Hoechst 33258 Staining을 이용한 웅성 생쥐 성세포의 간편 분류 (Simple Classification of Male Mouse Germ Cells using Hoechst 33258 Staining)

  • In the study for a differentiation and development of spermatogonial cells, the researchers should commonly require a simple, fast and reasonable method that could evaluate the developmental stage of male germ cells without any damage and also relentlessly culture them so far as a cell stage aiming at experimental applications. For developing the efficient method to identify the stage of sperm cells, the morphological characteristics of sperm cells were investigated by staining the cells with blue fluorescent dye Hoechst 33258, and a criterion for male germ cell classification was elicited from results of the previous investigation, then the efficiency of the criterion was verified by applying it to assort the germ cells recovered from male mice in age from 6 to 35 days. As morphological characteristics, spermatogonia significantly differed from spermatocytes in size, appearance and fluorescent patches of nucleus, and spermatids could also be distinguished from spermatozoa by making a difference in the volume and shape of nucleus and the shape and fluorescence of tail. Aforesaid criterion was applicable for classifying in vitro cultured sperm cells by verifying its efficiency and propriety for assorting the stages of testicular germ cells. However, the fluorescent staining showed that germ cells in mouse testis should be dramatically differentiated and developed at 21 days and 35 days of age, which were known as times of sexual puberty and maturity in male mice, respectively. In conclusion, the results indicated that this simple criterion for sperm cell classification using fluorescence staining with Hoechst 33258 may be highly efficient and reasonable for spermatogenesis study.

보기감비탕(補氣減肥湯)이 고지방사료 식이로 비만이 유발된 생쥐에게 미치는 영향 (Inhibitory Effects of Bogigambi-tang on the Obese-mouse Induced by High Fat Diet)

  • In order to investigate the effects of Bogigambi-tang(here in after referred to BGGBT) on the obese gene and obese inhibitory, C57BL/6 mice were induced by high fat diet. C57BL/6 mice were divided into three groups(normal, high fat diet with control, high fat diet with BGGBT extract) and fed for 15 weeks. Items of this experimental study are as follows. Body weight change, final inclose of body weight, the weight change of the adipocytes in body, the level change of ALT, AST, total cholesterol, LDL-Cholesterol, triglyceride, glucose, free fatty acid and creatinine, the expression of ${\beta}$3AR and leptin gene in primary adipocytes, the production change of leptin in primary adipocytes, the expression of ${\beta}$3AR and leptin in adipocytes tissue. The following results have been obtained All experimental group have shown that the weight and the final increase of weight have decreased considerably. All experimental group have shown that the amount of the adipocyte in weight has decreased considerably. All experimental group have shown that the amount of leptin has decreased considerably. All experimental group have shown that the revelation of ${\beta}$3AR in primary adipose cell and 3T3-L1 cell has increased considerably, and that the revelation of leptin in primary adipose cell and 3T3-L1 cell has decreased considerably, All experimental group have shown that the size of adipocyte in adipocytes tissue has decreased. The high density group have shown that the adipose vacuoles in liver tissue has decreased considerably, and that the cell nucleuses has similar with normal group.

Phamacopuncture and Dermal Application of Sebalgukhwa-san: Effects on Hair Growth in a Mouse Model of Alopecia

  • Background: This study was conducted to evaluate the effects of pharmacopuncture and dermal application of Sebalgukhwa-san extracts on hair growth in an alopecia mouse model. Methods: Twenty-one C57BL/6 mice were divided into 3 groups; control group-normal saline injection or vehicle solution application, positive control group-minoxidil (MNXD), experimental group-pharmacopuncture and applied Sebalgukhwa-san (SGS) extract. The effects of the treatment on hair growth, were determined through photographs, and phototrichogram analysis by folliscope. Hair follicle morphometry by hematoxylin-eosin staining was performed, and hair growth-related protein expression of vascular endothelial growth factor, insulin like growth factor-1, and transforming growth factor-beta 1 were monitored by Western blotting. Serum levels of aspartate aminotransferase and alanine aminotransferase were measured for liver function test. Results: Body weight increased consistently in all groups. Hair growth was improved in the MNXD and SGS groups compared with the control. Hair density and thickness improved statistically significantly in the MNXD and SGS groups compared with the control p < 0.05. The number of hair follicles improved in the MNXD and SGS groups compared with the control but the size did not. The expression of vascular endothelial growth factor and insulin like growth factor-1 increased, and there was a decrease in the expression of transforming growth factor-beta 1 in the MNXD and SGS groups compared with the control, however, there was no significant difference. Sebalgukhwa-san treatment had no toxicity in liver function tests. Conclusion: Pharmacopuncture and dermal application of Sebalgukhwa-san extract may be therapeutically beneficial for the treatment of alopecia.

