibrations and AE signal amplitudes decreased with increasing hBN contents. Also, measured surface roughness and profiles were improved with increasing hBN contents. As a result of this study, optimum machining conditions can be determined to fabricate desired products with AlN-hBN machinable ceramics based on the experimental results of this research.

Characteristics in Chemical Properties of Agricultural Groundwater in Gyeongnam Province (경남지역 농업용 지하수의 수질특성)

  • This survey was conducted to obtain basic data of the quality of groundwater for agriculture in Gyeongnam province. Groundwater samples from paddy 15, upland 15, and plastic film house 30 sites were collected on April, July, and October in every two years from 2002 to 2008. According to the result of water quality analysis, groundwater quality was suitable for irrigation purpose averagely. The $NO_3$-N contents by land use were in the order of plastic film house > upland > paddy field and its contents were 6.53, 4.80, and $3.68mg\;L^{-1}$, respectively. In annual changes of water quality, pH was no significant change in paddy, upland, and plastic film house by 6.6~6.9. EC was increased in upland and plastic film house in 2008 and majors factors were $NO_3$-N and $Cl^-$. In upland and plastic film house, $NO_3$-N contents were 4.72 and $6.52mg\;L^{-1}$ in 2002, respectively, whereas they were 5.63 and $8.70mg\;L^{-1}$ in 2008, respectively. Of the investigated sites, $NO_3$-N was exceeded water quality standards for agriculture by 3.3~15.0% in plastic film house and $Cl^-$ was exceeded water quality standards for agriculture by 2.2% in upland of 2004. The $NO_3$-N contents were decreased with well depth and their contents were $5.38mg\;L^{-1}$ from 3~10 m, $4.87mg\;L^{-1}$ from 10~20 m, and $2.58mg\;L^{-1}$ from above 30 m. The $NO_3$-N contents by soil texture were highest in sandy loam by $5.73mg\;L^{-1}$ and lowest in clay loam by $4.13mg\;L^{-1}$. The $NO_3$-N contents by crops category were in order of fruit vegetables > leaf vegetables > rice > fruits > beans, contents of fruit vegetables and leaf vegetables were 5.81 and $5.30mg\;L^{-1}$, respectively.
