Studies on the Effect of Cadmium on the Physiology of Silkworm, Bombyx mori L. I. With Reference to the Development of the Various Organs and Tissues (카드미윰이 누에의 생리에 미치는 영향 I. 누에의 각 조직기관의 발육과 카드미윰의 분포량)

  • Mulberry leaf sprayed on at the various concentrations of cadmium (Cd) was fed on the just molted fifth instar through the 6th day of the fifth instar and content of cadmium and zinc in the feces, organs and tissues involved were analyzed by spectrophotometer. Along with the effect of those heavy metals on development of the relevant organs and tissues sexwise. 1. The residual content of cadmium on mulberry leaf was about 50% of the total cadmium and the weight of feces disposed from the cadmium fed larvae was less than the control. There was distinctive differences in female-wise disposal of feces between cadmium treatment and control ; cadmium treatment female larvae disposed less feces than female of control. 2. Fourteen percent of the total dry weight of the anterior plus the middle silkgland and of the posterior silkgland in cadmium 100ppm treatment was decreased in female but only 9% of the anterior plus middle silkgland in male was made, showing no decrease the weight of the posterior silkgland in male. 3. Decrease of the dry weight of the silkgland, fat body and hindgut was made in cadmium 100ppm treatment and contrarily the weight of midgut and malphihian tube were increased. The total larval weight of the female and male were decreasd in cadmium 100ppm treatment and the decreasing ratio was higher in famale. 4. There wasa no detection of cadmium in all of the organs and tissues of the control and in those of the cadmium treatment the high residual content was in order of the midgut, hindgut, malphighian tube and head. A slight accumulation of cadmium was shown in fat body and silkgland. 5. Cadmium accumulation in the midgut was higher than in female but in the fat body, malphighian tube, trachea and integument was contrarily high in the female than male. 6. The total zinc content of the larvae was below 60ppm in the control and 80ppm in cadmium 100ppm treantment. The increase of zinc content in cadmium treatment was made on silkgland, malphigian tube and midgut but contrarily the decrease of zinc content in fat body was resulted. 7. In cadmium treatment, much more cadmium accumulation took place in the female and zinc accumulation was made much more in the male.

Purification and Biological Activity of Ecdysterone from Korean Achyranthes radix (韓國産 牛膝의 Ecdysterone 抽出과 그 生理活性에 관한 硏究)

  • It has been known that the insect molting hormone and its analogues exist also in plant kingdom and their concentration has been found to be about 0.1~2.0% of dry matter, which is equivalent to $10^3{\sim}10^5$ times of those in insects. This study was carried out; 1) to isolate the phytoecdysones from Korean Achyranthes radix and characterize their physico-chemical properties. 2) to investigate the biological activity of this phytoecdysone on Bombyx mori larvae. The resuls were summarized as follows; 1. The extraction method of phytoecdysones was optimized by three consecutive reflux for 1hr using 200g of dried and milled radix per 1l methanol. 2. The purification from the crude extract was made by a series of steps such as precipitation of gum-type polymer with n-Butyl acetate, adsorption on technical grade silica and chromatography with neutral alumina. The conditions of each step were optimized and the resulting crude crystal was about 500mg per kg dry radix. 3. The crude crystal from the cultivated Achyranthes(Achyranthes japonia) contained ecdysterone (20-hydroxyecdysone) and inokosterone in the proportion of one to one. In order to separate these, a series of processes such as acetylation, separation by alumina column chromatography deacetylation by alcoholysis, deionization and crystallization were introduced and optimized 125mg of ecdysterone and 18mg of inokosterone per kg dry radix were thus obtained. 4. The wild Achyranthes (Achyranthes obtusifolia) radix was found to contain the ecdysterone only. A 285mg of ecdysterone was crystallized per kg dry radix. 5. Isolated ecdysterone, inodosterone and acetylated compounds were characterized by IR., UV., NMR spectroscopy, mp, TLC and densitometry. 6. Ligation experiment was undertaken to confirm the biological activity of the purified ecdysterone; the ecdysterone could induce larval-pupal metamorphosis in the ligated abdomen of 4th instar larvae injecting 0.5~1.0${\mu}g$. 7. By ecdysterone feeding experiment using artificial diet, it was elucidated that the critical time of feeding would be the first half of each instar resulting in increased weight of silk layer. 8. The ecdysterone was fed to 5th instar silkworm at the level of 1, 2, 3, 5ppm of dry feed of artificial diet containing 5% mulberry leaves for 72hrs. At 2ppm of the concentration. body weight and silk layer weight were arrived at maximum. But at higher concentrations body weight and silk layer weight decreased than the control group. At 2ppm of the concentration, body weight was increased by 12.5%. 9. Feeding 2ppm of ecdysterone at the later half of 5th instar, the duration of larvae was shortened.

