Comparative Assessment of Diagnostic Performances of Two Commercial Rapid Diagnostic Test Kits for Detection of Plasmodium spp. in Ugandan Patients with Malaria

  • Prompt diagnosis of malaria cases with rapid diagnostic tests (RDTs) has been widely adopted as an effective malaria diagnostic tool in many malaria endemic countries, primarily due to their easy operation, fast result output, and straightforward interpretation. However, there has been controversy about the diagnostic accuracy of RDTs. This study was conducted to evaluate the diagnostic performances of the 2 commercially available malaria RDT kits, RapiGEN Malaria Ag Pf/Pv (pLDH/pLDH) and Asan $EasyTest^{TM}$ Malaria Ag Pf/Pv (HRP-2/pLDH) for their abilities to detect Plasmodium species in blood samples collected from Ugandan patients with malaria. To evaluate the diagnostic performances of these 2 RDT kits, 229 blood samples were tested for malaria infection by microscopic examination and a species-specific nested polymerase chain reaction. The detection sensitivities for P. falciparum of Malaria Ag Pf/Pv (pLDH/pLDH) and Asan $EasyTest^{TM}$ Malaria Ag Pf/Pv (HRP-2/pLDH) were 87.83% and 89.57%, respectively. The specificities of the 2 RDTs were 100% for P. falciparum and mixed P. falciparum/P. vivax infections. These results suggest that the 2 RDT kits showed reasonable levels of diagnostic performances for detection of the malaria parasites from Ugandan patients. However, neither kit could effectively detect P. falciparum infections with low parasitaemia (<$500parasites/{\mu}l$).

A New record of Elaeagnus (Elaeagnaceae) to Korean Flora: E. pungens Thunberg (한국 보리수나무속의 미기록 식물: 통영볼레나무)

  • Elaeagnus pungens Thunb., belongs to the family Elaeagnaceae, was recently recorded for the first time in Mt. Miruk, Tongyeong-si, Gyeongsangnam-do. The native distribution range of this species includes China and Japan in East Asia, while it is considered as an invasive species in North America. Elaeagnus pungens is related to E. glabra, but the species can be distinguished from its congeners by having revolute and repand leaf blades, silvery white and brown scales on the abaxial surface of the leaf, and a campanulate calyx. We propose the new Korean name 'Tong-yeong-bol-re-na-mu,' based on its first collecting site. A detailed description of vegetative and floral characteristics, trichome scanning electron micrographs, dichotomous keys, and the habitat of this species are also provided.

Antimicrobial efficacy of endophytic Penicillium purpurogenum ED76 against clinical pathogens and its possible mode of action

  • This study was aimed to evaluate the antimicrobial activity of Penicillium purpurogenum ED76 on several clinically important microorganisms. The endophytic fungus P. purpurogenum ED76 was previously isolated from Swietenia macrophylla leaf. The antimicrobial efficacy of P. purpurogenum ED76 dichloromethane extract was determined via disc diffusion and broth microdilution assay. A kill curve study was conducted and the morphology of extract treated bacterial cells were viewed under scanning electron microscope. The dichloromethane extract showed significant inhibitory activity on 4 test bacteria and 2 test yeasts. The minimal inhibitory concentration of the extract ranged from 125 to $1,000{\mu}g/ml$, which indicates the different susceptibility levels of the test microorganisms to the fungal extract. The kill curve study has revealed a concentration-dependent inhibition for all test microorganisms. With the increase of the extract concentration, the microbial growth was significantly reduced. The scanning electron micrograph of dichloromethane extract-treated Staphylococcus aureus cells showed the total damage of the cells. The cell wall invagination of the bacterial cells also indicates the loss of cellular materials and metabolic activity. The gas chromatography mass spectrometry analysis of the extract also showed that the major compound was stigmasterol, which constitutes 45.30% of the total area. The dichloromethane extract of P. purpurogenum ED76 exhibited significant inhibitory activity on several clinically important bacteria and yeasts. The study proposed a possible mode of action that the extract cause significant damage to the morphology of S. aureus cells.

