Proteome analysis of roots of sorghum under copper stress

  • Sorghum bicolor is considered as copper-tolerant species. The present study was conducted to understand the copper tolerance mechanism in Sorghum seedling roots. Morphological and effects of Cu on other interacting ions were observed prominently in the roots when the plants were subjected to different concentrations (0, 50, and $100{\mu}M$) of $CuSO_4$. However, the morphological characteristics were reduced by Cu stress, and the most significant growth inhibition was observed in plants treated with the highest concentration of $Cu^{2+}$ ions ($100{\mu}M$). In the proteome analysis, high-throughput two-dimensional polyacrylamide gel electrophoresis coupled with MALDI-TOF-TOF mass spectrometry was performed to explore the molecular responses of Cu-induced sorghum seedling roots. In two-dimensional silver-stained gels, a total of 422 differentially expressed proteins (${\geq}1.5-fold$) were identified using Progenesis SameSpot software. A total of 21 protein spots (${\geq}1.5-fold$) from Cu-induced sorghum roots were analyzed by mass spectrometry. Of the 21 differentially expressed protein spots from Cu-induced sorghum roots, a total of 10 proteins were up-regulated, and 11 proteins were down-regulated. The abundance of the most identified protein species from the roots that function in stress response and metabolism was significantly enhanced, while protein species involved in transcription and regulation were severely reduced. The results obtained from the present study may provide insights into the tolerance mechanism of seedling roots in Sorghum.

Comparative proteome analysis of seeds of proso millet (Panicum miliaceum) cultivars

  • Since the composition of proteins from the Korean cultivars of Proso millet is unknown; thereby, the present study was conducted to obtain a reference map of millet seed proteins and identify the functional characteristics of the identified proteins. Proteins extracted from the millet seeds of various cultivars, were investigated using proteomic techniques as 2D electrophoresis coupled with mass fingerprinting. The 1152 (differentially expressed) proteins were detected on 2-D gel. Among them, 26 reproducible protein spots were analyzed by MALDI-TOF-TOF mass spectrometry. Out of 26 proteins, 2 proteins were up-regulated towards all cultivars of millet, while 7 proteins were up-regulated and 13 proteins were down-regulated against only one cultivar. However, abundance in most identified protein species, associated with metabolism, transcription and transcription was significantly enhanced, while that of another protein species involved in polysaccharide metabolism, stress response and pathogenesis were severely reduced. Taken together, the results observed from the study suggest that the differential expression of proteins from the four cultivars of millet may be cultivar-specific. Taken together, a proteomic investigation of millet seeds from different cultivars, we sought to better understand the genetic variation of millet cultivars representing the future millet research, and the functional categorization of individual proteins on the basis of their molecular function.

Comparative proteome analysis of rice leaves in response to high temperature

  • The productivity of rice has been influenced by various abiotic factors including temperature which cause to limitations to rice yield and quality. Rice yield and quality are adversely affected by high temperature globally. In the present study, four Korean four cultivars such as Dongan, Ilpum, Samkwang, Chucheong were investigated in order to explore molecular mechanisms of high temperature at seedling stage. Rice seedlings grown at $28/20^{\circ}C$ (day/night) were subjected to 7-day exposure to $38/28^{\circ}C$ for high-temperature stress, followed by 2-D based proteomic analysis on biological triplicates of each treatment. The growth characteristics demonstrated that Dongan is tolerant while Ilpum is sensitive to high-temperature stress. High temperature has an adverse effect in the seedling stage both in high temperature sensitive and tolerant cultivar. Two-dimensional gels stained with silver staining, a total of 722 differential expressed protein spots (${\geq}1.5-fold$) were identified using Progenesis SameSpot software. However, a total of 38 differentially expressed protein spots were analyzed by LTQ-FT-ICR MS. Of these, 9 proteins were significantly increased while 10 decreased under high-temperature treatment. Significant changes were associated with the proteins involved in the carbohydrate metabolism, photosynthesis, and stress responses. Proteome results revealed that high-temperature stress had an inhibitory effect on carbon fixation, ATP production, and photosynthetic machinery pathway. The expression level of mRNA is significantly correlated with the results obtained in the proteome investigation. Taken together, these findings provide a better understanding of the high-temperature resistance by proteomic approaches, providing valuable insight into improving the high-temperature stress tolerance in the global warming epoch.

