Morphological and Molecular Diagnosis of Necator americanus and Ancylostoma ceylanicum Recovered from Villagers in Northern Cambodia

  • Human hookworm infections caused by adult Ancylostoma spp. and Necator americanus are one of the most important tropical diseases. We performed a survey of intestinal helminths using the Kato-Katz fecal examination technique targeting 1,156 villagers residing in 2 northern provinces (Preah Vihear and Stung Treng) of Cambodia in 2018. The results revealed a high overall egg positive rate of intestinal helminths (61.9%), and the egg positive rate of hookworms was 11.6%. Nine of the hookworm egg positive cases in Preah Vihear Province were treated with 5-10 mg/kg pyrantel pamoate followed by purging with magnesium salts, and a total of 65 adult hookworms were expelled in diarrheic stools. The adult hookworms were analyzed morphologically and molecularly to confirm the species. The morphologies of the buccal cavity and dorsal rays on the costa were observed with a light microscope, and the nucleotide sequences of mitochondrial cytochrome c oxidase subunit 1 (cox1) gene were analyzed. The majority of the hookworm adults (90.7%) were N. americanus, whereas the remaining 9.3% were Ancylostoma ceylanicum, a rare hookworm species infecting humans. The results revealed a high prevalence of hookworm infections among people in a northern part of Cambodia, suggesting the necessity of a sustained survey combined with control measures against hookworm infections.

한국산 망둑어과 (Gobiidae) 미끈망둑 (Luciogobius guttatus)의 재검토 및 첫기록종, L. martellii (A re-examination of Gobiid Fish, Luciogobius guttatus, with First Record of L. martellii in Korea)

  • 우리나라 전 해안을 대상으로 미끈망둑 (Luciogobius guttatus)에 대한 분류학적 재검토를 실시하였다. 그 결과 41개체 (31.3~54.4 mm SL)가 L. martellii로 분류되었다. 본 종의 형태학적 특징은 다음과 같다: 복추골 수 16개; 총 척추골 수 35~37개; 등지느러미 기저부의 길이가 미병부 상부의 길이보다 길거나 같음; 가슴지느러미 상부에 1개의 유리연조; 꼬리지느러미 끝이 하얀색인 특징이 있다. 그중에서 L. guttatus와 형태학적으로 가장 뚜렷한 차이점인 등지느러미 기저부 길이와 미병부 상부의 길이의 차이는 L. martellii의 총모식표본과도 잘 일치하였다. 본 종의 한국명으로 "검정미끈망둑"을 제안한다.

비-류마티스성 거대 좌심방의 심장 초음파 검사 증례 보고 (A Case Report of Echocardiography of Non-rheumaic Giant Left Atrium)

  • 본 증례에서는 비-류마티스성에 의한 GLA가 관찰되어 이를 초음파검사한 사례를 보고하고자 하였다. 증례 환자는 60세 남성으로 호흡곤란과 심계항진으로 응급실을 내원하였다. 영상의학적 검사는 흉부 엑스선 검사와 CT검사를 진행하였고 거대 좌심방을 확인하였다. 거대 좌심방의 원인을 찾기 위해 심장초음파 검사를 시행하였다. 심장 초음파 검사 좌심방의 크기와 볼륨을 simpson's method로 평가하였으며 일반적인 거대 좌심방보다 LVEDVI가 6배 큰 거대 좌심방을 평가 할 수 있었다. 또한 류마티스성 심질환이 없는 환자에게 발생한 거대 좌심방은 매우 드문 증례로 평가된다. 비-류마티스성 거대 좌심방은 기능적인 이첨판 역류증, 이완기 기능장애 등에 의해 발생할 수 있으므로 형태학 및 혈류역학적 검사가 모두 가능한 초음파검사가 유용한 사례임을 확인하였다.

