포유동물세포의 Forward Mutation을 지표로 한 Mouse Lymphoma Thymidine Kinase (tk+/-) Gene Assay (In vitro Mouse Lymphoma Thymidine Kinase (tk+/-) Gene Forward Mutation Assay in Mammalian cells)

  • The mouse lymphoma thymidine kinase (tk+/-) gene assay (MOLY) using L5178Y tk+/- mouse lymphoma cell line is one of the mammalian forward mutation assays. It is well known that MOLY has many advantages and more sensitive than the other mammalian forward mutation assays such as x-linked hyposanthine phosphoribosyltransferase (hprt) gene assay. The target gene of MOLY is a heterozygous tk+/- gene located in 11 chromosome of L5178Y tk+/- cell, so it is able to detect the wide range of genetic changes like point mutation, deletion, rearrangement, and mitotic recombination within tk gene or deletion of entire chromosome 11. MOLY has relatively short expression time (2-3 days) compared to 1 week of hprt gene assay. MOLY can also induce relatively high mutant frequency so a large number of events can be recorded. The bimodal distribution of colony size which may indicate gene mutation and chromosome breakage potential of chemicals according to mutation scale such as large normal-growing mutants and small slow-growing mutants can be observed in this assay. The statistical analysis of data can be performed using the MUTANT program developed by York Electronic Research in association with Hazelton as recommended by the UKEMS (United Kingdom Environmental Mutagen Society) guidelines. This report reviewed MOLY using the microtiter cloning technique (microwell assay).

Photo Diode Array형의 휴대용 근적외 분광기와 FT 근적외 분광기를 이용한 Hairless Mouse 피부 수분 정량 (Quantification of Skin Moisture in Hairless Mouse by using a Portable NIR System and a FT NIR Spectrometer)

  • In this study, the performance of a portable NIR system and a FT NIR spectrometer were compared to determine water content of hairless mouse skin. The stratum corneum parts wer e separated from the epidermal tissues by trypsin solution. NIR diffuse reflectance spectra of hairless mouse skin were acquired using a fiber optic probe. In the near infrared, water molecules show two clear absorption bands at 1450 nm from first overtone of O-H stretching and 1940 nm from the combination involving O-H stretching and O-H deformation. It was found that the variations of O-H absorption band according to water content. Partial least squares regression (PLSR) was applied to develop a calibration model. The PLS model showed a good correlation between NIR predicted value and the absolute water content of separated hairless mouse skin, in vitro. For both the portable and the FT NIR spectrometer, These studies showed the possibility of a rapid and nondestructive skin moisture measurement using NIR spectroscopy. The portable NIR spectrometer with a photodiode arrays-microsensor could be more rapidly applied for the determination of water content with comparable accuracy with the performance of a FT spectrometer .

User Identification Using Real Environmental Human Computer Interaction Behavior

  • In this paper, a new user identification method is presented using real environmental human-computer-interaction (HCI) behavior data to improve method usability. User behavior data in this paper are collected continuously without setting experimental scenes such as text length, action number, etc. To illustrate the characteristics of real environmental HCI data, probability density distribution and performance of keyboard and mouse data are analyzed through the random sampling method and Support Vector Machine(SVM) algorithm. Based on the analysis of HCI behavior data in a real environment, the Multiple Kernel Learning (MKL) method is first used for user HCI behavior identification due to the heterogeneity of keyboard and mouse data. All possible kernel methods are compared to determine the MKL algorithm's parameters to ensure the robustness of the algorithm. Data analysis results show that keyboard data have a narrower range of probability density distribution than mouse data. Keyboard data have better performance with a 1-min time window, while that of mouse data is achieved with a 10-min time window. Finally, experiments using the MKL algorithm with three global polynomial kernels and ten local Gaussian kernels achieve a user identification accuracy of 83.03% in a real environmental HCI dataset, which demonstrates that the proposed method achieves an encouraging performance.

마우스 형태에 따른 상지의 근활성도의 변화 (A Study of Muscle Activation in Upper Extremity According Mouse Shape)

  • Background : The purpose of the study was: to investigate muscle activation of upper arm according mouse shape. Methods : Twenty person(mean age : 23. 7) who have healthy condition was participated this study, we collected data of muscle activation using by EMG from upper trapezius(Tr), deltoid middle fiber(De), extensor digitorum(Ed), first dorsal interosseous(Di) during participants was performed click and drag according various mouse. Mouse shape was divided 4 level as follow shape 1 was very small, 2 was small, 3 was moderate, 4 was large. Data was analyzed ANOVA, independent t-test using by SPSS ver18.0. Results : There was significantly difference of muscle activation among each muscle according mouse shape in drag and click. In shape 1, 4, there was significantly difference of muscle activation of Tr, De, Ed between drag and click except Di. In shape 2, 4, there was significantly difference of muscle activation of all muscle between drag and click. Conclusion : We knew that extensor digitroum showed more higher muscle activation than other muscle in drag, first dorsal interoseous showed more higher muscle activation that other muscle in click. We suggest that mouse shape was very important factor in order to prevent skeletal muscular disorder for computer user, and mouse shape can reduce muscle fatigue during computer work.

