Survey and analysis of mulberry tree for mulberry production (오디 생산용 뽕나무 재배 현황 조사)

  • Though mulberry fruit is known to a by-product that was produced from mulberry tree after harvesting leaves for silkworm rearing, as a yield and consumption of mulberry fruit was increased, it has been fixing to a new income crop. But, a stable production of mulberry was issued on damage by the weather disaster, so it is requires that agricultural disaster insurance import. So, in this study as a basic research for a importation of agricultural disaster insurance, the cultivation of mulberry for produce mulberry fruit, the shape of tree, the number of a bearing fruit from each cultivar, the occurrence of mulberry fruit sclerotic disease, and a yield and the market price of mulberry fruits were investigated and that results obtained were as follows; The number of mulberry tree each 10a was abundant in Buan and Yangpyung region that cultivated a suitable cultivar on low cut shape, however, a large cultivated Gochang region was little. The number of a bearing fruit each the longest branch was abundant in the order of Gwasang No. 2, Suwonppong, Iksuppong, and Cheongilppong. Yield of mulberry fruit each 10a was abundant in the order of Cheongilppong, the second number of Gwasang No. 2, Suwonppong, Iksuppong, and yield of mulberry fruit each the age of three mulberry tree was abundant in older tree, however, the second number of Gwasang No. 2 was the opposite. Sale price of the mulberry fruit was highest in Gyeonggi Yangpyeong, and Jeonbuk Gochang was the opposite.

Pheophytin Content and Fibrinolytic Activity of Silkworm Feces in the Different Larval Stages of Silkworms

  • In order to find potential anticancer agents, we extracted pheophytin in the silkworm feces from various larval stages by water, chloroform and methanol extraction. The cytotoxicity of the pheophytin extracts of various silkworm feces was measured in the CT-26 cells originated from murine metastatic colon cancer, by dye uptake assay. The cytotoxicity of those pheophytins in 2nd, 3rd and 4th instars was better than remaining larval stages. The in vitro anticoagulant and fibyinolytic activities of ethanol extract from varietal mulberry leaves, mulberry branches and silkworm feces and pheophytin extracts from silkworm feces obtained at various larval stages were evaluated in order to find effective therapeutic drugs for the treatment of myocardial and cerebral thrombosis. The fibrinolytic activity was tested using the activated partial thromboplastin time (APTT) and thrombin time (TT) was measured for blood clotting activity. With regards to the fibrinolytic system, ethanol extracts of silkworm feces were better than varietal mulberry leaves and mulberry branches. The pheophytin extracts from 7th days of 5th instar contained the highest percentage of pheophytin and good fibrinolytic activity.

Antioxidative and Physiological Activities of Traditional Korean Teas (한국 전통차의 생리활성 및 항산화작용)

  • This study was investigated the antioxidative and antimicrobial activities of a water extract (70$^{\circ}C$) of traditional Korean teas (rose, chrysanthemum, pine needles, mulberry, persimmon leaves, and green tea). Total phenol contents in rose, chrysanthemum, pine needles, mulberry, persimmon leaves, and green tea were 272.8, 74.6, 153.5, 73.5, 69.5, and 260.8 mg tannic acid/g extract, respectively, whereas total flavonoid contents were 75.1, 47.8, 26.8, 40.0, 27.9, and 99.5 mg quercetin/g extract, respectively. The order of electron donating abilities of Korean traditional tea water extracts (1 mg/mL) were : rose (96.8%) > green tea (95.3%) > pine needles (71.3%) > chrysanthemum (36.8%) > mulberry (28.9%) ${\qeq}$ persimmon (28.8%). The order of nitrite-scavenging abilities at pH 1.2 (2 mg/mL) was green tea > rose > pine needles > chrysanthemum ${\qeq}$ mulberry ${\qeq}$ persimmon. The order of hydroxyl radical scavenging activity (5 mg/g) was chrysanthemum > mulberry > rose > persimmon leaves > pine needles > green tea. Rose extracts showed antimicrobial activity against Micrococcus luteus, Bacillus cereus, Salmonella enteritidis, and E. coli, whereas green tea extracts showed strong antimicrobial activity aginst S. enteritidis.

