Geft is a guanine nucleotide exchange factor, which can specifically activate Rho family of small GTPase by catalyzing the exchange of bound GDP for GTP. Geft is highly expressed in the excitable tissue as heart and skeletal muscle and plays important roles in many cellular processes, such as cell proliferation, migration, and cell fate decision. However, the in vivo role of Geft remains unknown. Here, we generated a Geft conditional knockout mouse by flanking exons 5-17 of Geft with loxP sites. Cre-mediated deletion of the Geft gene in heart using Mef2c-Cre transgenic mice resulted in a dramatic decrease of Geft expression. Geft knockout mice develop normally and exhibit no discernable phenotype, suggesting Geft is dispensable for the development of the second heart field in mouse. The Geft conditional knockout mouse will be a valuable genetic tool for uncovering the in vivo roles of Geft during development and in adult homeostasis.
To evaluate the effects of transportation system operation, usually measures of effectiveness(MOE) such as travel time, space mean speed, stop/delay ratio have been used. But, energy consumption as well as the existing MOE in transportation receives more attention as an alternative MOE in transportation operation. The purpose of this study is a development of procedure, which could measure the relative energy consumption for each alternative and compare the results. A mesoscopic simulator called INTEGRATION is used to evaluate the operation of high occupancy vehicle lane, signal optimization, lane expansion, and the application of ITS. Among those, the application of ITS shows the greatest effectiveness in energy reduction, and then lane expansion, signal optimization, and the operation of high occupancy vehicle lane in the order named. Because we don't consider the characteristics of vehicle class, Potential demand and the simulation time is just for an hour. it is recommended that a procedure for precise economic analysis and an improvement in methodology are needed in the future for the expanded application of this study.
The unloading effect from excavations can cause the deformation of adjacent tunnels, which may seriously influence the operation and safety of those tunnels. However, systematic studies of the deformation characteristics of tunnels located along side excavations are limited, and simplified methods to predict the influence of excavations on tunnels are also rare. In this study, the simulation capability of a finite element method (FEM) considering the small-strain characteristics of soil was verified using a case study. Then, a large number of FEM simulations examining the influence of excavations on adjacent tunnels were conducted. Based on the simulation results, the deformation characteristics of tunnels at different positions and under four deformation modes of the retaining structure were analyzed. The results indicate that the deformation mode of the retaining structure has a significant influence on the deformation of certain tunnels. When the deformation magnitudes of the retaining structures are the same, the influence degree of the excavation on the tunnel increased in this order: from cantilever type to convex type to composite type to kick-in type. In practical projects, the deformation mode of the retaining structure should be optimized according to the tunnel position, and kick-in deformation should be avoided. Furthermore, two methods to predict the influence of excavations on adjacent tunnels are proposed. Design charts, in terms of normalized tunnel deformation contours, can be used to quantitatively estimate the tunnel deformation. The design table of the excavation influence zones can be applied to determine which influence zone the tunnel is located in.
The purpose of this study was to investigate the relationships among density, moisture content, and modulus of elasticity in which are important characteristics in physical and mechanical properties of woods. In this study, the dynamic MOE was calculated through the combination with resonance frequency of transverse vibration method and density, and the estimated moisture contents were calculated with two different equations (1, 2) in order to compare with experimental moisture contents. The following results from this study were obtained: 1. According to the regression analysis with two different parameters (E and density), the two regression lines appeared to be straight intersecting at 0.6 density. As another factor, moisture contents in wood also influenced on the analysing regression at the below F.S.P. 2. When considering the relationship between moisture contents and E, the tendency of each moisture content and E showed very similar pattern suggesting that moisture contents in addition to density are very important parameter. 3. When together with moisture contents and density as parameters for multiple regression analysis, coefficiences of determinations are dramatically improved. Interestingly, the coefficiences of determinations are further increased when analysing at the below point of F.S.P. and when analysing higher and lower density separately. In summary, more correct estimation of the dynamic MOE of woods can be possible with only transverse vibration and density in wood. Therefore, with this indirect method, the calculation of MOE in all kinds of woods including timber, live tree and wood products can be feasible resulting in accelerating the efficiency of time and labor.
Backgroud: Sleep deprivation (SD) impairs learning and memory by inhibiting hippocampal functioning at molecular and cellular levels. Abnormal autophagy and apoptosis are closely associated with neurodegeneration in the central nervous system. This study is aimed to explore the alleviative effect and the underlying molecular mechanism of stem-leaf saponins of Panax notoginseng (SLSP) on the abnormal neuronal autophagy and apoptosis in hippocampus of mice with impaired learning and memory induced by SD. Methods: Mouse spatial learning and memory were assessed by Morris water maze test. Neuronal morphological changes were observed by Nissl staining. Autophagosome formation was examined by transmission electron microscopy, immunofluorescent staining, acridine orange staining, and transient transfection of the tf-LC3 plasmid. Apoptotic event was analyzed by flow cytometry after PI/annexin V staining. The expression or activation of autophagy and apoptosis-related proteins were detected by Western blotting assay. Results: SLSP was shown to improve the spatial learning and memory of mice after SD for 48 h, accomanied with restrained excessive autophage and apoptosis, whereas enhanced activation of phosphoinositide 3-kinase/protein kinase B/mammalian target of rapamycin signaling pathway in hippocampal neurons. Meanwhile, it improved the aberrant autophagy and apoptosis induced by rapamycin and re-activated phosphoinositide 3-kinase/Akt/mammalian target of rapamycin signaling transduction in HT-22 cells, a hippocampal neuronal cell line. Conclusion: SLSP could alleviate cognitive impairment induced by SD, which was achieved probably through suppressing the abnormal autophagy and apoptosis of hippocampal neurons. The findings may contribute to the clinical application of SLSP in the prevention or therapy of neurological disorders associated with SD.
