A Study on the Characterization and Dyeability of Mushroom Colorant (I)- Extraction, storage and analysis of mushroom colorant- (양송이 색소의 특성과 염색성에 관한 연구(I) - 색소의 추출과 보관 및 성분 분석을 중심으로 -)

  • The browning characteristic and dyeability of the mushroom were examined to establish the optimum condition for browning and extraction in the process of obtaining the natural dye, brown colorant. The composition of browning extracts from mushroom was also analyzed and the dyeability were investigated. The results of this study are as follows, 1. The optimum condition for obtaining the browning colorant from mushroom was the crushed phase of sample. 2. The browning reaction by enzymes in mushroom was efficient at a temperature of 35$^{\circ}C$, a duratron period of 2 hr and pH of 8. 3. The optimum condition for extraction of browning extracts from browned mushroom was at 95$^{\circ}C$ and Ihr. 4. To analyze the content of browning extracts in the mushroom, three fractions were obtained from gel filtration using Sephadex G-25 and the fraction 1 was melanin and the fraction 3 was dihydroxyphenylanine (DOPA) and glutaminyldihydroxybenzene (GDHB) . 5. The turbidity in enzymatic browning extraction of mushroom increased depending on refrigeration storage (4$^{\circ}C$) and UV radiation. The filtration of the extracts resulted in a decrease of absorbance. 6. The dyeability of the fraction 1 was greater compared to that of the fraction 3 and that of wool greater compared to the other two fabric materiales.

  • The mushroom pickles were prepared as a processed mushroom product. The changes in pH, total acidity, brix. color and texture of the mushroom pickles (Lentinus edodes, Pleurotus ostreatus, Agaricus bisporus and Pleurotus eryngii) were measured during storage at 4$^{\circ}C$. In addition, sensory evaluations on the mushroom pickles were performed. While the pH of Lentinus edodes and Pleurotus ostreatus pickles declined slowly, the pH of Agaricus bisporus and Pleurotus eryngii pickles declined rapidly during storage. The total acidity of Lentinus edodes pickles rose slowly, while the acidity Pleurotus ostreatus, Agaricus bisporus and Pleurotus eryngii pickles rose rapidly. As for the color of mushroom pickles, "L" , "a" and "b" values changed slightly. The changes in texture were determined by a rheometer Hardness, cohesiveness, springiness, gumminess and brittleness of Lentinus edodes pickles were lower than those of the other mushroom pickles. In the sensory evaluation of mushroom pickles, Lentinus edodes showed higher acceptability ratings than the other mushrooms in taste, color, clearness, texture and overall acceptability. This study showed that among the mushrooms tested, Lentinus edodes was the most acceptable mushroom pickle for storage.

Quality Characteristics of Processed Mushroom Products Using Pleurotus ostreatus, Agaricus bisporus and Flammulina velutipes

  • This study examined the effects of adding Pleurotus ostreatus, Agaricus bisporus and Flammulina velutipes on the quality characteristics of cookies and multiple purposes of wheat powders. The water content, crude protein content, mineral content, color value and sensory evaluation of mushroom cookies and multiple purposes of wheat powders were measured. In the case of cookies, the water content was 1.10-1.24% and crude protein content was 8.20-9.80%. General preferences, such as appearance, color, taste and texture, increased when mushroom was added to the cookies, especially flavor, which showed a much higher preference. In the studies where mushroom was added to wheat powders, the water content was 0.57-0.92% and crude protein content was 11.90-12.80%. The addition of mushroom to both of the cookies and multiple purposes of wheat powders resulted in a remarkable increase in the mineral content, especially Fe, K. In addition, an increase in the amount of added mushrooms resulted in an increase in the hunter L and a values; however, this also resulted in a decrease in the b value.

Analysis of management status of oak mushroom management in Chungcheongnam-do

  • This study attempted to estimate the degree of management of oak mushroom farms in Chungcheongnam-do and to provide information to establish oak mushroom cultivation-related policies. The oak mushroom management standard diagnostic table consists of three major categories, growing condition, inoculation management, cultivation management and management administration, along with 20 subcategories. Thus, 209 households of oak mushroom farms were surveyed from 2015 to 2018 in Gongju, Cheongyang, Buyeo and Seochun in Chungcheongnam-do. The average score for the 20 subcategories was 71.5 points (representing a significant level), indicating that these areas have excellent management conditions. The analysis of the management performance indicators revealed a high number of indicators with scores of five or above. The total score was higher, and the amount per bed log and the rate of top-grade products in the total output were also higher, indicating a significant correlation. These findings will provide consulting services to oak mushroom growers as they highlight the correlation between the higher scores of indicators in the oak mushroom management standard diagnostic table and the management performance of farmers. We found that the scores of the indicators for management administration, such as management record and analysis and fund plan were relatively lower than those of other indicators. It is assumed that the owners aging has led to negligence in recording the details on incomes, expenditures, and work and lowered the willingness to make substantial profits. Therefore, it is essential to overcome these problems for profitable oak mushroom farming.

