Protoplast Isolation and Differentiation of Marine Green Alga Monostroma nitidum (해산 녹조류 참홑파래, Monostroma nitidum의 원형질체 분리와 분화)

  • High yields of protoplasts were obtained following enzymatic digestion of the vagetative thalli of marine green alga Monostroma nitidum. The enzyme mixtures containing $4\%$ Cellulase R-10+$3\%$ Macerozyme R-10+$3\%$ Abalone acetone power produced $4.41\times10^6$ protoplasts per 300 mg of fresh tissue. The highest yield of protoplasts was obtained by 270 minutes treatment of the thalli in enzyme solution. Freshly isolated protoplasts were spherical in shape and ranged between $13\~33\mu$m in diameter. The high efficiency of differentiation were obtained by incubating freshly isolated protoplasts in 0.4 M mannitol f/2 medium for 7 days and then transferring to 0.2 M mannitol f/2 medium. Protoplasts began to form new cell walls three days after initial culture and began to germinate after 10 days, and then form a leafy thallus after further culture in f/2 medium. The addition of antibiotics in media inhibited the differentiation of protoplasts in culture.

  • A micro algal growth-enhancing polysaccharide compound was isolated from the green alga Monostroma nitidum using water extraction, molecular fractionation, a DEAE-cellulose column, and fast protein liquid chromatography using a Superose-12 column. The yield of the compound from the seaweed powder was 8.3$\times$l0$^{-3}$%. At 2 mg/mL concentration, the polysaccharide enhanced Tetraselmis suecica cell growth in f/2 medium by approximately 160%.

Immunomodulating Effects on Macrophage of Rhamnan Sulfate Extracted from Monostroma nitidum (홀파래에서 추출한 Rhamnan sulfate의 Macrophage에 의한 면역조절효과에 관한 연구)

  • The rhamnan sulfate extracted from green algae seaweed, Monostroma nitidum was characterized as activity in vitro culture assay with macrophages from mice. Rhamnan sulfate indicated that F-4-3 fraction enhanced glucose consumption, as well as the production of nitrogen dioxide and tumor necrosis factor(TNF). F-4-3 fraction was also augmented IL-1 secretion from those macrophages. Effects of the pretreatment of peritoneal macrophages with rhamnan sulfate F-4-3 fraction and several polysaccharides as relative standard on the production of H2O2 induced with unopsonized zymosan A were examined. Pretreatment with polysaccharides inhibited the zymosan A mediated H2O2 production by macrophages. The phorbol myristate acetate (PMA) mediated H2O2 production was not affected by the pretreament. These result suggested that pretreatment of rhamnan sulfate interfered with the interaction of macrophages zymosan A. Rhamnan sulfate inhibited zymosan A mediated production of H**O** by macrophages and F-4-3 Fraction was also activator of macrophages.

  • The anti-compliment activity of hemolytic complementary assay(TCH50) of rhamnan sulfate fraction obtained from water extracts of Monostroma nitidum was investigated Rhamnan sulfate Fraction, F-4-3 fraction appeared relatively strong anti-complementary activity which decreased TCH50 over 60% than that comparison with control, and F-4-3 considerably inhibited ACH50. F-4-3 inhibited formation of the classical pathway C3 convertase or C4 cleavage. The results also indicate the mode of complement activation by F-4-3 fraction shows not only the classical pathway but also the alternative pathway.