Anti-obesity effects of Tenebrio molitor larvae powder in high-fat diet-induced obese mice

  • Purpose: Obesity is a serious public health issue for the modern society and is considered a chronic health hazard. There are many surgical and pharmacological approaches to treat obesity. However, various potentially hazardous side effects remain the biggest challenge. Therefore, diets based on foods derived from natural products have gained increasing attention compared to anti-obesity drugs. Recently, research on edible insects as a food source has been a topic of considerable interest in the scientific communities. This study examined the anti-obesity effects of ingesting an edible insect by feeding a high-fat diet (HFD)-induced obese mouse models with a diet containing Tenebrio molitor larvae powder (TMLP). Methods: Six-week-old female C57BL/6J mice were divided into 4 groups according to treatment: 100% normal diet (ND), 100% HFD (HFD), HFD 99% + TMLP 1% (TMLP), and HFD 97% + TMLP 3% (TMLP 3%). TMLP was added to the HFD for 6 weeks for the latter two groups. Results: Compared to the HFD group, mice in the TMLP group showed weight loss, and micro-computed tomographic imaging revealed that the volume of the adipose tissue in the abdominal area also showed significant reduction. After an autopsy, the fat weight was found to be significantly reduced in the TMLP group compared to the HFD group. In addition, the degree of fat cell deposition in the liver tissue and the size of the adipocytes significantly decreased in the TMLP group. Reverse transcription polymerase chain reaction analysis for the mRNA expression of adipogenesis-related genes namely CCAAT-enhancer-binding proteins (C/EBP-β, C/EBP-δ), and fatty acid-binding protein 4 (FABP4) showed that the expression levels of these genes were significantly reduced in the TMLP group compared to the HFD group. Serum leptin level also decreased significantly in the TMLP group in the comparison with the HFD group. In addition, total cholesterol, triglyceride, and glucose levels in mouse serum also decreased in the TMLP group. Conclusion: Taken together, our results showed that TMLP effectively inhibited adipocyte growth and reduced body weight in obese mice.

Improvement of antithrombotic activity of red ginseng extract by nanoencapsulation using chitosan and antithrombotic cross-linkers: polyglutamic acid and fucoidan

  • Background: Red ginseng (RG) extract, especially ginsenoside Rg1 and Rb1 fractions has been reported to have antithrombotic activities. However, gastric instability and low intestinal permeability are considered to be obstacles to its oral administration. We hypothesized that stability, permeability, and activities of RG might be improved by encapsulation within nanoparticles (NPs) prepared with antithrombotic coating materials. Methods: RG-loaded chitosan (CS) NPs (PF-NPs) were prepared by complex ionic gelation with the antithrombotic wall materials, polyglutamic acid (PGA), and fucoidan (Fu). The concentrations of PGA (mg/mL, X1) and Fu (mg/mL, X2) were optimized for the smallest particle size by response surface methodology. Antithrombotic activities of RG and PF-NPs were analyzed using ex vivo and in vivo antiplatelet activities, in vivo carrageenan-induced mouse tail, and arteriovenous shunt rat thrombosis models. Results: In accordance with a quadratic regression model, the smallest PF-NPs (286 ± 36.6 nm) were fabricated at 0.628 mg/mL PGA and 0.081 mg/mL Fu. The inhibitory activities of RG on ex vivo and in vivo platelet aggregation and thrombosis in in vivo arteriovenous shunt significantly (p < 0.05) increased to approximately 66.82%, 35.42%, and 38.95%, respectively, by encapsulation within PF-NPs. For an in vivo carrageenan-induced mouse tail thrombosis model, though RG had a weaker inhibitory effect, PF-NPs reduced thrombus significantly due to the presence of PGA and Fu. Conclusion: PF-NPs contributed to improve the activities of RG not only by nanoencapsulation but also by antithrombotic coating materials. Therefore, PG-NPs can be suggested as an efficient delivery system for oral administration of RG.

