경화된 상아질의 접합 양상에 관한 주사전자현미경적 연구 (A SCANNING ELECTRON MICROSCOPIC STUDY OF BONDING ASPECTS TO THE SCLEROTIC DENTIN)

  • The changes of microstructures, morphology of sclerotic dentin and bonding aspects generated by an adhesive resin was investigated. Incisors and premolars showing natural cervical abrasions were collected and conditioned with 10 % phosphoric acid or 10 % maleic acid. The sclerotic dentin specimens were then rinsed and blot-dried and applied with dentin adhesive (All Bond 2) to the conditioned dentin surface. To examine the morphologic change of the sclerotic dentin specimen after etching and bonding procedure, the treated specimens were examined by SEM. To analyze the chemical composition of sclerotic dentin and crystals occluding dentinal tubules, the sclerotic dentin specimen was powdered and examined with X-ray Diffractometer. To investigate the Ca/P weight percent ratio within the dentinal tubules, the sclerotic dentin specimen was fractured perpendicularly to the long axis of the tooth from the center of cervical abrasion lesion and then examined with EDX(Energy Dispersive X-ray) microanalyzer. The results were as follows : 1. The increased width of peritubular dentin and the depositions of the irregular amorphous materials within the dentinal tubules were showed in the sclerotic dentin specimens. 2. After the treatment of sclerotic dentin specimen with 10 % phosphoric acid or 10 % maleic acid, the lateral side of tubules rather than cross-sectional tubule openings was showed exclusively at the incisal and gingival incline of the specimens. 3. After the treatment of sclerotic dentin specimen with 10 % phosphoric acid or 10 % maleic acid, the hybrid layer was not formed evidently and the resin tag was not formed or shortly penetrated into the tubules with the thinner diameter. 4. According to the results of XRD analysis of the sclerotic dentin specimen, Hydroxyapatite and Octacalcium phosphate were predominent, however, Whitlockite crystals were rare. 5. The mean Ca/P weight percent ratio analysed from 5 fractured sclerotic dentin specimens was $2.322{\pm}0.170$ at the intertubular dentin, $1.826{\pm}0.051$ within the dentinal tubule.

큰비쑥(Artemisia fukudo) 추출물의 암세포 증식 억제효과 (The Cytotoxicity of Artemisia fukudo Extracts Against HL-60 Cells)

  • 본 연구는 큰비쑥를 80% EtOH로 추출하고 극성에 따른 용매분획을 실시하여 혈핵암 세포주를 대상으로 분획별에 따른 암세포 증식억제(cytotoxicity) 효과와 이러한 혈액암 세포주에 대한 세포독성 효과가 세포자연사(apoptosis)에 의해 일어나는 것인지, 몇 가지 apoptosis 유도 실험을 통해 확인해 보았다. HL-60 세포에 대한 증식억제 효과는 전반적으로 모든 처리구에서 농도의존적인 세포증식 억제효과가 나타났으며, 특히 헥산과 디클로로메탄 분획물에서 가장 강한 세포성장 억제효과가 나타났다. 이러한 세포성장 억제효과가 apoptosis 유도에 의한 것인지 알아보기 위해 apoptosis 유도에 의한 세포사멸의 특징인 DNA 단편화 현상을 비롯하여 세포형태학적 변화 및 유동세포분석기를 통한 DNA content를 측정한 결과, 큰비쑥 추출물에 의한 암세포 증식억제효과는 apoptosis를 유도하여 세포자연사를 통해 세포의 성장을 억제하는 것으로 확인되었다.

불소 테이프의 법랑질 탈회 억제 효과에 관한 실험적 평가 (IN VITRO EVALUATION OF EXPERIMENTAL FLUORIDE TAPE IN INHIBITION OF ENAMEL DEMINERALIZATION)

  • 본 연구의 목적은 법랑질 표면 미세경도의 측정과 주사전자현미경 관찰을 통해 본 교실에서 개발한 2.26%의 불소를 함유한 폴리비닐알코올 테잎 (F-PVA tape)의 법랑질 탈회 억제 효과를 평가하기 위한 것이다. 60개의 법랑질 표본을 각 군당 15개 씩, 임의로 대조군, F-PVA tape 군, 불소바니쉬 군, CPP-ACFP 군으로 나누고, 각각의 재광화 제재를 도포한 후 pH-cycling 과정 거쳤다. 표본의 표면 미세경도를 측정하고 표면 미세경도 소실량을 계산하였다. 주사전자현미경 관찰을 위해, 각 군에서 5개의 표본을 추출하고 형태학적 특징을 분석하였고 다음과 같은 결론을 얻었다. 1. pH-cycling 후, F-PVA tape 군과 불소바니쉬 군의 법랑질 표면 미세경도 값은 CPP-ACFP 군과 비교 시 유의하게 높았으며, F-PVA tape 군과 불소바니쉬 군 사이에는 유의한 차이가 없었다. 2. 주사전자현미경 관찰 시, F-PVA tape 군과 불소바니쉬 군에서는 법랑질 표면에 형성된 수많은 구형 및 난원형의 결정 입자들이 관찰되었으며, 결정 입자의 밀도는 대조군 및 CPP-ACFP 군에서 보다 높았다. F-PVA tape은 법랑질의 탈회를 억제하는데 효과적이며, 그 효과는 불소바니쉬와 견줄만하고, CPP-ACFP 보다 우수하다.

