Isolation and Characterization of an Immunopotentiating Factor from Lactobacillus plantarum in Kimchi: Assessment of Immunostimulatory Activities

  • The immunostimulatory activities of Lactobacillus plantarum, the major microorganism in kimchi fermentations were investigated. Five strains of L. plantarum exhibited weak immunopotentiating activity, but L. plantarum PS-21 showed as strong a mitogenic activity as Bifidobacterium adolescentis M101-4, a known positive strain. It is of interest that, L. plantarum PS-21 stimulated proliferation of Peyer's patch cells, one of the most important tissues in the gut-associated lymphoreticular system. Cell' wall fractions from L. plantarum PS-21 also showed strong mitogenic activity compared with the soluble cytoplasmic fraction. A peptidoglycan fraction (PG) extracted from the cell wall of L. plantarum PS-21 was identified as an active mitogenic component when used in murine lymph node and spleen cell test systems. PG showed dose-dependent mitogenic activity and significantly enhanced antibody production in lymph node cells when studied in vitro. The lysosomal enzyme activity of murine peritoneal macrophages was increased when analyzed following injection of PG to the host animal. Furthermore, PG enhanced the production of cytokines such ($TNF-{\alpha}$ and IL-6) in the in vitro culture of RAW 264.7 macrophage cells.

Rhamnazin inhibits LPS-induced inflammation and ROS/RNS in raw macrophages

  • Purpose: The aim of this work was to investigate the beneficial effects of rhamnazin against inflammation, reactive oxygen species (ROS)/reactive nitrogen species (RNS), and anti-oxidative activity in murine macrophage RAW264.7 cells. Methods: To examine the beneficial properties of rhamnazin on inflammation, ROS/ RNS, and anti-oxidative activity in the murine macrophage RAW264.7 cell model, several key markers, including COX and 5-LO activities, $NO^{\cdot}$, $ONOO^-$, total reactive species formation, lipid peroxidation, $^{\cdot}O_2$ levels, and catalase activity were estimated. Results: Results show that rhamnazin was protective against LPS-induced cytotoxicity in macrophage cells. The underlying action of rhamnazin might be through modulation of ROS/RNS and anti-oxidative activity through regulation of total reactive species production, lipid peroxidation, catalase activity, and $^{\cdot}O_2$, $NO^{\cdot}$, and $ONOO^{\cdot}$ levels. In addition, rhamnazin down-regulated the activities of pro-inflammatory COX and 5-LO. Conclusion: The plausible action by which rhamnazin renders its protective effects in macrophage cells is likely due to its capability to regulate LPS-induced inflammation, ROS/ RNS, and anti-oxidative activity.

3-(4′-hydroxyl-3′, 5′-dimethoxyphenyl) Propionic Acid Suppresses NO Production and Elevates GSH Levels in Murine Macrophages

  • Previous studies have shown that kimchi and kimchi-derived 3-(4'-hydroxyl-3', 5'-dimethoxyphenyl) propionic acid have anti-oxidative and hypolipidemic effects in rats and rabbits. This study was designed to investigate whether chemically synthesized 3-(4'-hydroxyl-3', 5' -dimethoxyphenyl) propionic acid (HDMPPA) may ameliorate oxidative stress through the regulation of nuclear factor KB (NFkB) activation in lipopolysaccharide (LPS)-stimulated RAW 264.7 murine macrophage cells. Treatment of RAW 264.7 cells with 400 uM of HDMPPA significantly reduced LPS-stimulated nitric oxide (NO) production. Treatments with HDMPPA at 100 uM to 400 uM concentrations significantly elevated glutathione (GSH) level. However, cell viability and thiobarbituric acid-reactive substances (TBARS) concentrations were not affected by the concentrations of HDMPPA used. The specific DNA binding activities of NFKB, a transcription factor which is sensitive to oxidative stress, were not down-regulated by HDMPPA treatments. These results suggest that HDMPPA may have weak anti-oxidative activity against LPS challenge by scavenging NO and stimulating GSH production.