Identification of Botrytis cinerea, the Cause of Post-Harvest Gray Mold on Broccoli in Korea

  • In this study, we identified the causative agent of post-harvest gray mold on broccoli that was stored on a farmers' cooperative in Pyeongchang, Gangwon Province, South Korea, in September 2016. The incidence of gray mold on broccoli was 10-30% after 3-5 weeks of storage at $3^{\circ}C$. Symptoms included brownish curd and gray-to-dark mycelia with abundant conidia on the infected broccoli curds. The fungus was isolated from infected fruit and cultured on potato dextrose agar. To identify the fungus, we examined the morphological characteristics and sequenced the rDNA of the fungus and confirmed its pathogenicity according to Koch's postulates. The results of the morphological examination, pathogenicity test, and sequencing of the 5.8S rDNA of the internal transcribed spacer regions (ITS1 and ITS4) and three nuclear protein-coding genes, G3PDH, HSP60, and RPB2, revealed that the causal agent of the post-harvest gray mold on broccoli was Botrytis cinerea. To our knowledge, this is the first report of post-harvest gray mold on broccoli in Korea.

영남지역산(嶺南地域産) 붕어 Carassius auratus(Linnaeus)의 형태적(形態的) 변이(變異) (Morphological Variation of the Crusian Carp, Carassius auratus (Linnaeus) from Yongnam Area in Korea)

  • 영남지역(嶺南地域)의 여러 수계에서 채집한 C. auratus를 재료로 하여 수계에 따른 형태변이를 조사하였다. 계측 및 계수형질의 변이 범위는 수계 간에 서로 비슷하였으나 평균치에는 많은 차이가 있었으며 같은 수계 내에서도 서식 환경에 따라 각 형질의 변이가 크게 나타났다. 계수형질 가운데 배기극의 수(數)는 지금까지 3개로 기재하고 있으나 본 조사에서는 이것과 다르게 4-6개로 밝혀졌고 그 중 4개가 대부분이었다. 따라서 전체적으로 변이가 심하지만 일본산(日本産)의 C. auratus langsdorfii와 유사하였다. 또한 지금까지 알려진 것과는 다르게 새파가 길고 새파수에 있어서도 과거에 기재된 것 보다 더 많은 66-83개인 개체들이 나타나 일본산(日本産)인 C. auratus glandoculis와 유사하였다. 따라서 이들 개체의 분류에 대해서는 추후 세밀한 조사가 요망된다.

Comparison of Several Procedures for the Preparation of Synaptosomal Plasma Membrane Vesicles

  • Synaptosomal plasma membrane vesicles (SPMV) were isolated from fresh bovine cerebral cortex by several well-known procedures, and the best procedures was selected by enzymatic and morphological standards. The SPMV isolated by the modified procedure of Smith and Loh showed the highest purity. The specific activities of Na, K-ATPase, acetylcholinesterase and 5'-nucleotidase were about 5, 6-fold, 2, 5-fold and 3, 3-fold, respectively, enriched in the plasma membrane fraction as compared to those of the crude homogenate. Electron microscpic examination also showed that the membranes were in vesicular form.

Characterization of Coolia spp. (Gonyaucales, Dinophyceae) from Southern Tunisia: first record of Coolia malayensis in the Mediterranean Sea

  • This study provides the first report of the presence of Coolia malayensis in the Mediterranean Sea, co-occurring with C. monotis. Isolated strains from the Gulf of Gabès, Tunisia (South-eastern Mediterranean) were identified by morphological characterization and phylogenetic analysis. Examination by light and scanning electron microscopy revealed no significant morphological differences between the Tunisian isolates and other geographically distant strains of C. monotis and C. malayensis. Phylogenetic trees based on ITS1-5.8S-ITS2 and D1-D3/28S rDNA sequences showed that C. monotis strains clustered with others from the Mediterranean and Atlantic whereas the C. malayensis isolate branched with isolates from the Pacific and the Atlantic, therefore revealing no geographical trend among C. monotis and C. malayensis populations. Ultrastructural analyses by transmission electron microscopy revealed the presence of numerous vesicles containing spirally coiled fibers in both C. malayensis and C. monotis cells, which we speculate to be involved in mucus production.

Damping-off of Edible Aster Caused by Rhizoctonia solani AG-4

  • In August 2021, we surveyed diseases of wild vegetables grown in Taebaek, Gangwon Province, Korea. During the disease survey, we observed severe damping-off symptoms in young edible aster (Aster scaber) plants in a vinyl greenhouse investigated. The incidence of the disease in the plants ranged from 5% to 20%. Diseased plants of edible aster were collected from the vinyl greenhouse, and fungi were isolated from petiole lesions of the diseased plants. Rhizoctonia sp. was consistently isolated from the petiole lesions. We examined morphological characteristics and anastomosis groups of nine Rhizoctonia sp. isolates obtained from the petiole lesions. The examination results revealed that all the isolates corresponded to Rhizoctonia solani AG-4 based on the morphological characteristics and anastomosis test. Three isolates of R. solani AG-4 were tested for their pathogenicity on edible aster plants by artificial inoculation. Inoculation tests showed that the tested isolates caused damping-off symptoms on the inoculated plants. The induced symptoms were similar to those observed in the vinyl greenhouse investigated. Damping-off of edible aster caused by R. solani AG-4 is first reported in this study.

