Plum pocket caused by the dimorphic ascomycetous fungi, Taphrina spp., results in unsightly malformations and crop loss. In 2016, Japanese plums (Prunus salicina Lindl.) with plum pocket symptoms were found in Gimcheon. Three isolates were collected from symptomatic P. salicina fruits and identified as Taphrina deformans based on morphological characteristics and molecular sequence analysis of including internal transcribed space (ITS) and the mitochondrial small ribosomal subunit (SSU) regions of the three isolates. Pathogenicity test on plum fruits confirmed that, the present T. deformans isolates are causal agent of plum pocket. To the best of our knowledge, this is the first report of plum pocket caused by T. deformans in South Korea.

Korean fir (Abies koreana) is an evergreen coniferous tree species that is unique to South Korea. A. koreana is found in a limited sub-alpine habitat and is considered particularly vulnerable to climate change. Identification of populations vulnerable to climate change is an important component of conservation programs. In this study, a heat stress-induced transcriptome RNA-seq dataset was used to identify a subset of six genes for assessment as candidate marker genes for ecologically vulnerable populations. Samples of A. koreana were isolated from ecologically stable and vulnerable regions of the Halla and Jiri mountains, and the expression levels of the six candidate markers were assessed using quantitative real-time polymerase chain reaction. All six of the candidate genes exhibited higher expression levels in samples from vulnerable regions compared with stable regions. These results confirm that the six high temperature-induced genes can be used as diagnostic markers for the identification of populations of A. koreana that are experiencing stress due to the effects of climate change.
The aim of this study is to develope eco-printing method using natural pigments and chitosan as a natural binder. Three chitosans with different molecular weights were employed to find appropriate conditions including chitosan concentration and pigment/binder ratio. Dye uptake, color and fastnesses of the printed fabrics were evaluated to find optimum conditions within the range of experiments carried out in this study. The effectiveness of chitosan as a printing binder was examined in comparison with color, dye uptake, and fastnesses of conventional synthetic binder and guar gum. It was found that chitosans with low or medium molecular weight were appropriate. Using low molecular weight chitosan, optimum concentrations were 1.7% for charcoal, madder and chlorophyll, whereas 2.2% for ocher, yellow soil, indigo and cochineal. Regardless of molecular weight and concentration of chitosan, the color fastnesess of fabrics printed with mineral pigments were superior to those of the fabrics printed with plant and animal pigments. As pigment/chitosan ratio became higher, rubbing fastness was decreased by 1-3 grade. The colorfastness of printed fabric with chitosan binder was similar to that with synthetic binder, which was higher than that with guar gum.

Conventional culture-based methods of enumerating rumen microorganisms (bacteria, archaea, protozoa, and fungi) are being rapidly replaced by nucleic acid-based techniques which can be used to characterise complex microbial communities without incubation. The foundation of these techniques is 16S/18S rDNA sequence analysis which has provided a phylogenetically based classification scheme for enumeration and identification of microbial community members. While these analyses are very informative for determining the composition of the microbial community and monitoring changes in population size, they can only infer function based on these observations. The next step in functional analysis of the ecosystem is to measure how specific and, or, predominant members of the ecosystem are operating and interacting with other groups. It is also apparent that techniques which optimise the analysis of complex microbial communities rather than the detection of single organisms will need to address the issues of high throughput analysis using many primers/probes in a single sample. Nearly all the molecular ecological techniques are dependant upon the efficient extraction of high quality DNA/RNA representing the diversity of ruminal microbial communities. Recent reviews and technical manuals written on the subject of molecular microbial ecology of animals provide a broad perspective of the variety of techniques available and their potential application in the field of animal science which is beyond the scope of this treatise. This paper will focus on nucleic acid based molecular methods which have recently been developed for studying major functional groups (cellulolytic bacteria, protozoa, fungi and methanogens) of microorganisms that are important in nutritional studies, as well as, novel methods for studying microbial diversity and function from a genomics perspective.
Our previous work confirmed that Sph2/LA1029 was a sphigomyelinase-like hemolyisn of Leptospira interrogans serogroup Icterohaemorrhagiae serovar Lai. Characteristics of both hemolytic and cytotoxic activities of Sph2 were reported in this paper. Sph2 was a heat-labile neutral hemolysin and had similar hemolytic behavior as the typical sphingomyelinase C of Staphylococcus aureus upon sheep erythrocytes. The cytotoxic activity of Sph2 was shown in mammalian cells such as BALB/C mouse lymphocytes and macrophages, as well as human L-02 liver cells. Transmission electron microscopic observation showed that the Sph2 treated BALB/C mouse lymphocytes were swollen and ruptured with membrane breakage. They also demonstrated condensed chromatin as a high-density area. Cytoskeleton changes were observed via fluorescence confocal microscope in Sph2 treated BALB/C mouse lymphocytes and macrophages, where both cytokine IL-$1{\beta}$ and IL-6 were induced. In addition, typical apoptotic morphological features were observed in Sph2 treated L-02 cells via transmission electron microscope and the percentage of apoptotic cells did increase after the Sph2 treatment detected by flow cytometry. Therefore, Sph2 was likely an apoptosis-inducing factor of human L-02 liver cells.
Eurycoma longifolia (pasak bumi) is a popular medicinal plant in Indonesia and is widely used in various products. Its high economic value has caused illegal harvesting and product falsification. Using molecular techniques, the authentication and traceability of E. longifolia derivatives can be controlled to ensure consumer safety. Therefore, this study aimed to authenticate the products and derivatives of E. longifolia (pasak bumi) produced, marketed, and consumed in Indonesia using molecular identification techniques. Genomic DNA from 37 leaf samples collected from the Sumatran mainland and the Riau Islands and six E. longifolia products were amplified and sequenced using trnL-trnF and internal transcribed spacer (ITS) regions. The results revealed that all leaf samples were indeed E. longifolia based on the markers used, with the six products, only the herbal tea product (sample code TCPB) was most likely derived from E. longifolia based on the two regions, suggesting that not all products labelled as E. longifolia in the market are authentic. The results also indicated that several other plants species are used as substitutes or adulterants, including Simaba spp., Simarouba spp., Homalolepis spp., Vernonia gigantea, Elephantopus scaber, Gymnanthemum amygdalinum, Cyanthillium spp., Potentilla lineata, Ailanthus altissima, Geijera paniculata, Hannoa chlorantha, and Dalbergia spp. Klebsiella pneumoniae bacteria were also identified in this study on the outer wooden cup of E. longifolia products. Therefore, this molecular approach is effective in identifying the authenticity of E. longifolia products, with trnL-trnF and ITS as the recommended DNA markers.

