A Cyan Fluorescent Protein Gene (cfp)-Transgenic Marine Medaka Oryzias dancena with Potential Ornamental Applications

  • To evaluate their potential utility as an ornamental organism, novel transgenic marine medaka Oryzias dancena strains with a highly vivid fluorescent phenotype were established through transgenesis of a cyan fluorescent protein gene (cfp) driven by the endogenous fast skeletal myosin light chain 2 gene (mlc2f) promoter. The transgenic marine medaka strains possessed multiple copies of transgene integrants and passed their fluorescent transgenes successfully to subsequent generations. Transgenic expression in skeletal muscles at both the mRNA and phenotypic levels was, overall, dependent upon transgene copy numbers. In the external phenotype, an authentic fluorescent color was dominant in the skeletal muscles of the transgenic fish and clearly visible to the unaided eye. The phenotypic fluorescent color presented differentially in response to different light-irradiation sources; the transgenics displayed a yellow-green color under normal daylight or white room light conditions, a strong green-glowing fluorescence under ultraviolet light, and a cyan-like fluorescence under blue light from a light-emitting diode.

Comparative Analysis of the Growth for Selectively Bred Korean Turbot Scophthalmus maximus Population (한국 육종터봇(Scophthalmus maximus)의 성장률 비교 분석)

  • Turbot Scophthalmus maximus can be cultured at a higher density and has a faster growth rate than olive flounder Paralichthys olivaceus, making it an attractive alternative for flounder farmers in Korea. As it is a relatively new species to Korean farming environments, there is no general farming protocol, which takes multiple generations to develop, for turbot in Korea. Nevertheless, we applied breeding technology to develop a stock, which we considered first-generation turbot. Then we compared its growth rate (an important economic trait) to different populations (the original stock of Korean turbot, French turbot, and Chinese turbot). The four different populations were grown in similar culture conditions. First-generation Korean turbots grew about 28% faster than all others. We will continue to select for important traits such as color, disease resistance, and heat tolerance in this turbot broodstock; we believe that with the appropriate selection process, Korean turbot should become competitive in the markets, and will become a major farming species in the Korean aquaculture industry.

The Effect of Sub-division (Two or Three Sub-populations) of a Population on Genetic Gain and Genetic Diversity

  • Breeding efficiencies were compared among three population schemes: a single population, a population with two subpopulations and a population with three sub-populations. A simulation experiment of selection was carried out for 10 generations with 20 replications each by comparing average breeding values and inbreeding coefficients among the three population schemes. Phenotypes of three traits were generated with a model comprising 36 loci, each with additive genetic effects and residuals distributed normally. Among the three population schemes, the single population scheme was definitely superior to the other two with regards to selection response and inbreeding. The multiple sub-population scheme was, however, considered to be an alternative population scheme when the difference in economic weights of the traits was small among the sub-populations, assuming moderate inbreeding depression for traits and crossbreeding. The scheme with two sub-populations had a higher genetic value than that with three subpopulations; however, the genetic values of the schemes were comparable when maternal heterosis was taken into account. The choice of population schemes may depend on the cost-sharing policy between the breeding population and the commercial population rather than just the breeding efficiency.

  • Objectives : This study aims to : (i) characterize and differentiate between two different periods of baby boomers, (ii) study the utilization of their health care services, and (iii) establish effective ways of providing better health service utilization and preventive policy strategies for upcoming and older generations. Methods : A multiple regression analysis using descriptive statistics, frequency analysis, and dummy variables was utilized to access the presence of correlations between socio-demographic factors and health care service utilization. Results : Medical insurance type, marital status, and chronic disease were factors that influenced health care service utilization. Furthermore, the factors that influenced individual medical expenses were cohabitation, inpatient days, and chronic disease. Conclusions : Primary findings and exploratory statistics revealed that there were strong correlations and interaction among some of the predictor variables. Because of the chronologically limited nature of the sample data set gathered in 2012, it would be helpful to continue to develop or research related constructs that may capture relationships more effectively among extended populations.

  • The recent 3D printing industry has been focusing on developing 3D printers to fulfill the user's need to bring more colorful and realistic outcomes. Several 3D printers have deployed multiple extruders to print different colors. However, this method has a limit on its availability of colors. To solve this, recent research is focused on using mixing chambers to mix the possible colors to provide more color availability. In this paper, we discuss the environment and algorithm behind the development of G-code which allows a gradation effect of the mix of two filaments. The generation algorithm to make gradient G-code has been implemented in Cura using C ++ and Python.

  • Islanding phenomenon of photovoltaic system is undesirable because it leads to a safety hazard to utility service personnel and may cause damage to power generation and power supply facilities as a result of unsynchronized re-closure. Anti-islanding protection is an important technical requirement for grid-connected PV system. Until now, various anti-islanding methods for detecting and preventing islanding of photovoltaic and other distributed generations have been proposed. Most of them are focusing on the anti-islanding performance of single PV system according to the related international and domestic standard test procedures. There are few studies on the islanding phenomenon for multiple photovoltaic operation in parallel. This paper presents performance analysis of anti-islanding function for grid-connected PV inverter systems when several PV inverters are connected in parallel.

