The study of sawdust cultivation and the characteristics of mycelial growth of Pholiota nameko (맛버섯균의 균배양 특성 및 톱밥재배 기술에 관한 연구)

  • Pholiota nameko is one of the four major profitable mushrooms along with oak mushroom, winter mushroom, and oyster mushroom. It contains abundant proteins, carbohydrates, organic acids and vitamins. Its unique taste and flavor as well as its nutritional features make it widely favoured. Mushroom complete medium was the optimal medium for mycelial growth of Pholiota nameko. The optimal temperature and pH for the mycelial growth were $25^{\circ}C$ and 5.0, respectively. The best carbon sources for mycelial growth were glucose and mannose, and the best nitrogen sources were yeast extract, peptone, asparagine, etc. The 8:2 ratio mix of oak sawdust and wheat bran was the best for the bottle cultivation. The best mushroom was yielded after 30 days incubation. The best yield was produced with 850g of medium weight in a PP bag and bottle.

  • Nutritional characteristics of mycelial growth and fruit-body formation of Flammulina velutipes in synthetic media were investigated. The results are summarized as follows: 1. Among sugar substances, mannitol gave rapid mycelial growth and formation of fruit-body with higher yield, and among organic acids only succinic acid showed the fruit-body but poorly. Ethyl alcohol and glycerol comparatively promoted the formation of the fruit-body with higher yield. 2. Among nitrogen sources, peptone resulted in rapid mycelial growth and fruit-body formation with higher yield. and among amino acids glycine gave fast fruit-body formation with higher yield. However. nitrite nitrogen, lysine and methionine showed no mycelial growth at all. 3. The concentration of substances tended to affect the fruit-body formation and yield. Lower concentration of mannitol and peptone prompted fruit-body formation but with low yields, and higher concentration delayed the fruit-body formation with increased yields.

  • The optimal synthetic medium, temperature and pH range for the mycelial growth of Cordyceps scarabaeicola were PDA, $25^{\circ}C$, and pH $8.0{\sim}8.5$, respectively. The effects of carbon sources, nitrogen sources and mineral salts on mycelial growth were superior to lactose, ammonium phosphate and $CaCl_2$, respectively. Liquid spawn culture was selected out BM(brown sugar 3%, soybean flour 0.3%, potassium phosphate 0.05%, magnesium sulfate 0.05%) medium.

  • The compost dextrose(CD) medium was selected a an optimum liquid culture media for Portobella(Agaricus brunescens). The optimal conditions for the mycelial growth of Agaricus brunescens were 25 and pH 7.0. Sucrose and soluble starch were favorable to mycelial growth. Malt extract and ammonium nitrate appeared to be more favorable than the other kinds of nitrogen source. The optimum C/N ratio as about 10.When the collected Portobella trains were compared with growth characteristics, KME 49007 appeared the highest of $44.6kg/3.3m^2$.

  • This study was carried out to investigate the cultural characteristics and the genetic characteristics in Lepists nuda. The mycelia cultural characteristics which include the specificity for pH, carbon source, nitrogen source and medium were studied using petridish culture and liquid media culture respectively. The genetic characteristics were also investigated using random amplified polymorphic DNA (RAPD) analysis. To find out the mycelial growth rate by various medium, mycelia was cultured for 25 days in PDA, YM, MEA, GPB and Yamanaka. The Yamanaka was superior to the other media in supporting the mycelial growth. pH6 produced the best result in the test of an optimal pH. In experiment for optimal carbon source, starch showa 46.8$\pm$1.7mm of diameter of mycelial colony as the best and for nitrogen source, yeast extract showed a good effect as well. In investigation for genetic characteristics, Lepists nuda was amplified by 7 primers among 10 primers and Pleurotus ostreatus was also amplified by 7 primers. From the RAPD analysis between species, the band patterns were different.

  • In order to investigate the cultural characteristics of Lentinus lepideus, media, pH, carbon sources, nitrogen sources, vitamin and organic acids were tested in submerged culture. It grew well in GPB(broth), and the optimum temperature and pH was $25^{\circ}C$ and 4.2, respectively. The carbon sources such as galactose belong to monosaccharide and maltose belong to disaccharide were effective for mycelial growth. Peptone which is compound nitrogen source was good for mycelial yield but the other nitrogen sources were not effective. Among the various vitamins, pyridoxine is suitable for mycelial yield. Among the various organic acids, citric acid promoted the mycelial yield but acetic acid interrupted the mycelial growth of L. lepideus.

