Mycological Characteristics of Phytophthora nicotianae var. nicotianae Causing Phytophthora Rot of Strawberry and Resistance of Strawberry Cultivars to the Pathogen (딸기 역병균 Phytophthora nicotianae var. nicotianae의 균학적 특성 및 딸기 품종간 저항성)

  • Mycological characteristics of Phytophthora nicotianae var. nicotianae SPC10 (A1 type) causing Phytophthora rot of strawberry and the resistances of 11 strawberry cultivars against the pathogen were examined. Optimum temperature for the mycelial growth of the pathogen was obtained in the range of 30~35$^{\circ}C$, and the growth was completely stopped under 13$^{\circ}C$ or over 42$^{\circ}C$. Aerial mycelia were abundant on oatmeal agar (OMA), V-8 juice agar (V8A) and lima bean agar (LBA) medium, although there were slight differences, however, on cornmeal agar (CMA) medium, it was a shape of stellate without aerial mycelia. The colony shape on potato dextrose agar (PDA) medium was rough and irregular whereas the mycelial growth was slow, and some aerial mycelia were only produced in the middle of PDA medium. Optimum temperature for sporangial formation was 3$0^{\circ}C$, and zoospores were mostly released at $25^{\circ}C$ from the sporangia. Sporangia were more produced in C/Z solution with pH 5. 0~6.$0^{\circ}C$ than sterilized distilled water (DSW) and distilled water (DW), and zoospores were also released much more than other solutions. Eleven strawberry cultivars such as Reiko, Hokowase, Eyeberry, Akaneko, Sistakara, Toyonoka, Nyoho, Sulhong, Suhong, Myhong and Wonkyo #3104 revealed the disease incidence up to 88.9~100% by the leaf inoculation with mycelial disk. However, Nyoho and Suhong showed higher level of resistance against the pathogen by root inoculation.

Occurrence of Anthracnose on Chinese Mallow Caused by Colletotrichum malvarum

  • Anthracnose symptoms were frequently observed on leaves, petioles, and stems of Chinese mallow grown in Namyangju, Korea, during a disease survey performed in November, 2007. The disease incidence was as high as 30% in the 12 greenhouses investigated. A total of 38 isolates of the Colletotrichum species were obtained from the anthracnose symptoms, and all the isolates were identified as Colletotrichum malvarum based on their morphological and culture characteristics. Three isolates of the fungus caused anthracnose symptoms on leaves and stems following artificial inoculation, which were similar to those observed during the greenhouse survey. In this study, mycological and pathological characteristics of C. malvarum identified as causing anthracnose of Chinese mallow were clarified.

  • One fungus was isolated from lesions on imported chinese cabbage leaves in process of quarantine inspection from China. The fungus was identified as Mycosphaerlla brassicicola, based on morphology of perithecia, asci, ascospore, and curtural characteristics. In Korea, this fungus has been quarantine fungus, and not yet report to occur. Perithecia of the fungus were globose, dark brown with apical papilate ostioles. The size was 90-100 ${\times}$ 130-135$\mu\textrm{m}$. Asci were bitunicate, 8-spored and 38-43 ${\times}$ 15-l9$\mu\textrm{m}$. Ascospore were irregularly biseriate, hyaline, cylindrical, 2-celled, and rounded at the ends. Optium growth temperature of the fungus was at 20$^{\circ}C$ on PDA but did rarely grow over 24$^{\circ}C$. Colony on PDA was of black aerial mycelia.

Occurrence of Anthracnose on Welsh Onion Caused by Colletotrichum circinans

  • Anthracnose occurred frequently on leaf sheaths of Welsh onions grown in Gangwha island, Korea in November, 2007. The disease incidence was as high as 30% in five fields investigated. A total of 20 single spore isolates of Colletotrichum species were obtained from the affected plants, and all the isolates were identified as Colletotrichum circinans based on their morphological and cultural characteristics. Three isolates of the fungus caused anthracnose symptoms on the leaf sheaths of Welsh onions by artificial inoculation, which were similar to those observed during the field survey. In this study, the mycological and pathological characteristics of C. circinans identified as causing anthracnose of Welsh onions are clarified.

