Enzymatic Production and Adipocyte Differentiation Inhibition of Low-Molecular-Weight-Alginate (저분자 알긴산의 효소적 생산과 지방세포 분화 억제 효과)

  • In this study, we investigated the extraction condition of alginate from Laminaria japonica, the enzymatic degradation of the extracted alginate, and the inhibitory activity of the degraded alginate on the differentiation of 3T3-L1 preadipocytes. The optimal conditions for the efficient extraction, precipitation, and recovery of alginate from the brown seaweed L. japonica were 1% for Na2CO3 concentration, 80℃ for extraction temperature, and ethanol for precipitation solvent. In the enzymatic reaction for the production of low-molecular-weight alginate (LMWA) by using alginate lyase from Flavobacterium sp., the initial concentration of Laminaria alginate was 3%. The low-molecular-weight degree from alginate was independent with the enzyme concentration, and the optimal concentration of alginate lyase was found to be 5 unit/ml. Through the enzymatic reaction with 5 unit/ml of alginate lyase at 37℃ for 3 hr, the viscosity and molecular weight of LMWA were 4.5 cp and 307 kDa, respectively. Treatment with LMWA significantly suppressed the accumulation of lipid droplet and triglyceride in 3T3-L1 preadipocytes with a dose-dependent manner. Therefore, it seems that LMWA treatment could inhibit the differentiation of 3T3-L1 preadipocytes. These results indicate that LMWA or the degraded alginate produced by alginate lyase enzyme can be useful for the development of anti-obesity biosubstances.

Effect of chemical treatment and variations of the physical properties of waste water-soluble cutting oil (폐 수용성 절삭유의 화학처리효과 및 물성변화)

  • Waste water-soluble cutting oil was treated with WI type #1 and WI type #2. The properties of the original water-soluble cutting oil were pH=l0.4, viscosity=1.4cP, CODcr=44,750 ppm, and TOC=10,569 ppm. However, the properties of the oil used for more than 3 months were changed to pH=7.82, viscosity=2.1cP, CODcr=151,000 ppm, and TOC=74,556 ppm. It might be attributed to the fact that molecular chains were cut due to thermal oxidation and impurities such as metal chips were incorporated in to the oil during the operation processes. To prevent the putrefaction of oil, the sterilization effect of ozone and UV on the microorganism in the oil was investigated. Ozone treatment showed that 99.99% of the microorganism was annihilated with 30 minutes contact time and 60 minutes were necessary for the same effect when UV was used. Ozone treatment could cut molecular chains of the oil due to strong sterilization power, which was evidenced by the increase of TOC from 25,132 ppm at instantaneous contact to 28,888 ppm at 30 minutes contact time. However, UV treatment didn't show severe changes in TOC values and thus, seemed to cause of severe cut of molecular chains. When the activated carbon was used to treat the waste water-soluble cutting oil, TOC decreased to 25,417 ppm with 0.lg carbon and to 15,946 ppm with 5.0g carbon. This results indicated that the waste oil of small molecular chains could be eliminated by adsorption. From the results, it could be concluded that these treatment techniques could be proposed to remove the waste oil of small molecular chains resulting in the degradation of the oil properties. In addition, these experimental results could be used for the correlation with future works such as investigation of the molecular distribution according to the sizes, lengths, and molecular weight of the chains.

Fangchinoline Has an Anti-Arthritic Effect in Two Animal Models and in IL-1β-Stimulated Human FLS Cells

  • Fangchinoline (FAN) is a bisbenzylisoquinoline alkaloid that is widely known for its anti-tumor properties. The goal of this study is to examine the effects of FAN on arthritis and the possible pathways it acts on. Human fibroblast-like synovial cells (FLS), carrageenan/kaolin arthritis rat model (C/K), and collagen-induced arthritis (CIA) mice model were used to establish the efficiency of FAN in arthritis. Human FLS cells were treated with FAN (1, 2.5, 5, 10 µM) 1 h before IL-1β (10 ng/mL) stimulation. Cell viability, reactive oxygen species measurement, and western blot analysis of inflammatory mediators and the MAPK and NF-κB pathways were performed. In the animal models, after induction of arthritis, the rodents were given 10 and 30 mg/kg of FAN orally 1 h before conducting behavioral experiments such as weight distribution ratio, knee thickness measurement, squeaking score, body weight measurement, paw volume measurement, and arthritis index measurement. Rodent knee joints were also analyzed histologically through H&E staining and safranin staining. FAN decreased the production of inflammatory cytokines and ROS in human FLS cells as well as the phosphorylation of the MAPK pathway and NF-κB pathway in human FLS cells. The behavioral parameters in the C/K rat model and CIA mouse model and inflammatory signs in the histological analysis were found to be ameliorated in FAN-treated groups. Cartilage degradation in CIA mice knee joints were shown to have been suppressed by FAN. These findings suggest that fangchinoline has the potential to be a therapeutic source for the treatment of rheumatoid arthritis.

