Japanese pine sawyer beetle (Monochamus alternatus) is an economically important pest in coniferous trees. Ophiostoma ips was isolated from the beetle and identified based on analysis of morphological properties and the ${\beta}$-tubulin gene sequence. The fungus easily produced perithecia with a long neck on malt extract agar and its ascospores were rectangular shaped. This is first report of Ophiostoma species associated with the pinewood nematode vector beetle in Korea.

Movement of adult Monochamus alternatus was investigated on young black pine, Pinus thunbergii, trees for about one month in a screen cage. Number of pine trees to which M. alternatus females and males moved during the experimental period was $6.4{\pm}1.4$ and $7.2{\pm}1.5$ out of 15 trees, respectively. Adult females and males moved to $0.2{\pm}0.1$ and $0.3{\pm}0.1$ tree per day, respectively. A 74.6 and 80.7% of adult females and males moved at night (17:00-08:00) to other trees. After moving, most beetles (72.6% of females and 76.0% of males) stayed on the tree to which they moved for one to two days. Some beetles stayed for up to seven days.
We assessed efficacy of spraying pesticides from an unmanned helicopter to control two insect species, Monochamus alternatus and M. saltuarius, which are vectors of pine wood nematodes. Control efficacy of thiacloprid FL (33×), acetamiprid ME (33×), and flupyradifurone SL (33×) was determined by placing caged insects in the canopy of pine trees (Pinus sp). Water-sensitive paper was used to record the spray pattern of pesticide droplets and the degree of coverage; furthermore, we investigated peripheral scattering due to spraying. The three pesticides showed > 96% control efficacy against the targeted vectors, and pesticide droplet spray patterns were similar. Peripheral scattering was observed up to 20 m in front and 10 m to the left, right, and behind the targeted area. The coverage index of all the directions at 5 and 10 m distance was 6-7 and 2, respectively.
Esterase isozymes were investigated from digestive juice of M. saltuarius adults after pesticide treatment. Twelve esterase isozymes were separated on 12% native-PAGE gel and stained with three different substrates(${\alpha}$-naphthyl acetate, ${\beta}$-naphthyl acetate, and ${\alpha}$-naphthyl butyrate). Interestingly, the isozyme of Est1(${\alpha}$-naphthyl acetate) was strongly inhibited by the carbofuran and methomyl. The Est1 activity was completely inhibited by the chlorpyrifos and partially inhibited by methidation about 70 %. In addition, eserine suppressed esterase isozyme activities of Est1 about 70% and isozyme activities of Est2, Est3, and Est4 were weakly inhibited. ${\alpha}$-pinene did not suppressed esterase isozyme activities but activities of esterases were very weakly inhibited in camphor and bornyl acetate.
Distribution patterns of two pine sawyer species (Monochamus alternatus which is the main vector insect and M. saltuarius which is the potential insect vector of the pine wood nematode) were investigated in Korea. The data were collected at 89 study sites which were chosen to cover the whole region of South Korea. The selected pine trees were killed in early April and left for I year in the pine stands to be egg-laid by the pine sawyers. Emergence of the beetles from the dead pine trees was checked from early April to late July. M. saltuarius was the most abundant in the mid to northern areas of South Korea, whereas M. alternatus in Jeju-do, southernmost island of Korea. Considering temperature distribution patterns in areas where the two species occur, their thermal distribution boundary may be formed around $13.2^{\circ}C$ of annual mean temperature. The hypothesized distribution map of the two Monochamus species under the invasion of pine wilt disease is suggested on the base of thermal distribution of Korean peninsula.

Japanese pine sawyer (JPS), Monochamus alternatus is a vector insect of pine wood nematode, Bersaphlenchus xylophilus in Korea and Japanese pine forest. This study was to evaluate the possibility of biological control of JPS with Korean entomopathogenic nematodes (EPNs). Korean EPNs (Heterorhabditis sp. Gyeongsan, Steinernema carpocapsae GSN1, S. glaseri Dongrae and S. longicaudum Nonsan strain) were infested bate insect, Galleria mellonella in log of dead pine tree. Result showed that, S. carpocapsae GSN1 strain has the highest pathogenicity. Heterorhabditis sp. Gyeongsan and S. carpocapsae GSN1 had infested bate insect, which located in 7.5 cm depth of pine log. EPN has pathogenicity against larva and adult of JPS. Spray application of EPNs against adult of JPS, Heterorhabditis sp. Gyeongsan strain showed higher pathogenicity than S. carpocapsae GSN1 strain. EPN infested larva of JPS was detected on bark spray treatment with S. carpocapsae GSN1 strain in dead pine log that naturally infested larva of JPS however, no EPN infested JPS was detected on soaking of pine log with suspension of S. carpocapsae GSN1 strain. Though EPN had found with low efficacy against JPS but still possible to apply to control the boring insect pest as they can control log dwelling insect.
This study was performed to investigate the toxicity of 39 registered insecticides to the susceptibility, systemic effect, and residual effect and control effect against Pine sawyer beetle, Monochamus saltuarius. Eleven kinds of chemicals such as fenitrothion, fenthion, phenthoate, phosphamidon, dinotefuran, actamiprid, clothianidin, thiacloprid, thiamethoxam, esfenvalerate+fenitrothion, and fipronil were showed 100% insecticidal activity both in body spray and twig dipping bioassay. Among these chemicals, fenitrothion and fenthion were showed 100% insecticidal activity when sprayed at 4000 times diluted solutions, and phenthoate, thiacloprid, thiamethoxam and fipronil were showed 100% insecticidal activity when sprayed at 2000 times diluted solution. Root systemic effect was showed 100% mortality in phosphamidon, clothianidin, thiamethoxam, and 77.7% in thiacloprid. In residual effect, fenitrothion and thiamethoxam were showed 80% mortality fifteen days after treatment (DAT), and fenthion, phosphamidon, clothianidin were showed 80% mortality ten DAT, fenitrothion, thiamethoxam, fipronil showed 100% mortality in seven DAT, thiacloprid was showed 100% mortality in three DAT. Fenthion and phenthoate were showed 100% mortality one DAT. In the control effect, 6 kinds of chemicals were showed 100% mortality one DAT and all chemicals showed 100% mortality three DAT.
Susceptibility and persistence of Japanese pine sawyer, Monochamus alternatus adults, and acute contact toxicity, foliage contact toxicity, and residual toxicity to honeybee, Apis mellifera were evaluated by using 5 kinds of neonicotinoid pesticides. All neonicotinoids pesticides showed over 80% mortality 48 hour after body spray treatment. However, only 2 kinds of acetamiprid ME and acetamiprid+buprofezin showed over 80% mortality in the twig dipping treatment. All of the five neonicotinoides pesticides used in M. alternatus adults showed residual effect of 60-80% mortality, but the efficacy decreased rapidly after 1 day of treatment. Acetamiprid ME showed the lowest toxicity in the acute and foliage contact toxicity test of A. mellifera. The residual toxicity of leaves on A. mellifera was very low in acetamiprid ME and acetamiprid+buprofezin. However, the residual toxicity of all pesticides treatments decreased to 10-30% after 1 day of treatment and further decreased with time. These results indicate that acetamiprid ME among five neonicotinoid agents showed a high insecticidal activity in the M. alternatus adults, while it exhibited relatively low contact and residual toxicity in the A. mellifera. It is thought that acetamiprid ME can be effectively used for the control of the M. alternatus adults as vector of pine wilt disease.