Effect of Boron in Nutrient Solution on Root Development and Freezing Tolerance of Mulberry

  • The experiment was carried out to define the effect of the boron in nutrient solution on the development of mulberry root and cold damage. The length of shoots only attained about 50cm and then stopped in case of boron deficiency, while the mulberry trees developed vigorous and attained over 190cm long under boron supply condition. When boron is deficient, the measles appeared on surface of the stem and the necrosis appeared on the petiole, midribs and veins of leaves. At 70 days after planting, almost all new roots and old roots changed to brown, the lateral roots became necrosis by boron deficiency. After three days of stopping boron supply, many new roots changed to brown and after two days of boron resupply, new roots began to grow out and then new roots normally developed. Content of boron in leaves and barks increased by the increase of boron concentration in nutrient solution and maximum boron content in leaves and barks was obtained with boron-sufficient treatment of 0.5ppm. Treatment of 0.5ppm boron supply indicated that the leaves and barks contained more phospholipid, protein, sugar, RNA and proline than treatment of 0.01ppm boron supply in nutrient solution. The cuttings grown in boron supply nutrition have a sufficient tolerance at -10$^{\circ}C$ and -15$^{\circ}C$ for 24 hours while cuttings grown in boron-deficient nutrition have a weak tolerance at the same condition. As mentioned above, we can conclude that the effect of boron deficiency on root development is much severe and the relationship between the lack of boron and cold damage is very closely related.

Functional quality characteristics of extracts by sugar-leaching and lactic acid fermentation of mulberry leaves (Morus alba L.) (뽕잎의 당침 및 유산발효에 의한 추출물의 기능성 품질 특성)

  • This study was carried out to investigate functional quality characteristics of extract obtained after sugar-leaching for 12 weeks (SLE) and extract obtained after lactic acid fermentation for 8 weeks (LFE) of mulberry leaves. The yield, sugar content, pH, and total acidity of SLE were 27%, 43 $^{\circ}Brix$, 4.6, and 0.45%. The yield, sugar content, pH, and total acidity of LFE were 166%, 33 $^{\circ}Brix$, 3.6, and 1.17% respectively. The lactic acid bacteria viable numbers ($1.2{\times}10^{10}$ CFU/ml) of LFE were more than those of SLE ($2.8{\times}10^2$ CFU/ml). The LFE expressed activities of hydrolytic enzymes (amylase, cellulase, pectinase, protease), but SLE did not express. The contents of acetic acid, citric acid, and malic acid of SLE were higher than those of LFE, but lactic acid content of LFE was higher than that of SLE. The main free sugars of SLE were glucose (200.93 mg/g), fructose (236.32 mg/g), and sucrose (18.41 mg/g), but LFE did not detect all free sugars. The contents of polyphenol, anthocyanin, and piperidine alkaloid of LFE were higher than those of SLE. ${\alpha}$-Glycosidase activities were inhibited 3.4% and 16.2% by SLE and LFE. These results suggest that lactic acid fermentation extraction is an effective method to increase the yield and contents of functional quality of mulberry leaves extract.

Inhibitory Effects of Water and 80% Ethanol extracts from Mulberry Leaves (Morus alba L.) on Angiotensin Converting Enzyme and Xanthine Oxidase (뽕잎(Morus alba L.)의 물과 80% Ethanol 추출물의 Angiotensin Converting Enzyme과 Xanthine Oxidase에 대한 활성억제효과 탐색)

  • The objective of this research was to evaluate the ability of water and 80% ethanol extracts from one hundred eight mulberry leaves (Morus alba L.) to influence the inhibitory activity of angiotensin converting enzyme (ACE) and xanthine oxidase (XOase). The total phenol contents were that water extracts of ten species (Kakjayongsan (Morus alba L.), Daejungsun (Morus alba L.) etc.) and 80% ethanol extracts of twenty three species (Waryoung (Morus alba L.), Hasusang (Morus alba L.) etc.) showed more than 15 mg/g. The inhibitory activity on angiotensin converting enzyme (ACE) were that ten species (YamanakkadakKaskke (Morus alba L.), Mijiro (Morus alba L.) etc.) showed 100% inhibition rate both of water extracts and 80% ethanol extracts. The rest, water extracts of thirty four species (Cheongilppong (Morus alba L.) etc.) and 80% ethanol extracts of thirty four species (Wonjukojo (Morus alba L.) etc.) showed inhibitory activity (above 90%) on ACE. Also, to search of xanthine oxidase (XOase) inhibition were that water extracts of five species (Cheongsipjosaeng (Morus alba L.), Suwon 3 (Morus alba L.) etc.) and 80% ethanol extracts of Jeokmok (Morus alba L.) showed inhibitory activity (above 50%) on XOase. This result revealed, strong biological activity in spite of has a little total phenol contents. These water and 80% ethanol extracts from mulberry leaves (Morus alba L.) are expected good candidate for development into anti-hypertentive and anti-gout sources.