Gallic Acid Hindered Lung Cancer Progression by Inducing Cell Cycle Arrest and Apoptosis in A549 Lung Cancer Cells via PI3K/Akt Pathway

  • This study elucidates the anti-cancer potential of gallic acid (GA) as a promising therapeutic agent that exerts its effect by regulating the PI3K/Akt pathway. To prove our research rationale, we used diverse experimental methods such as cell viability assay, colony formation assay, tumor spheroid formation assay, cell cycle analysis, TUNEL assay, Western blot analysis, xenograft mouse model and histological analysis. Treatment with GA inhibited cell proliferation in dose-dependent manner as measured by cell viability assay at 48 h. GA and cisplatin (CDDP) also inhibited colony formation and tumor spheroid formation. In addition, GA and CDDP induced apoptosis, as determined by the distribution of early and late apoptotic cells and DNA fragmentation. Western blot analysis revealed that inhibition of the PI3K/Akt pathway induced upregulation of p53 (tumor suppressor protein), which in turn regulated cell cycle related proteins such as p21, p27, Cyclin D1 and E1, and intrinsic apoptotic proteins such as Bax, Bcl-2 and cleaved caspase-3. The anti-cancer effect of GA was further confirmed in an in vivo mouse model. Intraperitoneal injection with GA for 4 weeks in an A549-derived tumor xenograft model reduced the size of tumor mass. Injection of them downregulated the expression of proliferating cell nuclear antigen and p-Akt, but upregulated the expression of cleaved caspase-3 in tumor tissues. Taken together, these results indicated that GA hindered lung cancer progression by inducing cell cycle arrest and apoptosis, suggesting that GA would be a potential therapeutic agent against non-small cell lung cancer.

Webcam-Based 2D Eye Gaze Estimation System By Means of Binary Deformable Eyeball Templates

  • Eye gaze as a form of input was primarily developed for users who are unable to use usual interaction devices such as keyboard and the mouse; however, with the increasing accuracy in eye gaze detection with decreasing cost of development, it tends to be a practical interaction method for able-bodied users in soon future as well. This paper explores a low-cost, robust, rotation and illumination independent eye gaze system for gaze enhanced user interfaces. We introduce two brand-new algorithms for fast and sub-pixel precise pupil center detection and 2D Eye Gaze estimation by means of deformable template matching methodology. In this paper, we propose a new algorithm based on the deformable angular integral search algorithm based on minimum intensity value to localize eyeball (iris outer boundary) in gray scale eye region images. Basically, it finds the center of the pupil in order to use it in our second proposed algorithm which is about 2D eye gaze tracking. First, we detect the eye regions by means of Intel OpenCV AdaBoost Haar cascade classifiers and assign the approximate size of eyeball depending on the eye region size. Secondly, using DAISMI (Deformable Angular Integral Search by Minimum Intensity) algorithm, pupil center is detected. Then, by using the percentage of black pixels over eyeball circle area, we convert the image into binary (Black and white color) for being used in the next part: DTBGE (Deformable Template based 2D Gaze Estimation) algorithm. Finally, using DTBGE algorithm, initial pupil center coordinates are assigned and DTBGE creates new pupil center coordinates and estimates the final gaze directions and eyeball size. We have performed extensive experiments and achieved very encouraging results. Finally, we discuss the effectiveness of the proposed method through several experimental results.

연소 생성물 내 입자상 물질의 입도에 관한 연구 (A Research of Grain Size Analysis of Particulate Matter in Fire Effluent)

  • 현행 건축법은 건축물의 규모, 용도에 따라 화재확산방지와 재실자의 보호를 위하여 실내 마감 재료를 불연, 준불연, 난연재료로 시공하도록 규정하고 있으며, 가스 유해성 시험(KS F 2271)에 따라 해당 시료의 연소 생성물에 노출된 실험용 쥐의 평균행동정지시간을 측정하여 연소 생성물의 유해성을 평가하고 있다. 실험용 쥐를 대상으로 가스상, 입자상 물질의 복합적 유해효과를 분석한 결과인 평균행동정지시간이 15분의 노출 시험 중 9분 이내로 측정된 경우, 연소 유해성 평가 결과가 부적합한 것으로 판정한다. 이에 본 연구에서는 연소 생성물에 포함된 가스상, 입자상 물질 중에서 실험용 쥐의 행동정지에 미치는 영향이 큰 요소를 규명하기 위하여 연소 생성물에 포함된 입자상 물질의 입도 분석을 실시하여 위험도의 상관관계를 밝혔다.