Inhibitory effects of a new iridoids, patridoid I and II on TNF, iNOS and COX-2 expression in cultured murine macrophages

  • Possible role of anti-inflammatory effects of a new iridoids, patridoid I. II and II-A which were isolated from Patrinia saniculaefolia. examined by assessing their effects on tumor necrosis factor $\alpha$ (TN F$\alpha$) and 2 enzymes, inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) in the lipopolysaccaride (LPS)-stimulated murine macrophage-like cell line RAW 264.7. Among them. patridoid II consistently inhibited the production of TNF$\alpha$ and NO production in a dose dependent manner. But patridoid I and patrioid ll isomer palrioid ll-A. these compounds very weakly inhibited NO producion. Moreover. treatment of macrophage with these compounds, the decrease in NO products was accompanied by a decrease in iNOS protein level as assessed by Western Blot. But these compounds did not affect COX-2 protein expression in LPS-stimulated macrophage. Our results suggest that patridoid ll could become a leading compound for developing a novel of anti-inflammalory drugs.

  • Herein, we demonstrate that butein (1) can prevent swelling in a murine lymphedema model by suppressing tumor necrosis factor α (TNF-α) production. Butein derivatives were synthesized and evaluated to identify compounds with in vitro anti-inflammatory activity. Among them, 20 µM of compounds 7j, 7m, and 14a showed 50% suppression of TNF-α production in mouse peritoneal macrophages after lipopolysaccharide stimulation. Compound 14a, exhibited the strongest potency with an in vitro IC50 of 14.6 µM and suppressed limb volume by 70% in a murine lymphedema model. The prodrug strategy enabled a six-fold increase in kinetic solubility of compound 1 and five-fold higher levels of active metabolite in the blood for compound 14a via oral administration in the pharmacokinetics study. We suggest that the compound 14a could be developed as a potential therapeutic agent targeting anti-inflammatory activity to alleviate lymphedema progression.

마우스 Macrophage의 IL-1 및 TNF-${\alpha}$의 분비유도에 있어서 한국산 겨우살이 추출물이 미치는 영향 (Effect of Korean Mistletoe Extracts on the Induction of IL-1 and TNF-${\alpha}$ from Mouse Macrophages)

  • To investigate the effect of Korean mistletoe on stimulation of macrophage, the activity to induce interleukine-1(IL-1) and tumor necrosis factors-${\alpha}(TNF-{\alpha})$ from murine peritoneal macrophage by its extracts originated from oak was examined. From in vitro analysis of the cytokines using the culture supernatants of macrophages stimulated with its extracts for 1hr, it was found that Korean mistletoe induces IL-1 and $TNF-{\alpha}$ from murine macrophage. Furthermore, both extracts of Korean mistletoes that were extracted with distilled water and 2% acetic acid exhibited a significant activity to induce two cytokines. In the stimulation for 30 min, Korean mistletoe at concentration of $1{\sim}100\;\mu/ml$ showed a significant induction of IL-1 from macrophage until 24 hrs after stimulation, showing maximal activity on $5{\sim}10\;hrs\;at\;10{\sim}100\;\mu/ml$. On the other hand, $TNF-{\alpha}$ was induced on the early period, 2 hrs, after stimulation at a wide range of concentration, $1{\sim}500\;\mu/ml$. In addition, the fraction of Korean mistletoe from 80% saturated ammonium sulphate precipitation showed a significant activity to induce both cytokines from macrophage. The present study demonstrates that Korean mistletoe contains immunoregulatory factors responsible for stimulating murine macrophage to secrete IL-1 and $TNF-{\alpha}$ which play an important role in immune responses, and suggests that the activity of Korean mistletoe to induce two cytokines is functioned by a possible independent stimulation manner.

