Relationship between Genetic Variants of Mitochondrial DNA and Growth Traits in Hanwoo Cattle

  • Genetic variants of Hanwoo mtDNA in the region of cytochrome oxidase subunit I, II and III complex were detected using restriction enzymes. PCR primers were designed based on the bovine mtDNA sequence, and 6 primer sets (Mt4, Mt5, Mt6, Mt7, Mt8 and Mt9) were used. A total of 20 restriction enzymes were used, and 6 restriction enzymes, which were Hinf I, Pvu II, Rsa I, Eco RI, Bgl II, and Msp I, showed genetic polymorphisms. Significant associations between genetic variants and weight traits were observed at WT15 (p<0.05) and WT18 (p<0.01) with Pvu II for Mt9, Bgl II for Mt6 and Rsa I for Mt8 segments in the region of cytochrome oxidase subunit complex. Significant associations were also observed at Mt9-Pvu II and Mt6-Bgl II segments for WT9 (p=0.01), WT12 (p=0.02), respectively. These results suggest that genetic variants of mtDNA in the region of cytochrome oxidase subunit complex may be candidate segments for improvement of animal growth as weight traits.

Analysis of Deterioration Status on the Trails in the Gyeongju National Park - Focused on Mt. Toham, Mt. Nam, Mt. Danseok and Mt. Gumi District - (경주국립공원 탐방로의 훼손실태 분석 - 토함산, 남산, 단석산, 구미산 지구를 중심으로 -)

  • This study was carried out to systematically maintain and manage the trails by assessing the types of deterioration and naturalness of trails located in Mt. Toham, Mt. Nam, Mt. Danseok and in the Mt. Gumi district of the Gyeongju National Park. The route length of Mt. Toham was 2.7km, 2.3km of Mt. Nam, 1.0km of Mt. Danseok and 2.7km of Mt. Gumi district. The numbers of measuring points in Mt. Toham were 37 units, 29 units of Mt. Nam, 16 units of Mt. Danseok and 40 units of Mt. Gumi district. In characteristics of physical environments, Mt. Danseok was the widest and Mt. Gumi was the narrowest in the width of trail. In the bared width of trail, Mt. Danseok was the widest and Mt. Gumi was the narrowest. In depth of erosion, Mt. Danseok was the deepest and Mt. Gumi was the shallowest. Mt. Danseok was the steepest and Mt. Nam was the gentlest in the longitudinal slope. In the results of analysing the types of deterioration, Mt. Toham were 10 types, 14 types of Mt. Nam, 11 types of Mt. Danseok and 9 types of Mt. Gumi district. The times of appearance of deterioration types in Mt. Toham were 69 times, 87 times of Mt. Nam, 67 times of Mt. Danseok and 71 times of Mt. Gumi district. The most common type was the exposure of root in 4 districts of Gyeongju National Park. In the results of assessing the naturalness, Mt. Toham was 2.1 score(II degree), 2.6 score(II degree) of Mt. Nam, 2.9 score(III degree) of Mt. Danseok and 2.0 score(II degree) of Mt. Gumi district. The average naturalness was 2.4 score and II degree.

  • 한우의 mt DNA cytochrome oxidase subunit I, II, 및 III complex지역의 유전적 다형현상을 제한효소를 이용하여 검출하였다. PCR primer 6종에 대하여 20가지 제한효소를 처리하였으며, Pst I, Pvu II, Rsa I, Eco RI, Bgl II, and Msp I 제한효소를 사용하여 유전적 변이를 검출하였다. 검출된 변이체와 한우의 성장과의 관련성을 조사한 결과 cytochrome oxidase subunit III complex 지역의 유전염기서열을 근거로 제작한 primer Mt9 좌위에서 제한효소 PvuII를 이용한 절단형과 체중형질 인 WT15(P<0.05) 및 WT18(P<0.01)에서 고도의 유의성이 관찰되었다. 아울러 , Mt9-Pvu II(P=0.07), Mt6-Bgl II(P=0.05), and Mt8-Rsa I(P=0.05) 좌위 또한 WT9, WTl5, and WT15에서 각각 통계적 유의성이 관찰되었다. 따라서 본 결과는 cytochrome oxidase subunit III complex segments가 candidate gene으로서 기초적 유전정 보 제공은 물론 유전적 개량을 위해 사용될 수 있을 것으로 사료된다.

