Isolation and Identification of Cellulomonas fimi, Characteristics of its Cellulase and Conversion of the Sawdust into Ethanol (Cellulomonas fimi의 분리(分離) 및 동정(同定), cellulase 특성(特性)과 톱밥의 Ethanol 전환(轉換))

  • In the sheep and cattle's rumen, facultative anaerobic cellulolytic bacteria were isolated by using Hungate's roll tube technique. In the 21 isolated species, one was screened by its strong cellulolytic activity and identified as Cellulomonas fimi C-14 by investigate morphological, cultural, physiological characteristics and electron microgram. Optimum conditions of the cell growth and enzyme production were pH 6.5 an $30^{\circ}C$, Thiamine and biotin support a good growth of C. fimi C-14. In the enzyme activities, Crystalline cellulose hydrolyzing activity, CMCase activity and ${\beta}-glucosidase$ activity were 20.6, 226.6 and 0.56$(unit{\times}10^3/ml)$ at pH 6.0, $40^{\circ}C$. By addition of fungal cellulase, enzyme activity was increased. Simultaneous Saccharification Fermentation is better than two step fermentation in ethanol yield with Saccharomyces cerevisiae DY2.

Isolation, identification, and culture conditions of the strain producing antibacterial antibiotic (항세균성 항생물질을 생산하는 미생물의 분리, 동정 및 배양조건)

  • The strain with antibacterial activity was isolated among soil samples collected in Suwon area. The isolated strain was identified as Bacillus sp. YR-1 with respect to its morphological, cultural, and physiological characteristics. Optimal medium for the highest production of antibiotic was composed of sucrose 2.0%(w/v), peptone 2.0%(w/v) and NaCl 0.1%(w/v). The maximum production of antibiotic was shown at $35^{\circ}C$ for 48 hours with the initial pH 7.0.

  • A strain showing antibiotic activities against various bacteria and fungi was selected from approximately 2,000 microorganisms obtained from soil samples. This strain, designated as KH-431, was identified as a Clostridium sp. by its morphological, physiological and biochemical characteristics. The highest production of the antibiotics was achieved in a fermentation medium containing sorbitol, yeast extract, d-biotin and $CaCl_2$ The antibiotics were isolated from the culture broth by solvent extraction using ethyl acetate, silica gel column chromatography and recrystallization. Two kinds of antibiotics, KG-431A and KG-431B were obtained after the purification procedure, and only KG-431B was successful to recrystallize.

Diversity of Fungal Endophytes in Various Tissues of Panax ginseng Meyer Cultivated in Korea

  • Endophytic fungi were isolated from various tissues (root, stem, petiole, leaf, and flower stalk) of 3- and 4-year-old ginseng plants (Panax ginseng Meyer) cultivated in Korea. The isolated endophytic fungi were identified based on the sequence analysis of the internal transcribed spacer (ITS), 1-5.8-ITS 2. A morphological characterization was also conducted using microscopic observations. According to the identification, 127 fungal isolates were assigned to 27 taxa. The genera of Phoma, Alternaria and Colletotrichum were the most frequent isolates, followed by Fusarium, Entrophospora and Xylaria. Although 19 of the 27 taxa were identified at the species level, the remainder were classified at the genus level (6 isolates), phylum level (Ascomycota, 1 isolate), and unknown fungal species (1 isolate). Endophytic fungi of 13 and 19 species were isolated from 3- and 4-year-old ginseng plants, respectively, and Phoma radicina and Fusarium solani were the most frequently isolated species colonizing the tissues of the 3- and 4-year-old ginseng plants, respectively. The colonization frequency (CF%) was dependant on the age and tissue examined: the CFs of the roots and stems in the 3-year-old ginseng were higher than the CF of tissues in the 4-year-old plants. In contrast, higher CFs were observed in the leaves and petioles of 4-year-old plants, and endophytic fungi in the flower stalks were only detected in the 4-year-old plants. In conclusion, we detected diverse endophytic fungi in ginseng plants, which were distributed differently depending on the age and tissue examined.

Morphological Study of Microfilariae Found in the Area of Yongju-Gun (영주지방에 분포하는 사상충 자충에 대한 형태학적 연구)

  • The object to this study is to identify the genus and species of the microfilariae which were recently found in the area of Youngju-Gun. THe identification of the microfilariae was made on the morphologicla aspects. 1. Blood samples were collected through vena punction from known microfilariae carriers living in the newly confirmed filaria endemic area of Youngju0Gun in Jyongsang Pukdo Province. Youngju-Gun is located in the mountainous central part of Korean Peninsula. 2. The following fixation and staining techniques were applied. (1) Fixation by drying in the air, followed by staining with Azeo or Giemsa. (2) Knott's fixation method (2% formalin), followed by staining with Azur II. 3. A comparative study of the body length of the microfilariae after different fixation and staining techniques were applied. (1) Knott's fixation method followed by staining with Azur II : average body length found was 28.4$\mu$. (2) Dry fixation followed by staining with Giemsa : average body length found was 209.4$\mu$. (3) Dry fixation followed by staining with Azeo : average body length found was 205.4$\mu$. 4. The locations of the different body cells were measured in 60 individuals of microfilariae in the wet preparation fixed by Knott's method and stained with Azur II. The distance of the different body cells to cephalic apex of microfilariae was measured and calculated as a percentage of the total body length. The average results are as follows : BNC , 3.04% ; N, 22.74%, EP, 31.4% ; EC, 37.77%, G1 cell , 67.94%; G2 cell, 73.54% : G3 cell , 75.55% ; G4 cell, 77.65% ; AP, 82.02%%. 5. As a result of the above findings the microfilariae found in the above mentioned area could be identified as Brugia malai(BRUG, 1927) BUCKLEY, 1960.

