A New Report on Edible Tropical Bolete, Phlebopus spongiosus in Thailand and Its Fruiting Body Formation without the Need for a Host Plant

  • Phlebopus spongiosus is a well-known edible ectomycorrhizal mushroom indigenous to southern Vietnam. The mushroom specimens collected from northern Thailand in this study were identified as P. spongiosus. This identification was based on morphological characteristics and the multi-gene phylogenetic analyses. Pure cultures were isolated and the relevant suitable mycelial growth conditions were investigated. The results indicated that the fungal mycelia grew well on L-modified Melin-Norkans, and Murashige and Skoog agar all of which were adjusted to a pH of 5.0 at 30 ℃. Sclerotia-like structures were observed on cultures. The ability of this mushroom to produce fruiting bodies in the absence of a host plant was determined by employing a bag cultivation method. Fungal mycelia completely covered the cultivation substrate after 90-95 days following inoculation of mushroom spawn. Under the mushroom house conditions, the highest amount of primordial formation was observed after 10-15 days at a casing with soil:vermiculite (1:1, v/v). The primordia developed into a mature stage within one week. Moreover, identification of the cultivated fruiting bodies was confirmed by both morphological and molecular methods. This is the first record of P. spongiosus found in Thailand and its ability to form fruiting bodies without a host plant.

  • To find antitumor components in the hot water extract of the cultured mycelia of Ganoderma lucidum, protein-bound polysaccharides were purified and fractionated (Fr. I-V) by DEAE-cellulose ion exchange column chromatography and Sepbarose CL-4B gel filtration. When a dose of 20 mg/kg/day of each was, i.p., injected into ICR mice, the inhibition ratios against the solid form of sarcoma 180 were $64.2{\sim}75.8%$. The antitumor component was examined for immunological activity. It increased the amount of superoxide anion released by induced macrophages in peritoneal cavity to 1.8 times and the count of hemolytic plaque-forming cells (PFC) was increased to 4.4 times as compared with those of the control group. It contained 68.6% polysaccharide which consisted of mannose, glucose, galactose, fucose and xylose and 5.1% protein consisting of 17 amino acids. The contents of hexosamine were 0.78%. The molecular weight of Fr. V that showed the highest antitumor activity was $5.8{\times}10^4$ dalton by Sepharose CL-4B gel filtration. It was named lucidan.

액체종균(液體種菌) 접종(接種)에 의한 표고톱밥재배(栽培) 효과(效果) (Effect of Sawdust Culture on Oak Mushroom, Lentinula edodes (Berk.) Pegler by Inoculation of the Liquid Spawn)

  • For cultivation on sawdust-bed of oak-mushroom until present time, inoculation of spawn on sawdust bed has been performed by sawdust spawn. But, liquid spawn may have advantages for rapid mass production of spawn, and now, sawdust-cultivation by liquid spawn inoculation should be applied instead of sawdust spawn. Therefore, investigations were performed to evaluate the effect of sawdust-cultivation by liquid spawn inoculation. The results were as follows: 1. When 11 kinds of liquid media were applied, the oak-mushroom culture medium was the most excellent in growth. Most suitable temperature at PDA was $25^{\circ}C$, and $22.5\sim27.5^{\circ}C$ in range were optimal for liquid culture. In liquid culture, amount of mycelial growth increases rapidly up to 40 days of cultivation. Incubation at fermentor brought yield of 106mg dry mycelia per 40ml media after 17 days. 2. In 1l-spawn bottle, growth of mycelium by inoculation of 20ml-liquid spawns were faster than 6g-sawdust spawn in spread of mycelia. On 2kg-bag culture, inoculations of 10ml-, 20ml- and 30ml-liquid spawns were all slower than 20g-sawdust spawn in mycelial spread. So, amount increasement in ampunt of liquid spawn should be discussed. Yields of mushrooms until third sproutings of 2kg-bag culture were 580g in 30ml-liquid spawn inoculation, but 510g, 486g and 470g from 20g-sawdust spawn, 20ml-liquid spawn and 10ml-liquid spawn, respectively. Thus, 30ml-liquid spawn inoculation was highest in yield.