Four Species of Montagnulaceae Unrecorded in Korea and Isolated from Plant Litter in Freshwater

  • Freshwater is a diverse and complex environment for fungi and provides several types of habitat including plant litter, sediment, and carcasses. We collected plant litter from the main stream and branch streams of Nakdong River, Sohancheon in Samcheok, and Geumoreum in Jeju. From several samples of plant litter, we isolated 8 fungal strains belonging to 4 Montagnulaceae species unrecorded in Korea: Paraconiothyrium archidendri, Paraphaeosphaeria sporulosa, Paraphaeosphaeria michotii, and Paraphaeosphaeria viridescens. These fungi were identified by phylogenetic analysis using the internal transcribed spacer (ITS) region and examination of morphological characteristics. Montagnulaceae is known as one of the families in Pleosporales and includes coniothyrium-like fungi. In this study, we described phylogenetic analysis and mycological characteristics of these species, and this is the first report of these taxa in Korea.

Mycological Features of Trichophyton verrucosum Isolated in Cattle

  • In this study, the author observed the fungal characteristics of T. verrucosum which is commonly known as the cattle ringworm fungus in the farms of Hoengseong, Gangwon-do. After isolating 20 strains of T. verrucosum from cattle, they were cultured on SDA, PDA media and the fungal characteristics were concluded through visual observation of the colonies, microscopic findings, hair perforation test and urease test. The size of the colonies cultured on SDA media at 37 C was on an average 5 mm at 1 week, 33 mm at 2 weeks, 42 mm at 3 weeks and 58 mm after 4 weeks. Observing the characteristics of the colonies, 17 strains of T. album showing central bold radial folds, 2 strains of T. ochraceum having l throughout the colonies and 1 strain of T. discoides with rapid growth rate and gray-white cotton patterns were found. On microscopic observations of the cultured colonies on SDA, PDA media, macroconidia and microconidia were not found in T. verrucosum and hyphae and chlamydospore were only seen in T. album types. Out of 20 strains of T. verrucosum, hair perforation test was positive on only 3 strains and urease test was positive on all of the 20 strains.

Spot Anthracnose Disease Caused by $Colletotrichum$ $gloeosporioides$ on Tulip Tree in Korea

  • The tulip tree ($Liriodendron$ $chinense$) has been widely cultivated in Korea as a street or garden tree for its large flowers, which have a superficial resemblance to tulips. Occurrence of anthracnose disease on the leaves of tulip trees growing on the campus of Gyeongsang National University, Jinju, Korea, has been observed. Based on mycological characteristics, pathogenicity, and internal transcribed spacer sequence, the causal fungus was identified as $Colletotrichum$ $gloeosporioides$. This is the first report on anthracnose disease caused by $C.$ $gloeosporioides$ on tulip trees in Korea.

Downy Mildew of Astragalus membranaceous Burge Caused by Peronospora trifoliorum de Bary

  • A severe down mildew of Astragalus membranaceus occurred in Gangwon province in 1999 and 2000. Symptoms developed on young leaves and shoots, showing grayish white mycelium on the lower leaves. The infected plants had reduced internodes and twisted leaflets when the disease was severe. Peronospora trifoliorum was identified as the causal agent of the disease based on mycological characteristics. Pathogenicity of the fungus was confirmed by artificial inoculation. This is the first record of downy mildew on astrgal plant caused by Peronospora trifoliorum in Korea.

  • This paper is the eighth contribution towards taxonomic studies on Cercospora and allied genera, and contains ten species of Korean cercosporoid fungi; viz., Cercospora brassicicola, C. flagellaris, C. sesami, C. zinniae, Passalora amurensis, Pseudocercospora balsaminae, P. humuli, P. puderi, P. salicina, and Stenella dianthi. Morphological characteristics of taxonomic value are described and illustrated for these species to contribute towards a mycological monograph of Korean cercosporoid fungi.
