Characteristics of fruiting body growth according to alternative substrates of king oyster mushroom (Pleurotus eryngii) (큰느타리 대체배지 종류에 따른 자실체 생육 특성)

  • The present study aimed at selecting a cost-effective substrate for king oyster mushroom based on the growth characteristics of fruiting body for use as a basic resource to ensure stable production on farms. Compositional analysis of substrates manufactured with different materials in each process revealed that the total carbon content was 42.24-48.22% higher and the total nitrogen content was 1.7-2.29% higher in the processed lot than in the control (i.e., substrate used by the farmhouse; 40.86% and 1.39%, respectively). Meanwhile, the carbon-to-nitrogen ratio was the highest in the control (27.9% vs. 19.12-27.88% in the processed lot). When cultured for 28 days, the mycelium growth was 11.5 and 11.3 mm in substrates 1 and 6, respectively, indicating the fastest growth; meanwhile, the values were 10.1-10.3 mm in the control and substrate 11, showing a similar tendency. Mycelial density did not differ significantly among the processed lots. Yield per bottle was higher in substrates 8 (205.95 g/bottle), 7 (178.51 g/bottle), and 11 (170.63 g/bottle) than in the control (152.2 g/bottle). Fruiting body quality was comparable to controls in all processed lots. Overall, economic effects, such as substrate material prices, should be analyzed, and stability evaluations, such as residual pesticide and harmful microorganisms, should be undertaken along with further detailed examination to ensure safe and stable production on farms.

  • 'Baekseung', a new variety of Flammulina velutipes, was bred by mating two monokaryotic strains isolated from KMCC 4210 and KMCC 4216 in the Mushroom Research Division, Baekseung ARES in 2016. The Baekseung and Uri1ho strains showed fast mycelial growth and mycelial density on malt extract agar media after 7 days of incubation. The spawn running period on the sawdust substrate required a cultivation period and temperature of 30 days and $25^{\circ}C$, respectively, for primordia formation where in fruiting body development occurred from $11{\pm}1days$ at $14^{\circ}C$ and $14{\pm}1days$ at $7^{\circ}C$. The length of the pilei and stipes of the Baekseung harvested in optimal stag were $11.3{\pm}0.4$ and $89.2{\pm}7.1mm$, respectively, whereas the values for Uri1ho were $10.7{\pm}1.0$ and $91.3{\pm}20.8mm$, respectively. The yield of the Baekseung and Uri1ho strain grown on the sawdust substrate was $153.7{\pm}12.5$ and $139.8{\pm}17.8g$, respectively, per 850 ml in bottle cultivation. The inferred tree exhibited a phylogenetic relationship between the Korean white fruiting body strains of Baekseung, Uri1ho and Fv-14-a-38, Fv-14-a-51, and the Japanese white fruiting body-forming strains of KMCC 4226, and these were confirmed to be genetically related.

Culture Maturity of Lentinula edodes on Sawdust-Based Substrate in Relation to Fruiting Potential (표고 톱밥배지의 성숙도와 자실체형성 포텐셜)

  • Culture maturity assessment can be used to control fruiting body flush timing. Culture maturity of sawdust-based substrate was evaluated by using oak mushroom, (Lentinula edodes (Berk.) Pegler). The influence of substrate water potential (${\psi}$) on the growth and fruiting of three genotypes of L. edodes was also investigated. Glucosamine content revealed a peak at the fruiting body senescent stage. Glucosamine increased steadily to the sporophore senescent stage, and sharply declined at crop treatment. Lipid phosphate and ergosterol contents peaked at pinning and button break stages, respectively. Therefore lipid phosphate and ergosterol contents would be considered as the convenient measurement for judging culture maturity and fruiting potentials. The substrate pH values before inoculation and on the fruiting stage were varied from 6.3 to 4.0. This pH changes were detected as changes in color from bluish purple to yellow by direct bromphenol blue(BPB) spraying, and shown a good correlation with fruit body yield of the 1 st flush. Concerning water potential of the cultures, a slight reduction of water potential, -0.5MPa, stimulated mycelial and colony growths on liquid, agar and sawdust-based substrates. The water potential of well-colonized matured substrate was -0.7MPa and -4.0MPa, before and after the fruiting, respectively. Excellent water providing capacity (higher ${\psi}$) is expected to well-matured cultures with a high density of mycelial colonization. Also, the substrate water potential significantly affected by the interaction between genotypes and spawn run time.

  • The objective of this study was to achieve biological control of green mold disease in Pyogo mushrooms using antagonistic microorganisms. Bacillus subtilis BSM320 cells inhibited mycelial growth by 48-60% against three Trichodermaisolates including T. hazianumisolated from the substrates of Lentinula edodes, showing their antifungal activity.The bacteria were cultured to a high density of 4.2 × 109±113.7 cfu/mlin aqueous extract of composted spent mushroom substrates of L. edodes containing 1% glucose and showed a higher growth rate than that observed when using the commercial medium, Luria-Bertani broth. The bacterial culture showed a 75% protective effect without damaging the mushroom fruiting bodies. These results suggest that B. subtilis BSM320culture is suitable for biological control of green mold disease during mushroom cultivation.