  • 콩고오지 제조중(製造中)에 일어나는 콩 단백질(蛋白質)의 형태적(形態的) 변화(變化) 및 대두단백질(大豆蛋白質)이 Aspergillus soya가 분비(分泌)하는 protease의 작용(作用)을 받아 생성(生成)되는 peptide의 총체적(總體的)인 동향(動向)을 알기 위(爲)하여 제국중(製麴中) 경시적(經時的)으로 채취(採取)한 시료(試料)에 대(對)하여 불용성단백태질소(不溶性蛋白態窒素) 수용성단백태질소(水溶性蛋白態窒素) peptide 태질소(態窒素)를 측정(測定)한 다음 peptide군(群) 액(液)을 cross linkage가 각각(各各) 다른 여러 가지 Dowex-50을 사용(使用)하여 분자사별(分子篩別)을 한 각(各) fraction에 대하여 전질소(全窒素) amino태(態) 질소(窒素), 및 average peptide lingth을 측정계산(測定計算)하여 다음과 같은 결과(結果)를 얻었다. 1. 불용성단백태질소(不溶性蛋白態窒素) 및 peptide 태(態) 질소(窒素)는 균사발생시(菌絲發生時)까지 별변화(別變化)가 없었으나 균사발생시(菌絲發生時)부터 불용성단백태질소(不溶性蛋白態窒素)는 급격(急激)히 감소(減少)되고, peptide 태(態) 질소(窒素)는 급격(急激)히 증가(增加)되었다가 포자발생시(胞子發生時)를 한계점(限界點)으로 하여 감소율(減少率) 및 증가율(增加率)이 완화(緩和)되었다. 수용성단백태질소(水溶性蛋白態窒素)는 콩고오지 전면(全面)을 균사(菌絲)가 피복(被覆)할때까지 일정(一定)하나 그후 포자발생시(胞子發生時)를 한계점(限界點)으로 하여 증가(增加)되었다가 다시 서서(徐徐)히 감소(減少)하였다. 2. Peptide 군(郡)을 분자식별(分子篩別)한 결과(結果) 제국초(製麴初)는 각(各) fraction의 비(比)에 별(別) 변동(變動)이 없으나 균사(菌絲) 발생후(發生後)에는 X-16, X-12, X-8, X-4, X-2 fraction은 상당(相當)한 증가(增加)를 보이다가 포자잘생후(胞子發生後)에는 큰 변동(變動)이 없고 effluent fraction은 출국(出麴)까지 계속(繼續)하여 증가(增加)되고 있다.

樹種, 接種量 및 接種方法이 茯笭 菌絲生長과 結笭에 미치는 영향 (Effect of Tree Species, Inoculation Amount and Inoculation Methods on Mycelium Growth and Sclerotia Formatino of Poria cocos Wolf)

  • When medicinal mushroom, Poria cocos, is cultured , inoculation method of spawn is cross slice inoculation of which the both sides of pine tree were peeled and spawn of P.cocos was inoculated. However, this method required lots of inoculation amount. This study was carried out to improve the culturing method of P. cocos. A good growth of P.cocos was observed in MCM(mushroom complete medium), showing proper mycelia growth and density. In inoculation amount, conventional method(cross slice inoculation) requires 20 bottles of spawn. In contrast, short log method required 8 bottles of spawn and drilling inoculation method 2~3 bottles, which could save by 60% and 85-90% respectively. In the selectrion of tree species, pine and larch had better condition for spawn culture and sclerotia formation condition.In terms of yield , pine was 33.7kg/3.3$m^2$. In the yield of pine, conventional method was 23.4kg/3.3$m^2$, drilling inoculation 29.4kg/3.3$m^2$, short log inoculation 31.7kg/3.3$m^2$, therefore drilling inoculation could increase by 25% and short log inoculation 35%, In addition, management cost was also saved.

