Anti-inflammatory Effect of an Ethyl Acetate Fraction from Myagropsis yendoi on Lipopolysaccharides-stimulated RAW 264.7 Cells (LPS로 유도된 RAW 264.7 대식세포에 대한 애기외톨개 모자반(Myagropsis yendoi) 에틸아세테이트 분획물의 항염증 효과)

  • An ethanolic extract from Myagropsis yendoi was fractionated using several solvents. Among these, an ethyl acetate fraction (Myagropsis yendoi ethyl acetate fraction: MYE) showed the highest anti-inflammatory activity based on inhibition of lipopolysaccharides (LPS)-induced nitric oxide (NO) production in RAW 264.7 cells. We thus investigated the molecular mechanisms underlying MYE's inhibitory effects. Pretreatment of cells with up to $30{\mu}g/mL$ of MYE significantly inhibited NO production and inducible nitric oxide synthase expression in a dose-dependent manner (P<0.05). Similarly, MYE markedly reduced the production of pro-inflammatory cytokines, such as interleukin (IL)-$1{\beta}$, IL-6, and tumor necrosis factor (TNF)-${\alpha}$, as well as their mRNA levels. While the nuclear translocation of nuclear factor-kappa B (NF-${\kappa}B$) was strongly suppressed by MYE, the activation of a nuclear factor erythroid 2-related factor (Nrf2) was increased. Moreover, MYE significantly reduced the phosphorylation of JNK, p38 MAPK, and phosphatidylinositol 3-kinase/Akt in LPS-stimulated cells. These results indicate that MYE contains anti-inflammatory compounds, and that it might be used as a dietary supplement for the prevention of inflammatory diseases.

  • Marine brown algae have been identified as a rich source of structurally diverse bioactive compounds. Whether Myagropsis yendoi ethanolic extracts (MYE) inhibit inflammatory responses was investigated using lipopolysaccharide (LPS)-stimulated microglia BV-2 cells. MYE inhibited LPS-induced nitric oxide (NO) production in a dose-dependent manner and suppressed the expression of inducible nitric oxide synthase in BV-2 cells. MYE also reduced the production of pro-inflammatory cytokines in LPS-stimulated BV-2 cells. LPS-induced nuclear factor-${\kappa}B$ (NF-${\kappa}B$) transcriptional activity and NF-${\kappa}B$ translocation into the nucleus were significantly inhibited by MYE treatment through preventing degradation of the inhibitor ${\kappa}B-{\alpha}$. Moreover, MYE inhibited the phosphorylation of AKT, ERK, JNK, and p38 mitogen-activated protein kinase in LPS-stimulated BV-2 cells. These results indicate that MYE is a potential source of therapeutic or functional agents for neuroinflammatory diseases.

  • This study examined the antimicrobial activity of Myagropsis yendoi (MY). The antimicrobial activity of ethanol and aqueous MY extracts were measured using a paper disc assay. The MY ethanol extract had the best antimicrobial activity. It inhibited the growth of Candida tropicalis and Gram-positive bacteria such as Bacillus subtilis, Listeria innocua, L. monocytogenes, and Staphylococcus aureus at 4 mg/mL. The minimum inhibitory concentration (MIC) of the MY ethanol extract ranged from 0.1 to 0.5%. The MY ethanol extract inhibited the growth of B. subtilis and L. innocuaat concentrations of 0.15% and 0.1%, respectively. The antimicrobial activity of the MY ethanol extract was not affected by heating at $121^{\circ}C$ for 15 min or pH 2.10. Therefore, the main substances responsible for the antimicrobial activity of the MY ethanol extract are believed to be stable with changing heat and pH.

Variation of Species Composition of Benthic Algae and Whitening in the Coast of Dokdo Island during Summer (하계 독도 연안 해조류의 종조성 변동과 갯녹음현상)

  • This study was to clarify the variations in a marine ecosystem through examining the species composition of benthic algae in the coast of Dokdo Island during summer (June & August, 2003). The total number of species found in benthic algae was 45 composed of 11% green algae, 29% red algae and 60% brown algae. The dominant species were Enteromorpha linza, Cladophora spp. and Codium frgile for green algae, Ecklonia cave, Eisenia bicyclis, Sargassum confusum, S. patens, S. horneri, Myagropsis myagroides for brown algae and Amphiroa cphedrace. A pusilla, Lithophyllum okamurae, Corallina spp. Laurencia pinnata, L. hamata, Chondria crassicaulit, C. dasyphylla, Gelidium amansii, Neosiphonia yendoi and Polysiphonia yendoi for red algae. The number of benthic algal species decreased more in this study than in previous researches. The whitening of the rocky shore might provoke the decrease of benthic algal biomass and species.

Studies on Bioactive Substances and Antioxidant Activities of Marine Algae from Jeju Island (제주도 연안 서식 해조류에 대한 주요 활성성분 및 항산화효능 연구)

  • The objective of this study was to investigate the useful components and potential antioxidant activities of 30 marine algae, collected from Jeju Island in Korea. The contents of bioacitve substance of extracts from marine algae, such total polyphenol and carbohydrate, were determined. The extract of Distromium decumbens had the highest amount of total polyphenol content (21.27%), and that of Gracilaria incurvata Okamura had the highest amount of total carbohydrate content (10.18%). The antioxidant activities of extracts obtained from algae were tested through the evaluation of DPPH radical and hydroxyl radical scavenging activity. The extracts of Distromium decumbens, Sargassum hemiphyllum (Turner) C.Agardh, Sargassum serratifolium (C. Agardh) C. Agardh and Acrosorium yendoi Yamada were found to have more than 80% DPPH radical scavenging activity and that of Dictyota okamurae (Dawson) $H{\ddot{o}}tning$, Schnetter, et Prud'homme van Reine, Myagropsis myagroides (Martens ex Turner) Fensholt, Sargassum serratifolium (C. Agardh) C. Agardh and Cladophora wrightiana Harvey showed more than 50% hydroxyl radical scavenging activity. These results suggest that algae collected from Jeju Island would be good raw materials for antioxidant.