  • An antagonistic bacteria, Stenotrophomonas maitophilia BW-13 strain which was effectively inhibited mycerial growth of Bottom rot pathogen, Rhizoctonia solani PY-1 strain was isolated from the rhizosphere of the lettuce in Uiryeong-Gun, Gyeongsangnam-Do from 2002 to 2003. For the biological control, the most suitable inoculum and its density of pathogen, PY-1 strain ware tested prior biological control test, For the pathogenicity test, A inoculum (wheat bran)sawdust+rice bran+PDB) showing disease incidence of 100% was selected as the most suitable inoculum, which showed more effective than B inoculum (sawdust+rice bran+DW) and mycelial disc. also, In selection of the amount of inoculum (40g, 50g, 60g, 70g, 80g), most suitable amount of inoculum of pathogen determined as 40g showing disease incidence of 80%. For the selection of effective microorganism to control bottom rot on lettuce, about 200 isolates were isolated from the diseased soil and lettuce leaves, and examined their antifungal activity to the pathogen on PDA. As the pots assay, BW-13 strain showed the highest control value as 90%, and followed by R-13 and R-26 strain as 80% and 60%, respectively. Selected BW-13 isolates identified as 5. maltophilia (GeneBank accession no. AJ293473.1, 99%) by 16S rRNA sequencing. This is the first report on the biological control using by S. maltophilia to the bottom rot pathogen, Rhizoctonia solani PY-1 strain.

  • This study was carried out to investigate the conditions of liquid culture and fruiting body formation of Cordyceps militaris using grain mediacontained the brown rice and black purple rice. Liquid mycelial growth and density of Cordyceps militaris were the highest in the medium of malt extract 0.7% and pH 5.0~5.5. The optimal condition of formation the fruiting bodies were shown to be temperature of $17^{\circ}C$, light intensity of 600 lux, $C0_2$ of 500ppm and humidity intensity of 95% produced highest amount of fruit bodies.

  • This study was carried out to select oyster mushroom strains tolerant to salinity and characteristic cultivation by different NaCl concentration. Among the 64 strains, Kimjae-10ho, Nongki-2-1ho, Myungyul, Byungneutari-1ho, Bupyungsoyae-1ho, Sambok and Chunchu-2ho resulted in higher mycelial growth and density on PDA medium containing NaCl. The maxium NaCl concentration possible to fruit body formation was 0.5% NaCl in Myungyul, 1.0% NaCl in Kimjae-10ho and Bupyungsoyae-1ho, 1.5% NaCl in Nongki-2-1ho, Byungneutari-1ho, Sambok and Chunchu-2ho, respectively. Increased NaCl concentration on sawdust medium prolonged culture period, while decreased total quantity and come to be short and thin in Length and thickness of stipe. Higher NaCl concentration in the medium decreased the uptake rate of $K_2O$ and CaO, whereas increased the NaCl uptake rate.

Selection of Biocontrol Agents against Phytophthora Blight of Pepper and Its Root Colonization Ability (고추역병 생물적방제 근권세균의 선발 및 근권정착 능력 연구)

  • Four promising biocontrol agents against Phytophthora capsici were selected from 507 bacterial isolates collected from rhizosphere soils and roots of pepper plants. In vitro experiment, these four biocontrol agents inhibited mycelial growth, germination of cystospores, and formation of zoosporangia and zoospores of Phytophthora capsici. In the pot experiment, the four biocontrol agents showed control efficiency higher than 70%. In greenhouse experiment, the isolates G28-6 gave the control value of 79.4%. These four biocontrol agents successfully colonized in the population density beyond 105 cfu/g on roots of pepper in vitro. The isolates G28-6 was identified as Pseudomonas aurantiaca, based on its cultural, morphological, and biochemical characterization and 16S rRNA gene sequence analysis.

  • The medium used for bottle cultivation of mushrooms is made according to a specific workflow; this includes mixing the raw materials, adjusting the moisture content, placing the mixture into bottles, and sterilizing and cooling the medium. Medium composition should be kept constant throughout the year. placed in the bottle, choice of materials, moisture content, weight of the medium, bulk density, nitrogen content, three-phase medium, such as suitable conditions. To achieve constant medium composition a comprehensive model has been established in accordance with the technology developed in the work process with respect to medium compositions piecemeal. By comprehensively applying the various methods to establish medium composition, our country can expect to grow at over four seasons and overcome the many difficulties in mushroom mycelial growth periods, cultivation periods, quantity, and quality.

  • This study was conducted to elucidate the optimal nitrogen content for each variety of oyster mushroom (Pleurotus ostreatus) by investigating the change in the characteristics of fruiting body according to the nitrogen content of the sawdust media, and to use the results as the basic data for cultivation of high quality oyster mushroom. The nitrogen content was adjusted to 1.3%, 1.6%, 1.8% and 2.3%, and at each condition, mycelial growth was observed, which showed that the nitrogen content did not affect the rate of mycelial growth and density substantially. In the investigation of the characteristics of fruiting body, at the 1.8% nitrogen content, Chunchu No.2 showed the pileus diameter of 27 mm, and the stipe diameter and length of 11 mm and 61 mm, respectively, and the measurement of its color values revealed that the brightness of the pileus and the stipe was 31 and 80, respectively, offering the best commercial value, and yielding 25 valid stipes, which was the largest number. Likewise, at the 1.8% nitrogen content, Heuktari also showed the excellent growth characteristics including the pileus diameter of 29 mm and the stipe diameter and length of 10 mm and 68 mm, respectively, and the superb color values of the brightness of 37 at the pileus and 78 at the stipe. Moreover, the number of valid stipes was also found to be the highest with 19. In terms of morphological characteristics such as the pileus diameter and color, Soltari also produced the best mushrooms at the 1.8% nitrogen content and the number of valid stipes was high with 22. In summary, irrespective of varieties of oyster mushroom, high quality products could be cultivated at the nitrogen content of 1.8%.
