Nondestructive Prediction of Fatty Acid Composition in Sesame Seeds by Near Infrared Reflectance Spectroscopy

  • Near infrared reflectance spectroscopy (NIRS) was used to develop a rapid and nondestructive method for the determination of fatty acid composition in sesame (Sesamum indicum L.) seed oil. A total of ninety-three samples of intact seeds were scanned in the reflectance mode of a scanning monochromator, and reference values for fatty acid composition were measured by gas-liquid chromatography. Calibration equations were developed using modified partial least square regression with internal cross validation (n=63). The equations obtained had low standard errors of cross-validation and moderate $R^2$ (coefficient of determination in calibration). Prediction of an external validation set (n=30) showed significant correlation between reference values and NIRS estimated values based on the SEP (standard error of prediction), $r^2$ (coefficient of determination in prediction) and the ratio of standard deviation (SD) of reference data to SEP. The models developed in this study had relatively higher values (more than 2.0) of SD/SEP(C) for oleic and linoleic acid, having good correlation between reference and NIRS estimate. The results indicated that NIRS, a nondestructive screening method could be used to rapidly determine fatty acid composition in sesame seeds in the breeding programs for high quality sesame oil.

  • In recent years, a miniature spectrometer has been extensively developed due to the marriage of fiber optics and semiconductor detector array. This type of miniature spectrometer has advantages of low price and robustness due to the capability of mass production and no moving parts are required such as lenses, mirrors and scanning monochromator. These systems are ideal for use in teaching labs, process monitoring and field analyses. A portable near infrared (NIR) system has been developed for qualitative and quantitative analysis. This system includes a tungsten halogen lamp for light source, a fiber optics connected a light source, and a sample module to the microspectrometer, The size of spectrometer can be as small as 2.5 cm x 1.5 cm x 0.1 cm. Wavelength ranges can be chosen as 360-800 nm, 800-1100 nm and 1100-1900 nm depending on the type of detector. The software consists of various tools for multivariate analysis and pattern recognition techniques. To evaluate the system, long and short-term stability, wavelength accuracy, and stray light have been investigated and compared with conventional scanning type NIR spectrometer. This developed system can be sufficiently used for quantitative and qualitative analysis for various samples such as agricultural product, herbal medicine, food, petroleum, and pharmaceuticals, etc.

Development of Spectral Irradiance Measurement System (분광복사조도 측정 장치 개발)

  • Spectral irradiance measurement system was developed to measure the spectral irradiance of optical sources in the wavelength range from 250[nm] to 1600[nm]. Our system is composed of source system, fore-optics, monochromator system, optical detector system, and automatic control system. Optical detector system with PMT, Si, InGaAs, and IR enhanced InGaAs detectors is used to measure the wide spectrum of optical sources in ultraviolet visible, and infrared wavelength regions. Spectral irradiance of the 1[kW] quartz-halogen tungsten lamp was measured and compared in the wavelength range from 250[nm] to 1600[nm]. The differences between our results and those reported by NIST are below 3[%], 3.5[%], and 5[%] in the wavelength range of 450∼700[nm], 700∼1600[nm], 250∼400[nm], respectively.

  • We investigated visible light radiated from Plasma focus device by time-resolved analyzed method and time-integrated analyzed method. Plasma focus consisted of two coaxial electrodes is a device that translated from electric energy of maximum 40 kV/20 kJ in capacitor banks into visible light by electric discharge. Spectral analysis is using Monochromator(f =0.5m). Time-resolved spectrum is analyzed with a oscilloscope the light pulse of constant wavelength and time-integrated spectrum does with densitometer the film which developed a constant range of wavelength. The optimum condition of visible emission was that the discharging voltage was 17kV and the gas pressure 0.5 torr Ar.

  • Geographical origin of the Korean, Chinese and Japanese sesame seeds were identified very high accuracy by NIR spectroscopy. The NIR instrument of filter type showed the same accuracy of the monochromator scanning type to identify the geographical origin of the sesame seeds. In case of adulteration between the Korean and Chinese sesame seeds, the ratio of addition could be determined about 10% error level. The reason of identification of geographical origin by NIR spectroscopy, it was supposed to the difference, of oil cake substance.