어미 크기 및 복부팽만도에 따른 능성어, Epinephelus septemfasciatus의 배란유도 효과 (Ovulation Induction Effect of Sevenband Grouper, Epinephelus septemfasciatus in Size and Abdomen Inflation Degree of Mother Fish)

  • 능성어, Epinephelus septemfasciatus의 우량 수정란을 생산할 수 있는 어미 선별기준을 설정하기 위하여 해상가두리에서 6년간 키운 시험어의 현태적 특징을 관찰하고 호르몬 처리하여 난질을 관찰 분석하였다. 시험어는 4개의 체중 범위(3.0~4.0, 4.0~5.0, 5.0~6.0, 6.0~7.0 kg)와 뒷지느러미를 기준으로 복부팽만한 정도를 I~IV 단계로 나눈 후 산란빈도, 채란량, 부상률, 수정률, 난발생 생존율 및 부화율을 조사하고, 수정란과 유구의 크기를 측정하였다. 또한 생식소 발달을 확인하기 위하여 복부팽만도 단계별 생식소 중량지수(GSI)를 구하고, 조직학적으로 관찰하였다. 그 결과 어체중이 5.0 kg 이상일 때 모든 개체에서 배란이 유도되었다. 또한 복부팽만도 I, II 단계에서는 배란이 유도되지 않고, III, IV 단계에서만 배란이 유도되었다. 난질은 III 단계가 우수하였고 그 때의 GSI는 $4.68{\pm}0.40%$였다.

Brca1 결손 세포주에서 nocodazole 처리에 의한 spindle checkpoint 활성화 연구 (Impaired Spindle Checkpoint Response of Brca1-deficient Mouse Embryonic Fibroblasts (MEFs) to Nocodazole Treatment)

  • 항암유전자 Brca1의 변이는 유방암 및 난소암에 대한 감수성을 증가시키며, Brca1은 DNA손상신호후 세포주기 조절에 필수적인 역할을 한다. 연구결과, Brca1이 세포주기 S기와 G2/M 조절점에서 중요한 역할을 담당함이 밝혀졌다. 그러나, Brca1의 spindle checkpoint 관여여부는 알려져 있지 않다. 본 연구에서는 spindle checkpoint를 활성화시키는 nocodazole를 처리하여 야생형, $p53^{-/-}$ 그리고 $p53^{-/-}\;Brca1^{-/-}$ 세포주의 세포주기 변화를 조사하였다. 야생형과 $p53^{-/-}$ 세포주는 신속한 mitosis기 정지가 나타난 반면, $p53^{-/-}\;Brca1^{-/-}$ 세포주의 경우 모든 세포가 M기에서 정지하지 않았다. Double-thymidine block 기법에 의한 세포주기 동조화후 nocodazole 처리시에도 $p53^{-/-}\;Brca1^{-/-}$ 세포주에서는 일부세포가 M기 조절점을 통과하여 계속 G1기로 진행하였다. 형태학적 분석에서도 nocodazole 함유배지에서 계속 증식하는 세포형태가 관찰되었다. 이와 같은 결과들은 Brca1이 spindle checkpoint가 정상적으로 작동하는데 중요한 역할을 담당한다는 것을 의미하고 있다.

칼슘수용액으로 처리한 상아질과 합착용 글래스아이오노머의 전단결합강도에 관한 연구 (A STUDY ON THE SHEAR BOND STRENGTH OF LUTING GLASS IONOMER AND DENTIN TREATED WITH CALCIUM SOLUTION)

