Protease Inhibitor Production using Streptomyces sp. SMF13

  • The aim of the current study is to evaluate the effects of medium compositions on the production of protease inhibitor in Streptomyces sp. SMF13. The production of protease inhibitor was counter-currently linked to extra-cellular protease, which were regulated by the culture conditions. Nitrogen source was the most critical ingredient affecting the production of protease inhibitor and protease. Carbon source was an important factor to determine the culture pH which affected very clearly the formation of protease and protease inhibitor. Inorganic phosphate inhibited the protease inhibitor production which was linked to the cell growth rate, although the optimal conditions for the production of protease inhibitor were not favouring to the cell growth.

Synthesis and Properties of Triblock and Multiblock Copolymers Consisting of Poly(L-lactide) and Poly(oxyethylene-co-oxypropylene)

  • Both A-B-A triblock and multiblock copoly(ester-ether)s consisting of poly(L-Lactide) and poly(oxyethylene-co-oxypropylene) were prepared and characterized. The preparation of the triblock copolymer was done by ring-opening copolymerization of L-lactide with a commercially available telechelic copolyether, Pluronic$\^$TM/(PN) by catalysis of stannous octanoate. The molecular weight and unit composition of the produced copolymers were successfully controlled by changing the L-lactide/PN ratio in feed. However, a high molecular weight copolymer incorporating PN in large amount was not obtained because the molecular weight of the resulting copolymer was limited at a high L-lactide/PN composition. The multiblock copolymer was synthesized by the copolycondensation of oligo(L-lactic acid) prepared by thermal dehydration of L-lactic acid, PN, and dodecanedioic acid as carboxyl/hydroxyl adjusting agent. This polycondensation proceeded by catalysis of stannous oxide to give multiblock copolymers with high molecular weight and wide range of compositions.

선형 및 분지화된 폴리카보네이트 블렌드의 기계적 성질과 유동특성 (Flow and Mechanical Properties of Linear and Branched Polycarbonates Blends)

  • 본 논문은 선형 폴리카보네이트와 분지화된 폴리카보네이트를 블렌드한 시스템에 대해서 연구하였다. 여러가지 조성으로 두 성분을 블렌드하여 각 조성에 대해서 상용성, 기계적 성질 그리고 유동특성을 조사하였다. 본 블렌드 시스템에서 상분리는 관찰되지 않았고 상용성도 있었다. 기계적 성질은 블렌드의 각 조성에 대해서 인장강도, 굽힘강도 그리고 충격강도 등을 측정하여 검토하였으며, 흐름특성 역시 블렌드의 각 조성에 대해 용융점도, 저장탄성률 및 손실탄성률을 여러 온도에서 측정하여 검토하였다. 그리고 용융점도의 분자량에 대한 의존성을 조사하였다. 선형 폴리카보네이트와 분지화된 폴리카보네이트의 조성에 따라 기계적 물성은 거의 변화가 없었으나, 흐름특성은 큰 변화가 있었다. 본 블렌드 시스템에서 분지화된 폴리카보네이트 함량이 증가할수록 점도의 분자량에 대한 의존성이 컸으며, shear thinning 현상은 두드러지게 나타났다. 따라서 재료의 기계적 물성의 변화없이 흐름특성이 다른 블렌드 시스템을 설계할 수 있다.

유기폴리머 용액에 혼합한 몬모릴로나이트의 응집 거동 및 특징 (Flocculation Behavior and properties of Montmorillonites Mixed with Organic Polymer Solutions)

  • Four organic polymers were mixed with mothmorillonite. Two cationic polymers a hi로 molecular weight polyacrylamide (494C) and a low molecular weight polymer (587C).Two anionic polymers include a high molecular weight polymer (aerotil). Each clay supension series were allowed to stand for 24 hours and were centrifuged, and the clay plugs were washed and dried. The dried samples investigated by XRD, IR and CEC measurement. The suspended clay containing anionic polymers was not flocculated at any concentratuons of polymer. But the suspendions containing two cationic polymers were rapidly flocculated at almost all concentrations. the d(001) spacings of Na-montmorillonite after being with cationic polymer 587C show about 15$\AA$ suggesting the polymers may have entered the interlayer spaces. The polymer 494C-treated sample produced double peaks of about 12 and 15$\AA$ in XRD. It indicates that the high molecular weight polymer. And cationic polymer 494C may be adsorbed mainy on the outside surface of clay, and some polymers may peretrate into olny interlayers in the margin of montmorillonite particles because of its high molecular weught. CEC of polumer 587-treated sample was reduecd mmarkedly suggesting polymer blocks CEC sites. The d(001) spacings of Ca-montmorillonite after being treated with cationic polymers show about 15$\AA$ suggesting that the interlayer spaces have not been expanded. In the experiment using a dilute Ca-bearing solution, the suspended caly containinf anionic polymers was flocculated. The results indicate that the flocculation behavior of montmorillonite-polymer supension depends on not only polymer properties such as concentration, electric charge and molecular weight but also compositions of solvent.