  • Larval development of Chirona cristatus Ren and Riu, 1978, found in the low part of rocks in the intertidal clone or the shell of scallops, was described in detail and compared with those of other known barnacles. Durations from nauplius through cyprid to pinhead stage are three weeks at 20$\pm$0.5$^{\circ}C$. Trilobed labrum bearing three groups of slender hairs and frontolateral horns folded under the anterior cephalic shield margin are diagnostic features through all nauplius stages. The posterior border of the cephalic shield bears a pair of cephalic shield spines in nauplius stages IV,V and Ⅵ. There is no specific hispid sets at the fourth group of the antennal endpodite. The dorsal thoracic spine, abdominal process and posterior shield spine haute numerous small spines Morphological features such as the cephalic shield, labrum, abdominal process, antennules, antennae and mandibles in all nauplius and cyprid stages are illustrated and described. The numerical setations of antennule in this species are found to be practically helpful for intraspecific identification of barnacle nauplius stages without dissection.

Relationship Between EUV Coronal Jets and Bright Points Observed by SDO/AIA

  • We have investigated the relationship between EUV coronal jets and bright points observed by Solar Dynamic Observatory (SDO)/Atmospheric Imaging Assembly (AIA). For this we consider 39 EUV coronal jets from May 2010 to July 2011 in 171 A identified by Heliophysics Events Knowledgebase (HEK) which provides an automatic identification of coronal jets. We look for coronal jet-bright point pairs as follows. First, we select the size of event area as 360 arcsec * 360 arcsec where the coronal jets are located at the center of the area. Second, we select jet-bright point pairs in case that they are located at the same position or just adjacent. Third, we select jet-bright point pairs that are connected by loops each other. Otherwise, we select jet-bright points pairs as the nearest one. As a result, we present 19 coronal jet-bright point pairs. The mean distance of these pairs is 77.24 arcsec. According to their distance and morphological connection, we classify the following three groups: 1) Adjacent (6 events), 2) Loop connected (5 events), and 3) Not connected in appearance (8 events). The histogram of mutual distance has two peaks; the first peak corresponds to the first group and the other one to the second group. We compare these events with previous observations and theoretical models as well as discuss possible physical connections between jets and bright points.

  • Among 110 isolates of actinomycetes isolated from ginseng pathogen-suppressive soils, the three actinomycetes showing the effective controls to Fusarium solani or Cylindrocarpon destruc­tans causing ginseng root rots were identified according to their morphological, cultural and physio­logical characteristics on various culture media. Spore chains of K 6-2, S 2-1 and Y 2-2 were Spira (S), Retinaculum-apertum (RA) and Rectus-flexibilis (RF), respectively. Spore surfaces of K 6-2 were spiny, whereas S 2-1 and Y 2-2 were all smooth. Aerial mass colors of 3 isolates were gray series. As a result of various tests, they were identified as Streptomyces variabilis, Streptomyces virgi­niae and Streptomyces griseo/us, respectively.

Plant Growth Promoting Fungi Isolated from Rhizosphere of Zoysiagrass in Korea (잔디 근권에서 분리한 식물생장촉진 균류)

  • Fifteen PGPF (plant growth promoting fungi) isolates were selected from 728 fungal isolates collected from rhizosphere of zoysiagrass in Korea. Identification of the 15 isolates was based on their morphological characteristics. They were classified as Gliocladium sp. (n=1), Penicillium sp. (n=5), Trichoderma sp. (n=3), Fusarium sp. (n=3), and unidentifed species (n=3). Of the 15 isolates, six (PF-31, PF-136, PF-238, PF-255, PF-395, PF-420) significantly promoted the growth of tomato seedlings, and three (PF-31, PF-101, PF-255) also promoted the growth of hot pepper and two (PF-31, PF-225) also promoted the growth of cucumber, The 15 PGPF isolates were divided into 4 groups based on root colonizing ability. Isolates PF-17, PF-101 and PF-225 were included in the group 1, which had high root colonizing ability.

New Species of Termitomyces (Lyophyllaceae, Basidiomycota) from Sabah (Northern Borneo), Malaysia

  • The genus Termitomyces (Lyophyllaceae, Basidiomycota) is often associated with fungus-feeding termites (Macrotermitinae) due to their strong symbiotic relationships. The genus is widely found exclusively in certain regions of Africa and Asia. They are recognized as edible mushroom within Southeast Asia as well. But it is often misidentified based on morphology by the local communities especially in Malaysia for Chlorophyllum molybdites which is a highly poisonous mushroom. Thus, it is necessary to study the genus for Malaysia with the synergy of using both morphological and molecular identification. In this study, we aim to describe another new species as an addition to the genus Termitomyces found within Sabah, Malaysia. We generated two new sequences (nrLSU and mtSSU) for the new species and a total of 28 nrLSU and mtSSU sequences were retrieved from GenBank for the phylogenetic analysis using maximum likelihood and Bayesian inferences. We identified that the new collection from Sabah province is a new species and named as Termitomyces gilvus based on the termites found in the mound. A phylogeny tree made from the concatenated genes of LSU and mtSSU suggests that T. gilvus is closely related to T. bulborhizus from China. According to our results, the combination of molecular and morphology proved to be a robust approach to re-evaluate the taxonomic status of Termitomyces species in Malaysia. Additional surveys are needed to verify the species diversity and clarify their geographic distribution.
