Phyllostachys consists of high and fast growing trees and is a genus in the family Gramineae. The genus has many species in Asia, with main distribution being in India and China. One of the most popular sequences for phylogenetic inference at the generic and infrageneric levels in plants is the internal transcribed spacer (ITS) region of the 18S-5.8S-26S nuclear ribosomal cistron. We evaluated four taxa with the ITS region to estimate phenotypic relationships within the genus Phyllostachys in Korea. Alignment of the DNA sequences required the addition of numerous gaps. Sequence variation within the Phyllostachys was mostly due to natural selection, although several indels and inserts were found. Within the genus Phyllostachys, P. nigra and P. nigra var. henonis were the relatives in the three phylogenetic analyses (MP, ML, and NJ). However, some external nodes were poorly supported. Morphological traits and simple repeats (ISSR) represented the result of a relationship similar to the that of ITS sequences in the genus Phyllostachys. This suggests that ITS sequences are very informative for identification of these taxa.
The removal of nitrogen compounds from a wastewater is essential and it is often accomplished by bio-logical process. An aerobic nitrate-removing bacterium was isolated from a municipal sewage treatment plant and soil. On the basis of its morphological, cultural and physiological characteristics and 16S rRNA sequencing data, this strain was identified as Pseudomonas fluorescens, and named as P. fluorescens K4. The optimal conditions of the initial pH and temperature of media for its growth were $7.0{\sim}8.0$ and $30^{\circ}C$, respectively. P. fluorescens K4 was able to remove 99.9% of nitrate after 24 h in a culture. The strain could grow with a nitrate concentration up to 800 mg/l and was able to remove 99.9% of nitrate after 104 h of incubation. The optimal electron donor was sodium citrate for a nitrate removal. The strain K4 showed a capability of a complete nitrate removal when the initial C/N ratio was 1.0. An effect of the initial seed concentration was observed for a cell of 10% (v/v) for a nitrate removal. Especially P. fluorescens K4 could completely remove 200 mg/l ammonium for 3 days.
The purpose of this study was to investigate eco-friendly measures to manage major diseases which cause heavy economic damages to ginseng. Morphological, physicochemical, and molecular biological species identification was carried out after isolating useful antagonistic bacteria from ginseng fields. In addition, optimal conditions for mass culture were established, and he efficacy of the bacteria in the prevention of the diseases was verified in the field. The results showed that about 150 bacteria were extracted from 150 ginseng fields in the whole county. Among them, B. pyrrocinia LA101 was finally selected, which had a strong antagonistic potency against Alternaria panax, Botrytis cinerea, Rhizoctonia solani, and Cylindrocarpon destructans on agar media. The B. pyrrocinia LA101 is a baculiform gram-negative bacterium identified as Burkholderia pyrrocinia according to results from an API(Analytical Profile Index) kit, 16S rRNA, and gyrase gene sequencing analysis. It was donated to the microbe bank of the Agricultural Genetic Resources Center at the National Academy of Agriculture Science under the Rural Development Administration on September 28, 2011 (Donation No. KACC91663P). A patent for the mass culture technology was granted in August 2012 (Patent No. 10-1175532).
Chemo- and numerical taxonomic studies on the isolate SL-387 producing novel aminopeptidase M inhibitors MR-387A and B were carried out The genus of the isolate was determined as Streptomyces by cultural and morphological data and chemical indices. Forty one taxonomic unit characters were tested for determining the species of the isolate, and the data were analyzed numerically using a computer program as called TAXON. The isolate was best matched to Streptomyces neyagawaensis in the major cluster 18 of Streptomyces with $S_{SM}$ value of 75.67%. On the base of chemotaxonomic data and TAXON analysis, the isolate SL-387 was identified to be a member of Streptomyces neyagawaensis.

Natural clonal stock of triploid crucian carp, Carassius aurahs was identified and its cytogenetic, molecular genetic and morphological traits were studied. Cytogenetic analysis of the clonal crucian carp revealed that they were natural triploidy, evidenced by 1.5-fold increases of cell size, DNA content, and chromosome number. Multi-locus DNA fingerprinting using $(GATA)_4$ probe showed that they had an identical fingerprint profile, indicating the clonal propagation of the population. External morphology and morphometric characteristics of triploid individuals were much uniform compared to those of diploids. Natural triploid crucian carp was proven to be all-female in this study.