The Biocontrol Activity of Chromobacterium sp. Strain C-61 against Rhizoctonia solani Depends on the Productive Ability of Chitinase

  • A chitinolytic bacterium, Chromobacterium sp. strain C-61, was found strongly antagonistic to Rhizoctonia solani, a causal agent of damping-off of eggplant. In this study, the biocontrol activity and enzymatic characteristics of strain C-61 were compared with its four Tn5 insertion mutants (C61-A, -B, -C, and -D) that had lower chitinolytic ability. The chitinase activity of a 2-day old culture was about $76\%,\;49\%\;and\;6\%$ level in C61-A, C61-B and in C61-C, respectively, compared with that of strain C-61. The $\beta-N-acetylhexosaminidase$(Nahase) activity was little detected in strain C-61 but increased largely in C-61A, C61-B and C61-C. Activities of chitinase and Nahase appeared to be negatively correlated in these strains. Another mutant, C-61D, produced no detectable extracellular chitinase and Nahase. The in vitro and in vivo biocontrol activities of strain C-61 and its mutants were closely related to their ability to produce chitinase but not Nahase. No significant differences in population densities between strain C-61 and its mutants were observed in soil around eggplant roots. The results of SDS-PAGE and isoelectrofocusing showed that a major chitinase of strain C-61 is 54-kDa with pI of approximately 8.5. This study provides evidence that the biocontrol activity of Chromobacterium sp. strain C-61 against Rhizoctonia solani depends on the ability to produce chitinase with molecular weight of 54-kDa and pI of 8.5.

Morphological Variations, Genetic Diversity and Pathogenicity of Colletotrichum species Causing Grape Ripe Rot in Korea

  • Ripe rot was frequently observed on fruits, leaves and stems of grape growing in eight locations in Korea from 2004 to 2006. All 30 isolates of Colletotrichum sp. were obtained from lesions of the ripe rot on grape plants. Out of the isolates, 19 isolates were identified as Colletotrichum acutatum and the others as Colletotrichum gloeosporioides based on morphological and cultural characteristics. Inter and intra specific variations of the Colletotrichum spp. isolates were investigated using RAPD and sequences of rDNA ITS and $\beta$-tubulin-2. Isolates of C. acutatum and C. gloeosporioides were distinctly differentiated by molecular analyses. Phylogenetic trees of ITS and$\beta$-tubulin-2 showed that Korean isolates of C. acutatum were clustered into groups A2 and A3 among the eight global groups. A2 included non-chromogenic isolates and A3 chromogenic ones. Both C. acutatum and C. gloeosporioides isolates were tested for pathogenicity to grape leaves. All isolates tested induced lesions on the leaves of grape by artificial inoculation. There was no difference in pathogenicity between C. acutatum and C. gloeosporioides isolates. This is the first report that C. acutatum except C. gloeosporioides causes grape ripe rot in Korea.

Calcification on the Surface of Silicone Nasal Implants: Regional Properties of Calcification Deposits (실리콘비삽입물 표면의 석회화: 부위별 석회침착물의 특성)

  • The silicone rubber implants are widely used in plastic surgery because of various advantages; however, calcification in surface of implant(as a chemical resistance) may transform or destroy the high molecular biomaterial when it stays too long within the human body. The purpose of this study is to determine the relationship between calcification and the histological disparities of the tissues surrounding the area adjoining the silicone nasal implant by examining the regional characteristics of calcium deposits in the silicone nasal implant via elemental analyses using EDX(energy-dispersive X-ray analysis) and ultrastructural analyses using SEM(scanning electron microscopy). The subjects of the study were 19 silicone nasal implants removed by revision rhinoplasty, all displaying calcification. According to the tissue characters, the implant surface was divided into 4 zones with the rhinion as the basis. For each zone, elemental and ultrastructural analyses were performed. Elemental analysis revealed that the calcium deposits consisted of Ca and P only. There were no statistically significant disparities among the ratios between Ca and P according to the zones. Ultrastructural analysis showed acellular mineral-like deposits coalesced to create amorphous deposits in all zones; however, in zones 1 and 3(more pressurized zones by periosteum or nasal bone), additional flaky cylinder-shaped calcium deposits were detected. Thus, it seems that the histological disparities in the surrounding tissues do not affect the components and their proportions in the calcification process. However, it can be inferred that the physical environment due to the histological disparities in the surrounding tissues affects the ultrastructures of calcium deposits.