Judgement of Resistant Cultivar by Screening method for Resistance of Oyster Mushroom to Trichoderma disease in vitro (푸른곰팡이균의 저항성 품종 검정방법에 의한 느타리버섯 균주의 저항성 판별)

  • In coculturing with strains of Trichoderma and oyster mushroom, we could detect the difference in the resistance of oyster mushroom against Trichoderma with the phenomena of barrage reaction, overgrowth and lysis. We selected the isolates ASI 2183, ASI2504 and ASI 2477 as varieties that showed the resistance. The isolates ASI 2240, ASI 2479 and ASI 2181 were the best in their resistance against Trichoderma in the method using culture filtrate. In common, the isolates ASI 2479 and ASI 2240 were selected in both methods. In post-inoculation method, the isolates ASI 2479, ASI 2333 and ASI 2181 were selected and ASI 2302 was susceptible. For the same isolate of Trichoderma, the resistance varied depending on the isolates of oyster mushroom used in the experiments. Because we could detect the interactions between Trichoderma and oyster mushroom, it is possible to detect the level of the resistance that differs in the varieties. However, there were the cases of detecting the level of the resistance in repetitions with the same isolate, which may be caused by the vitality of isolates of Trichoderma and oyster mushroom. It is efficient to test the resistance with the resistant isolate of Pleurotus salmoneostramine and the susceptible isolate of ASI 2302.

Biological control of mushroom mites using predatory mite Stratiolaelaps scimitus in button mushroom (Agaricus bisporus) cultivation (양송이에서 포식성 스키미투스응애 (Stratiolaelaps scimitus)를 이용한 버섯응애류의 생물적 방제 효과)

  • Mushroom mites have recently caused severe damage to sawdust beds, mushroom mycelia, and fruiting bodies; therefore, they have reduced the production of the button mushroom, Agaricus bisporus, in greenhouses. There are currently no registered pesticides for mushroom mites. It is necessary to selectively control mushroom flies and mites without affecting the growth of the mushroom. We examined biological control of mushroom mites using predatory mite Stratiolaelaps scimitus in button mushroom cultivation. As a result, a three times treatment (1 treatment after water cleaning, 1 treatment after fungus inoculation, and 1 treatment before or after casing) was most effective at controlling mushroom mites, with 3.000 predatory mites (3 bottles) scattered evenly over $165-230m^2$ every 1-2 m. Predatory mite Stratiolaelaps scimitus could control mushroom flies and mites at the same time and could be used at any time during cultivation.

Insecticidal Activity of Bacillus thuringiensis 656-3 Strain to Mushroom Flies in Oyster Mushroom House

  • Bacillus thuringiensis 656-3 which was isolated from a soil sample of mushroom house and showed high toxicity to mushroom flies, Lycoliella mali and Coboldia fuscipes, was surveyed for insecticidal effect in the oyster mushroom house.3. thutingiensis 656-3 was mass-cultured in the fermenter containing soybean cake(2%) and wheat bran (2%) as media source. Semi-for-mutation of B. thuringiensis 656-3 was performed with metamorphic starch only. When the formulation suspension containing $5{\times}{10^7}$ cfu was sprayed on the mushroom in mushroom house, the insecticidal effect of B. thuringiensis 656-3 to mushroom flies,1. mali and C. fuscipes, was maintained over 90% by the fifth day after starting spraying. The yield of oyster mushroom house with B. thuringiensis 656-3 was significantly increased compared to control. B. thuringiensis 656-3 represents a powerful biological insecticide for the control of mushroom flies.

Characteristics and breeding of a new golden oyster mushroom variety 『Sunjung』 (느타리버섯의 신품종 『순정』 육성 및 특성)

  • 'Sunjung', a new variety of golden oyster mushroom was bred in Mushroom Research Institute, Gyeonggido Agricultural Research & Extension Services in 2006. This oyster mushroom was selected one which mate with monokaryon collected from KME20064 and KME20066. The optimum temperature for the mycelial growth was about 26 ~$28^{\circ}C$ and that for the primordia and growth of fruitbody was about $18{\sim}23^{\circ}C$. Compared to the control which took 25 days of incubation period, Sunjung required 21~22days. It grew primordia after developed a lump of mycelium. In the characteristics of fruit body, pileus was infundibuliform of yellow-color and stripe was slender/long shape of white color. The yield was 112.9g/850cc bottle and 275.5g/2kg P.P bag.

  • Feeding rate and growth rate of earthworm population on the variously pretreated spent materials of the agaric-mushroom cultivation were investigated. When the spent mushroom substrates with different aging periods were supplied to earthworm, feeding rate and growth rate of earthworm population on spent mushroom substrates aged less than 10 days were higher than that on spent mushroom substrates aged more than 20 days. Feeding rate and growth rate were not increased when the spent mushroom substrate mixed with vermicasts or nitrogenous fertilizer was supplied. Feeding rate and growth rate on the ground mushroom substrate were higher than that on the non-ground mushroom substrate. Especially when the ground mushroom substrate was mixed with rice bran and supplied to earthworms, growth rate was much higher than that on the non-ground spent mushroom substrate; it increased 1.85 times.

Development of a new brown button mushroom cultivar 'Hogam' (갈색 양송이 신품종 '호감' 육성)

  • A new brown button mushroom cultivar, 'Hogam', C34 line, was made by crossing homokaryons, ASI1164-37 and ASI1175-66, selected by RAPD analysis and by cultivating three times. Mycelium of 'Hogam' on CDA (compost dextrose agar) media grew well at $25^{\circ}C$. The optimum pin-heading temperature of new variety and optimum growing temperature was $14-18^{\circ}C$. The thickness of the mature cap and stipe were thicker than a control, 'Dahyang' that developed in 2010. The color of pileus was light brown and lighter than 'Dahyang'. Days required from casing to first harvesting were three days longer than control strain, but the weight of harvested fruiting body increased by 1.35 times. 'Hogam' cultivar are expected to contribute to the diversification of domestic mushroom cultivars.