  • We isolated Actinomycetes strain No. 1882-5, which produces the inhibitor on the bleb formation of K562 cell induced by phorbol ester, from soil sample. Through solvent extraction, Amberlite XAD-4, silica and Lobar low pressure LC, antifungal antibiotic MT 1882-1 and bleb forming inhibitor MT 1882-II were purified from strain No. 1882-5. MT 1882-1 was identified as piericidin $A_{1}$($C_{25}H_{37}0_4N$, M.W. 415) and MT 1882-11 as glucopiericidin A($C_{31}H_{47}0_9N$, M.W. 577) from the analysis of physico-chemical properties and UV, $^1H-NMR$, $^13C-NMR$, and mass spectra of these compounds.

Effect of RFLP Marker of the Mitochondrial DNA D-Loop Region on Milk Production in Korean Cattle (한우 Mitochondrial DNA D-Loop 영역의 RFLP Marker가 산유량에 미치는 영향)

  • This study was performed to determine sequence variation and RFLP of the mt DNA D-loop region using Southern blot hybridization analysis and to develop mt DNA marker affecting milk production traits in Hanwoo cows. The PCR was used to amplify an 1142 bp fragment within the D-loop region of mt DNA using specific primers. Mt DNA were digested with seven restriction enzymes and hybridized using DIG-labeled D-loop probe. The mt DNA RFLP polymorphisms were observed in the four enzymes, BamHI, RsaI, XbaI and HpaII. Nucleotide substitutions were detected at positions 441 (G/C), 469 (T/C), 503 (C/T), 569 (G/A), 614 (C/A) and 644 (C/T) of the mt DNA D-loop region between two selected lines. Significant relationship between the XbaI RFLP type and breeding value was found(p<0.05). Cows with A type had higher estimated breeding values than those with B type (P<0.05) between high and low milk production lines. Therefore, the RFLP marker of mt DNA could be used as a selection assisted tool for individuals with high milk producing ability in Hanwoo.

Vegetation Structure of Abies nephrolepis Forest in Mt. Doota and Mt. Cheongok, Gangwon-do, Korea (강원도 두타산과 청옥산 일대 분비나무림의 식생 구조 분석)

  • This study was conducted to identify the vegetation structure of the Abies nephrolepis forests by analyzing species composition in Mt. Doota and Mt. Cheongok in the Gangwon region with the phytosociological method. In order to perform a field investigation, 18 survey areas with an area of about 400 m2 were set up for the stands where A. nephrolepis grow wild. Communities were classified through TWINSPAN and visualized using DCA (Detrended Correspondence Analysis). Species diversity analysis, importance value analysis, and DBH class analysis were employed to identify the characteristics of each community. As a result, the A. nephrolepis forests in Mt. Doota and Mt. Cheongok have been classified into 4 communities; GroupI, II, III and IV. GroupIis Quercus mongolica-A. nephrolepis competitive forest dominated by Q. mongolica and A. nephrolepis in the order. Group II is A. nephrolepis-Betula ermanii competitive forest dominated by A. nephrolepis, Sorbus commixta, and B. ermanii in the order. Group III is dominated by A. nephrolepis, but is expected to have competition between A. nephrolepis and Q. mongolica in the future due to the mass occurrence of Q. mongolica saplings. A. nephrolepis, S. commixta, and Q. mongolica are the dominant species in Group IV, and A. nephrolepis is the major dominant species in all layers. The species diversity of the A. nephrolepis forests in Mt. Doota and Mt. Cheongok is lower than other A. nephrolepis forests in Mt. Seorak, Maruguem, and Mt. Odae. It is necessary to establish conservation measures and to continuously monitor the A. nephrolepis forests in Mt. Doota and Mt. Cheongokis due to the harsh environment and the competition that may occur between A. nephrolepis and deciduous trees such as Q. mongolica in the future.

