백강균(Beauveria bassiana)을 이용한 식물병원성세균 생장억제효능 검증 (Effects of the growth inhibition against plant pathogenic bacteria using Beauveria bassiana)

  • 뽕나무병원성세균에 대한 Beaveria bassiana 6균주의 항균활성을 paper disc법으로 조사하였고 최소성장저해농도(MIC)를 액체배지희석법으로 조사하였다. 항균활성은 B. bassiana J200에서만 나타났으며, Erwinia rhapontici KACC 10407에서는 13mm, Pseudomonas syringae KACC 10390와 Xanthomonas campestris KACC 12134에서는 각각 17 mm의 항균활성이 나타났다. MIC 측정 결과 E. rhapontici KACC 10407, P. syringae KACC 10390 및 X. campestris KACC 12134 모두 4.0%에서 MIC를 보였다. 이상의 결과로 B. bassiana의 뽕나무병원성세균에 대한 생물학적 방재제로서의 이용 가능성을 확인하였다.

Mulberry Low-Fat Ice Cream Supplemented with Synbiotic: Formulation, Phytochemical Composition, Nutritional Characteristics, and Sensory Properties

  • For this study, we designed and produced mulberry low-fat ice cream supplemented with synbiotics (MLF-ISS). The sensory characteristics and physical, chemical, and microbiological qualities of MLF-ISS were then determined. Mulberry juice inoculated with or without probiotic (Lactobacillus plantarum TISTR 926 and Saccharomyces boulardii CNCM I-745) was also tested at 37℃ for 24 h to determine probiotic growth rate, pH, total anthocyanin content (TAC), total phenolic content (TPC), and antioxidant activity (AA). Only the TAC of mulberry juice inoculated with S. boulardii CNCM I-745 increased considerably (p < 0.05) among these parameters. MLF-ISS was produced with varied mulberry fruit concentrations (0, 10, 20, 30, or 40%) (w/w). The MLF-ISS prepared with 30% mulberry fruit (w/w) (30-MLF-ISS) had a higher score in appearance, color, and sweetness (p < 0.05) when sensory qualities were measured using the 9-point hedonic scale method. In the CIE lab system (L*, a*, b*), the color values of 30-MLF-ISS were 27.80 ± 0.26, 12.99 ± 0.59, and 1.43 ± 0.05, respectively. The 30-MLF-ISS was also subjected to a proximate analysis. The melting rate of 30-MLF-ISS was 0.29 ± 0.03 g/min and the time it took for the first drop to fall was 37.00 ± 7.00 min. TAC, TPC, and AA of 30-MLF-ISS were observed to alter significantly (p < 0.05) during varied intervals of storage at - 18℃ (0, 30, and 60 days). The viability of probiotics in 30-MLF-ISS slightly decreased after storage at -18℃ for 8 weeks, but remained about 6 log CFU/g. During storage at -18℃ for 0 and 120 days, no pathogenic bacteria were detected in 30-MLF-ISS. These findings show that 30-MLF-ISS has nutritional and functional value, is free of foodborne pathogenic bacteria, is safe for consumers' health, and is suitable for application in the ice cream and related food industries.

뽕잎 장아찌로부터 분리된 Lactobacillus plantarum 균주의 유해균 증식 억제 활성 (Antipathogenic Activity of Lactobacillus plantarum Isolated from Pickled Mulberry Leaf)

  • 전국 각지에서 수집된 발표식품으로부터 젖산균을 분리하고 유해균인 B. cereus, L. monocytogenes, S. Typhimurium, S. aureus 및 S. epidermidis 균주에 대하여 생육 억제효능이 강한 L2167 균주를 선발하고, 16S rDNA 분석법을 이용하여 L. plantarum로 동정하였다. L2167 균주의 단백질 분획물을 제작하여 S. aureus 등 5종의 유해균 배양액에 첨가한 후, 전자현미경으로 관찰하였다. L. plantarum L2167 균주의 균체 단백질 분획물은 유해균의 생육을 억제하였으며, 생육 억제활성은 균체 내부에 함유된 단백질이 유해균의 세포막을 파괴하는 것에서 기인하는 것으로 추정되었다. L. plantarum L2167은 pH 5.5로 조절한 MRS 배지에서 35℃로 배양하였을 때, 가장 우수한 비성장속도를 나타냈으며, 대조구로 사용된 L. plantarum KCTC21004보다 NaCl에 대하여 민감한 특징을 나타내었다. 본 연구를 통하여 선발한 L. plantarum L2167 균주는 추가연구를 통하여 유해균의 생육을 효율적으로 억제할 수 있는 스타터 균주로서 개발될 수 있을 것으로 기대된다.

Investigation on Antibacterial and Antioxidant Activities, Phenolic and Flavonoid Contents of Some Thai Edible Plants as an Alternative for Antibiotics

  • This study was aimed to examine the antibacterial and antioxidative properties of seven edible plants from Thailand to develop alternative antibiotics as feed additives. The plants include Citrus aurantifolia Swingle (Lime) fruits and its leaves, Sesbania grandiflora L. (Agati sesbania) leaves, Piper sarmentosum Roxb (Wild betal) leaves, Curcuma domestica Valeton (Turmeric) roots, Morinda citrifolia L. (Beach mulberry) leaves, Cassia siamea britt (Siamea cassia) leaves, and Cocos nucifera L. (Coconut) peels. The plants were extracted by methanol, n-hexane, chloroform, ethyl acetate, butanol and water. Antibacterial activities with minimum inhibitory concentration (MIC) were determined by agar diffusion assay against Escherichia coli, Burkholderia sp., Haemopilus somnus, Haemopilus parasuis, and Clostridium perfringens that were considered pathogenic strains in livestock infection. Methanol extracts of C. aurantifolia Swingle fruits and leaves showed the broadest spectrum of antibacterial activities except for C. perfringens. Butanol extract of S. grandiflora L. leaves showed the strongest activity against Burkholderia sp. with MIC, $135{\mu}g/mL$. P. sarmentosum Roxb leaves showed antibacterial activities against E. coli, Burkholderia sp. and H. parasuis. Ethyl acetate and water extracts from C. domesitca Valeton roots showed MIC of $306{\mu}g/mL$ and $183{\mu}g/mL$, respectively against only C. perfringens. Antioxidative activity was determined by 2-diphenyl-2-picryl hydrazyl photometric assay. The methanol extracts of C. aurantifolia Swingle fruits and P. sarmentosum Roxb leaves showed the highest antioxidant activity among all the extracts with 3.46 mg/mL and 2.70 mg/mL effective concentration 50% ($EC_{50}$) values, respectively. Total contents of phenolics and flavonoids were measured from the plant extracts. Methanol extracts of S. grandiflora L. and chloroform extracts of C. domestica Valeton were found to have the highest amount of total phenolics, 41.7 and $47.8{\mu}g/mL$, respectively. Flavonoid content of methanol extracts in S. grandiflora L. T was $22.5{\mu}g/mL$ and the highest among plant extracts tested. These results indicated that C. aurantifolia Swingle, S. grandiflora L., P. sarmentosum Roxb, and C. domestica Valeton have antibacterial and antioxidant activities and can be used as alternative antibiotics or potential feed additives for the control of animal pathogenic bacteria.