  • We present the characteristics of the 2K CCD camera at the Bohyunsan Optical Astronomy Observatory of the Korea Astronomy Observatory at the time of its development. The purpose of this paper is to support the observers who may need detailed information on the characteristics of the camera and to provide helpful information on the optimization' of a CCD camera for those who try to develop their own camera. The 2K CCD camera was optimized to have a gain of $1.8e^-/ADU$ and a read out noise of $7e^-$ from an experiment using radioactive $^{55}Fe$ X-ray source. The charge transfer efficiency was measured as 0.9999976 for serial and 0.9999942 for parallel direction, which means $0.5\%$ charge loss along the serial direction and $1.2\%$ along the parallel direction across the chip. The quantum efficiency of the camera was measured from an experiment using a homogeneous light source consisting of a halogen lamp and an integrating sphere with a monochromator. The resulting quantum efficiency of the camera peaked at the wavelength range 600-700 nm with the value of $\~0.89$.

Determination of Protein Content in Pea by Near Infrared Spectroscopy

  • Near infrared reflectance spectroscopy (NIRS) was used as a rapid and non-destructive method to determine the protein content in intact and ground seeds of pea (Pisum sativum L.) germplasms grown in Korea. A total of 115 samples were scanned in the reflectance mode of a scanning monochromator at intact seed and flour condition, and the reference values for the protein content was measured by auto-Kjeldahl system. In the developed ground and intact NIRS equations for analysis of protein, the most accurate equation were obtained at 2, 8, 6, 1 math treatment conditions with standard normal variate and detrend scatter correction method and entire spectrum (400-2,500 nm) by using modified partial least squares regression (n=78). External validation (n=34) of these NIRS equations showed significant correlation between reference values and NIRS estimated values based on the standard error of prediction (SEP), $R^2$, and the ratio of standard deviation of reference data to SEP. Therefore, these ground and intact NIRS equations can be applicable and reliable for determination of protein content in pea seeds, and non-destructive NIRS method could be used as a mass analysis technique for selection of high protein pea in breeding program and for quality control in food industry.

  • 고전압 펄스 플라즈마를 액체 속에서 발생시켜 수소 스펙트럼의 광학적 특성을 연구하였다. 고전압 펄스 발생 장치인 막스 제네레이터는 용량이 $0.5{\mu}F$인 축전기 5개로 이루어져 있다. 각각의 축전기는 전원 장치를 이용하여 저항을 통해 병렬로 충전되며, 방전 시에는 불꽃 방전 스위치에 의해 동시에 직렬로 연결되어 고전압을 발생시킨다. 따라서, 출력 전압과 전류는 40kV, 3 kA이며 총 에너지는 약 125 J이다. 직육면체 모양의 폴리카보네이트 용기 내부의 양쪽면에는 탐침 모양의 전극이 구성되어 있으며 전극 사이에서 고전압을 가진 플라즈마가 형성된다. 실험에서 액체로는 증류수를 사용하였다. 액체 방전 시 발생하는 수소 스펙트럼을 관측하기 위해 초점거리 30 cm의 monochromator를 이용하였고, 수소 알파선의 656.3 nm와 수소 베타선의 434.1 nm를 관측하였다. 전자 밀도의 측정법으로는 Stark broadening법을 이용하여 측정하였으며, 전자 온도는 Stark profile의 상대적인 전자 밀도의 비를 이용하여 계산하였다. 전자밀도는 실험조건에서 약 $3{\times}10^{15}cm^{-3}$, 전자온도는 약 2.5 eV가 측정되었다.

Real-time malfunction detection of plasma etching process using EPD signal traces (EPD 신호궤적을 이용한 플라즈마 식각공정의 실시간 이상검출)

  • This paper presents a novel method for real-time malfunction detection of plasma etching process using EPD signal traces. First, many reference EPD signal traces are collected using monochromator and data acquisition system in normal etching processes. Critical points are defined by applying differentiation and zero-crossing method to the collected reference signal traces. Critical parameters such as intensity, slope, time, peak, overshoot, etc., determined by critical points, and frame attributes transformed signal-to symbol of reference signal traces are saved. Also, UCL(Upper Control Limit) and LCL(Lower Control Limit) are obtained by mean and standard deviation of critical parameters. Then, test EPD signal traces are collected in the actual processes, and frame attributes and critical parameters are obtained using the above mentioned method. Process malfunctions are detected in real-time by applying SPC(Statistical Process Control) method to critical parameters. the Real-time malfunction detection method presented in this paper was applied to actual processes and the results indicated that it was proved to be able to supplement disadvantages of existing quality control check inspecting or testing random-selected devices and detect process malfunctions correctly in real-time.