Investigation into Whether Agronomic Traits Are Fixed for the Breeding of Drought Tolerance and BPH-resistant Crosses (내건성 및 BPH 내성 계통의 육성을 위한 농업형질 고정여부 조사)

  • Late in the 1960s, South Korea established cultivation technology and began breeding new "Tongil" type rice cultivars by crossing indica and japonica. To date, this cultivation technique has been used to produce a wide variety of cultivars to meet consumer preferences and adapt to extreme weather conditions. Once major genetic traits are no longer segregated through advanced generations, varieties obtained from a cross between different crops may become a new variety. In this study, we confirmed the agronomic traits of F4, F5, and F6 by advanced generation a population in which drought tolerance and brown planthopper (BPH) resistance were crossed. HV23, Ilmi/HV23, and Drimi2ho/HV23 were used. HV23 was drought-tolerant, and Drimi2ho was resistant to BPH. As a result, it was possible to consider that the agronomic traits were fixed because none of them showed a significant difference from the others. While it takes more than 10 years for the pedigree method of breeding, this study confirmed that the agronomic traits were fixed in 4-6 years. In the future, we will investigate the homology of the CaMsrB2 gene and the Bph1 gene, to confirm that both genes are closely related to each other, and analyze the stable inheritance of the introduced gene for multiple successive generations.

Estimation of Genetic Gains in Commercial Layer Breeding Population (상업용 난용종계 집단에서의 유전적 개량량 추정에 관한 연구)

  • This study was carried out to investigate the effectiveness of the selection for commercial populations of layer chickens subjected to multiple objectives. For that purpose, predicted selection responses obtained from estimated genetic parameters and observed values were compared. Data for two layer pure lines, Line-w(Single Comb White Leghorn) and Line-B (brown layer) which have been maintained at the Mani Breeding Farm were collected from 1980 to 1985 during 5 generations. The annual genetic gains were estimated by the moth of selection index in retrospect on the basis of selection differentials, phenotypic and genetic variance-covariance for the traits considered. SM was found to be shorten by 1.67 days and 1.14 days per generation in Line-W and Line-B. EN300 and EN400 were found to be increased by 4.15 and 5.92 per generation in Line-W and 257 and 3.73 per generation in Line-B. The annual gains realized were found to be significantly different from expected gains predicted on the basis of the annual selection differentials and genetic parameters in later generations, but it could not be concluded that the differences were attributed to genetic effects.

A Novel COMP Gene Mutation in a Korean Kindred with Multiple Epiphyseal Dysplasia

  • Multiple epiphyseal dysplasia (MED) is a clinically and genetically heterogeneous chondroplasia, characterized by delayed development of the ossification centers and, deformities of the extremities that involve only the epiphysis and result in mild short stature. Mutations in the cartilage oligomeric matrix protein (COMP) gene are most commonly found, and most of the mutations are located in the calmodulin-like repeats and the C-terminal domain. We report a Korean kindred of 12 family members with MED in four generations who were found to have a novel mutation in the COMP gene. A pedigree showed early onset osteoarthritis requiring arthroplasty that was an autosomal dominant inherited trait. Radiological examinations demonstrated the presence of osteochondral defects in the medial femoral condyles, and the knee and hip joints showed variable degrees of precocious degenerative changes. Mutation analysis of the COMP gene in the proband and five other affected family members identified a novel missense mutation, c.1280G>C (p.Gly427Ala) in exon 12, which was not found in three unaffected family members. Direct sequencing of the COMP gene may yield pathogenic mutations in dominantly inherited MED cases, and may provide opportunities of carrier detection among high-risk family members, leading to genetic counseling for early diagnosis and intervention before the onset of complications.

A Case of Retronychia on Toenail (발톱에 발생한 역행조갑 환자의 치험례)

  • Purpose: Retronychia is the embedding of the nail into proximal nail fold. Retronychia starts with disruption of the longitudinal growth of the nail. With the growth of a new nail, the old one is pushed upwards and backwards. This leads to embedding of the top nail into the ventral aspect of the proximal nail fold and results with chronic paronychia. We present a case of retronychia that was rarely reported in the literature. Methods: A 46-year old female presented with a 3-month history of painful right first, 2nd, 3rd toenail changes. Although she was initially treated with broad spectrum antibiotics, she did not response to therapy. Later, she presented to our department because of progressively worsening pain that impaired her walking. Physical examination revealed with proximal nail fold erythema, painful swelling, yellowish nail discoloration, and distal onycholysis. Bacterial and fungus culture showed no organism. Treatment was surgical nail avulsion under local anesthesia. Results: The postoperative course was uneventful. 10 months later, the patient had a normal growing nail and was free of symptoms. Conclusion: We report a case of retronychia on toenail. Retronychia is a proximal nail plate ingrowth into the proximal nail fold which is associated with multiple generations of nail plate misaligned beneath the proximal nail. Management consisted of simple avulsion of superimposed nail. Retronychia is suspected with a persistent paronychia, particularly in the setting of trauma. Avulsion of the top nail confirms the diagnosis and may be curative if the underlying nail appears healthy.
