Integumentary Ultrastructure of the Blenny, Pholis nebulosa (Teleostei: Pholidae) (베도라치 (Pholis nebulosa) 피부계의 미세구조)

  • To provide basic information on the integumentary system of the blenny, Pholis nebulosa, ultrastructures of epidermal and dermal layer were examined by means of the light and transmission electron microscope. The skin of the blenny consisted of epidermal and dermal layer. Epidermal layer consisted of supporting cell and unicellular gland. The supporting cells were classified into superficial cell, intermediated cell and basal cell, and the gland cells were classified into mucous secretory cell and club cell. The cytoplasm of supporting cells was divided into cortex and medullar part. In the cortex and medullar part, microfilaments and cell organelles were well developed, respectively. Superficial cell of epidermal layer was cuboidal and contained nucleus of horseshoe shape. Intermediated cell had a nucleus of irregular form and the electron density was higher than the other supporting cells, Basal cell was columnar, but nucleus was situated in the upper cytoplasm. Cell organelles of the basal cell were poor than the other supporting cells, but membrane interdigitations were well developed. The cytoplasm of mucous secretory cell had a well-developed ovoid secretory granules, which reacted to red with AB-PAS reaction. The club cell had a we31-developed round secretory granules and endoplasmic reticulum. figment cells were classified into two type. The one contained pigment granules of electron dense, and the other contained reflecting platelets. The cytoplasm of fibrocyte had n well developed rough endoplasmic reticulum.

Ultrastructure of the Submandibular Gland in the Lesser White-Toothed Shrew, Crocidura suaveolens (작은땃쥐 Crocidura suaveolens 악하선의 미세구조)

  • The ultrastructure of submandibular gland was examined in the lesser white-toothed shrew, Crocidura suaveolens. The submandibular gland of C. suaveolens was a mixed gland composed of serous and mucous acinar cells. Secretory granules from the acini were discharged through the salivary ducts into the oral cavity. Serous and mucous acinar cells had well developed rough endoplasmic reticulum and mitochondria and large amount of granules. In case of serous acinar granules, an immature granule was formless and had only dense specks, and a matured granule was a complete round type delimiting by a single membrane and had a homogeneous dense center with dense specks on the border. In case of mucous acinar granules, while an immature granule was a round type and had an only homogeneous matrix and an indistinct limiting membrane, a mature granule was an even round type having a variety of pattern with several dense bands into the homogeneous matrix and had a distinct membrane. Therefore, a mature mucous acinar granule of C. suaveolens was not only distinct from those of the other mammalian species to have a variety of pattern but also from those of C. lasiura to have an even round type. A great serous-like secretory granules and Myelin-like body were observed in the cytoplasm and lumen of granular duct cells. Myelin-like body, a characteristic structure only reported in salivary gland of three shrews, was discharged from secretory cell into lumen by the manner of exocytosis which has little differences from discharging manner of secretory granules.

Clinical and Cytomorphological Studios on Bovine Leukosis (우백혈병의 임상 및 세포형태학적 연구)

  • Clinical and cytomorphological studies were carried out in 32 leukotic cattle from Tokachi and Kushiro districts in Hokkaido during the 12 year period from 1969 to 1980. The leukotic cattle were examined :and divided into four types(15 cases of the adult, 11 cases of the thymic, 4 casas of the calf and 2 cases of the skin types). The results obtained were as follows : 1. As for the frequency of the main clinical signs in each type, In the adult type, the main clinical signs (of decreasing order) are as follows: swelling of the superficial lymph nodes>depression and loss of weight>tachycardia>anorekia, anemia of the visible mucous membrane and tachypnea. Those of the thymic type were swelling of the thymus>swelling of the medial iliac lymph nodes> swelling of the superficial lymph nodes>tachypnea. Those of the calf type were swelling of the auperficial lymph nodes>depression and emaciation>tachypnea>anorexia, tachycardia, anemia of the visible mucous membrane and recumbency. Those of the skin type were generalized urticaria-like lesions in skin and swelling of superficial lymph nodes>and depression and loss of weight in the decreasing order of frequency. In addition, large tumor mass in the pelvic cavity and swelling of the medial iliac lymph nodes were detected through rectal palpation in 33.3% and 100% in the adult type cases, respectively. 2. As for the hematological findings, The frequency of occurrence of decreased erythrocyte counts in the decreasing order were as follows : adult>calf>thymic>and skin types. The increase in the total leukocyte count in the order of decreasing frequency were as follows: calf>thymic>adult>and skin types. The increase in the absolute lymphocyte counts was found to be at a low rate, 62.5% of all the cases examined. By contrast, the increase of 5% or more of abnormal lymphocyte rates was observed at a high rate, 96.9% of the total cases. 3. Abnormal lymphocytes were found in all cases examined for lymph nodes biopsied. 4. From the cytomorphological point of view, leukotic cells were divided into 3 types: reticulum cell, lymphoid cell and monocytic cell types. The adult type leukotic cattle were divided with reticulum cell type (66.7%), the lymphoid cell type(22.6%) and monocytic cell type(6.7%). The thymic type was lymphoip cell type(72.7%) and reticulum cell type(27.3%). In the calf type, all were lymphoid cell type while all of the skin type were reticulum cell type only. 5. The leukotic cattle had higher NP frequency in the blood and lymphoid tissue than non-1 eukotic cattle. Especially the adult type had the highest NP frequency. However, it was not recognized that NP were characteristic of leukotic cattle alone. 6. The above findings lead to the conclusion that the most effective diagnostic methods for bovine leukosis are the confirmation of swelling of the superficial and internal lymph nodes and thymus in addition to appearance of abnormal neoplastic cell in the peripheral blood and lymph nodes biopsied.