셀레늄 강화 팽이버섯과 폐배지의 셀레늄 형태 및 팽이버섯내 셀레늄 축적대사에 관한 연구 (Studies on the Selenium Type and Metabolism of Selenium Accumulation in the Selenium-Enriched Mushroom, Flammulina Velutipes, and Its Spent Mushroom Composts)

  • This study was conducted to determine the total amount and type of seleniwn (Se) in the Se-enriched mushroom and its spent mushroom composts (SMC), and to investigate the metabolism in relation to Se accwnulation in the mushroom. Mushrooms, Flammulina velutipes, used in this study were grown for 60 days by adding 2 rng of inorganic Se (Na2Se03) per kg of mushroom composts (MC) on as-fed basis and were compared with normal mushrooms grown on the non Se-supplemented Me. Total Se contents for Se-treated mushrooms were significantly increased (P < 0.0001) by 20-fold (4.51 $\mu$/ g of dry) compared to Se-untreated (0.23 $\mu$/ g of dry). On the contrary, organic Se ratio was significantly lower (P < 0.0001) in the Se-treated mushroom (72.3 %) than the Se-untreated one (100 %, not analytically detected of inorganic Se). Se distribution upon a length in the Se-treated mushrooms was the highest in the bottom part (6.86 $\mu$/ g of dry) near to MC, and top and middle parts were significantly lower (3.71 and 3.01 $\mu$/ g of dry, respectively; P < 0.001) than the bottom. In the SMC from Se-treated mushrooms, the significant amount of Se (5.04l1g/g of dry) was remained, but that from the Se-untreated mushrooms was significantly low (P$\mu$ / g of dry. Se-treated SMC showed a high ratio of organic Se (65.67 %), suggesting that the significant amount of inorganic Se in the SMC was converted to organic Se by mushroom mycelia. Prior to mycelia inoculation in the mushroom culture, the sterilization of MC brought approximately 18% of Se loss in the MC. Apparent and net accumulation rates (%) for Se into mushrooms were 14.81 and 10.14 %, respectively, resulting from the Se volatilization into the air via metabolic process of mushroom itself. The result of this study shows that inorganic Se addition to MC for mushroom improved the organic Se contents in the mushroom and SMC. This study showed the possibility that Se in Se-enriched mushroom and SMC could be utilized as Se sources of food for human as well as feed for livestock.

Lentinus edodes IY105 알칼리 추출물의 보체계활성 및 항종양효과 (Anticomplementary and Antitumor Activities of the Alkali Extract from the Mycelia of Lentinus edodes 1'11105)

  • 경기도 광릉에서 채집한 자실체로부터 분리한 Lentinus edodes IY105균을 배양하여 얻은 균사체를 알칼리로 추출하여 얻은 물질이 면역계와 sarcoma 180이 이식된 mouse에 미치는 영향을 검토하였다. Lentinus edodes IY105 배양균사체의 알칼리 추출물은 항보체활성과 alternative complement hemolytic 활성을 가지고 있었고, carbon clearance 활성을 가지고 있었고, carbon clearance 활성을 나타내는 것으로 보아 mouse의 망내계에 존재하는 macrophage를 활성화시켜 이 물질에 대한 탐식기능을 증가시킴을 알수 있었다. 또한, 이물질은 mouse의 용혈반 형성 세포수를 증가시키는 것으로 보아 항체생산 자극효과가 있는 것으로 추측되었으며, 종양세포인 sarcoma 180이 이식된 mouse의 암을 50 정도 억제하는 효과를 나타내었다. 알칼리 추출물의 성분을 분석한 결과, 당 17, 단백질 42로 구성된 단백질다당체가 주성분으로 단백다당체는 16종의 아미노산과 52종류의 당으로 구성되어 있었다.

Possible Negative Effect of Pigmentation on Biosynthesis of Polyketide Mycotoxin Zearalenone in Gibberella zeae

  • We investigated a possible coordination between the biosyntheses of two polyketides in the cereal head blight fungus Gibberella zeae, zearalenone (ZEA) and aurofusarin (AUR), which are catalyzed by the polyketide synthases (PKS) PKS4/PKS13 and PKS12, respectively. To determine if the production of one polyketide influences that of the other, we used four different transgenic strains of G zeae; three were deficient for either ZEA or AUR or both, and one was an AUR-overproducing strain. The mycelia of both the wild-type and ${\Delta}PKS4$ strain deficient for ZEA produced AUR normally, whereas the mycelia of both the ${\Delta}PKS12$ and ${\Delta}PKS4::{\Delta}PKS12$ strain showed no AUR accumulation. All the examined deletion strains caused necrotic spots on the surface of com kernels and were found to produce the nonpolyketide mycotoxins trichothecenes to the same amount as the wild-type strain. In contrast, the AUR-deficient ${\Delta}PKS12$ strains produced greater quantities of ZEA and its derivatives than the wild-type progenitor on both a rice substrate and a liquid medium; the AUR-overproducing strain did not produce ZEA on either medium. Furthermore, the expression of both PKS4 and PKS13 was induced earlier in the ${\Delta}PKS12$ strains than in the wild-type strain, and there was no difference in the transcription of PKS12 between the two strains. Therefore, these results indicate that the ZEA biosynthetic pathway is negatively regulated by the accumulation of another polyketide (AUR) in G zeae.