Effect of the Seaweed (Monostroma nitidum) Extract on Triton WR-1339 Induced Hyperlipidemia in Mouse (Triton WR-1339 복강주사로 유도된 생쥐의 고지혈증에 대한 참홑파래 추출물의 영향)

  • An antihyperlipidemic effect was observed by intraperitoneal injection of the seaweed Monostroma nitidum extract. Hyperlipidemia was induced by intraperitoneal injection of Triton WR-1339 $(60\;{\mu}g/g-body\;weight)$ into a mouse. Maximum level of blood cholesterol was reached at 20 hours after Triton injection. By simultaneous injection of the M. nitidum extract $(30\;{\mu}g/g-body\;weight)$ with the Triton into each right and left sides of the peritoneal cavity, levels of total cholesterol and low density lipoprotein were decreased to $24\%\;and\;14\%$ compared to Triton injection only. For histochemical changes, hepatic tissues obtained at 40 hours after injection of the Triton and the M. nitidum extract were fixed in fromol-calcium solution. The meshlike cytoplasms disappeared and hapatic plates were recovered in mice injected with the M. nitidum extract. Numbers of lipid drops and cholesterol particles also decreased in the portal space of the hepatic cytoplasm. This indicates that the accumulation of lipid, including cholesterol, caused by Triton was prevented by the antihyperlipidemic effect of extract from the seaweed M. nitidum.

  • Cell growth of the marine microalga Tetraselmis suecica was regulated by the addition of seaweed extracts in its culture medium. Of 26 species of seaweed tested, water-soluble extracts from Monostroma nitidum and Pachymeniopsis elliptica and methanol-soluble extract from Sargassum confilsum enhanced the growth of T suecica cells. The water extract of M. nitidum was the most effective, producing up to a 2-fold increase in cell density with the addition of 1 mg/mL of extract to the culture medium. Cell size, gross biochemical composition, fatty acids, and digestion efficiency all differed marginally between cultures of T suecica grown with and without the M. nitidum extract.

Ecology and Early Growth of Monostroma nitidum (Ulvales, Chlorophyta) in Korea (녹조류 참홑파래 (Monostromu nitidum)의 생태와 초기생장)

  • This study investigated the ecology and growth of Monostroma nitidum Wittrock in both its natural habitat and the laboratory. The maximum length, width, and weight of M. nitidum in March were 9.0$\pm$4.7 cm, 9.6$\pm$3.6 cm, and 1.52$\pm$1.13 g, respectively. Yellowish-green or yellowish-brown reproductive thalli began to appear in January, and over 80% of the thalli matured by March. The male and female spores were ca. 6 $\mu$m long, and elongate and ovoid in shape. The spores had two flagella and one-eye spot, and tended to swim toward light. Maximum number of spores released from matured thalli was 236 cells/mL after 70 minutes at a light intensity of 100 $\mu$mol/m$^2$/s. The zygote diameter ranged from 3.4-6.0 $\mu$m (mean 4.2 $$m) and increased to 69.8 $\mu$m 14 weeks after culture. The mass release of zoospores was observed from thalli in the dark (3 to 12 days), after 30 min under dry conditions in the shade, at 25$^{\circ}C$, and a light intensity of 100 $\mu$mol/m$^2$/s. The maximum number of zoospores released was 109.8 cells/mL after 60 min of induction. M. nitidum fronds on the net increased to 6.8-7.2 cm in length, and 6.6-8.9 cm in width during the winter.

Characterization of Rhamnan Sulfate Purified from Monostroma nitidum (홑파래에서 분리정제한 Rhamnan Sulfate의 특성)

  • The rhamnan sulfat extracted from green algae seaweed, Monostroma nitidum was characterized on sugars, sulfate compositions and molecular structure. Rhamnan sulfate was extracted with boiling water, and purified with two steps of cetylpyridinium chloride and ion exchange chromatography. The yield of crude rhamnan sulfate was about 2% from raw material. Rhamnan sulfate fraction, F-4 was composed of 30% rhamnose, 0.9% arabinose, 2.5% xylose, 2% glucose and 32.6% sulfate. Rhamnan sulfate F-4-3 obtained from F-4 fraction was composed of 36.8% rhamnose, 3.6% xylose, 2.7% glucose, 1.4% galactose and 30.8% sulfate. The molecular weight of F-4-3 fraction was estimated as 10,000-10,300 dalton with Sephacryl S-200 gel filtration chromatography.