Induction and Characterization of Precocious Trimolting Silkworm, Bombyx mori (조숙 3면잠의 유발과 실용형질)

  • For fine-denier cocoon filament production, the precocious trimolting silkworms were induced by the treatments with both the imidazole compound "KK-42" hating anti-juvenile hormone activity and high temperature, and their growth, dietary efficiency, cocoon and cocoon filament qualities were compared with those of normal tetramolters as control. 1. The percentage of precocious trimolters was higher in the application of KK-42 than the treatment of high temperature. The effective concentration of KK-42 was 10$\mu\textrm{g}$/1arva. The high temperature treatment for 48 hour was more effective than 36 hour treatment, and silkworm larvae fed on mulberry leaves were more sensitive than artificial diet to induce trimolters. 2. The larval duration of the trimolters induced by KK-42 was 5.17 day shorter than that of normal tetramolters in mulberry leaves rearing, and the increasing pattern of body weight in the 4th inster larvae of trimotlers was similar to the 5th instar larvae of normal tetramolters. 3. The qualities such as cocoon weight, cocoon layer weight and cocoon layer ratio of precocious trimolter induced by KK-42 were much lower those of normal tetramolters: the cocoon weight and cocoon layer ratio were 0.78g, 14.2cg and 18.4% in mulberry leaves rearing, and 0.86 cg, 10.3cg and 12.3% in artificial diet rearing, respectively. 4. The size of cocoon and cocoon filament was smaller in the precocious trimolter, both KK-42 and high temperature, as compared with that of the normal tetramolters. 5. The efficiency of cocoon layer production of the precocious trimolters by the KK-42 was lower than that of the normal tetramolters: amount of the cocoon layer production per 1g of dry mulberry leaves ingested was 7.97cg in the precocious trimotors, while 9.20cg in the normal tetramolters.

Quantitative Analysis of 1-deoxynojirimycin in Mulberry Leaves (뽕나무 유전자원의 1-deoxynojirimycin 함량 변이)

  • Countries of the world are competing for the collection and utilization of genetic resources, which is a resource of value-added industrialization. We are building database of silkworm and mulberry resources with morphological, genetic characteristics data etc. Moreover, recent bioactive compound has been receiving increasing attention. So we analyzed 1-deoxynojirimycin(1-DNJ) content as a this item in addition to the basic information using 363 strains(varieties) of mulberry genetic resources. They were grown under the same environment and conditions. Mulberry leaves were collected and then freeze-dried and powdered for 1-DNJ test. As a result, 1-DNJ mean content of 363 mulberry strains was $0.176{\pm}0.077%$, and the coefficient of variation (CV) was 43.5%. The variation between the strains was greatly severe. Among the tested strains, 'C1D89/29' was showed the highest content of 0.47%, whereas 'Pumbo 24' and 'Turkey E' were showed the lowest content of 0.05% respectively. The content of 1-DNJ of 16 mulberry varieties for silkworm rearing were compared. Average concentration was $0.17{\pm}0.04%$, and the coefficient of variation (CV) was 22.8%. Variation among cultivars was not significant. Finally we selected 1-DNJ high-containing 11 strains. They are as follows. 'C1D89/29', 'Hiihak', 'Jeokasibmunja', 'Gweonchil', 'Botongsibmunja', 'Jeokchuk', 'bulguksang', 'Geunsookgojo', 'Busanggeum', 'Guksang 20' and 'Taekwang' which are more than twice of the overall average content.

The Characteristic of Natural Dyeing with Mulberry Leaf and Coffee Powder using Zinc Mordant (아연 매염제를 이용한 뽕잎, 커피 천연염재의 염색특성)

  • The use of natural organic dyes obtained from renewable resources such as plants and trees has the potential for not only preserving petrochemicals but also all-endangered environment for coming next generations. In spite of inferior fastness, natural dyes can be employed in the colouration of natural as well as synthetic fibers. Recently, the potentiality of using natural dyes in colouration with additional UV-protection and antimicrobial properties has been reported. This study is aimed to investigate the effect of Zinc compounds compared to another metallic compounds as mordants on the dyeing properties of natural dye extracted from mulberry leaves and coffee. In this study, the results showed Zinc compounds was expose that is more similar than other mordants in washing fastness through a repeated experiment. Also, Zinc is the environmental impact is less material compared with other mordants, essential edibility minerals ingredient that is necessary in human body.

Breeding of an Autotetraploid, Sawonppong 28, Derived from a Thick Leaf Mulberry Variety, Huyeoprok (Morus sup.) (厚葉性 뽕품종 厚葉綠의 同質4倍體 ″4原뽕 28號″의 創成)

  • An autotetraploid, Sawonppong 28 was derived from a thick-leaf and highly productive mulberry variety, Huyeoprok, by colchicine solution treatment on growing point and its characteristics were as follows. The number of somatic chromosomes of the derived variety was 2n=56 of an autotetraploid and green tip sprouting stage was medium budding variety with triangular oval shape and thick leaves contained rich chlorophyll. The length of branches was comparatively short and the number of branches was also small. However the branches were thick and non-budding length percentage at basal part of branch was high and cold hardiness was weak.