Based on comparative studies on standards and grading procedures of machine graded lumber in Korea and other countries, this study proposed a procedure of determining the grade classification and design strengths of domestic machine graded lumber. Differences between machine stress rated lumber and E-rated laminations were detailed in order to clarify the need for the procedure improvement. To this improvement the use of average MOE requirement for grading was introduced instead of the fixed minimum MOE requirement which is currently used in the Korean standards. It was found that the fixed minimum MOE requirement method was easier for an inspector to grade but, less efficient as a strength predictor than the average MOE requirement method. The advantage of average MOE requirement method is statistically MOR-MOE regression-based MOR prediction and highly efficient in quality control though it requires a computer-aided operation system in an initial setup. A major weakness of the current Korean grading system was found that different strength characteristics depending on wood species were not reflected on the grade classification and the tabulated allowable design stress. The proposed procedures were developed taking advantages of respective merits of both methods and based on MOR-MOE regression analysis. Through this procedure, the grades of machine stress rated lumber should be revised to become interchangeable with E-rated lamination, which would be beneficial to the cost competitiveness of domestic machine graded lumber and glued laminated timber industry.
Chlamydiae, obligate intracellular bacteria, are associated with a variety of human diseases. The chlamydial life cycle undergoes a biphasic development: replicative reticulate bodies (RBs) phase and infectious elementary bodies (EBs) phase. At the end of the chlamydial intracellular life cycle, EBs have to be released to the surrounded cells. Therefore, the interactions between Chlamydiae and cell death pathways could greatly influence the outcomes of Chlamydia infection. However, the underlying molecular mechanisms remain elusive. Here, we investigated host cell death after Chlamydia infection in vitro, in L929 cells, and showed that Chlamydia infection induces cell necrosis, as detected by the propidium iodide (PI)-Annexin V double-staining flow-cytometric assay and Lactate dehydrogenase (LDH) release assay. The production of reactive oxygen species (ROS), an important factor in induction of necrosis, was increased after Chlamydia infection, and inhibition of ROS with specific pharmacological inhibitors, diphenylene iodonium (DPI) or butylated hydroxyanisole (BHA), led to significant suppression of necrosis. Interestingly, live-cell imaging revealed that Chlamydia infection induced lysosome membrane permeabilization (LMP). When an inhibitor upstream of LMP, CA-074-Me, was added to cells, the production of ROS was reduced with concomitant inhibition of necrosis. Taken together, our results indicate that Chlamydia infection elicits the production of ROS, which is dependent on LMP at least partially, followed by induction of host-cell necrosis. To our best knowledge, this is the first live-cell-imaging observation of LMP post Chlamydia infection and report on the link of LMP to ROS to necrosis during Chlamydia infection.

Significant soil erosion and water quality degradation issues are occurring at highland agricultural areas of Kangwon province because of agronomic and topographical specialities of the region. Thus spatial and temporal modeling techniques are often utilized to analyze soil erosion and sediment behaviors at watershed scale. The Soil and Water Assessment Tool (SWAT) model is one of the watershed scale models that have been widely used for these ends in Korea. In most cases, the SWAT users tend to use the readily available input dataset, such as the Ministry of Environment (MOE) land cover data ignoring temporal and spatial changes in land cover. Spatial and temporal resolutions of the MOE land cover data are not good enough to reflect field condition for accurate assesment of soil erosion and sediment behaviors. Especially accelerated soil erosion is occurring from agricultural fields, which is sometimes not possible to identify with low-resolution MOD land cover data. Thus new land cover data is prepared with cadastral map and high spatial resolution images of the Doam-dam watershed. The SWAT model was calibrated and validated with this land cover data. The EI values were 0.79 and 0.85 for streamflow calibration and validation, respectively. The EI were 0.79 and 0.86 for sediment calibration and validation, respectively. These EI values were greater than those with MOE land cover data. With newly prepared land cover dataset for the Doam-dam watershed, the SWAT model better predicts hydrologic and sediment behaviors. The number of HRUs with new land cover data increased by 70.2% compared with that with the MOE land cover, indicating better representation of small-sized agricultural field boundaries. The SWAT estimated annual average sediment yield with the MOE land cover data was 61.8 ton/ha/year for the Doam-dam watershed, while 36.2 ton/ha/year (70.7% difference) of annual sediment yield with new land cover data. Especially the most significant difference in estimated sediment yield was 548.0% for the subwatershed #2 (165.9 ton/ha/year with the MOE land cover data and 25.6 ton/ha/year with new land cover data developed in this study). The results obtained in this study implies that the use of MOE land cover data in SWAT sediment simulation for the Doam-dam watershed could results in 70.7% differences in overall sediment estimation and incorrect identification of sediment hot spot areas (such as subwatershed #2) for effective sediment management. Therefore it is recommended that one needs to carefully validate land cover for the study watershed for accurate hydrologic and sediment simulation with the SWAT model.