  • The objective of this paper was to evaluate the shear bond strength of luting glass ionomer cement with defferent calcium based solution treatment on dentin surface. 120 extracted human teeth were classified into 12 group based on presence of smear layer on dentin surface and type of treatment solution. Smear layer remove on dentin surface was done using 6% citric acid for 60 seconds. Five different dentin surface treatment solutions(calcium acetate, calcium carbonate, clacium chlorided, calcium hydroxide, and calcium phosphate) were evaluated in this study. After surface modification, metal ring(inner diameter : 3mm, depth : 1mm) was placed to expose the same dentin surface area and inner space was filled with luting glass ionomer cement according to the recommended procedure for stadard clinical procedure. The shear bond strength of glass ionomer cement was determined after 24 hours. SEM was used for the evaluation of the surface morphologic changes and EDAX analysis was done for determination of the change of the calcium contents of treated dentin. Follwing conclusion can be drawn : 1. In the group of the dentin surface with smear layer, the calcium carbonate solution was the most effective for the increase of the clacium content and the shear bond strength of glass ionomer cement to dentin surfaces. 2. In the group of the calcium carbonate treated dentin with msear layer, the shear bond strength was increased twice compared to the control group and cohesive failure mode was observed. 3. The shear bond strength of cement was increased significantly be the removal of smear layer using 6% citric aicd. However, additional calcium solution treatments were not effective for further bond strength increase. 4. The shear bond strength of cement was significantly improved by both of the removal of smear layer and the calcium solution treatment, and the former was more effective for bond strength improvement. 5. The smear layer removed/calcium solution treated groups showed dentinal tubule obstruction and crystal attachment in SEM evaluation. However, the shear bond strengths of these groups were not increased compared to the smear layer removed/no dentin treatment group.

산 표면처리 후 상아질 표면의 습윤이 하이브리드층 형성에 관한 전자현미경적 연구 (ELECTRON MICROSCOPIC STUDY OF RESIN-DENTIN HYBRID ZONE PRODUCED BY THE MOISTENING OF ACID CONDITIONED DENTIN SURFACE)

  • The effect of moistening and air-drying of acid-conditioned dentin before priming on the formation of resin-dentin hybrid zone was investigated, Freshly extracted human molars were used and divided at random into 5 groups, Groups 1 - 3 consisted of specimens conditioned with 10 % phosphoric acid for 20 seconds; Group 1 served as a control in which the conditioned dentin was simply blot-dried with a damp facial tissue; Group 2 was air dried for 30 seconds ; Group 3 was air dried for 30 seconds and immediately remoistened for 10 seconds with air-water syringe. and then the specimen was blot-dried with a damp facial tissue. Groups 4-5 were not acid conditioned ; In group 4, the smear layer on the dentin was blot dried before primer placement; Group 5 was air dried only for 30 seconds, The acetone-based primer and bonding agent of All Bond 2 (Bisco. Inc., USA) and composite resin (Z-100, 3M Dental products, USA) were applied for acid conditioned dentin and non-conditioned dentin. The morphologic ultrastructure of resin-dentin hybrid zone was examined by the use of SEM and TEM. and the existence of inorganic material and analysis of Ca/P weight-percent ratio in the resin-dentin hybrid zone were revealed by the EDAX, The results were as follows : 1. In the moistened specimens from acid-conditioned groups, the resin penetrated about 3-$4{\mu}m$ into dentin and the denatured collagen smear layer was not present at the surface. The resin tag was formed to a thickeness of 3-$4{\mu}m$ at the upper part of dentinal tubule and compactively connected to each other by means of many lateral branching. 2. In the air-dried specimens from acid-conditioned groups, the resin penetrated about 2.0-$2.5\;{\mu}m$ into dentin and an upper thin black layer to a thickness of 30-35nm was identified between adhesive resin and demineralized collagen layer. The resin tag to have a diameter of $2.5{\mu}m$ was formed at the upper part of dentinal tubule. However the funnel shape of the tag was not notable compared to the moistened specimens. 3. In the remoistened specimens from acid conditioned groups, the resin penetrated about 2.0-$2.5{\mu}m$ into dentin and an upper black layer was not present. The resin tag at the upper part of dentinal tubule was formed less than $2{\mu}m$ and was weakly connected to each other by means of few lateral branching. 4. In the non-conditioned groups, the smear layer was formed to a thickness of $0.5{\mu}m$ at dentin surface. However, the resin-dentin hybrid zone was not identified by TEM. The evidence of resin penetration into intertubular and intratubular dentin did not show. 5. All the acid-conditioned groups showed that the detected calcium and phosphorus weight percent ratios at the $2{\mu}m$ upper portion from the resin-dentin interface into the resin were much higher than that at the $2{\mu}m$ lower portion from the resin-dentin interface to dentin. Morphologic Alterations in Amygdala Subregions of Adult Patients with Bipolar Disorder