  • Chemical analysis, measurement of pumping rates of 60 production wells and depth to water tables of 57 monitoring wells were carried to protect depletion of water resources and deterioration of water quality for the commercial portable ground-water. Borehole depth of production well averages 149m(31 boreholes), casing depth is 28m(29 boreholes), production rate is 70 $m^3$/day and depth to water table of monitoring well is 23.26m, respectively. The geology of 60 wells can be divided into Daebo granite(20), Okchun metarmorphic complex(18), Precambrian granitic gneiss(15), Bulguksa granite(4), Cheju volcanics(2), Cretaceous sedimentary rock(1). Average electrical conductivity and pH are 152$\mu$S/cm, and 7.35, respectively. The contents of major cation and anion predominantly $Ca^{2+}$>N $a^{+}$>M $g^{2+}$> $K^{+}$ and HC $O_{3}$$^{-}$ >S $O_{4}$$^{2-}$>Cl ̄>F ̄. Water type is predominantly $Ca^{2+}$-HC $O_{3}$$^{-}$(81.7%). It's possible that water chemistry of some wells were affected not only by the geology of boreholes penetrated but by inflows of surface water or shallow ground-water. Therefore, it is strongly necessary to steadily monitor the water quality and hydrogeologic conditins of production wells.ells.ls.ells.

Tetrathyridia of Mesocestoides lineatus in Chinese Snakes and Their Adults Recovered from Experimental Animals

  • Morphological characteristics of Mesocestoides lineatus tetrathyridia collected from Chinese snakes and their adults recovered from experimental animals were studied. The tetrathyridia were detected mainly in the mesentery of 2 snake species, Agkistrodon saxatilis (25%) and Elaphe schrenckii (20%). They were 1.73 by 1.02 mm in average size and had an invaginated scolex with 4 suckers. Adult tapeworms were recovered from 2 hamsters and 1 dog, which were orally infected with 5-10 larvae each. Adults from hamsters were about 32 cm long and those from a dog were about 58 cm long. The scolex was 0.56 mm in average width with 4 suckers of 0.17 by 0.15 mm in average size. Mature proglottids measured 0.29 by 0.91 mm (av.). Ovaries and vitellaria bilobed and located in the posterior portion of proglottids. The cirrus sac was oval-shaped and located median. Testes were follicular, distributed in both lateral fields of proglottids, and 41-52 in number per proglottid. Gravid proglottids were 1.84 by 1.39 mm (av.) with a characteristic paruterine organ. Eggs were 35 by $27{\mu}m$ in average size with a hexacanth embryo. These morphological characteristics of adult worms were identical with those of M. lineatus reported previously. Therefore, it has been confirmed that the tetrathyridia detected in 2 species of Chinese snakes are the metacestodes of M. lineatus, and 2 snake species, A. saxatilis and E. schrenckii, play the role of intermediate hosts.

Influence of para-orientating Methoxyl Units on the Electronic Structures and Light Absorption Properties of the Triphenylamine-based dyes by DFT Study

  • The geometries, electronic structures and absorption spectra of the two organic triphenylamine-based dyes TA-St-CA and TA-DM-CA, containing identical electron donors and acceptors but the different conjugated bridges, were studied by density functional theory (DFT) at the B3LYP and PBE1PBE levels, respectively. The influence of para-orientating methoxyl units on the electronic structures and light absorption properties of the dyes and the consequent photovoltaic performance of the dye-sensitized solar cells (DSSCs) were investigated in detail. The results indicate that the introduction of the para-orientating methoxyl units into the conjugated bridge induces the increased absorption wavelength as well as the more negative EHOMO corresponding to the bigger driving force $(E_{I^-/I^-_3}-E_{HOMO})$ for dye reduction, which together improve the photovoltaic performance of TA-DM-CA, although there is a decline of the open circuit voltage caused by the more negative $E_{LUMO}$.