Structure and Histochemistry of the Skin of a Flat-headed Goby, Luciogobius guttatus (Gobiidae, Pisces) from Korea (한국산 미끈망둑 Luciogobius guttatus (망둑어과) 피부의 구조 및 조직화학적 특성)

  • To investigate the skin of the flat-headed goby, Luciogobius guttatus, it was used 8 body regions such as the head, the upper and lower jaw, the chin, the back, lateral region, abdomen, and the operculum. The epidermis consisted of three layers: the outermost layer, middle layer and stratum germinativum (basal layer). The outermost layer consisted of rather flattened cells arranged in 1 to 4 layers and mucous cells. The middle layer consisted of large epidermal cells occupying the height of the epidermis with 1 to 10 layers, causing a web-shaped structure. Due to the large epidermal cells of the middle layer, L. guttaus had a thick epidermis. The large epidermal cell contains tonofilaments, lucent contents, and desmosome. The basal layer was comprised of a single layer having cuboidal cells. A large number of fine blood capillaries were found just under the basal layer. The dermis consisted mostly of stratum compactum with numerous blood capillaries but had no scale.

Changes of Biomarker in Manila clam, Ruditapes philippinarum Exposed to Lead (납(Pb)에 노출된 바지락, Ruditapes philippinarum의 생물지표 변화)

  • This study was conducted to find out biological response of Manila clam, Ruditapes philippinarum exposed to lead (Pb). Experimental period was four weeks. Experimental groups were composed of one control condition and three lead exposure conditions (0.25, 0.50 and 1.00 mg/l). The results of the study confirmed that lead induces reduction of survival rate and oxygen consumption rate and histopathology of organ structure of the bivalve. Oxygen consumption rate was observed exposure groups lower than control decline by 25%-72%. Histological analysis of organ system illustrated expansion of hemolymph sinus, disappearance of epidermal layer and degeneration of connective tissue layer of the mantle. Also, histological degenerations as epithelial necrosis and hyperplasia of mucous cells are recognized in the gill and it was observed expansion of hemolymph sinus, disruption of epithelial layer, decrease of mucous cell and degeneration of connective tissue layer in the foot. In the digestive diverticulum, it was showed atrophy of basophilic cell and degeneration of epithelial cell in the digestive tubules, and as the concentration of lead increased the accumulation of lipofuscin increased.

  • Integumentary structures of the rockfish, Sebastiscus tertius were examined by means of the light and transmission electron microscopy. Stratified epidermal layer consists of supporting cells, unicellular glands, granular cells and mitochondria-rich cells. The epidermal layer could be classified into superficial, intermediate and basal layer by morphology and structure of the supporting cells. Mucous cells of unicellular gland were observed in the superficial and intermediate layer of the epidermis. The mucous materials were identified as acidic and carboxylated mucosubstance by histochemical methods. Club cell has well-developed central vacuole, rough endoplasmic reticula and Golgi complex in the cytoplasm. Granular cells were observed in the superficial layer and contained numerous granules of high electron density. Mitochondria-rich cells are characterized by well-developed microfilaments in cortex and numerous tubular mitochondria in medullar cytoplasm. Three types of pigment cells in the dermal layer could be distinguished with electron density of cytoplasmic inclusions.

The Effects of Sasammaickmoondong-tang against Colonic Mucosal Lesions (사삼맥문동탕이 Indomethacin으로 유발된 mouse의 대장 점막 손상에 미치는 영향)

  • Objectives: This study was carried out to investigate 1he effects of Sasammaickmoondong-tang(SME) on colonic mucosal lesions induced by indomethacin in mouse. Methods: The normal group is 1hat no inflammation elicitated mouse. Control group is that gastro-inflammation elicitated mouse. Sample group is that SME administered mouse after gastro-inflammation elicitation. Results: In the common morphology and histochemical change, control group was observed various injury-mucous surface cell, micro-villi, paneth cell, surface epithelial cell, goblet cell-by hemorrhagic erosion, while sample group was as same as normal group. In the immunohistochemical change, 1he distributions of COX-1, Bcl-2, and BrdU treated with SME noticeably increased than control group(P<0.05). The distributions of TUNEL, $NF-{\kappa}B$, COX-2, $IL-2R-\alpha$, NK-1.1, ICAM-1, and CD11b/18 treated with SME noticeably decreased than control group(P<0.05). And the distribution of SBA was as same as normal group. Conclusions: According to the above results, it is supposed that Sasammaickmoondong-tang is applicable to colonic mucosal lesions.