  • Objectives Previous studies have revealed inconsistent results on amygdala volume in adult bipolar disorder (BD) patients compared to healthy controls (HC). Since the amygdala encompasses multiple subregions, the subtle volume changes in each amygdala nucleus might have not been fully reflected in the measure of the total amygdala volume, causing discrepant results. Thus, we aimed to investigate volume changes in each amygdala subregion and their association with subtypes of BD, lithium use and clinical status of BD. Methods Fifty-five BD patients and 55 HC underwent T1-weighted structural magnetic resonance imaging. We analyzed volumes of the whole amygdala and each amygdala subregion, including the anterior amygdaloid area, cortico-amygdaloid transition area, basal, lateral, accessory basal, central, cortical, medial and paralaminar nuclei using the atlas in the FreeSurfer. The volume difference was analyzed using a one-way analysis of covariance with individual volumes as dependent variables, and age, sex, and total intracranial volume as covariates. Results The volumes of whole right amygdala and subregions including basal nucleus, accessory basal nucleus, anterior amygdaloid area, and cortico-amygdaloid transition area in the right amygdala of BD patients were significantly smaller for the HC group. No significant volume difference between bipolar I disorder and bipolar II disorder was found after the Bonferroni correction. The trend of larger volume in medial nucleus with lithium treatment was not significant after the Bonferroni correction. No significant correlation between illness duration and amygdala volume, and insignificant negative correlation were found between right central nucleus volume and depression severity. Conclusions Significant volume decrements of the whole amygdala, basal nucleus, accessory basal nucleus, anterior amygdaloid area, and cortico-amygdaloid transition area were found in the right hemisphere in adult BD patients, compared to HC group. We postulate that such volume changes are associated with altered functional activity and connectivity of amygdala nuclei in BD.

The role of the iliotibial band cross-sectional area as a morphological parameter of the iliotibial band friction syndrome: a retrospective pilot study

  • Background: Iliotibial band friction syndrome (ITBFS) is a common disorder of the lateral knee. Previous research has reported that the iliotibial band (ITB) thickness (ITBT) is correlated with ITBFS, and ITBT has been considered to be a key morphologic parameter of ITBFS. However, the thickness is different from inflammatory hypertrophy. Thus, we made the ITB cross-sectional area (ITBCSA) a new morphological parameter to assess ITBFS. Methods: Forty-three patients with ITBFS group and from 43 normal group who underwent T1W magnetic resonance imaging were enrolled. The ITBCSA was measured as the cross-sectional area of the ITB that was most hypertrophied in the magnetic resonance axial images. The ITBT was measured as the thickest site of ITB. Results: The mean ITBCSA was 25.24 ± 6.59 ㎟ in the normal group and 38.75 ± 9.11 ㎟ in the ITBFS group. The mean ITBT was 1.94 ± 0.41 mm in the normal group and 2.62 ± 0.46 mm in the ITBFS group. Patients in ITBFS group had significantly higher ITBCSA (P < 0.001) and ITBT (P < 0.001) than the normal group. A receiver operator characteristic curve analysis demonstrated that the best cut-off value of the ITBT was 2.29 mm, with 76.7% sensitivity, 79.1% specificity, and area under the curve (AUC) 0.88. The optimal cut-off score of the ITBCSA was 30.66 ㎟, with 79.1% sensitivity, 79.1% specificity, and AUC 0.87. Conclusions: ITBCSA is a new and sensitive morphological parameter for diagnosing ITBFS, and may even be more accurate than ITBT.

Reduced Volume of a Brainstem Substructure in Adolescents with Problematic Smartphone Use

  • Objectives: Despite the growing concern regarding the adverse effects related to problematic smartphone use (PSU), little is known about underlying morphologic changes in the brain. The brainstem is a deep brain structure that consists of several important nuclei associated with emotions, sensations, and motor functions. In this study, we sought to examine the difference in the volume of brainstem substructures among adolescents with and without PSU. Methods: A total of 87 Korean adolescents participated in this study. The PSU group (n=20, age=16.2±1.1, female:male=12:8) was designated if participants reported a total Smartphone Addiction Proneness Scale (SAPS) score of ≥42, whereas the remaining participants were assigned to the control group (n=67, age=15.3±1.7, female:male=19:48). High-resolution T1 magnetic resonance imaging was performed, and the volume of each of the four brainstem substructures [midbrain, pons, medulla, and superior cerebellar peduncle (SCP)] was measured. Analysis of covariance was conducted to reveal group differences after adjusting for effects of age, gender, whole brainstem volume, depressive symptoms, and impulsivity. Results: The PSU group showed a significantly smaller volume of the SCP than the control group (F=8.273, p=0.005). The volume of the SCP and the SAPS score were negatively correlated (Pearson's r=-0.218, p=0.047). Conclusion: The present study is the first to reveal an altered volume of the brainstem substructure among adolescents with PSU. This finding suggests that the altered white matter structure in the brainstem could be one of the neurobiological mechanisms underlying behavioral changes in PSU.
