Studies on the Hydrolysis of Milk Fat by Microbial Lipases (미생물에서 추출된 Lipase의 유지방 분해)

  • To utilize microbial lipases for hydrolysis of milk fat, optimum reaction conditions and characteristics of enzymatic reactions of lipases originated from Rhizopus delemar, Mucor sp., and Candida cylindracea were investigated. Optimum pH and temperature were pH 5.6 and $45^{\circ}C$ for Rhizopus delemar lipase, pH7.5 and $35^{\circ}C$ for Mucor sp. lipase, and pH7.5 and $35^{\circ}C$ for Candida cylindracea lipase. Optimum lipase concentration and optimum substrate concentration were $600{\sim}800\;units/ml$ and 20% milk fat, regardless of their origin. Km values were 6.06% milk fat for Rhizopus delemar lipase, 7.69% for Mucor sp. lipase and 7.99% for Candida cylindracea lipase. Rate of lipid hydrolysis was Rhizopus delemar lipase>Mucor sp. lipase>Candida cylindracea lipase. As the reaction time was extended, liberation of short chain fatty acids was increased. After 8 hours reaction, capric acid content significantly increased with Candida cylindracea lipase, palmitic acid with Mucor sp. lipase and butyric acid with Rhizopus delemar lipase.

Production of the Fungal Lipid Containing ${\gamma}-Linolenic$ Acid from Mucor sp. KCTC 8405P (Mucor sp. KCTC 8405P에 의한 ${\gamma}-Linolenic$ Acid 함유 곰팡이 유지의 생산)

  • Mucor sp. KCTC 8405P was cultivated in a jar fermentor for the production of fungal lipid containing ${\gamma}-linolenic$ acid with feeding the glucose solution periodically. The transition of the fungal growth into the mycelial phase from yeast-like growth was achieved by pH shift after the first two day of cultivation in the low pH medium and then lipid accumulation was accelerated until the seven day of cultivation, when the glucose in the culture broth was almost consumed. With the culture conditions applied in this experiment, biomass of 99.3 g/l by the dry cell weight and the total extractable lipid of 38.0 g containing 3.5 g/l ${\gamma}-linolenic$ acid were obtained.

  • In order to produce ${\gamma}-linolenic$ acid by Mucor sp. KCTC 8405P. the fungus was cultivated in fed-batch culture with two phases. i.e., growth in yeast-like form and induction to hyphal growth by pH shift of the culture medium during cultivation. The synchronous growth of the fungus into the appropriate sizes was important for the high density cell culture of this dimorphic fungus. Dissolved oxygen concentration in the medium did not affect degree of unsaturation of fatty acids and ${\gamma}-linolenic$ acid content. Under the culture conditions applied in this experiment. the fungus was found to produce 100 g/l dry mycelia containing 40% of the lipids, where ${\gamma}-linolenic$ acid comprised about 9% of the total extractable fatty acids.

  • For a studyon the production of ${\gamma}$-linolenic acid(GLA) by fungi 3 fungal strains were isolated from soil. Their cell growth lipid content and fatty acid composition were compared in shake flask culture. Among these fungi the fungus designated as FA-007 has high lipid content(21.1%) and GLA content(15.6% of total fatty acids) The fungal strain FA-007 was tentatively identified as Mucor sp. on the basis of morphological characteristics, Fungal oil produced by this fungus was composed of 75.2% neutral lipid 5.3% glycolipid and 19.5% phospholipid. Although the GLA content in phospholipid was higher than it in neutral lipid the GLA content in neurtal lipid was high as 15.5%.

  • In order to select ${\gamma}-Linolenic$ acid (GLA)-producing fungi, a total of forty-four strains of 4 genera such as Phytophthora, Pythium, Mucor and Rhizopus were obtained from Koran Agricultural Culture Collection (KACC) and then analysed by using GC-FID and GC-MS. GLA was detected on 39 fungal strains, and the highest rate of GLA was found as 24.8% of total fatty acids on Mucor hiemalis f. sp. hiemalis KACC 40264. Total GLA content of Zygomycota was comparatively high - Mucor (14.2%) and Rhizopus (14.3%), whereas that of Oomycetes was low - Phytophthora (3.3%) and Pythium (3.0%). Moreover, total fatty acids of the Zygomycota fungi such as Mucor (15.4 mg/100 ml) and Rhizopus (7.1 mg/100 ml) were higher compared with the Oomycetes such as Phytophthora (2.6 mg/100 ml) and Pythium (4.5 mg/100 ml). Thus, two genera such as Mucor and Rhizopus have higher potential as an useful microbial resource. The total fatty acid content varies even within the strains of the same genus e.g. Mucor. M. blumbeus KACC 40935 showed the highest values on productivity (18.2%) of GLA and total fatty acid contents (50.8 mg/100 ml liquid medium).