Analysis of Fatty Acyl Groups of Diacyl Galactolipid Molecular Species by HPLC/ESI-MS with In-source Fragmentation

  • The structures of molecular species of galactolipids, such as monogalactosyl diacylglycerol (MGDG) and digalactosyl diacylglycerol (DGDG), isolated from wheat flour have been investigated using negative-ion electrospray ionization (ESI) mass spectrometry interfaced with high performance liquid chromatography (HPLC). According to the result of HPLC analysis, MGDG and DGDG were found to consist of mixtures of five and four molecular species, respectively. The galactolipids have been also analyzed to determine their fatty acid compositions, using HPLC/ESI-MS combined with in-source (or cone voltage) fragmentation. HPLC/ ESI-MS is very useful for one-step analysis of mixtures of galactolipids with a small sample quantity. Especially, the carboxylate anions produced in in-source fragmentations of the negative-ion of each component separated by HPLC provide valuable information on the composition of its fatty acyl chains.

  • A sequential copolypeptide with repeating unit sequences, in which a glycyl residue is flanked progressively by two ${\gamma}$-benzyl-L-glutamyl residues, has been synthesized by poymerizing p-nitrophenyl ester of ${\gamma}$-benzyl-L-glutamyl-${\gamma}$ -benzyl-L-gutamyl-glycine. Polymers obtained were characterized by viscosity and infra-red spectral data. The highest molecular weight obtained was 21,000. Molecular conformation in solid state was found to be a mixed form of and |${\beta}$-structure. Polymers obtained were insoluble in the most of the organic solvents except in a strong acid like dichloroacetic acid, but in binary mixtures of solvents such as dichloroacetic acid-ethylene dichloride or dichloroacetic acid-chloroform, they were soluble within certain ranges of solvent compositions.

The Interface Between ER and Mitochondria: Molecular Compositions and Functions

  • Mitochondria and endoplasmic reticulum (ER) are essential organelles in eukaryotic cells, which play key roles in various biological pathways. Mitochondria are responsible for ATP production, maintenance of $Ca^{2+}$ homeostasis and regulation of apoptosis, while ER is involved in protein folding, lipid metabolism as well as $Ca^{2+}$ homeostasis. These organelles have their own functions, but they also communicate via mitochondrial-associated ER membrane (MAM) to provide another level of regulations in energy production, lipid process, $Ca^{2+}$ buffering, and apoptosis. Hence, defects in MAM alter cell survival and death. Here, we review components forming the molecular junctions of MAM and how MAM regulates cellular functions. Furthermore, we discuss the effects of impaired ER-mitochondrial communication in various neurodegenerative diseases.

Prediction of Gas Permeability by Molecular Simulation

  • The research and development of high-performance polymer materials with excellent gas barrier properties has gained considerable attention from the viewpoint of expanding their applications in various fields, including tire automobile parts and the polymer film industry. Natural rubber (NR) has been widely used as a rubber material in real-life, but its application is limited owing to its poor gas barrier properties. In this paper, we study the gas barrier properties of NR, epoxidized natural rubber (ENR), and their blend compositions by using molecular simulation. The results show that ENR-50 has superior oxygen barrier properties than those of NR. Moreover, the oxygen barrier properties of a blend of NR/ENR-50 improve with increasing volume fraction of ENR-50. The trend of improved oxygen barrier properties of NR, ENR-50, and their blend is in good agreement with experimental observations.

바인더의 공중합체 조성에 따른 안료잉크의 발색성 및 견뢰도 연구 (Color Strength and Fastness of Pigment Ink with Various Binder Monomer Compositions)

  • The binder polymers for digital textile printing(DTP) pigment inks were prepared using miniemulsion polymerization with various monomer compositions to study effects of monomer compositions on particle size distribution, average molecular weight, Tg, and color strength and rubbing fastness of the dyed fabrics with the prepared binder based pigment ink. The monomers used were MMA(Methyl methacrylate), BA(Butyl acrylate), MAA(Methacrylic acid), NMA(N-methylol acrylamide), NEA(N-ethylol acrylamide) and the ratios of the monomers were changed. The particle size was the smallest with 136nm when the MMA to BA weight ratio was 4:16 and the largest with 290nm when the MMA, BA, MAA, NEA ratio was 2.5:17:0.25:0.25. However, the glass transition temperature was lowest with $-41.90^{\circ}C$ and the color strength and rubbing fastness of the resulting sample were the best when the MMA, BA, MAA, NEA ratio was used. This suggested that the introduction of the NEA monomer to the binder polymer for the pigment ink could be an efficient way to enhance the rubbing fastness of the DTP pigment inks present.

바인더 단량체 조성 변화에 따른 안료 잉크의 마찰견뢰도 연구 (Investigation on Rubbing Fastness of Pigment Ink with Polymer Binders having Various Comonomer Compositions)

  • To improve rubbing fastness of the printed fabrics, the binder polymers for Digital Textile Printing(DTP) pigment inks were synthesized with miniemulsion polymerization using various acrylic monomers, which are MMA(Methyl methacrylate), BA(Butyl acrylate), and Self-crosslinking monomers, such as NEA(N-Ethylol acrylamide) and MAA (Methacrylic acid). The acrylic monomer compositions were varied when synthesizing the binder polymers and their particle size distributions, average molecular weights, and Tgs were investigated. The prepared binder polymers were applied to prepare Cyan, Black, Yellow and Magenta pigment ink for DTP and the prepared inks were used to dye cotton fabrics. Then, color strength, and rubbing fastness were also investigated to study the effect of the comonomer compositions of the binder polymer on the color strength and rubbing fastness of the resulting pigment inks.