mtDNA Analysis of 5 Species of the genera Moroco and Phoxinus(Pisces, Leuciscinae) (황어아과어류 2속 5종의 mtDNA분석)

  • 한국산 담수어류의 잉어목, 황어아과(Leuciscinae) 어류 2속 5종의 계통적 유연관계를 구명하기 위하여 mtDNA분석을 실시하였다 6 base를 인지하는 10개의 제한효소를 처리하여 얻어진 강tDNA의 크기는 16.5-17.5 Kb였으며 Bcl I, Bgl I, Bgl II, Hin dIII, Pvu II, Xba I 등은 종간 차이가 뚜렷하였다. 각종의 집단간 mtDNA분화정도는 매우 낮았으나(p=1% 미만) M. oxyephalus의 무주집단과 제주집단은 예리적으로 큰 차이를 보였다(p=5.3%) Moroco속의 종간 분화정도를 비교한 결과 M. oxycephafus와 M. lagowsk서 사이가 평균 f=7.2%로 근면관계가 제일 가까웠고 M. keumkang과 M. semotilus는 타종들과 근연관계가 제일 멀었다. Moroco속과 Phoxinus속간의 평균 유전적 분화정도는 f=13.7%로 현저한 차이를 보였다. Brown 등(1979)의 공식을 이용하여 이들 황어아과 2속 5종의 분화시기를 추정한 결과 이들은 후기 선신세(Pliocene)와 흥적세(Pleistocene) 사이에 분화된 것으로 추정되었으며 이 결과는 동위효소 연구에서 얻어진 결파(Yang and Min, 1989)와 잘 일치한다.

  • In this study, we investigated the effect of cadmium on genes expression related to zinc homeostasis in HT22 hippocampal neuron cell line by RT -PCR and western blotting technics. In the time-course effect, cadmium up-regulated the relative levels of MT -I and MT -II to~b-actin at 4 hr after treatment. These effects were consistent with MT -I/II protein contents by western blot analysis. But MT -III, a specific MT isoform in brain, was not affected by cadmium. (omitted)

  • During the course of the field trip to Mt. Baekdu and Manchurian region especially along the border of North Korea, young flower buds were collected and fixed to analyze the meiotic configuration of chromosomes and to discuss the cytotaxonomic characteristics of these areas. A total of 29 taxa with 31 populations of vascular plants were analyzed chromosomally. Among them, 26 taxa with 28 populations were collected from Mt. Baekdu which include Papaver radicata var. pseudoradicata, Bupleurum euphorbioides, Oplopanax elatus, Silene jenisseensis, and Gentiana jamesii and three taxa collected from Manchurian region which include Scrophularia buergeriana, Ostericum grosseserratum and Callistephus chinensis.   • Sampels of Korean field mice (apodemus peninsulae peninsulae Thomas ) from six localities in Korea were used for the analyses of mitochondrial DNa (Mt DNA) fragment patterns resulted from the digestion with eight restriction enzymes. A total of 29 fragments were recognized and seven mtDNA clones were revealed. The nucleotide-sequence divergences (p) among the seven mtDNA clones ranged from 0.42% to 2.01%. Moreover, the seven clones were grouped into three major subgroups with the mean divergence value of 1.52% among them. One subgroup was composed of three clones of 18 sample from three localities (16, Cheongu: 1, Mt. Sobaek : 1, Mt. weolak) L another subgroup, three clones of eight samples from four localities (2, Cheongju ; 2 , Mt. Weolak ; 2, Mt. Gaya ; 2, haenam) ; and the last subgroup, one clone of two samples from Cheongju. Three subgroups were also distinct with one another in their mtDNA genotypes of Stu I and the former two subgroups differed from the last subgroup in their genotypes with Pvu II. Further analyses with additional samples from various localities in Korea appeared to be necessary in order to clarify the taxonomic status of the distinct mtDNA subgroups.