In August 2013, brown rot was observed on cherry fruits (Prunus avium L.) in Hwaseong city, Korea. Fruit rot first appears as small, circular brown spots that increase rapidly in size causing the entire fruit to rot. Grayish spores appear in tufts on rotted areas. Based on these morphological characteristics, the two isolates were identified as Monilinia fructicola. Molecular analysis of 5.8S subunit and flanking internal transcribed spacers (ITS1 and ITS2) was performed to confirm the identification. The ITS sequences had 100% identity with those of other reference M. fructicola isolates of NCBI GenBank. Pathogenicity was tested using spore suspension inoculation on wounded or unwounded cherry fruits. The lesions were observed on wounded and unwounded fruits after inoculation for 7 days. This is the first report on M. fructicola causing brown rot on cherry fruits in Korea.

The crustose lichen genus Pertusaria comprises over ca. 800 species worldwide. In total, 20 Pertusaria species were localized to the Mt. Sorak and Jeju-do in Korea. To date, information regarding the distribution of Pertusaria species in the South Korean peninsula is scarce. In this study, we collected Pertusaria species across South Korea and identified them based on morphological, chemical, and molecular characteristics. Of the 387 samples collected, we identified 24 taxa and 1 variety, of which 17 were previously recorded, and 6 taxa were newly found in South Korea (P. leioplaca, P. leucosora var. violascens, P. texana, P. thiospoda, P. thwaitesii, and P. xanthodes), 2 known species were transferred to Varicellaria (Varicellaria lactea and V. velata), one species was transferred to Variolaria as a new record (Variolaria multipunctoides) and one was a new species (P. jogyeensis J. S. Park & J.-S. Hur, sp. nov.). Characteristics of the newly discovered species, P. jogyeensis, are as follows: smooth to bumpy thallus, scattered to crowded poriform apothecia, blackish ostioles, definitely sunken, thin yellowish green rims around ostioles, 8-spored ascus, and the presence of perlatolic acid and thiophaninic acid (chlorinated xanthone). Phylogenetic studies on P. jogyeensis based on the mitochondrial small subunit sequence revealed proximity to P. flavicans and P. texana, and supported its classification as a new species within the genus Pertusaria. Additionally, we describe the chemical composition and morphology of all listed species in detail and provide an artificial key for identification.
Four actinomycetes which produces extracellular chitinase were isolated from soil and organic matter all over the Chonnam provincial area. The chemical composition, morphological, cultural and physiological properties of isolated strain S-25, S-42, S-172 and S-267 were studied in relation to the toxonomical properties. All of strains contained phospholipids such as phosphatidyl ethanolamine, phosphatidyl inositol and diphosphatidyl glycerol. The components of cell wall in all strains have L, L-DAP, Glutamic acid, Alanine, Glycine and Glucosamine. The surface of spore is smooth and colony is grey in all strains, Based on the results obtained in these experiments all of strains are identified as Streptomyces sp.S-25, Streptomyces sp.S-42, Streptomyces sp.S-l72 and Streptomyces sp. S-267.
Taxonomic composition and diversity were wxamined by statistical analysis for bacterial communities in surface waters of the Sudong Stream, a tributary of North Han River. For total 282 isolates, Flavobacterium, Aeromonas and Enterobacteriaceae was identified by the deterministic schemes as a major group above 50% of total isolates in all sampling sites. Morphological, biochemical and physiological characteristics were numerically analyzed for bacterial isolates from each site and clustered into 15-28 groups. Not all statistically clustered groups were identical to the groups derived from deterministic identification. Especially, consistent relationship was not found in dendrograms for the groups with each a single strain which gas peculiar sugar-degrading activity. At a level of 80% similarity, bacterial diversity (H) was ranged as 2.37-3.14, and it was suggested that the research area was oligotrophic-mesotrophic status. Regional distribution of bacterial community was most heterogeneous at the site where large input of allochthonous materials or bacteria were occurred. And that was the significant factor for the compositions of bacterial communities in the Sudong stream.
The genetic relationship of Bletilla striata native to Korea and Japan was investigated using random amplified polymorphic DNA (RAPD). The 156 reproducible DNA bands, consisted of 58 polymorphic and 98 monomorphic bands, were obtained by polymerase chain reaction (PCR) with selected 10 random primers. The 8 Bletilla lines have been classified into three groups according to the similarity coefficient obtained by RAPD analysis. The dendrogram showed overall correlationship between similarity coefficient of 0.48 and 0.84. The first group included A (Bletilla striata native to Korea), B (Bletilla striata variegated and native to Korea in Mokpo), C (Bletilla striata variegated and native to Korea), D and E (Bletilla striata native to Japan). In this group, it was showed that B and C had the most similar genetic relationship. The similarity coefficient between D and E was 0.77. D and E had a very close resemblance in plant height and flower color with A native to Korea, respectively. The second group included only G (dwarf Bletilla native to Japan) and had a different morphological character compared to other cultivars. The last group included F and H (dwarf and variegated Bletilla native to Japan) and they had a similarity of variegation.