Effect of Extraction Temperature and pH on the Chemical Properties of Crude Porphyran Extracted from Porphyra yezoensis (추출온도 및 pH가 김에서 추출한 Crude Porphyran의 화학특성에 미치는 영향)

  • The effect of extraction temperature ($60\~100^{\circ}C$) and pH ($2.0\~13.5$) on the yield and chemical compositions of crude porphyran from Porphyra Yezoensis was investigated. The yield and chemical compositions of crude porphyran were greatly affected by extraction pH and temperature. Yield was highest between pH $3\~4$ at all temperature ranges. Crude porphyran extracted in acidic conditions showed more sulfate and less protein content than extracted in neutral conditions, but molecular weights was decreased. Crude porphyran extracted in alkaline conditions showed low sulfate and high protein content. For high yield and low molecular weight, acidic condition, particulary pH $3\~4$, was effective. But in order to avoid molecular weight degradation, neutral conditions were effective.

A novel preparation of polyaniline in presence electric and magnetic fields

  • We have described primary studies on conductivity and molecular weight of polyaniline separately in the electric and magnetic fields when it is used in a field effect experimental configuration. We report further studies on doped in-situ deposited polyaniline. First we have chemically synthesized polyaniline by ammonium peroxodisulfate in acidic aques and organic solutions at different times. Then we measured mass and conductivity and obtained the best time of polymerizations. In continue, we repeated these reactions separately under different electric and magnetic fields in constant time and measured mass and conductivity. The polyaniline is characterized by gel permeation chromatography (GPC), UV-Visible spectroscopy and electrical conductivity. High molecular weight polyanilines are synthesized under electric field, $M_w$ = 520000-680000 g/mol, with $M_w/M_n$ = 2-2.5. The UV-Visible spectra of polyanilines oxidized by ammonium peroxodisulfate and protonated with dodecylbenzenesulfonic acid (PANi-DBSA), in N-methylpyrolidone (NMP), show a smeared polaron peak shifted into the visible. Electrical conductivity of polyanilines has been studied by four-probe method. The conductivity of the films of emeraldine protonated by DBSA cast from NMP are higher than 500 and 25 S/cm under 10 KV/m of potential) electric field and 0.1 T magnetic field, respectively. It shows an enhanced resistance to ageing too. By the next steps, we carried chemical polymerization at the best electric and magnetic fields at different times. Finally, resulted in finding the best time and amount of the fields. The longer polymerization time and the higher magnetic field can lead to degradation of polyaniline films and decrease conductivity and molecular mass.

Kinetics of the Low-temperature Pyrolysis of Mixed Plastics in the Batch Reactor (회분식반응기에서의 혼합 플라스틱의 저온열분해 kinetics)

  • Pyrolytic characteristics of mixed plastics containing 22 wt.% HDPE, 17 wt.% LDPE, 27 wt.% PP, 12 wt.% PS, 16 wt.% ABS, 6 wt.% PVC have been studied in the batch-type microreactor of stainless steel. Thermal degradation experiments were performed at temperature of $410{\sim}450^{\circ}C$. The yield of each pyrolytic products were obtained by the weight measurement and molecular weight distribution of pyrolytic liquid products determined by the GC-SIMDIS method. It was shown that the yield and molecular weight of pyrolytic liquid product were decreased with the increase of reaction temperature and time. It was know that 20wt% of PVC composing of the mixed plastics was converted to the gas products of chloride during the pyrolysis process. The chain-end scission rate parameter was determined to be 50.2 kcal/mole of mixed plastics by the Arrhenius plot.

Protease Inhibitors in Porcine Colostrum: Potency Assessment and Initial Characterization