Hydrocarbon Plasma of a Low-Pressure Arc Discharge for Deposition of Highly-Adhesive Hydrogenated DLC Films

  • Plasma generator based on non-self-sustained low-pressure arc discharge has been examined as a tool for deposition of highly-adhesive hydrogenated amorphous diamond-like carbon(DLC) films. Since the discharge is stable in wide range of gas pressures and currents, this plasma source makes possible to realize both plasma-immersion ion implantation(PIII) and plasma-immersion ion deposition(PIID) in a unified vacuum cycle. The plasma parameters were measured as functions of discharge current. Discharge and substrate bias voltage parameters have been determined for the PIII and PIID modes. For PIID it has been demonstrated that hard and well-adherent DLC coating are produced at 200-500 eV energies per deposited carbon atom. The growth rates of DLC films in this case are about 200-300 nm/h. It was also shown that short(∼60$\mu\textrm{s}$) high-voltage(> 1kV) substrate bias pulses are the most favorable for achieving high hardness and good adhesion of DLC, as well as for reducing of residual intrinsic stress are.

  • The catalytic exchange of hydrogen isotopes between hydrogen and water has been known to be a very useful process for the separation of tritium from tritiated water. For the process, a highly active hydrophobic catalyst is needed. This study provides an effective fabrication method of size-controlled platinum/poly[styrene-divinylbenzene-tri(propylene glycol) diacrylate] [Pt/poly(SDB-TPGDA)] hydrophobic catalyst beads with a narrow size distribution. Platinum nanoparticles were prepared by ${\gamma}$-ray-induced reduction in the aqueous phase first, and then uniformly dispersed in SDB-TPGDA comonomer after the hydrophobization of platinum nanoparticles with alkylamine stabilizers. The porous Pt/poly(SDB-TPGDA) hydrophobic catalyst beads were synthesized by the UV-initiated polymerization of the mixture droplets prepared in a capillary-based microfluidic system. The size of as-prepared catalyst beads can be controlled in the range of $200-1,000{\mu}m$ by adjusting the flow rate of dispersed and continuous phases, as well as the viscosity of the continuous phase. Sorbitan monooleate and cyclohexanol were used as coporogens to control the porosities of the catalyst beads.

  • This study was performed to evaluate the characteristics of current air quality and to examine the monthly and yearly variation of $SO_2,\;O_3,\;CO,\;NO_2$, and fine particulate matter(PM-10) concentration in Seosan for the period of 1998-2006. During the recent 3 years, from 2004 to 2006, the yearly mean concentration of $SO_2,\;NO_2$, and PM-10 remained stability but CO and $O_3$ are gradually decreased. The monthly mean concentration of $NO_2$, CO, and $SO_2$ were high in the winter season and decreased steadily to the summer season, but the maximum concentration of $O_3$ and PM-10 appeared in the springtime. The number of days exceeding 24-hour standards for PM-10 were 54 times at Dokgodri and 25 times at Dongmundong. The number of sampling days exceeding the 1-hour and 8-hour standards for $O_3$ were 68 and 210 times at Dongmundong and at Dokgotri 93 and 213 times, respectively.

  • Jicon scallop (Chlamys farreri), as one of new culture species, has been taken a growing interest in Korea recently. Artificial seeds are needed since seeds in natural waters are absolutely short. To develop the better methods for the artificial spawning and the rearing larvae on a large scale, experiments were carried out from July 1993 to May 1994. Mother shells were collected from the area near Tong-yeong and Geoje-do, the southern part of the Korean Penninsula. It was estimated that a period of 2-years was needed to be mother shells after the fertilization. Generally, spawning occourred intermittently from March to August and the maximum occurred in July. A best way for the artificial spawning was a chemical incentive, a 'serotonin injection' in parallel with the exposure and the temperature stimulation. Densities of the fertilized eggs for the normal development was from 30 to 40 per mililiter. It took 27-days from the fertilization to the settling larva averaging $155{\mu}m$ in size with water temperature 11.5\~13.0^{\circ}C$. Isochrysis galvana and Chaetoceros calcitrans with a density of $1,500\times10^4$ cells per ililiter were supplied for rearing the larvae. A proper density of larvae was 10 individuals per mililiter, and the survival rate of larvae from the flowing water system was much better than that of larvae from the still water.