Physiological Characteristics and Optimized Culture Condition of the Mycelia of Inonotus mikadoi. (황갈색시루뻔버섯(Inonotus mikadoi)의 균사체 배양 최적 조건 및 생리학적 특성)

  • The culture condition and medium composition for the enhanced mycelial growth of Inonotus mikadoi IMSNU 32058 were investigated. The optimal temperature and pH for the mycelial growth were $27^{\circ}C$ and pH 4.5, respectively. Among the complex media tested, the malt extract medium and Phellinus igniarius medium were very good for mycelial growth of I. mikadoi. When Czapek-Dox medium was used as a minimal medium for cultivation of mycelia, xylose, raffinose and carboxymethyl cellulose were very excellent as a carbon and energy source. With respect to carbohydrate, sucrose and glucose were very good carbon sources. In general, organic complex nitrogen sources were better than other inorganic ones. As the organic complex nitrogen sources tested, yeast extract, soytone, proteose peptone and bacto peptone were the best as a source of organic nitrogen. When ammonium sulfate as an inorganic source of nitrogen was used, the enhanced mycelial growth was shown. p-Aminobenzoic acid was proved to be most appropriate source of vitamin. After the mycelia of I. mikadoi was cultivated at $27^{\circ}C$ for 5 days in MEM broth (pH 4.5), the activities of both exomycelial and endo-mycelial enzymes were determined. Among endomycelial enzymes assayed, the specific activity of laccase was much higher than those of other enzymes. When the fungus was grown in MEM broth, exomycelial specific enzyme activity of laccase was comparatively high. However, little or no enzyme activities of protease, chitinase and lipase were found.

Phylogenetic Relationships and Cultural Characteristics among Inonotus obliquus Strains Collected in Korea (국내 수집 차가버섯 균주의 배양특성과 유전적 유연관계 분석)

  • Fruiting bodies of Inonotus obliquus were collected from the trunk of Betula ermani at 1,100 m of Mt. Odae. Diameter range of the trees at breast height (DBH) was $10{\sim}50$ cm and size range of the sclerotia was $8{\times}5{\sim}20{\times}16cm$. Relationships between the examined strains and Inonotus obliquus strain registered in National Center for Biotechnology Information (NCBI) were very near. And all of 10 strains except strains registered in NCBI showed high homologous characteristics by neighbour joining analysis of ITS sequence. Mycelial growth showed a big difference among strains. Mycelial growth of KFRI 744 was fastest and KFRI 739 was slowest. Difference of mycelial growth between KFRI 735 and 738 was slight, but the difference of mycelial growth between KFRI 744 and 739 was almost twice. Also weight reduction rate among strains showed some difference. KFRI 744 was highest and KFRI 741 was lowest. Vegetative incompatibilities were observed in all mycelial pairings except for KFRI 740-741 and KFRI 742-743 combinations.

On the Cultural Characteristics and Wood-Decayedness of Wood-Decaying Fungi in Korea (목재부후균의 배양적 특성과 부후성에 관한 연구)

  • In order to investigate early identification of species of the wood-decaying fungi in the the mycelial stage, the authors isolated of 41 species, 28 genus, 9 family at 8 locations in Korea and cultivated these isolates on the various kinds of solid media. After investigating such cultural characteristics as oxidase reactions with tannic and gallic acid, various morphological features of colony and growth grade, appeared on the various media, the authors obtained the following results : 1. The oxidase reactions with tannic and gallic acid in the PDTA, DTA, PDGA and DGA media are available for identificantion of the wood-decaying fungi. 2. The oxidase reactions with guaiacol, pyrogallol and hydroquinone in the PDGUA, PDPA and PDHA media are not so much available for identification of the wood-decaying fungi. 3. Morphological features of colonies such as mycelium color, floccose, floccose-powdery, mycelloid, powdery-mycelloid, velvet, radiate, contoured, rosulate and growth grade on the PDA, PSA and PXA media are useful for identification of wood-decaying fungi. 4. It is believed that early identification in species level of wood-decaying fungi using cultural characteristics in the mycelial stage is possible. 5. The key for the identification of 41 species of wood-decaying fungi is proposed by the cultural characteristics using several solid media.

Development of Neutaribeosut Varieties (Pleurotus ostreatus) Chongpung, Myongwol

  • This study was carried out to find a useful mushroom at Chungnam Agricultural Research And Extention Service. Twenty materials used were collected from domestic and exotic area. These races were compared bontanical characteristics to leading varieties by PCR-RAPD methods. Mycelial growth temperature of Chongpung and Myongwol were at 20 to 25 $^{\circ}C$ and 25 to 30 $^{\circ}C$ at PDA medium, respectively mycelial growth of these varieties were similiar at pH 6.5 to 7.5. In case of mushroom cultivation temperature ranges, Chongpung was at 5 to 26$^{\circ}C$ and Myongwol was at 7 to 28$^{\circ}C$, but the optimum temperature range of these were appeared at 15 to 19$^{\circ}C$. Culture temperature of these was 23$^{\circ}C$ and period of mycelial culture was needed 23 to 24 days under 850cc/pp, while was needed 11 to 12 days at waste cotton medium. Cap color of these at first inducing mushroom was all dark blue, but at late growing stages Chongpung was shown as grey, and Myongwol was shown as dark grey. Yield of Chongpung was appeared as 46kg/3.3$m^2$ and that of Myongwol was 41kg /3.3$m^2$, while Chunchu No2 as check was 40kg/3.3$m^2$. Results from PDA medium and PCR-RAPD analysis two of these were different from others.