Effects of Level of Dietary Protein or Carbohydrate on the Economic Characters and Amylase Activity of the Silkworm, Bombyx mori (인공사료의 단백질 및 탄수화물 수준이 가잠의 실용형질과 Amylase활성에 미치는 영향)

  • A series of experiment was conducted to investigate the effects of the level of dietary Protein (soybean meal) and carbohydrate (sucrose) on the growth rate, feed efficiency, nutrients digestibility and amylase activity of larvae at 5th instar. The results obtained from the experimentation are summarized as follow: (1) It was found that the body weight gain, cocoon quality and feed efficiency of the 5th instar larvae were significantly affected by the level of protein and carbohydrate in the artificial diet. Present data revealed that the proper level of dietary protein and carbohydrate would be the most important factor for the optimum growth of larvae. (2) In the amount of diet consumption, the group of larvae gained more body weight consumed more diet than the group gained less amount of body weight. (3) Digestibility of nutrients was improved as the level of protein in the diet increased, although the level of dietary carbohydrate did not exert any positive results. (4) It was observed that the amylase activity in digestive juice was stimulated by the increasement of dietary carbohydrate. However, no correlation between the level of dietary protein and carbohydrate was detected in the amylase activity. (5) Analytical data indicated that the amount of protein or carbohydrate excreted through feces and urine was proportional to the content of protein or carbohydrate in artificial diet. The group o( larvae consumed the diet contained high protein excreted more protein and less nitrogen free extract than did the group received low protein diet. Likewise, the group of larvae fed high dietary carbohydrate excreted more nitrogen free extract and less protein through excreta than the group consumed low carbohydrate diet group. (6) Although the amylase activity of the larvae received the artificial diet was higher than the larvae received natural diet (mulberry leaf), the rate of body weight gam and the quality of cocoon produced from the natural diet group was better than the artificial diet group. It is, thus, concluded that feeding the larvae by the artificial diet may net be recommendable. (7) It was determined that the best level of protein (soybean meal) and carbohydrate (sucrose) in artificial diet was found to be 40g and 0g, respectively when it was fed to the 5th instar larvae. (8) It may be concluded from the results obtained that the artificial diet that could stimulate the activity of amylase nay not be recommendable for the practical larvae feeding Purpose due simply to the Poor economic return from this diet than natural diet.

Effect of the Meteorological Conditions on the Cocoon Production in silkwom, in Kyungsang-Nam-Do, Korea (제기상조건이 잠견생산량에 미치는 영향(경상남도를 중심으로))

  • The statistical interpretations are on the correlations and the linear regressions between the various meteorological conditions the branch Pusan-Observatory of the National-Central-Meteorological-Observatory had measured from 1930 to 1962, and the production of silk worm cocon that the Section of Sericulture in the Ministry of Agriculture & Fishery had recorded from 1930 to 1962 in Gyung Sang Nam Do Korea. These were most in proportion to the sun shine time, an inverse proportion to the atmosphere humidity and the lowest temperature, in proportion to the mean temperature, the highest temperature and the amount of the rain fall. The correlations and linear regressions between the various meteorological factors to control the production of the silkworm cocoon of both per a box of the silk worm egg and per 10a of mulberry field was as follows. (1) The production of the spring silk worm cocoon was dicided by the various meteorological factors. 1. This was in proportion to the total sun shine time between 5 and 6 month. r=0.40*-(0.42$^{**}$) Y=0.0269$\chi$+6.46-(0.2795$\chi$+10.25) 2. This was an inverse proportion to the total amount between 5 and 6 month atmosphere humidity. r=-0.49$^{***}$-(-0.40*) Y=-0.2989$\chi$+62.77-(0.1474$\chi$+31.86) 3. This wasin proportion to the total rain fall amount between 6 and 7 month. r=0.47$^{***}$-(0.44$^{***}$) Y=0.0037$\chi$+16.42-(0.0035$\chi$+8.35) (2) The production of the autumn silk worm cocoon was decided by the various meteorological factors. 1. Studies on the Nature and Pathogenicity of Nuclear Polyhedrosis Virus of the Fall Webworm, Hyphantria cunea (Drury) (흰불나방 핵다각체병바이러스의 성상과 병원성에 관한 연구)