The Effects of Sasammaickmoondong-tang against Gastric Mucosal Lesions (Mouse의 위점막 염증에 대한 사삼맥문동탕의 면역반응 연구)

  • Objectives: This study was carried out to investigate the effects of Sasammaickmoondong-tang (SME) on gastric mucosal lesions induced by indomethacin in mice. Methods: The normal group was no inflammation-induced mice. The control group was gastro-inflammation-induced mice. The sample group was mice administered SME after gastro-inflammation elicitation. Results: In the common morphology and histochemical change, the control group was observed with various injury-mucous surface cell, micro-villi, paneth cell, surface epithelial cell, goblet cell - by hemorrhagic erosion, while the sample group was as same as the normal group. In the immunohistochemical change, the distributions of COX-1, Bcl-2, and BrdU treated with SME were noticeably higher than in the control group (p<0.05). The distributions of TUNEL, NF-B, COX-2, IL-2R-, NK-1.1, ICAM-1, and CD11b/18 in those treated with SME were noticeably lower than in the control group (p<0.05). Finally, the distribution of SBA was the same as in the normal group. Conclusions: According to the above results, it is supposed that Sasammaickmoondong-tang is applicable to gastric mucosal lesions.

Desquamative Diseases of Gingiva (임상가를 위한 특집 3 - 잇몸이 벗겨지는 질환)

  • The gingiva consists of an epithelial layer and an underlying connective tissue layer. The oral epithelium is a keratinized, stratified, squamous epithelium. The epithelium can be divided into the following cell layer: basal layer, prickle cell layer, granular cell layer and keratinized cell layer. The desquamative disease of gingiva means exfoliative diseases of epithelial layer on the gingiva. The chronic desqumative gingivitis is usually related to the dematologic disorders that produce cutaneous and mucous membrane blisters. The cicatricial pemphigoid and lichen planus are representative diseases of the dermatologic cases. Patients may be asymptomatic or symptomatic. When symptomatic, their complaints range from a mild burning sentation to an severe pain. The clinical examination must be considered with a thorough history, and routine histologic and immunofluorescence studies. A systemic approach needs to achieve accurate diagnosis and treatment of the gingival desquamative diseases.

An Ultrastructural Study on the Epithelial Cells of the Rabbit Gallbladder after Common Bile Duct Ligation (총담관 결찰후 집토끼 담낭상피세포의 미세구조적 변화)

  • The gallbladder is known to have the function of the storage and the concentration of the bile produced by the liver. This function is carried out by the removal of water and inorganic electrolytes. Extrahepatic cholestasis or the impairment of excretion of the bile leads to the distension and loss of the function of the gallbladder. The purpose of this study was to examine the ultrastructural characteristics of the normal gallbladder epithelial cells, and their structural changes induced by the ligation of common bile duct of the rabbit. Common bile duct ligation was performed under ether anesthesia. The rabbits were sacrificed on the 1st, 3rd, 5th, 7th and 14th day, respectively after operations. The tissue blocks of the gallbladder were fixed in 2.5% glutaraldehyde-1.5% paraformaldehyde prior to fixation in 1% osmium tetroxide, and embedded in the araldite mixture, and observed with JEM 100 CX-II electron microscope. The results were as follows: 1. The normal gallbladder epithelium of adult rabbit demonstrated two cell types, the ordinary epthelial cell and the dark cell. The dark cells have electron dense cytoplasm, and were found much infrequently, whereas ordinary epthelial cells were found quite numerous. 2. The ordinary epthelial cells of normal gallbladder were provided with the regular microvilli at the free surface and the images of pinocytotic activities in the apical cytoplasm, and exhibit highly convoluted lateral surfaces with elaborated microfolds. These figures of the cells suggest that they are resorptive in functional activity. 3. In the early stages (1st, 3rd, 5th day groups) following the ligation, the apical cytoplasm of some cells is protruding from the free surface and lost their microvilli. Numerous mucous granules filled in the apical and supranuclear cytoplasm compactly. 4. In the late stages (7th, 14th day groups) following the ligation, many light cells containing mumerous mucous granules are seen, between the ordinary epthelial cells. Mucous granules are fused each other, and are discharged into the lumen from the apical cytoplasm. The lateral membranes are straight or undulating without any interdigitations. From the above results, it was concluded that in the cholestasis induced by the common bile duct ligation, there is a tendency for the mucosal epithelium of the rabbit gallbladder to have secretory rather than an absorptive function.
