  • The present study was carried out to investigate the effects of season on semen characteristics, frozen-thawed sperm viability and testosterone concentration in Yorkshire boars. There were no significant differences in the semen volume and sperm concentration on Yorkshire boars among spring, summer, autumn and winter. However, the pH of sperm-rich and sperm-poor fractions in winter season was higher than in spring, summer and autumn season in Yorkshlre boars. Sperm motiliy and normal acrosome of raw semen in Yorkshire boars did not differ significantly among spring, summer, autumn and winter. However, motility and normal acrosome of frozen-thawed sperm were higher in spring season than in summer, autumn and winter. Serum testosterone concentrations in Yorkshire were higher in spring than summer, autumn and winter. In conclusion, we found out that serum testosterone concentrations were very important role for frozen-thawed sperm viability in Yorkshire boars.

  • The present experiments were undertaken to find out the effects of ova and cummulus cells on the motiliy and movement of mouse sperms in a capillary tube modified from the microtube culture system (Cho, 1974). The results obtained were as follows: 1. The motility of the mouse sperms cultured in vitro was decreased gradually as the culture period was prolonged or the concentration of sperms was diluted with the culture medium. 2. The ova whose cummulus cells were removed have some effects of reducing the sperm motility, but this effect seems to disappear at 8 hours of culture, whereas ova-cummulus cells complex showed a motility supprression effect only after 8 hours of culture. 3. Cummulus cells or ova-cummulus complex stimulated the movement of the sperm through a capillary tube by some degree. It is, therefore, assumed that cummulus cells secrete some factors which induce the movement of the sperm toward them.

  • The purpose of this research is to develop the simple freezing method for sperm cryopreservation in the Sevenband grouper and Kelp grouper. The motility and survival rate were checked to investigate the effects of diluent and cryoprotectant on sperm cryopreservation. In cryopreservation experiments, 300 mM glucose and 15% dimethyl sulfoxide (DMSO) were used as a diluent and a cryoprotectant, respectively. As a results of cryopreservation, the motility rate of Sevenband grouper was found to be more than 90%, and that of Kelp grouper was found to be more that 60% in sperm 5 months after cryopreservation.