  • An inclusion forming virus isolated from a fan webworm, Hyphantria cunea, in 1975 was identified as a nuclear polyhedrosis virus. With the virus isolated in Korea, it was considered that the virus would be one of the valuable microorganism in microbial control. In this connection, 1) the shape and size of the virus for identification, 2) susceptibility of the various instar larvae to the virus, 3) the effects of storage condition on the pathogenicity and the cross infection of the virus to the larvae of Bombyx mori were examined. The results are summarized as follows; 1. The polyhedron was tetrahedron or hexahedron of $2\mu$ in size and the rod-shaped virus particles consisting of $2\~14$ rods in a bundle were $330m{\mu}\times35m{\mu}$ in size. 2. The hexagonal nuclear polyhedra were found only in the nucleus of the midgut cells but were variable in size. 3. The $LD_{50}$ values for the various instar larvae of H. cunea were $8.377\times10^4\;PIBs/ml$ for the second, $4.974\times10^5\;PIBs/ml$ for the fifth instar larvae. The $LT_{50}values$ for $10^6\;PIBs/ml$ were 9.6 days for the second, 11.5 days for the third, 12.0 days for the fourth and 17 days for the fifth instar larvae. 4. The susceptibility of H. cunea to the nuclear polyhedrosis virus was greater in the first generation than in the second generation. 5. The effect of the storage conditions on the pathogenicity of the nuclear polyhedra was less in refrigerator $(5^{\circ}C)$ and in freezing $(-80^{\circ}C)$ than in room temperature $(18.5^{\circ}C)$, especially as air-dried polyhedra than as suspension. The pathogenicity of the polyhedra seemed to decrease by sunlight during storage as cadavers, since rather greater decrease in pathogenicity was found in sunny condition than in shady condition. 6. The effective spray concentration was $6.4\times10^7\;PIBs/ml$ in the field and its $LT_50$ values for the third and the fifth instar larvae were 4.8 days and 14.2 days, respectively. 7. No cross infections were found in the nuclear polyhedrosis virus between H. cunea and B. mori. larvae.