지주막하출혈에 의한 뇌기저동맥의 형태학적 변화에 미치는 양격산화탕(凉膈散火湯)의 효과 (The Effect of Yangkyuksanhoa-tang Extracts on the Morphological Changes of the Basilar Artery after Cerebral Subarachnoid Hemorrhage)

  • The present study was performed to investigate the effect of Yangkyuksanhoa-tang on the morphological changes of the basilar artery after experimentally induced subarachnoid hemonrrhages(SAH). Yangkyuksanhoa-tang has been used freguently for cerebrovascular accident Sprague Dawley rats weighing between 350-400 g were used. The 6 normal rats and 24 SAH elicited rats were used, The SAH induced by injection of the fresh autologus heart blood (0.3-0.4 ml) into the cisterna magna through the posterior atlanta-occipital membrane, Sample group was given 3.3 ml/kg/day of Yangkyuksanhoa-tang extracts for 2 days after SAH. The experimental animals were killed at 48hrs after SAH. The morphological changes of the arterial walls were examined by light and electron microscopy. Following are the obtained results: 1. In SAH elicited rats, the size of the lumen in basilar artery was diminished by about 45% and the thickness of arterial wall was increased by about 82%. In SAH elicited rats with Yangkyuksanhoa-tang treatment, the size of the lumen in basilar artery was merely diminished by about 18% and the thickness of arterial wall was merely increased by about 19%. 2. In light microscopic examination, the endothelium was swollen into a cuboid shape and the layer of smooth muscle was increased in the basilar artery of SAH elicited rats. In SAH elicited rats with Yangkyuksanhoa-tang treatment, the size of the lumen in basilar artery was enlarged and the thickness was decreased than in SAH elicited rats. The endothelium was flattened into a squamous shape and the layer of smooth muscle was decreased more than in SAH elicited rats. 3. In electron microscopic examination, the endothelial cells with fragmentation nuclei were changed into a cuboid shape and the internal elastic lamina were folded at the basilar artery of SAH elicited rat. The nuclei of smooth muscle cells were changed into a round or crumpled shape. The length of smooth muscle was shorten and thickness was increased. But all kinds of morphologic changes were diminished in SAH elicited rats with Yangkyuksanhoa-tang treatment. Conclusion : Yangkyuksanhoa-tang extracts were effective to treat cerebral vasospasm after experimentally induced subarachnoid hemorrhage in rats.

랫드에서 cyclophosphamide에 의해 유발된 흉선세포의 apoptosis (Thymocyte Apoptosis Induced by Cyclophosphamide in Rats)

  • Cyclophosphamide(25, 50 or 100 mg/kg), orally administered to male Sprague-Dawley rats, caused a time- and dose-dependent thymic atrophy. In the light microscopic examination of the atrophic thymus, thymocytes with condensed or fragmented nucleus were multifocally observed in the cortical region, started to increase 8 hr after CPA treatment and reached to the maximal level at 16 hr, although such cells were not seen after 48 hr when the severe depletion of thymocytes were marked. In agarose gel electrophoresis to analyze the DNA changes, DNA extracted from atrophic thymus showed a oligonucleosomal laddering at the corresponding time to morphological changes. In an additional supportive experiment, thymocytes showing morphological changes, nuclear condensation or apoptotic body, exhibited a positive reaction to immunoperoxidase staining using in situ apoptosis detection kit. Separately, agarose gel electrophoresis of DNA from bone .marrow cells was performed to investigate the involvement of bone marrow cells in the process of thymocyte apoptosis. Although DNA laddering was slightly increased 2 and 4 hr after treatment, no clear correlation was inferred. Taken togather, it is concluded that thymocytes showing morphological changes in thymic atrophy induced by cyclophosphamide administration represent an apoptosis having biochemical nature of programmed cell death.