First Report of Peach Fruit Rot Caused by Fusarium avenaceum in Korea (Fusarium avenaceum에 의한 복숭아 신규 과실 썩음병 발생 보고)

  • In July 2019, typical rot symptom was observed on peach fruits harvested from the fields at Andong, Korea. As the disease progressed, white and purple colored mycelial mat developed on the surface of the infected fruits. A causal pathogen was isolated from the infected fruit and cultured on potato dextrose agar media for identification. Fungal colonies on potato dextrose agar produced 3 pigments, including purple, yellow, and white colors. The isolate incited fruit rot symptoms on artificially inoculated peach fruits, from which the same fungus was isolated, fulfilling Koch's postulates. Based on the morphological characteristics and sequence analysis of rDNA internal transcribed spacer, translation elongation factor 1-alpha, and β-tubulin, the causal agent of the disease was identified as Fusarium avenaceum. This study is the first report of fruit rot of peach fruits caused by Fusarium avenaceum in Korea.

Risk assessment and distribution characteristics of N-nitrosamines in drinking water treatment plants (나이트로사민류의 국내 정수장 분포 특성 및 위해성 평가)

  • A nationwide survey of 8 N-nitrosamines in finished water samples from drinking water treatment plants (DWTPs) in Korea was conducted. The samples were pre-treated by solid-phase extraction (SPE) and analyzed using a gas chromatography coupled with tandem mass spectrometry (GC-MS/MS). According to the study results, four N-nitrosamines (NDMA, NDEA, NMOR, NDBA) were detected for three consecutive years, NMEA and NPYR were only found in samples collected in 2013. Two of these N-nitrosamines, N-nitrosodimethylamine (NDMA) and N-nitrosodiethylamine (NDEA), have received attention and were the most commonly detected. The concentration of NDMA and NDEA in this study ranged from $0.002{\mu}g/L$ to $0.013{\mu}g/L$ and in $0.001{\mu}g/L$ to $0.008{\mu}g/L$, respectively. In comparison to studies performed in EPA(UCMR2), the concentrations of NDMA (from $0.002{\mu}g/L$ to $0.630{\mu}g/L$) and NDEA (from $0.005{\mu}g/L$ to $0.100{\mu}g/L$) observed in the this study were low.

The study on photoreflectance characteristics of the $Al_xGa_{1-x}As$ epilayer grown by MBE method (MBE 법으로 성장시킨 $Al_xGa_{1-x}As$ 에피층의 Photoreflectance 특성에 관한 연구)

  • We analyzed photoreflectance (PR) characterization of the $Al_xGa_{1-x}As$ epilayer grown by molecular beam epitaxy (MBE) method. The band-gap energy $(E_0)$ satisfying low power Franx-Keldysh (LPFK) due to GaAs buffer layer is 1.415 eV, interface electricall field $(E_i)$ is 1.05$\times$$10^4$V/cm, carrier concentration (N) is $1.3{\times}10^{15}\textrm{cm}^{-3}$. In PR spectrum intensity analysis at 300 K the $A^*$ peak below $(E_0)$ signal is low and distorted because of residual impurity in sample growth. The trap characteristic time ${\tau}_i$ of GaAs buffer layer is about 0.086 ms, and two superposed PR signal near 1.42eV consist of the third derivative signal of chemically eteched GaAs substrate and Franz-Keldysh oscillation (FKO) signal due to GaAs buffer layer.

Identification of water mold from wild brook lamprey, Lethenterone reissneri (자연산 다묵장어, Lethenterone reissneri에서 발생한 물곰팡이병 원인체의 동정)

  • Saprolegnia isolate from wild brook lamprey was identified on the basis of its morphological and molecular characteristics. The isolates showed aseptic hyphae and clavate zoosporagium. Zoospores discharge was typically saprolegnoid. Neither oogomia nor antheridia was observed in this study. ITS sequence obtained from the isolate was compared with other Saprolegnia spp. to analyse their phylogenetic relationships. Results showed that the isolate belongs to clade I including Saprolegnia parasitica. Based on the asexual organs, zoospore discharge manner and ITS sequence analysis, the isolate was identified as S. parasitica.