  • Porcine colostrum and milk were separated into the acid-soluble and casein fractions by acidification followed by centrifuge. The acid-soluble fraction of porcine colostrum was further separated by liquid chromatography and anisotropic membrane filtration. Trypsin and chymotrypsin inhibitory capacity in porcine colostrum, milk and their components was determined by incubating bovine trypsin or chymotrypsin in a medium containing their corresponding substrates with or without addition of various amounts of porcine colostrum, porcine milk or their components. The inhibition of insulin-like growth factor I (IGF-I) and epidermal growth factor (EGF) degradation in pig small intestinal contents by porcine colostrum was measured by incubating iodinated IGF-I or EGF with the intestinal contents with or without addition of porcine colostrum. Degradation of labeled IGF-I or EGF was determined by monitoring the generation of radioactivity soluble in 30% trichloroacetic acid (TCA). The results showed that porcine colostrum had high levels of trypsin and chymotrypsin inhibitory activity and increased the stability of IGF-I and EGF in pig intestinal contents. The inhibitory activity declined rapidly during lactation. It was also found that trypsin and chymotrypsin inhibitory activity and the inhibition on IGF-I and EGF degradation in the acid-soluble fraction were higher than that in the casein fraction. Heat-resistance study indicated that trypsin inhibitors in porcine colostrum survived heat treatments of $100^{\circ}C$ water bath for up to 10 min, but exposure to boiling water bath for 30 min significantly decreased the inhibitory activity. Compared with the trypsin inhibitors, the chymotrypsin inhibitors were more heatsensitive. Separation of the acid-soluble fraction of porcine colostrum by liquid chromatography and anisotropic membrane filtration revealed that the trypsin and chymotrypsin inhibitory capacity was mainly due to a group of small proteins with molecular weight of 10,000-50,000. In conclusion, the present study confirmed the existence of high levels of protease inhibitors in porcine colostrum, and the inhibition of porcine colostrum on degradation of milk-borne growth factors in the pig small intestinal tract was demonstrated for the first time.

  • Drag reduction in polymer solutions is the phenomenon where by extremely dilute solutions of high molecular weight polymers exhibit frictional resistance to flow much lower than the pure solvent. This effect, largely unexplained as yet, has attracted the attention of polymer scientists and fluid flow specialists. Although applications are beginning to appear, the principle interest to data has been in attempting to relate the effect to the fluid mechanics of turbulent flow. Drag reduction in two phase flow can be applied to the transport of crude oil, phase change system such as chemical reactor, and pool and boiling flow. But the research on drag reduction in two phase flow is not intensively investigated. Therefore, experimental investigations have been carried out to analyze the drag reduction produced by polymer addition in the single phase and two phase flow system. The objectives of the proposed investigation are primarily in identifying and developing high performance polymer additives for fluid transportations with the benefits of turbulent drag. Also we want to is to evaluate the drag reduction in horizontal flow by measuring pressure drop and mean velocity. Experimental results show higher drag reduction using co - polymer(A611P) then using polyacrylamide (PAAM) and faster degradation using PAAM than using A611P under the same superficial velocity.

  • Proposals were mode on how to differentiate radiation effects in morphological phases of polyethylene and discussions were developed with the results obtained on a low density polyethylene, SOCAREX, specified by number average molecular weight; overbar Mn=5,400, density; 0.92, and degree of branch; 3.4/100 carbon atom, which was irradiated to Co$^{60}$ .gamma. ray at the dose rate of 0.5 Mrad/hr in ambient temperature under the pressure of 10$^{-5}$ Torr. or 1 atm. respectively. The effect to crystalline phase in possibly deduced from dose dependent variation of relative area between (110) and (200) peaks on X ray diffraction spectrum and that, the effects to amorphous phase can be understood through dose dependent relaxation behaviours of .betha. peak on internal friction characteristics of the specimen. The results obtained thus far indicate that, in crystalline phase, relative crystallinity shows a rather rapid decrease up to 20 Mrad with increasing dose, however, little change of crystallinity can be observed in the region between 20-200 Mrad, and degradation appears to be more predominant than crosslinking up to 60 Mrad. While in amorphous phase the indication also shows that degradation is only predominant up to 20 Mrad. Furthermore several correlations can be seen with amenable explanation between dose dependent behaviours observed in both phases.

Relation between Surface degradation and Anti-pollution Characteristics in RTV Silicone Rubber (RTV 실리콘 고무의 표면열화와 내오손 특성과의 상관관계)

  • In this paper we investigated the relation between the surface degradations and anti-pollution characteristics of Room Temperature Vulcanized(RTV) silicone rubber coating that has different roughness through immersing into saline water. We utilized several analytic techniques such as atomic force microscopy(AFM) scaning electron microscopy(SEM) contact angle Salt Deposit Density(SDD) and average leakage current under the condition of salt fog. It is found that the surface roughness of treated RTV silicone rubber increased and the hydrophobicity of sample surface decreased with increasing the duration o immersion into water due to the erosion of base polymer the melting down alumina trihydrate(ATH) and the diffusion of Low Molecular weight(LMW) fluid. Despite the roughness of surface had been increased by water immersion excellant anti-pollution and recovery characteristics were maintained and SDD saturated to 0.1~0.14mg/cm$^2$. The average leakage current under salt fog increased with surface roughness. Measurement of average leakage current will be helpful to investigate surface degradation and lifetime expectation of RTV silicone coating.