아까시재목버섯 유래 카르테노이드 계열 항산화 물질 생산 (Development of carotenoid production process using perenniporia fraxinea)

  • 이 연구에서는 Perenniporia fraxinea를 사용하여 배지가 카르테노이드 생성 및 균사 성장에 미치는 영향을 분석했습니다. 0.1% 미만의 펩톤을 함유한 맥아추출물은 카르테노이드의 생성을 촉진했으며 0.2% 이상의 펩톤이 그 생산을 억제하는 것으로 나타났다. 맥아추출물이 없으면 Perenniporia fraxinea는 소량의 펩톤 첨가에도 불구하고 카르테노이드를 생산하지 못했다. 아카시재목버섯의 배양을 통하여 생산된 균사체를 원심분리로 배양액을 분리하여 상층액을 카르테노이드 용액으로 회수가 가능하였다. 균사체 내부에 축적된 카르테노이드는 에탄올을 사용하여 추출하였다. 배양액에서 분리 또는 추출한 카르테노이드 용액은 우수한 DPPH 라디칼 소거 활성을 나타냈다. 본 연구에서 생산된 항산화 카르테노이드는 천연물 유래 물질로 독성 및 부작용이 없으며 항산화 효과가 우수하여 활성산소에 의해 생성된 산화물을 효과적으로 제거할 수 있다.

큰느타리버섯에서 석충 페리틴 단백질 유전자의 발현 최적화 및 생물학적 활성 (Optimization of the Expression of the Ferritin Protein Gene in Pleurotus eryngii and Its Biological Activity)

  • 큰느타리버섯에서 철 저장과 관련된 페리틴 단백질의 발현 및 분비를 최적화하기 위해, T-Fer 벡터에 EcoRI 및 HindIII처리를 해 페리틴 유전자를 얻은 후, BamHI으로 처리된 선형의 pPEVPR1b 분비 벡터에 클로닝하여pPEVPR1b-Fer 재조합 벡터를 구축한 다음 Agrobacterium tumefaciens LBA4404 로 도입하였다. Agrobacterium tumefaciens-mediated transformation 방법에 의해 Pleurotus eryngii로 형질전환하고 kanamycin함유된 MCM 배지에서 올바른 형질전환체를 선별하였고, 단백질 발현은 SDS-PAGE 및 항원항체 반응에 의한 western blot으로 확인하였다. 페리틴 단백질의 분비 발현은 batch culture 및 20 L airlift type fermenter에서 배양 시간 및 온도와 같은 배양 조건에 의해 최적화되었다. 페리틴 생산을 위한 배양 조건은 MCM 배지에서 25℃ 및 8 일 배양에 의해 최적화되었다. 페리틴 단백질의 양은 정량적 단백질 분석에 의해 2.4 mg/g mycelium으로 측정되었다. 그러나, PR1b (32 amino acid)의 분비서열은 큰느타리버섯 내부의 peptidase에 의해 정확하게 processing되지 않았지만, 페리틴 단백질은 균사체에서 최대로 전체단백질의 24.7% 발현되었고, 배양액에서는 검출되지 않았다. 철 결합 활성은 7.5% non-denaturing gel에서 Perls' staining에 의해 확인되었으며, 다량체 페리틴(24 subunits)이 P. eryngii 균사체에서 형성되었음을 보여준다. 생물학적 활성 측정을 위하여 페리틴을 함유한 분말을 제조하여 육계의 사료 첨가제로서의 사용 가능성에 대해 시험하였으며, 결과적으로 페리틴은 육계의 성장을 촉진하고 사료 효율 및 생산 지수를 향상시키는것으로 확인되었다.