Sequential analysis of cell differentials and $IFN-{\gamma}$ production of splenocytes from mice infected with Toxopluma gondii

  • To assess the relationship between the changes of cellular components and the production of Th 1 cytokine in the immune tissue, inbred C57BL/6 mice were orally infected with 40 cysts of 76K strain of Toxoplosma gondii. The sequential change of cell differentials and $IFN-{\gamma}$ production of splenocytes were analyzed by Diff-Quik stain and RT-PCR. There were no significant proportional changes of cellular components of splenocytes until day 4 postinfection (Pl) as compared to those of day 0, and the relative percentage of macrophages and neutrophils/eosinophils increased significantly (p<0.01) thereafter. The expression of $IFN-{\gamma}$ mRNA of $CD3^{-}$ cells was observed from day 1 Pl at a low level. However, $IFN-{\gamma}$ production of $CD3^{+}$ cells increased significantly from day 4 Pl (p<0.01) which progressively increased thereafter. These findings provide the relative percentages of granulocytes and macrophages were increased in conjunction with increase of total number of splenocytes after oral infection with T. gondii in the susceptible murine hosts, and lymphocytes were the major cellular components and the important source of $IFN-{\gamma}$.

  • Hydrogen peroxide is one of major chemicals mediating antitumor and antimicrobial activities of macrophages. We searched natural products enhancing hydrogen peroxide generation from murine macrophage-like cell line J774. Among 21 methanol extracts of Korean medicinal plants, the extract from Cornus officinalis was the most effective. The active component from the fractions was searched by activity guided fractionation, and identified as ursolic acid by spectral data.

Anti-inflammatory Activity of Dichloromethane Extract of Auricularia auricula-judae in RAW264.7 Cells

  • The present study investigated the anti-inflammatory effects of dichloromethane extract of Auricularia auricula-judae. Dichloromethane extract of Auricularia auricula-judae inhibited Lipopolysaccharide (LPS)-induced nitric oxide (NO) production significantly in a dose-dependent manner in the concentration ${\geq}\;10\;{\mu}g/ml$ (p < 0.05). Furthermore, RT-PCR results of this study indicated that the extract markedly reduced the expressions of inflammatory cytokines (IL-6, TNF-$\alpha$ and IL-$1{\beta}$) mRNA in LPS-treated murine RAW 264.7 macrophages, which could possibly ameliorate the inflammation. Nevertheless, dichloromethane extract of Auricularia auricula-judae did not show complete inhibition of IL-6 mRNA expression. The inhibition of IL-$1{\beta}$ cytokine at protein level was also observed in a dose dependent manner. In conclusion, the current study revealed the previously unknown effect of dichloromethane ethyl extract of Auricularia auricula-judae inhibitions of the production of NO, IL-6, TNF-$\alpha$ and IL-$1{\beta}$ in LPS-stimulated macrophages.

귀비탕이 Stress 부하 후 혈중 호르몬 및 비특이적 면역반응에 미치는 영향 (Effects of Kwibi-tang on Serum Levels of Hormone and the Non-Specific Immune Response after Immobilization Stress in Mice)

  • To investigated the effects of Kwibi-tang water extract (KBT) on the non-specific immune response in C57BL/6 mice stressed by immobilization, we evaluated the changes in the contents of serum histamine and corticosterone and the phagocytic activity of macrophages. The level of serum histamine and corticosterone was determined with spectrofluorometer. The cell viability was determined by a MTT assay method. The subpolpulation of lymphocytes was determined by a flow cytometry. The phagocytic activity was determined with luminometer. KBT decreased the serum level of histamine and corticosterone increased by immobilization stress. Also, KBT enhanced the phagocytic activity and decreased the level of nitric oxde in murine peritoneal macrophages decreased by immobilization stress. These results indicate that KBT may be useful for the prevention and treatment of stress via suppression of serum histamine and corticosterone level and enhancement of the non-specific immune response.