  • For a study on the production of ${\gamma}-linolenic$ acid(GLA) by microorganisms, fifteen strains of Mucorales obtained from culture stocks and ten isolate strains were compared in their cell growth, lipid content, fatty acid composition and pellet size formed in shake flask culture. Among the fungi examined, the isolated fungus, designated as FB-354, was found to be the most suitable one for the production of GLA mainly due to its high contents of lipid, 29.9% of dry cell weight and GLA, 16.8% of the total fatty acids. The strain FB-354 was tentatively identified as Mucor sp. on the basis of morphological characteristics. Fungal oil produced by Mucor sp. FB-354 was fractionated into 81.1% of neutral lipid, 7.2% of glycolipid, and 11.8% of phospholipid. Although the GLA content in the phospholipid fraction was as high as 21.4%, most of the GLA was found in the neutral lipid fraction.

  • A potent fungus producing milk clotting enzyme with fairly weak proteolytic activity was isolated from various soil and sewage, which the selected strain, SA-101, was identified as Mucor sp. with microbiological characteristics. Its milk clotting enzyme production was maximized when grown on 10g of wheat bran media added to 8ml of tap water containing 0.1M HCl for 60hrs at $30^{\circ}C$. This enzyme production was stimulated by addition of 6% lactose, 0.05% NaCl and reached a maximal level of 9810 unit/g wheat bran. The crude enzyme product could be produced effectively by salting out with ammonium sulfate fractionation and lyophilization. The ratio of milk clotting activity to proteolytic activity of crude enzyme product was lower than Hansen rennet, but resembled to Meito rennet. The optimal temperature of milk clotting activity of crude enzyme product was abound $60^{\circ}C$ on a substrate of 10% reconstituted skim milk containing 1/100M $CaCl_2$.

  • In order to study the influence of culture conditions on cell growth and lipid formation by Mucor sp, various carbon and nitrogen sources initial pH and C./N ratio of medium were investigated. Glucose was found to be suitable carbon source in terms of lipid yield and ${\gamma}$-linolenic acid(GLA) content. When NH4Cl and (NH4)2SO4 were used as nitrogen source lipid content was high(19-21%) but GLA content was low(15-17%) On the other hand when NaNO3 and KNO3 were used lipid content was low(about 13%) but GLA content was high(22-23%). The highest production of lipid was obtained at a C/N ratio of 40 using glucose and (NH4)2SO4 as carbon and nitrogen source respectively. it was found that lipid yield was high at pH4.6 Also this fungus did not grow at 35$^{\circ}C$ and lipid yield was highr at 15$^{\circ}C$ than $25^{\circ}C$.

  • As a way to determine the optimal culture conditions for the production of ${\gamma}$-linolenic acid by Mucor sp. KCTC 8405P, the influence of different carbon and nitrogen sources, initial pH, and C/N ratio of medium was investigated. Glucose was found to be the best carbon source in terms of lipid content and ${\gamma}$-linolenic acid yield. Ammonium sulfate and organic nitrogen sources such as urea and peptone resulted in relatively increased lipid and ${\gamma}$-linolenic acid production. The highest accumulation of lipid was obtained at a C/N ratio of 56.6 using glucose and (NH$_4$)$_2$SO$_4$ as carbon and nitrogen source, respectively. It was found that the lipid content increased significantly with increasing initial pH of medium up to pH 9.0. The influence of mixed carbon source on the ${\gamma}$-linolenic acid yield was also investigated. High accumulation of lipids, 315 mg/100 ml medium, and 13-14% of ${\gamma}$-linolenic acid content in the cellular lipid were obtained in a shaking culture containing 3% of glucose and 2% sodium acetate as carbon source and 0.1% of (NH$_4$)$_2$SO$_4$ as nitrogen source at pH 8.0.

  • - The effects of sodium acetate on the production of $\gamma$-linolenic acid (GLA) and the secretion of the mycelial lipid into the culture medium with noninnic surfactants were studied with Mucor sp. KCTC 8405P. In the addition of 2.0%) sodium acetate to the basal medium, dry cell weight and total lipid content were increased from 7.8 g/l and 2.46 g/l to 16.0 g/1 and 4.77 g/l, but GLA content was decreased from 18.6% to 13.85%. The growth of Mucor sp. KCTC 8405P was greatly dependent on both the initial pH and the concentration of sodium acetate of culture medium, which was considered as the results of the formation of acetic acid because the fungal growth was completely inhibited at the concentration of acetic acid higher than 22 mM. With the decrease of the oxygen supply, the cell growth, total lipid, and GLA content were sharply decreased in the presence of 2.0% sodium acetate. For the secretion of mycelial lipid into the culture medium, the effects of the various nonionic surfactants were examined. In the addition of 0.5% Tween 80 or Span 80 to the basal medium, 194 mg/l or 263 mg/l of GLA was obtained in the culture medium.