  • The purpose of this study is to develope a computer mouse using the head movements and eye blink in order to help the disability persons who can't move the hands or foot because of car accident or cerebral apoplexy. The mouse is composed of two gyro-sensors and photo sensor. The gryo-sensors detect the head horizontal and vertical angular velocities, respectively. The photo sensor detect the eye blink to perform click, double click, and to reset the head position. In the results, we could control the mouse points in real time using the proposed system.

Effects of Heating on Hydroxyl Radical-Generated Toxicity in Mouse Forebrain Tissue Culture

  • This experiment was carrid out to know the effects of heating and serum on hydroxyl radicals in embryonic mouse forebrain (cerebrum) culture. The heating to mouse embryonic cerebrum cells in culture was done in a water bath at 43${\circ}C$ for 60min. After that, two supernatants were prepared at 20 hrs and 48 hrs respectively after heat treatment to the brain cells. To find out the heating effects on neuron cells, mouse cerebrum cells (13 embryonic day) were cultured in hydroxyl radical generation system composed of 20mU/ml glucose oxidase (GO system), using condition of normal culture media (MEM, 5% serum, 5% $CO_2$or supernatant prepared after heating at 43${\circ}C$ for 60 min in a water bath. Supernatant prepared at 20 hrs after heat treatment had a greater protective effects against hydroxyl radical than supernatant prepared at 48 hrs after heat treatment . Otherwise, the protective effect of serum against hydroxyl radicals in the cultured brain cells is higher than that in the heat treatment. These results indicated that serum in culture media reduced cytotoxicity of hydroxyl radicals in mouse forebrain culture, also that heat treatment showed the protective effects against hydroxyl radicals generated with 20mU/ml GO system in mouse forebrain culture.

VDT 작업시 짧은 휴식시간과 마우스 클릭이 주관적 불편도에 미치는 영향에 관한 연구 (The Effects of Short Break Time and Mouse Clicks on the VDT Work by using Subjective Discomfort)

  • This paper suggests the best work conditions, including short break time and number of mouse clicks on a computer, to prevent MSDs(Musculoskeletal Disorders) on VDT(Visual Display Terminal) work. Discomfort measures are calculated according to the Borg's CR-10 Scale. The short break time conditions are grouped into 7, 15, and 30 seconds after every 10-minute work period and the number of mouse clicks are divided into 10 clicks/min, 20clicks/min, and 30clicks/min. The result of the ANOVA on the shift value of subjective discomfort shows the following: 1) Regarding the break time and the number of mouse clicks, there are statistical differences between the measured values for the neck and the wrist(p<0.05). 2) Regarding the number of mouse clicks, there are statistical differences between the measured values for the shoulder and the forearm(p<0.1). 3) Regarding the break time and the number of mouse clicks, there are no statistical differences between the measures values for the eyes, upper arms and back(p<0.1).

  • Phenotypic analysis of gene-specific knockout (KO) mice has revolutionized our understanding of in vivo gene functions. As the use of mouse embryonic stem (ES) cells is inevitable for conventional gene targeting, the generation of knockout mice remains a very time-consuming and expensive process. To accelerate the large-scale production and phenotype analyses of KO mice, international efforts have organized global consortia such as the International Knockout Mouse Consortium (IKMC) and International Mouse Phenotype Consortium (IMPC), and they are persistently expanding the KO mouse catalogue that is publicly available for the researches studying specific genes of interests in vivo. However, new technologies, adopting zinc-finger nucleases (ZFNs) or Transcription Activator-Like Effector (TALE) Nucleases (TALENs) to edit the mouse genome, are now emerging as valuable and effective shortcuts alternative for the conventional gene targeting using ES cells. Here, we introduce the recent achievement of IKMC, and evaluate the significance of ZFN/TALEN technology in mouse genetics.

Characterization of Brain Tumor Cell using Vasopressin-SV40 T Ag Transgenic Mouse

  • In previous reports, pVPSV.IGR2.1 transgenic mouse were described that brain tumor and lymphoma by reason of Vasopressin-SV40 T antigen. In this study, we produced pVPSV.IGR3.6 transgenic mouse that used pVPSV.IGR3.6 vector. Expression of transgene was vary different in transgenic mouse. We obtained 6 transgenic mouse line, moreover they had died at the age of 2~6 weeks without transmitting the transgene to their offspring, and had tumorigenesis on same location with pVPSV.IGR2.1 transgenic mouse. Only a founder mouse was investigated for expression of fusion gene. Here we extended this transgenic approach to the study of tumor progression. From the mouse, we confirmed brain tumor cell, after then cultured for investigate characterization. In this report, we demonstrate that reduction of survival rate in transgenic mouse fused vasopressin gene length, acquisition of brain tumor cell, composition with astrocyte cells and neuronal cells. Finally, cells had no change with increase of passage.

Insights into granulosa cell tumors using spontaneous or genetically engineered mouse models

  • Granulosa cell tumors (GCTs) are rare sex cord-stromal tumors that have been studied for decades. However, their infrequency has delayed efforts to research their etiology. Recently, mutations in human GCTs have been discovered, which has led to further research aimed at determining the molecular mechanisms underlying the disease. Mouse models have been important tools for studying GCTs, and have provided means to develop and improve diagnostics and therapeutics. Thus far, several genetically modified mouse models, along with one spontaneous mouse model, have been reported. This review summarizes the phenotypes of these mouse models and their applicability in elucidating the mechanisms of granulosa cell tumor development.