인간 골육종 세포주에서 Zoledronic acid의 종양 억제에 대한 생체내 실험 (Tumor Suppressive Effect of Zoledronic Acid on Human Osteosarcoma Cells in Vivo)

  • 목적: Bisphosphonate(BPs)는 endogenous pyrophosphates의 유사체로 Paget's disease, 골다공증, 종양 유발성 골용해와 같은 골격계 질환의 치료에 쓰이고 있으며, 유방암의 골파괴성 전이에 대한 치료제로 사용되어 지고 있다. 골전이의 양상이 골흡수성 및 골생성이 혼합되어 나타나는 전립선암의 전이에서도 치료제의 하나로 이용되고 있다. BPs는 파골세포성 골흡수가 과하게 일어난 질환에 대해서는 비교적 널리 알려져 있지만, 골종양 세포에 대한 직접적인 효과에 대해서는 아직 밝혀져 있지 않다. 가장 강력한 질소 함유(nitrogen-containing) BPs인 Zoledronic acid(ZOL)로서 골육종이 발현된 nude mouse model을 이용하여 ZOL의 종양 억제능을 생체내 실험으로 확인하고자 하였다. 대상 및 방법: 인간 골육종 세포주로는 MG-63과 HOS 골육종 세포주를 이용하였고, 종양의 크기 변화를 육안으로 확인하기 위하여 GFP 유전자가 형질 전환된 MG-63-GFP, HOSGFP 세포를 6주령된 수컷 마우스 10마리에 각각 피하주사하여 종양의 조각이 $3{\times}3{\times}3$ mm이 될 때까지 사육한 후, ZOL을 120 ug/kg의 농도로 일주일에 2번 피하에 주사하였다. 종양의 크기를 일주일에 두 번씩 측정하고, 형광조명을 이용하여 촬영하였다. 결과: HOS 골육종 세포주를 이용한 생체실험에서 대조군의 종양의 평균 크기는 2,520 $mm^3$이며 ZOL 투여군은 131 $mm^3$로서 94%의 감소를 보이며, MG-63 골육종 세포주를 이용한 생체실험에서는 대조군의 종양의 평균 크기는 2,866 $mm^3$이며 실험군은 209 $mm^3$로서 72%의 감소를 보였다(P<0.05). 결론: Nude mouse를 이용한 생체실험에서 ZOL은 골육종의 세포사멸에 직접적인 영향을 미치며 이것은 앞으로 골육종 치료의 약제중 하나로 선택되어 질수 있다고 판단되며, 종양세포주에 따라 ZOL의 영향이 다를 수 있으므로 ZOL에 감수성 있는 종양세포를 찾아 적용하는 것이 좋을 것으로 사료된다.

Lewis 폐암 마우스 모델에서 톡소포자충에 의한 항암 및 항전이 효과 (Antitumor and Antimetastatic Effects of Toxoplasma Gondii in Mice with Lewis Lung Carcinoma)

  • 배 경: 종양은 우리나라뿐만 아니라 선전 국가에서도 주요 사망 원인의 하나로, 새로운 치료법의 개발이 절실히 요구된다. 최근 면역종양학의 발전으로 면역강화요법에 의한 종양의 면역치료에 대한 관심이 높아지고 있다. 이에 본 연구는 Lewis 폐암 마우스 모델을 대상으로 원충의 일종인 톡소포자충 (Toxoplasma gondii)에 의한 비특이적 면역증강요법에 의한 폐암의 성장 및 전이 억제 효과를 평가하고자 시행하였다. 방 법: C57BL/6 마우스 톡소포자충 충체 (마우스당 5개의 씨스트를 복강내로 주사) 혹은 Lewis폐암세포 (마우스당 $1{\times}10^6$씩 대퇴근육에 주사)를 여러 조합으로 처치하여 각 군별 생존기간, 주사부위 근육의 종양크기, 근육 및 폐장의 조직병리 소견을 조사하였다. 또한 각 군별 마우스를 톡소포자충 항원(마우스당 $50{\mu}g$)혹은 lymphokine(마우스당 0.5ml)으로 추가 면역한 다음 항암 및 항전이 효과를 비교하였다. 결 과: 톡소포자충 충제만을 감염시킨 마우스는 실험기간중 한 마리도 죽지 않았으나, 폐암세포만을 주입한 마우스(폐암대조군)의 평균 생존기간은 $29.1{\pm}4.4$일이었다. 톡소포자충 감염 후 2주에 폐암세포를 주입한 마우스 (전감염대조군), 충체와 폐암세포를 동시에 주입한 마우스 (동시감염대조군) 및 폐암세포 주입 후 충체를 감염시킨 마우스(후감염대조군)의 생존기간은 각각 $32.4{\pm}3.3$$30.9{\pm}5.0$일 및 $34.9{\pm}2.9$일로 폐암대조군에 비하여 모두 유의하게 증가하였으며(0.0001