  • $Nguy{\tilde{\hat{e}}}n$ Cư Trinh has two faces in the history of territorial expansion of Vietnam into the Mekong delta. One is his heroic contribution to the $Nguy{\tilde{\hat{e}}}n$ family gaining control over the large part of the Mekong delta. The other is his role to make the eyes of readers of Vietnamese history be fixed only to the present territory of Vietnam. To the readers, $Nguy{\tilde{\hat{e}}}n$ Cư Trinh's achievement of territorial expansion was the final stage of the nam $ti{\acute{\hat{e}}n$ of Vietnam. In fact, however, his achievement was partial. This study pays attention to the King $V{\tilde{o}}$ instead of $Nguy{\tilde{\hat{e}}}n$ Cư Trinh in the history of the territorial expansion in the Mekong delta. King's goal was more ambitious. And the ambition was propelled by his dream to build a new world, and its order, in which his new capital, $Ph{\acute{u}}$ $Xu{\hat{a}}n$ was to be the center with his status as an emperor. To improve my assertion, three elements were examined in this article. First is the nature of $V{\tilde{o}}$ Vương's new kingship. Second is the preparation and the background of the military operation in the Mekong Delta. The nature of the new territory is the third element of the discussion. In 1744, six years after this ascending to the throne, $V{\tilde{o}}$ Vương declared he was a king. Author points out this event as the departure of the southern kingdom from the traditional dynasties based on the Red River delta. Besides, the government system, northern custom and way of dressings were abandoned and new southern modes were adopted. $V{\tilde{o}}$ Vương had enough tributary kingdoms such as Cambodia, Champa, Thủy $X{\tilde{a}}$, Hoả $X{\tilde{a}}$, Vạn Tượng, and Nam Chưởng. Compared with the $L{\hat{e}}$ empire, the number of the tributary kingdoms was higher and the number was equivalent to that of the Đại Nam empire of the 19th century. In reality, author claims, the King $V{\tilde{o}}^{\prime}s$ real intention was to become an emperor. Though he failed in using the title of emperor, he distinguished himself by claiming himself as the Heaven King, $Thi{\hat{e}}n$ Vương. Cambodian king's attack on the thousands of Cham ethnics in Cambodian territory was an enough reason to the King $V{\tilde{o}}^{\prime}s$ military intervention. He considered these Cham men and women as his amicable subjects, and he saw them a branch of the Cham communities in his realm. He declared war against Cambodia in 1750. At the same time he sent a lengthy letter to the Siamese king claiming that the Cambodia was his exclusive tributary kingdom. Before he launched a fatal strike on the Mekong delta which had been the southern part of Cambodia, $V{\tilde{o}}$ Vương renovated his capital $Ph{\acute{u}}$ $Xu{\hat{a}}n$ to the level of the new center of power equivalent to that of empire for his sake. Inflation, famine, economic distortion were also the features of this time. But this study pays attention more to the active policy of the King $V{\tilde{o}}$ as an empire builder than to the economic situation that has been told as the main reason for King $V{\tilde{o}}^{\prime}s$ annexation of the large part of the Mekong delta. From the year of 1754, by the initiative of $Nguy{\tilde{\hat{e}}}n$ Cư Trinh, almost whole region of the Mekong delta within the current border line was incorporated into the territory of $V{\tilde{o}}$ Vương within three years, though the intention of the king was to extend his land to the right side of the Mekong Basin beyond the current border such as Kampong Cham, Prey Vieng, and Svai Rieng. The main reason was $V{\tilde{o}}$ Vương's need to expand his territory to be matched with that of his potential empire with the large number of the tributary kingdoms. King $V{\tilde{o}}^{\prime}s$ strategy was the variation of 'silkworm nibbling' and 'to strike barbarians by barbarians.' He ate the land of Lower Cambodia, the region of the Mekong delta step by step as silkworm nibbles mulberry leave(general meaning of $t{\acute{a}}m$ thực), but his final goal was to eat all(another meaning of $t{\acute{a}}m$ thực) the part of the Mekong delta including the three provinces of Cambodia mentioned above. He used Cham to strike Cambodian in the process of getting land from Long An area to $Ch{\hat{a}}u$ Đốc. This is a faithful application of the Dĩ Man $C{\hat{o}}ng$ Man (to strike barbarians by barbarians). In addition he used Chinese refugees led by the Mạc family or their quasi kingdom to gain land in the region of $H{\grave{a}}$ $Ti{\hat{e}}n$ and its environs from the hand of Cambodian king. This is another application of Dĩ Man $C{\hat{o}}ng$ Man. In sum, author claims a new way of looking at the origin of the imperial world order which emerged during the first half of the 19th century. It was not the result of the long history of Đại Việt empires based on the Red River delta, but the succession of the King $V{\tilde{o}}^{\prime}s$ new world based on $Ph{\acute{u}}$ $Xu{\hat{a}}n$. The same ways of Dĩ Man $C{\hat{o}}ng$ Man and $T{\acute{a}}m$ Thực Chi $K{\acute{\hat{e}}}$ were still used by $V{\tilde{o}}^{\prime}s$ descendents. His grandson Gia Long used man such as Thai, Khmer, Lao, Chinese, and European to win another man the '$T{\hat{a}}y$ Sơn bandits' that included many of Chinese pirates, Cham, and other mountain peoples. His great grand son Minh Mạng constructed a splendid empire. At the same time, however, Minh Mạng kept expanding the size of his empire by eating all the part of Cambodia and Cham territories.