DNA markers have been studied and used intensively to identify plant species based on molecular approaches. The genus Medicago belongs to the family Fabaceae and contains 87 species distributed from the Mediterranean to central Asia. Five species of Medicago are known to be distributed in South Korea; however, their morphological characteristics alone cannot distinguish the species. In this study, we analyzed the phylogenetic relationships using collected five species of Medicago from South Korea and 44 taxa nucleotide information from NCBI. The constructed phylogenetic tree using gibberellin 3-oxidase 1 and tRNALys (UUU) to maturase K gene sequences showed the monophyly of the genus Medicago, with five species each forming a single clade. These results suggest that there are five species of Medicago distributed in South Korea. In addition, we designed polymerase chain reaction primers for species-specific detection of Medicago by comparing the plastid sequences. The accuracy of the designed primer pairs was confirmed for each Medicago species. The findings of this study provide efficient and novel species identification methods for Medicago, which will assist in the identification of wild plants for the management of alien species and living modified organisms.
This study was carried out to determine the pathogen-causing diarrhoea in sheep Ovis aries in the Qinghai Tibetan Plateau Area, China. A trophozoite was identified as species of ciliate alveolates infecting the sheep based on morphological characteristics examined by microscope. It was mostly spherical, colourless and transparent, with many vesicles. Macronucleus and contractile vacuoles could not be distinguished. Size of the trophozoite was $80-180{\times}70-150{\mu}m$ and its surface was covered with cilia. Molecular analysis based on sequences of 18S rRNA and ITS genes confirmed the ciliate species as Balantidium coli. According to the literature, there have been many epidemiological investigations of B. coli infection in pigs, monkeys and humans. To our knowledge, this was the first report of B. coli infections in sheep in the Qinghai Tibetan Plateau Area of China, or eleswhere around the world. Importantly, the sheep case was rare but raised our concern that B. coli may spread across species and expand its host range.
The discovery of deep-sea hydrothermal vents and their ecosystems is a monumental landmark in the history of Ocean Sciences. Deep-sea hydrothermal vents are scattered along the global mid-ocean ridges and back-arc basins. Under sea volcanic phenomena related to underlying magma activities along mid-ocean ridges generate extreme habitats for highly specialized communities of animals. Multidisciplinary research efforts during past three decades since the first discovery of hydrothermal vents along the Galapagos Rift in 1977 revealed fundamental components of physiology, ecology, and evolution of specialized vent communities of micro and macro fauna. Heterogeneous regional geological settings and tectonic plate history have been considered as important geophysical and evolutionary factors for current patterns of taxonomic composition and distribution of vent faunas among venting sites in the World Ocean basins. It was found that these communities are based on primary production of chemosynthetic bacteria which directly utilize reduced compounds, mostly $H_2S$ and $CH_4$, mixed in vent fluids. Symbioses between these bacteria and their hosts, vent invertebrates, are foundation of the vent ecosystem. Gene flow and population genetic studies in parallel with larval biology began to unveil hidden dispersal barrier under deep sea as well as various dispersal characteristics cross taxa. Comparative molecular phylogenetics of vent animals revealed that vent faunas are closely related to those of cold-water seeps in general. In perspective additional interesting discoveries are anticipated particularly with further refined and expanded studies aided by new instrumental technologies.

The Asiatic black bear, Ursus thibetanus, is among the most threatened or endangered species in Asia. For its conservation and management, sex identification of U. thibetanus using non-invasive samples (e.g., hair and/or feces) is potentially valuable. In this study, a non-invasive molecular method for sex identification of U. thibetanus samples collected from various countries was first utilized, and it was based on polymerase chain reaction (PCR) amplification of the amelogenin gene via PCRs. Thirty-three bear DNA samples, extracted not only from blood (n=9) but also from hair (n=18) and feces (n=6), were used. We performed sex-specific PCR amplifications of the amelogenin gene using a primer set, SE47 and SE48. The primer set could successfully amplify a single X-specific band for females and both X- and Y-specific bands for males from all blood (100%) and hair (100%) samples. In addition, the primer set could distinguish the sex of bears in four out of a total of six fecal samples (approximately 67%). This study's findings suggest that this molecular method can be applied to sex identification of Asiatic black bears from various Asian regions using non-invasive samples, such as hair and feces.
